High potassium solution is one of the most commonly used cardioplegic solution, but the mechanism of action is still poorly defined. In the present study, isolated rat hearts were utilized to investigate the protective effects and mechanism of action of high potassium against ischemia/reperfusion injury. The results showed that high potassium (22 mmol/L) apparently improved the recovery of contraction amplitude (P < 0.01), inhibited the rise of resting tension (P < 0.01) and abolished ventricular fibrillation during reperfusion after global ischemia for 40 minutes. Moreover, high potassium could preserve myocardial Na+, K(+)-ATPase activity (P < 0.01) and inhibit sodium and calcium overload (P < 0.01) during reperfusion. The results indicate that small amount of high potassium solution (5 ml) administered even after ischemic arrest of rat heart has remarkable protective effects against ischemia/reperfusion injury at 37 degrees C. Its mechanism of action is at least partially by preserving Na+,K(+)-ATPase activity and inhibiting sodium and calcium overload.

A Fe-containing superoxide dismutase (EC 1.15.1.1) has been isolated from the soil denitrifying bacterium Pseudomonas aeruginosa and characterized. An apoprotein has been obtained and protein reconstruction by Fe ions carried out. The absorbance in the visible part of the spectrum is fully determined by the Fe-containing cluster which does not influence the absorption in the UV region. The effects of inorganic anions on the enzyme activity and ESR spectra have been studied. The mechanisms of inhibition have been found to be different for F- and CNS-. Based on the data on the pH dependence of the shape of ESR spectra it is assumed that the environment of Fe-containing centers in the protein molecule is different.
The effects of the application of potassium ferrate to remove possible toxic compounds are presented. Potassium ferrate (K2FeO4) is shown in this work to be an effective means to remove toxic metals and nonmetals from aqueous solution. The toxic material present in water is precipitated from aqueous solution and readily removed. Potassium ferrate removes itself from solution. Discolored contaminated water may be made clear by utilizing potassium ferrate. In addition, turbidities of solutions induced by dissolved substances are eliminated by the action of potassium ferrate. The efficacy of potassium ferrate in cleaning contaminated water shows great potential in application to municipal and industrial waste water.

OBJECTIVE: To test the hypothesis that shear stress relaxation functions of dental ceramics can be determined from creep functions measured in a beam-bending viscometer. METHODS: Stress relaxation behavior was determined from creep data for the following materials: (1) a veneering ceramic-IPS Empress2 body ceramic (E2V); (2) an experimental veneering ceramic (EXV); (3) a low expansion body porcelain-Vita VMK 68 feldspathic body porcelain (VB); (4) a high expansion body porcelain-Will Ceram feldspathic body porcelain (WCB); (5) a medium expansion opaque porcelain-Vita feldspathic opaque porcelain (VO); and (6) a high expansion opaque porcelain-Will Ceram feldspathic opaque porcelain (WCO). Laplace transform techniques were used to relate shear stress relaxation functions to creep functions for an eight-parameter, discrete viscoelastic model. Nonlinear regression analysis was performed to fit a four-term exponential relaxation function for each material at each temperature. The relaxation functions were utilized in the ANSYS finite element program to simulate creep behavior in three-point bending for each material at each temperature. RESULTS: Shear stress relaxation times at 575 degrees C ranged from 0.03 s for EXV to 195 s for WCO. SIGNIFICANCE: Knowledge of the shear relaxation functions for dental ceramics at high temperatures is required input for the viscoelastic element in the ANSYS finite element program, which can used to determine transient and residual stresses in dental prostheses during fabrication.
We report kinetic data of penicillin hydrolysis catalyzed by beta-lactamase entrapped in reverse micelles formed with cetyl trimethylammonium bromide (CTAB), n-octane, hexanol and aqueous buffer. The K(cat) of this diffusion-limited reaction can be improved in aqueous buffer by a factor of 1.1-1.2 just by increasing the phosphate buffer concentration from 50 to 100 mM. In reverse micelles, increasing the buffer concentration has little effect on K(cat) when the size of the empty micelle is below the size of the protein. However, in larger micelles, the effect is enhanced and the K(cat) improves several fold, changing the form of the curve of K(cat) versus Wo from bell-shaped to almost hyperbolic. The results indicate that micellar exchange and internal diffusion may limit the reaction in reverse micelles and provide further evidence that the form of the curve depends on other factors besides the relationship between the size of the enzyme and that of the empty reverse micelle.
The medium composition for bacteriocin production by Lactococcus lactis ATCC 11454 was optimized using response surface methodology. The selected six factors based on CM medium were sucrose, soybean peptone, yeast extract, KH(2)PO(4), NaCl, and MgSO(4).7H(2)O. Fractional factorial designs (FFD) and the path of steepest ascent were effective in searching for the main factors and approaching the optimum region of the response. By a 2(6-2) FFD, sucrose, soybean peptone, yeast extract, KH(2)PO(4) were found to be significant factors and had positive effects on cell growth, however, only soybean peptone and KH(2)PO(4) were shown to be the two significant factors for bacteriocin production and had negative and positive effects, respectively. The effects of the two main factors on bacteriocin production were further investigated by a central composite design and the optimum composition was found to be 1% sucrose, 0.45% soybean peptone, 1% yeast extract, 2.84% KH(2)PO(4), 0.2% NaCl, and 0.02% MgSO(4) x 7H(2)O. The optimal medium allowed bacteriocin yield to be doubled compared to CM medium.
