[Study on mitochondrial ultrastructure, trace elements and correlative factors of gastric mucosa in patients with spleen deficiency syndrome].

Eighty-eight gastropathic patients with Spleen deficiency syndrome by using transmission electron microscope (TEM), X-ray energy disperse analysis system (EDAX), histochemical staining and radioimmuno methods were examined. The authors found that the gastric mucosa cAMP, SOD level, the quantity of mitochondria and its crista, the ratio of diameter between ventricle and cavity of mitochondria and the content of Zn, Cu of mitochondria were reduced in the trend of healthy control group, Spleen Qi deficiency group, Spleen deficiency with Qi stagnation group; chronic superficial gastritis group, chronic atrophic gastritis group, gastric cancer group: complete small intestinal metaplasia group, incomplete small intestinal metaplasia group, complete colonic intestinal metaplasia group, incomplete colonic intestinal metaplasia group (P < 0.05-0.001). While the degeneration rate of mitochondria, the Cu/Zn ratio of mitochondria, the metaplasia rate of gastric, the rate of incomplete colonic intestinal metaplasia and the content of serum LPO were increased in the above turn. It is suggested that the comprehensive effect of the degeneration of mitochondria and the quantitative changes of its correlative factors is the physiopathologic base for inducing Spleen deficiency disease, gastric mucosa metaplasia and canceration. Much attention must be paid in clinic to the cancerization trend of gastric disease with Spleen deficiency syndrome.

[Evaluation on glycine chelated iron (II) in improving nutritional anemia in rats].

OBJECTIVE: In order to evaluate the efficacy of glycine chelated iron (II) in improving nutritional anemia in rats. METHODS: After weaning wistar rats, weighing 50 - 60g, were fed the iron deficient diet for three weeks, whole blood was taken from rat's tail to measure Hb, then the rats were randomly divided into five groups by Hb, that is the control group fed with iron deficient diet, three groups fed the diets supplemented with different glycine chelated iron (II), and another group fed with the diet added with lactate iron. Each group was fed respective diets for four weeks and indicators related to iron-deficient nutritional anemia were evaluated. RESULTS: The addition of glycine chelated iron (II) did improve the growth and development of rats, increase the hemoglobin level and the iron retention in spleen, decrease free erythrocyte protoporphyrin (FEP) content in whole blood, and there was no significant difference between three supplemental groups with glycine chelated iron (II) and the group with lactate iron. CONCLUSION: glycine chelated iron (II) can be used as a reasonable iron supplement in improving iron-deficient nutritional anemia.

[Efficiency of NaFeEDTA fortified soy sauce on anemia prevention].

OBJECTIVE: Study the efficiency of the NaFeEDTA fortified soy sauce on anemia prevention among the children from iron deficiency anemia (IDA) prevalence region. METHODS: A double blind controlled random intervention in Bijie, Guizhou was conducted for 18 months. The finger hemoglobin (Hb) was detected at baseline and after 6, 12, 18 months intervention. Anthropometrics were also taken at baseline and after 12 months intervention. RESULTS: The Hb level increased significantly in the intervention group [(111.08 +/- 11.31) g/L at baseline and (122.99 +/- 8.43) g/L after 12 months], the anemia prevalence decreased markedly (58.0% at baseline, 17.6% after 12 months). After 12 months intervention, a significantly gain of the weight for age Z-score (WAZ) was observed in the intervention group. The abdomen and triceps skin folders were thicker in the intervention group, and the intervention group had greater gains of the abdomen and triceps skin folders than those in the control group. CONCLUSION: The soy sauce fortified with NaFeEDTA can increase the Hb level and decrease the prevalence of anemia in the children, and the NaFeEDTA fortified soy sauce can enhance the weight gain and the pull up of the subcutaneous fat. It is suggested that using NaFeEDTA fortified soy sauce in high IDA prevalence population was an efficient way for IDA preventing.

[Studies of gene networks and singal transduction of kidney deficiency syndrome by syndrome differentiation through drug effects].

Three kinds of networks were summarily described in this review including the small intracellular molecular networks, the middle-scale networks of hypothalamus-pituitary-adrenal-thymus (HPAT) axis and the large network, neuroendocrine-immune (NEI) network, covering the whole organism and linking multiple systems together. The hypothesis was expressed that the "disease" or "syndrome" formed in the human body by the intervention from outside world is based on the changes of multi-molecular network. In this paper, the pattern and ability of signal transduction channel and the methods of studying changes in it were also described, and raised, herefrom, "to determine syndrome by drug effects (DSDE)" is the intervention means for studying syndrome in the light of systemic biological methods. We found Kidney-yang deficiency syndrome covered the NEI network and the regulating center located in hypothamus with Compound Bushen Recipe (CBR, Kidney-tonifying recipe). By intervention with EF, an effective component of CBR, it was found that EF can activate the immune system and the three networks, including growth axis, sex hormone axis and lymphocyte apoptosis network in HPAT axis through the downward pathway of NEI network to play its efficiency of molecular network. There are many regulation patterns of EF on networks. For example, in the network mechanism of lymphocyte apoptosis and proliferation, EF can reconstruct the balance of the opposite apoptosis related genes and proliferation related genes; EF can assemble and integrate co-stimulating molecules, transform growth factors (TGF), and several oncogenes to form an upstream factor network for initiating the proliferation and anti-apoptosis promotion; EF can simultaneously up-regulate the two opposite genes expression of IkappaB and NFkappaB in NIK/IKK/IkappaB/Rel/NFkappaB signal transduction channel, which could not only control the rising of NFkappaB in a moderate range, but also guarantee its predominant status to exert its hinge role in molecule regulating network, by which gene network regulation atlas in HPAT axis of Kidney-deficiency syndrome was observed.

[Idiopathic thrombocytopenic purpura in children treated with replenishing qi and tonifying kidney and the changes in thrombocyte aggregative function].

UNLABELLED: Forty-one children with idiopathic thrombocytopenic purpura (ITP) were reported in this paper. Of all, 36 with petechia and ecchymosis, 28 with nasal hemorrhage, 5 with hematochezia and 1 with subcutaneous hematoma. According to the TCM differentiation, 24 and 17 of all belonged to the type of impairment of Yin due to blood-heat and both deficiencies of Qi and tonifying Kidney and its basic prescriptions were: dried Radix Rehmanniae, Radix Angelicae Sinensis, Radix Paeoniae Rubra, Radix Rubiae, Fructus Psoraleae, stir-frying Radix Astragali, Semen Cuscutae, Caulis Spatholob; Chinese Jujubae, Radix et Rhizoma Rhei. The average treating duration was 5.02 months. RESULTS: 24 cases cured, 6 recovered, 10 improved, 1 failed, with a total effective rate of 97.6%. 22 of the cured were followed-up without recurrence in average 10.4 months after withdrawal. In 32 cases, the thrombocyte aggregative function was tested with ADP induction, meanwhile in 20 of 32 cases aggregation with adrenaline induction was tested. The results before and after treatment showed an evident difference, with low function before treatment and normal after treatment. It is concluded that the replenishing Qi and tonifying Kidney may have a better effect on both acute and chronic children ITP without recurrence after withdrawal.

Arrayed primer extension: a robust and reliable genotyping platform for the diagnosis of single gene disorders: beta-thalassemia and thiopurine methyltransferase deficiency.

Mutation screenings, which were conventionally carried out individually because of different assay conditions, are usually time consuming and not cost effective. Using microarray technology, simultaneous molecular diagnosis of multiple mutations on a single platform is possible. To evaluate this idea, we developed a DNA chip platform to simultaneously detect 23 mutations of the beta-globin gene and 9 mutations of thiopurine methyltransferase (TPMT) gene based on the principle of arrayed primer extension (APEX). A blinded test consisting of 200 DNA samples with known genotypes was performed to validate this strategy. High genotyping accuracy of 97.3% and 100% for beta-globin and TPMT genes, respectively, were achieved. Further analysis on the fluorescent intensity demonstrated clear separation between the real signal and the background noise, which enabled us to set two cutoff values (V(lower) = 4.0 and V(upper) = 12.0) to determine the genotype quantitatively. Our results showed that APEX is a highly reliable genotyping strategy to detect mutations that cause beta-thalassemia or TPMT enzyme deficiency.

Steroid 5alpha-reductase 2 deficiency in two generations of a non-consanguineous Chinese family.

We report a Chinese family in which two family members were diagnosed to have steroid 5alpha-reductase 2 deficiency. The proband was an 8 year-old boy presenting with isolated micropenis. Mutational analysis revealed a homozygous mutation in exon 4 of the SRD5A2 gene, changing codon 227 from CGA to CAA, i.e. R227Q. The paternal granduncle of the proband has incomplete virilization in adulthood. Besides having the mutation R227Q, he has another mutation in exon 5 of the SRD5A2 gene, changing codon 246 from CGG to CAG, i.e. R246Q. To our knowledge, this is the first reported Chinese family diagnosed with steroid 5alpha-reductase 2 deficiency with identified molecular defects.

Contrasting roles for STAT4 and STAT6 signal transduction pathways in murine renal ischemia-reperfusion injury.

Recent data support a modulatory role for CD4 T cells in experimental renal ischemia-reperfusion injury (IRI). CD4 T cells can functionally differentiate to either a Th1 (IFN-gamma producing) or the counterbalancing Th2 (IL-4) phenotype. The enzymes signal transducers and activators of transcription (STAT) 4 and STAT6 regulate Th1 or Th2 differentiation and cytokine production, respectively. We therefore hypothesized that mice that were STAT4 deficient would be protected from renal IRI and that STAT6-deficient mice would have a more severe course. Intracellular cytokine staining of splenocytes from STAT4-/- or STAT6-/- exhibited distinct IFN-gamma and IL-4 cytokine expression profiles. STAT6-/- had markedly worse renal function and tubular injury postischemia compared with wild type. STAT4-/- had only mildly improved function. Renal phagocyte infiltration and ICAM-1 upregulation were similar in STAT4-/-, STAT6-/-, and wild type. To evaluate if the mechanism of the marked worsening in the STAT6-/- mice could be due to IL-4 deficiency, IL-4-deficient mice were studied and had similar postischemic phenotype to STAT6-/- mice. These data demonstrate that the STAT6 pathway has a major protective role in renal IRI. IL-4 deficiency is a likely mechanism underlying the STAT6 effect. A "yin-yang" role for inflammation is emerging in renal IRI, similar to recent observations in atherosclerosis.

Objective auscultation for traditional chinese medical diagnosis using novel acoustic parameters.

The goal of this work is to propose novel acoustic parameters of voice for the purpose of providing a quantitative analysis of auscultation in traditional Chinese medical diagnosis. There is rare amount of available literature related to this topic. Four novel acoustic parameters, the average number of zero-crossings, the variations in local peaks and valleys, the variations in first and second formant frequencies, and the spectral energy ratio, are presented to analyze and identify the characteristics among non-vacuity, qi-vacuity, and yin-vacuity subjects. Among these acoustic parameters, two temporal parameters, the average number of zero-crossings and the variations in local peaks and valleys, outperformed other parameters in classifying both non-vacuity and deficient subjects. The spectral energy ratio was adequate for the classification between qi-vacuity and yin-vacuity patients. This research is a first step in an ongoing effort to modernize the auscultation in traditional Chinese medical diagnosis.

Mechanistic study of malononitrileamide FK778 in cardiac transplantation and CMV infection in rats.

BACKGROUND: FK778 is a malononitrilamide, a class of immune suppressive compounds with antiviral features and experimental activity in chronic rejection, a potentially interesting combination for organ transplantation. The goal of this project was to study the tolerability, immune suppressive efficacy, and anti-cytomegalovirus (CMV) activity of FK778 and to assess the in vivo relevance of its previously described inhibition of de novo pyrimidine synthesis. METHODS: Heart transplants were performed in rats (Brown Norway [BN] to Lewis) and treated with varying doses of FK778 or leflunomide for 28 days. At 28 days, at the time of rejection or at the death of the animal, the allograft and other vital organs were obtained for study by light microscopy and immunohistochemistry. In separate experiments, Lewis rats were given sublethal irradiation, inoculated with rat CMV (Maastricht strain), and treated with varying doses of FK778 and leflunomide. In both the transplant and CMV studies, IP uridine was given at 250 mg/kg to cohorts or animals receiving FK778 and leflunomide. RESULTS: FK778 controls acute rejection and inhibits CMV replication at 20 mg/kg but is toxic at 25 mg/kg. Toxicity is manifested as anemia, changes in hepatic and intestinal histology, and mortality. The toxicity but not the immune suppressive or antiviral efficacy, is reduced significantly by exogenous uridine administration. CONCLUSION: FK778 has both immune suppressive and antiviral activities, neither of which is entirely dependent on inhibition of pyrimidine synthesis. These, and other published observations, suggest that the antiviral activity and a considerable part of the efficacy of the malononitrilamide family of drugs is attributable to activities other than drug induced pyrimidine deficiency.
