2-aminoethoxydiphenyl borate activates and sensitizes the heat-gated ion channel TRPV3.

Six of the mammalian transient receptor potential (TRP) ion channel subtypes are nonselective cation channels that can be activated by increases or decreases in ambient temperature. Five of them can alternatively be activated by nonthermal stimuli such as capsaicin [transient receptor potential vanilloid 1 (TRPV1)] or hypo-osmolarity (TRPV2 and TRPV4). No nonthermal stimuli have yet been described for TRPV3, a warmth-gated ion channel expressed prominently in skin keratinocytes. Here, we demonstrate that 2-aminoethoxydiphenyl borate (2-APB), a compound used to inhibit store-operated Ca2+ channels and IP3 receptors, produces robust activation of recombinant TRPV3 in human embryonic kidney 293 cells with an EC50 of 28 microm. 2-APB also sensitizes TRPV3 to activation by heat, even at subthreshold concentrations. In inside-out membrane patches from TRPV3-expressing cells, 2-APB increases the open probability of TRPV3. Also, whereas heat alone is capable of activating TRPV3-mediated currents in only a small proportion of primary mouse keratinocytes, 2-APB activates heat-evoked, TRPV3-mediated currents in the majority of these cells. Together, these findings identify 2-APB as the first known chemical activator of TRPV3 and enhance the notion that TRPV3 participates in the detection of heat by keratinocytes.

Derivatization of tributyltin with sodium tetrakis(4-fluorophenyl)-borate for sensitivity improvement of tandem mass spectrometry.

Derivatization of tributyltin for tandem mass spectrometry is described. Tributyltin (TBT) and triphenyltin (TPT) were derivatized with sodium tetrakis(4-fluorophenyl)borate. After optimization of their MS/MS conditions, derivatization conditions were examined. Under the optimum conditions using in-situ derivatization, the calibration curves for the TBT and TPT were linear in the ranges of 0.4 - 200 and 1.2 - 200 pg of Sn, respectively. The detection limits for TBT and TPT were 0.07 and 0.43 pg of Sn, respectively. In the case of TBT, the detection limit with 4-fluorophenylation was improved about five times compared with that with pentylation (0.35 pg). This improvement is ascribed to the bond-dissociation energy of Sn-aryl being stronger than that of Sn-alkyl. Namely, the selective fragmentation of 4-fluorophenyl TBT resulted in high sensitivity. The relative recoveries of TBT and TPT from seawater were 99 and 109%, respectively. The method was successfully applied to the seawater samples.

2-Aminoethoxydiphenyl borate directly inhibits store-operated calcium entry channels in human platelets.

In this study, we examined 2-aminoethoxydiphenyl borate (2APB) as an inhibitor of Ca(2+) influx in human platelets. 2APB was found to inhibit thrombin-mediated intracellular Ca(2+) mobilization rapidly in platelets incubated in the absence of extracellular Ca(2+). This result supports an intracellular action of 2APB on inositol 1,4,5-trisphosphate (IP(3))-receptor Ca(2+) channels. 2APB was without effect on the ability of thapsigargin to mobilize intracellular Ca(2+). This result suggests that the efflux of Ca(2+) from the endoplasmic reticulum mediated by thapsigargin is not via IP(3) Ca(2+) channels. However, 2APB was able to prevent the entry of Ca(2+) and Sr(2+) through thapsigargin-activated, store-operated Ca(2+) channels (SOCC). This result supports a direct inhibitory effect of 2APB on SOCC. 2APB was also able to block the entry of Sr(2+), Ba(2+), and Mn(2+) entry into unstimulated platelets, which suggests that 2APB was inhibiting the Ca(2+) influx channels directly. The capacity of 2APB to prevent Ca(2+) influx and Sr(2+) influx was rapid because it occurred immediately upon addition to the platelets. The inhibition of Ca(2+) and Sr(2+) influx by 2APB was similar to that seen with the cell-impermeable nonselective Ca(2+)-channel blocker La(3+) or the Ca(2+) chelator EGTA. Diphenylboronic anhydride and 2,2-diphenyltetrahydrofuran, two compounds that are structurally similar to 2APB, also inhibited Ca(2+) influx. It was concluded that 2APB was a rapid and effective direct inhibitor of SOCC in human platelets; as such, it cannot be used to support the involvement of IP(3) receptors in the activation of SOCC.

2-Aminoethoxydiphenyl borate (2-APB) inhibits capacitative calcium entry independently of the function of inositol 1,4,5-trisphosphate receptors.

Capacitative calcium entry (CCE), the mechanism that replenishes intracellular calcium stores after depletion, is essential to intracellular calcium signaling. CCE is mediated by the channels in the plasma membrane generally referred to as "store operated channels (SOCs)". However, the molecular identity of the SOCs has never been determined, and the mechanism of the activation of SOCs remains to be elucidated. Recent studies have demonstrated that 2-aminoethoxydiphenyl borate (2-APB), which has been found to be an antagonist of inositol 1,4,5-trisphosphate receptors (IP3Rs), inhibits CCE, suggesting that IP3Rs channel activity is essential to the generation of CCE. However, CCE has also been reported to occur normally in IP3R-deficient cells. In order to resolve this discrepancy, we investigated the effect of 2-APB on CCE in IP3Rs-deficient cells. In response to store depletion with thapsigargin or N,N,N',N'-tetrakis (2-pyridylmethyl) ethylene diamine (TPEN), CCE was generated in IP3Rs-deficient cells the same as in wild-type cells, however, 2-APB abolished CCE in IP3Rs-deficient cells, despite the fact that this cell line does not possess functional IP3Rs. We also examined the effect of 2-APB on several types of TRP Ca2+ channels, which exhibit properties similar to those of SOCs. 2-APB had a different inhibitory effect on spontaneous and thapsigargin-induced Ba2+ influx in cells that transiently expressed individual TRP subtypes. These results suggest that the channel activity of IP3Rs is not essential to the generation of CCE in this cell line and that 2-APB inhibits CCE independently of the function of IP3Rs.

Control of glycoprotein synthesis. The purification by preparative high voltage paper electrophoresis in borate of glycopeptides containing high mannose and complex oligosaccharide chains linked to asparagine.

The heterogeneity of glycopeptides prepared from pronase digests of various glycoproteins has previously been demonstrated primarily by lengthy paper chromatography of oligosaccharides released from the glycopeptides by enzymatic or chemical means. We describe the use of high voltage paper electrophoresis in borate for the resolution of seven glycopeptides from ovalbumin and seven glycopeptides from human immunoglobulin G. This technique has been used in the preparation of glycopeptide substrates required for the study of certain glycosyltransferases and glycosidases.

Regulation of glutaminase B in Escherichia coli. II. Modulaltion of activity by carbosylate and borate ions.

Glutaminase B is both activated and inhibited by L-glutamate, the product of the reaction. The activation process is time- and temperature-dependent. Activation by L-glutamate alters the Vmax, So.5, and shape of the substrate saturation curve. The activation decays as a first order process with time after separation of L-glutamate from glutaminase B. L-Glutamate inhibits both glutaminase B and the glutamate-activated enzyme. Like L-glutamate, borate activates and inhibits glutaminase B. Inhibition of the enzyme by glutamine and glutamate analogs is also examined and similarities between the glutamate activation and activation by warming at 23 degrees are noted.
