[Hydrogen-adduct radicals in thymidine and thymidine 5'-monophosphate photosensitized by proflavin. Protection by sulfur compounds (author's transl)].

At pH values 5 and 9, the influence of cysteine, cystine, 3-mercaptopropionic acid and S-methylcysteine on the induction of H-adduct radicals in thymidine and thymidine 5'-monophosphate photosensitized by proflavin is investigated. The results obtained indicate that efficient protection is given when the electrostatic interactions between the charged groups of the molecules present allow the close proximity of a thiol or a disulfide bridge and the proflavin - DNAs complex constituent. The scheme proposed for the mechanism involves the capture by the protector of an electron or a proton, indispensable intermediates in the formation of the H-adduct radicals.

Variation of Se, Zn, Co, Fe and Rb distribution in rats after injection with selenium or sulfur compounds.

Contents of Se, Zn, Co, Fe and Rb in liver, kidney, heart, spleen, brain, pancreas and testicle of Wistar rats were determined 2 h after i.p. injection with selenodiglutathione, selenomethionine, selenocystine, selenocystamine, methionine, cystine and cystamine. Instrumental neutron activation analysis (INAA) was applied as the analytical method. It was found that Se was incorporated to all examined rat organs. High Se incorporation was observed after injection with selenomethionine and selenocystine. Variations in the Zn, Co, Fe and Rb contents were observed in all the investigated organs after the injections. These variations depended upon injected compounds.

Monitoring of reduced sulfur compounds in the atmosphere of Gosan, Jeju Island during the Spring of 2001.

The atmospheric concentrations of dimethylsulfide (DMS) and carbon disulfide (CS2) were measured concurrently with relevant environmental parameters at Gosan, Jeju Island, Korea during 5-26 April 2001. The mean concentrations for these two compounds were 18.7+/-17.9 and 6.4+/-9.9 pptv, respectively. Results of our analysis indicated that relative temporal variations between DMS and CS2 can be best described by dividing the whole data set into three different periods which reflect the variable transport patterns of air masses into the study area. (Periods I, II, and III denote: 5-10, 10-18, and 19-26 April.) The environmental conditions during those three periods varied greatly. The effects of continental and/or oceanic processes were evident for certain periods, yielding diverse relationships between DMS and CS2 in both absolute and relative terms. Most observed variations were best explained in terms of an interplay between source/sink processes and air mass transport patterns. The sea-to-air flux of DMS, when estimated using our measurement data during this study period, was approximately 4 micromole m(-2)d(-1).

Synthesis and radiation stability of novel biologically active sulfur compounds derived from 1,2-bis(4-amino-5-mercapto-s-triazol-3-yl)ethane.

Some novel 1,2-bis(s-triazolo[3,4b][1,3,4]thiadiazino-3-yl)ethane (4-7); 1,2-bis(s-triazolo[3,4b][1,3,4]thiadiazol-3-yl)ethane (16a,b) and 1,2-bis(s-triazolo[3,4b][1,3,4]thiadiazepino-3-yl)ethane (17) were synthesized via reaction of 1,2-bis(4-amino-5-mercapto-s-triazol-3-yl)ethane (3) with different reagents. Identification of the new compounds was established by elemental analyses, IR, 1H NMR and mass spectral data. Compounds 12, 13, 16b and 17 were promising antifungal activity. The biologically active compounds 13, 16b and 17 were radioresistant retaining their structures unchanged up to 40 k Gy. Radiosterilization of these compounds in the dry state may prove to be applicable.

Palladium(0)-catalyzed enantioselective O,S-rearrangement of racemic O-allylic thiocarbamates: a new entry to enantioenriched allylic sulfur compounds.

Reaction of (+/-)-(E)-pent-3-en-2-ol, (+/-)-(E)-hept-4-en-3-ol, (+/-)-(E)-2,6-dimethylhept-4-en-3-ol, (+/-)-cyclohex-2-en-1-ol, and (+/-)-cyclohept-2-en-1-ol with methyl, ethyl, n-propyl, isopropyl, n-butyl, tert-butyl, and benzyl isothiocyanate gave the corresponding racemic O-allylic thiocarbamates of medium to good thermal stability in good yields. The palladium(0)-catalyzed rearrangement of the (+/-)-(E)-pent-3-en-2-ol-, (+/-)-(E)-hept-4-en-3-ol-, (+/-)-cyclohex-2-en-1-ol-, and (+/-)-cyclohept-2-en-1-ol-derived O-allylic thiocarbamates at room temperature in methylene chloride by using Pd2(dba)3*CHCl3 (dba = dibenzylideneacetone) as precatalyst and (+)-(1R,2R)-1,2-bis-N-((2-(diphenylphosphino)benzoyl)-1,2-diaminocyclohexane as ligand for the palladium atom proceeded quantitatively and gave the corresponding acyclic (R)-configured S-allylic thiocarbamates and the cyclic (S)-configured S-allylic thiocarbamates with ee values ranging from 85% to > or = 99% in yields of 76-94%. Rearrangement of the O-allylic thiocarbamates carrying a methyl group at the N atom not only was the fastest but also proceeded with the highest enantioselectivity. No rearrangement was observed under these conditions in the case of the racemic N-methyl O-allylic thiocarbamate derived from (+/-)-2,6-dimethylhept-4-en-3-ol, which has a branched carbon skeleton. (S)-cyclohex-2-enethiol of 97% ee was obtained through hydrolysis of the corresponding N-methyl S-allylic thiocarbamate. 2-((R)-(E)-1-methylbut-2-enylsulfanyl)pyrimidine of 91% ee and 2-((S)-cyclohex-2-enylsulfanyl)pyrimidine of 97% ee were synthesized in one synthetic operation from the corresponding N-methyl S-allylic thiocarbamates and 2-chloropyrimidine. Similarly, (S)-cyclohex-2-enylsulfanyl)benzene of 97% ee was obtained in one synthetic operation from the corresponding N-methyl S-allylic thiocarbamate through a palladium(0)-catalyzed substitution of iodobenzene in the presence of a base. The palladium(0)-catalyzed enantioselective rearrangement of O-allylic carbamates to S-allylic carbamates has been extended from the solution phase to the solid phase by using a methyl thioisocyanate polystyrene resin. In the case investigated the enantioselectivity of the rearrangement on the solid phase was considerably lower than that in solution.

Bruguiesulfurol, a new sulfur compound from Bruguiera gymnorrhiza.

A new cyclic 4-hydroxy-dithiosulfonate, bruguiesulfurol (1), as well as two known 4-hydroxydithiolane 1-oxides, brugierol (2) and isobrugierol (3) were isolated from the flowers of Bruguiera gymnorrhiza. With stably-transfected HepG2 cells, the three isolates activated antioxidant response element (ARE) luciferase activation with (EC(50)) values of 56.7, 3.7 and 1.8 microM, respectively. Compounds 2 and 3 also inhibited phorbol ester-induced NF-kappaB (nuclear factor-kappaB) luciferase activity with IC (50) values of 85.0 and 14.5 microM, respectively. In addition, compound 2 inhibited enzyme cyclooxygenase-2 (COX-2) activity with an IC(50) value of 6.1 microM. The structures of these isolates were determined by spectral data, and that of compound 1 was confirmed by X-ray crystallographic analysis.

Dependence of synergistic fungicidal activity of Cu2+ and allicin, an allyl sulfur compound from garlic, on selective accumulation of the ion in the plasma membrane fraction via allicin-mediated phospholipid peroxidation.

Allicin was effective in decreasing the lethal concentration of Cu (2+) against various fungal strains including a plant pathogen, Fusarium oxysporum, so that the minimum fungicidal concentration (MFC) of the ion for the fungus could be reduced to 2 % of that detected without allicin. In Saccharomyces cerevisiae, Cu (2+) was not apparently taken up by cells when added alone at a non-lethal concentration, whereas the ion was efficiently incorporated into cells in the presence of allicin, as in the case of cells treated with the ion at a lethal concentration. Although allicin likely increased cellular permeability to Cu (2+) due to its promotive effect on plasma membrane phospholipid peroxidation, these cell-surface events did not result in endogenous reactive oxygen species (ROS) production, a typical toxic effect of the ion. Cu (2+) was detected in the cytoplasmic fraction of cells that had been treated with the ion at a lethal concentration, whereas the ion was entrapped in the plasma membrane fraction upon their treatment with the ion at a low concentration in combination with allicin. Cu (2+) could be solubilized from the plasma membrane fraction by a procedure for the extraction of hydrophobic proteins rather than the extraction of phospholipids, suggesting its complexation with a plasma membrane protein as a result of allicin treatment. Such a subcellular localization of Cu (2+) resulted in the selective leakage of intracellular K (+), but not in the disruptive damage on the plasma membrane, and was considered to underlie the synergistic fungicidal activity of Cu (2+) and allicin.
