Progesterone in the uterus. VIII. Uptake of corticosterone and incorporation of progesterone under concurrent injection of corticosterone into rat uterus.

A study of the subcellular distribution of radioactivity in rat uterus after injection of labelled corticosterone showed that the radioactivity was observed in all fractions from 5 min. to 120 min. A maximum uptake was observed 10 min. after application of the labelled steroid. Competitive uptake of radioactive progesterone and unlabelled corticosterone was assayed 10 min. after injection of the hormone mixture. The ratio between radioactive progesterone and unlabelled corticosterone was 1:1 and 1:2 (moles:moles), respectively. Compared with control experiments with rats which had received radioactive progesterone alone, the results gave evidence that progesterone found in all subcellular fractions and in the total homogenate was not depressed by unlabelled corticosterone. However, unlabelled progesterone reduced the tritiated progesterone in uterine tissue. This observation demonstrates that the uptake of progesterone by rat uterus is specific.

Effect of total or partial uterus extirpation on uterus-projecting neurons in porcine inferior mesenteric ganglion. C. Changes in expression of apoptosis-associated (Bcl-2 and Bax) and regeneration-associated (GAP-43) proteins.

The expression of Bcl-2 and Bax proteins was studied with immunohistochemistry, immuoblotting and RT-PCR in the uterine horn- and uterine cervix-projecting neurons of the inferior mesenteric ganglion (IMG) of the sexually immature gilts after partial or total hysterectomy. Additionally, the expression of regeneration-associated protein GAP-43 was studied in these neurons with immunohistochemistry. The uterus-projecting neurons were identified with retrograde fluorescent tracer Fast Blue (FB). The weak immunoreactivity to Bcl-2 and GAP-43 and moderately intense immunoreactivity to Bax was revealed in all FB+ (FB+) neurons of control and hysterectomized pigs. No difference in the intensity of immunostaining for Bcl-2, Bax and GAP-43 was found between control and hysterectomized gilts. Immunoblotting revealed the presence of Bcl-2 and Bax proteins in IMGs of control and hysterectomized animals and no difference in the band intensities between control and experimental groups was detected. RT-PCR detected weak induction of bcl-2 and bax only in the ganglia of animals which had undergone total hysterectomy. It was found that the axotomy of the uterus-projecting neurons located in the porcine IMG did not change the expression of the studied substances (Bcl-2, Bax and GAP-43) at protein level and only the induction of bcl-2 and bax at the level of RNA was visible.

Uterus-derived growth substances: possible implications for human uterus physiology and pathophysiology (review). The Group for Gynecological Investigation.

The presence of mitogens and inhibitors in myometrial, endometrial and leiomyoma extracts was documented. Growth substances of human uterus extracts were sensitive to tryptic digestion and were acid stable. Mitogens and inhibitors were retained and could be eluted from cartridges of octadecylsilyl-silica by 80% acetonitrile. Reverse-phase high performance liquid chromatography (r-HPLC) could separate mitogens from inhibitors. In addition, r-HPLC profiles revealed eluates of myometrial and leiomyoma extracts containing mitogens with preferential activity for either osteoblasts or myoblast-like cells, respectively. The presence of mitogens and inhibitors in human uterine extracts suggests a possible involvement of these growth substances in the regulation of various physiological and pathophysiological processes of human uterus.

Uterus unicornis with uterus solidaris.

Five patients are presented who manifested a heretofore unreported uterine anomaly: uterus unicornis with uterus solidaris. The condition is illustrated and described in detail. Patients with this anomaly have a good fertility potential, although they may expect a high fetal wastage, frequent premature labor and delivery, increased complications of pregnancy and delivery, and an increased incidence of infants that are small for the pregnancy duration. The importance of laparoscopy in diagnosing this anomaly is emphasized.

Acute puerperal inversion of the uterus--treatment by a new abdominal uterus preserving approach.

OBJECTIVE: Inversion of the uterus is still a rare (1, 2) but serious and life-threatening obstetric complication. It is said to be complete when the fundus uteri protrudes through the cervix and into the vagina. Within minutes a state of shock is reached due to pulling forces on the peritoneum as well as blood loss. METHODS: The vagina was entered by a longitudinal incision (3 m) below the contraction ring. Through this opening it was possible to advance two fingers (second and third finger of the left hand) into the vagina above the invaginated corpus uteri. The invaginated cavum uteri was loaded on these two fingers and, exerting counterpressure with the right hand, the inside was turned out. CONCLUSION: The present operative method guarantees easy reposition of the uterus in cases of failed vaginal manual repositioning. Furthermore, the cavum uteri remains intact and hysterectomy can be avoided.

[The detection of the carcinoma in situ of the cervix of the uterus and of the invasive carcinoma of the cervix of the uterus (author's transl)].

Report of the detection of 203 cases (87 of a carcinoma in situ of the cervix of the uterus, 116 of carcinoma of the uterus) out of 7769 cytologically examined patients at the ob.-gyn. deptm. of the Hospital Vienna-Lainz (Austria). The accuracy of cytology with respect to the detection of the carcinoma of the cervix: 95%. The importance of a gynecological examination performed at least once a year together with a cytological test and the examination of the breast is hereby stressed.

Uterus didelphys with fibroid uterus and ovarian cyst--rare Muellerian malformation.

Persistent Muellerian duct syndrome is a very rare anomaly. A woman of 46-year-old was admitted with the history of lower abdominal pain, bleeding per vaginum and irregular menstrual cycles for last 6 months. An irregular non-tender mass of 16 x 10cm in size was palpable in suprapubic region. Per vaginal examination revealed two cervices and an incomplete vaginal septum. Ultrasound abdomen showed one uterus with endometrial cavity clearly delineated with a fibroid and an ovarian cyst in the left lumbar region. On laparotomy, 2 uterii along with a fibroid from the anterior wall of left uterus and a left ovarian cyst was seen. Patient had undergone total abdominal hysterectomy and adnexa removal. The specimen showed florid adenomyosis with leiomyoma of fibroid.

Quantitative study on the uterus-inhibiting activity of sodium naproxen in rat isolated uterus.

Action of Sodium Naproxen on spontaneous and field stimulated motility of isolated rat uterus was studied. Sodium Naproxen inhibited both spontaneous and field stimulated motility. The IC50 on spontaneous motility was lower than the IC50 on stimulated motility. The inhibition produced on the field stimulated uterus was more regular in relationship to concentration used of Sodium Naproxen. The IC50 of Sodium Naproxen in these experimental conditions was 8.22 X 10(-4) M (pD2 = 3.09 +/- 0.02) and the maximum inhibitory effect was 100%. These results show that prostaglandin synthesis plays a necessary role in uterine contraction, spontaneous or induced by field stimulus.

[Differences in right-left distribution of foetuses in uterus cornua and relevance of differentiated germ implantation in uterus? -experiments with laboratory mice (author's transl)].

Corpora lutea were counted in laboratory mice, and the production of the right ovary was found to be slightly higher than that of the left. Rates of implantation, consequently, were higher in the right cornu of the uterus. No right-left difference between uterus cornua was observed regarding foetal weight and sex distribution. Any impact of implantation site on germ survival was not safely established.

Septate uterus: new idea on the histologic features of the septum in this abnormal uterus.

OBJECTIVE: Our purpose was to examine some of the classic concepts about the histologic features of the uterine septum. STUDY DESIGN: Four biopsy specimens were taken from 16 patients with septate uteri during Tompkins metroplasty: biopsy A from the septum near the serosal layer, biopsy B from the midpoint of the septum, biopsy C from the tip of the septum, and biopsy D from the left posterior aspect of the uterus away from the septum. Thirteen characteristics were examined in these specimens by means of mean ridit analysis and Bonferroni criteria for multiple comparisons in relation to connective tissues (four characteristics), muscles (four characteristics), and vessels (five characteristics). RESULTS: In the septum the mean ridit score was less for the amount of connective tissues (p < 0.001) and higher for the amount of muscle tissue (p < 0.025), amount of muscle interlacing (p < 0.05), and vessels with a muscle wall (p < 0.005). However, the Bonferroni criterion for multiple comparisons was only significant for the amount of connective tissue (p < 0.001). CONCLUSION: Our findings of less connective tissue in the septum, which was confirmed by the Bonferroni criterion for multiple comparisons and the mean ridit results for the three other significant findings, were contrary to the classic view about the histologic features of the septum.

Progesterone in the uterus. V. Correlation of the in vitro progesterone metabolism with the progesterone binding in rat uterus.

Incubations of rat uterine segments with varying 3H-progesterone concentrations were performed to study the hormone uptake by the tissue. The radioactivity of the uterus and the nutrient medium were plotted in form of a SCATCHARD plot. Additionally, the binding capacity of the uterine cytosol was measured. In both systems, the hormone was found to be associated with two components which differ from each other in their association constants. The progesterone metabolism occuring at a hormone concentration of 10(-6)M and more in the incubation medium is discussed with respect to the affinity and the capacity of the hormone binding components.

Progesterone in the uterus. X. Dependence of the in vitro progesterone metabolism on progesterone binding in rat uterus.

The effect of estrogen pretreatment was stud-ed on the in vitro metabolism and binding of progesterone in uteri of ovariectomized rats in order to prove the dependence of the metabolism of progesterone on its binding. For this purpose, the extent of progesterone binding was varied in uterine tissue by different estrogen treatment of the rats and compared with the metabolism under the same conditions. The protein content determined in 100 mg tissue was used as parameter indicating the success of the pretreatment. Estrogen exposure of the rats for 30 or 45 hrs. caused a rise of protein amount in uterine tissue which was accompanied by an increase of binding sites of progesterone binding components. The binding sites were determined by charcoal adsorption technique and SCATCHARD-analysis. Under nearly the same success of estrogen pretreatment, the increase of the portein amount and with it the rise of binding sites reduced the amount of progesterone metabolites in uterine tissue. The metabolites were determined by quantitative TLC-analysis of the recovered compounds from uterine segments after incubation with radioactive progesterone. Additionally, an enlarged metabolic rate could be observed after saturation of binding components. It is concluded from the results of these experiments that progesterone binding components are factors limiting the enzymatic conversion of progesterone in rat uterus.

Prospective comparison of ultrasound and computed tomography in the evaluation of the size of the uterus: can these methods be used for intracavitary treatment planning of carcinoma of the uterus?

A prospective evaluation of computed tomography and ultrasound was performed on 34 patients with Stage I and II endometrial carcinoma. All patients underwent immediate surgery following intracavitary treatment directed to vaginal mucosa. Pathologic measurements of the uterus were compared to those obtained by imaging technologies. The results of the study suggest that all but height measurements were rather accurately determined by both ultrasound and CT scan. However ultrasound was significantly better in determining the size of the cervix. Therefore ultrasound measurements could be used routinely for intracavitary treatment planning in endocervical carcinoma and endometrial carcinoma.

The nuclear oestrogen receptor in the female rat. Effects of oestradiol administration during the oestrous cycle on the uterus and contrasting effects of progesterone on the uterus and hypothalamus.

Oestradiol administration to immature or ovariectomized rats has been reported to increase the uterine content of long-term nuclear oestrogen receptors. However, in the intact adult female rat, oestradiol administration did not increase the concentration of long-term nuclear oestrogen receptors at all phases of the oestrous cycle. Progesterone administration to rats in late dioestrus did not affect the concentration of uterine nuclear oestrogen receptors 24 h later, although it did prevent the normal cyclic increase at pro-oestrus in the concentration of hypothalamic nuclear oestrogen receptors. Our results therefore show that in the intact adult rat, factors other than the concentration of progesterone or oestradiol determine the nuclear concentration of oestrogen receptors in the uterus. They also demonstrate differences between neural and non-neural tissues in the regulation of oestrogen-receptor interactions.

[Histological and biochemical studies of the uterus of ovariectomized gilts during the first two months of pregnancy after different hormone substitutions. 4. Effect of the conceptus on the activities of alkaline and acid phosphatase and the concentration of glycogen in the uterus during different supplies of progesterone and norgestrel].

Forty-five ovariectomised gilts in early gravidity received injections of progesterone doses between 120 mg and 40 mg as well as 250 micrograms/die of oestradiol-benzoate and oral application of 12 mg/die of norgestrel, the latter being administered in addition to the 40 mg dose of progesterone. The animals were killed between the 20th and 24th or on the 35th days of gravidity, before endometrial activities were determined of alkaline and acid phosphatases at various points of the uterus (centre and sides of ampullae and in-between ampullae). Under conditions of sufficient progesterone supply (120 mg/die and 100 mg/die), endometrial activities of both alkaline and acid phosphatases in the centre of ampullae were found to be higher than those in-between them. Activities were lower with inadequate progesterone supply (40 mg/die), and differences between points of sampling were less strongly pronounced for alkaline phosphatase. Endometrial glycogen concentrations in the centre of ampullae were lower than those in-between. Values in response to inadequate progesterone administration were lower than those following sufficient supply. With regard to glycogen concentrations, differences between points of sampling in the myometrium were below those in the endometrium.

[Hysteroplasty for hemi-uterus and divided uterus. Apropos of 45 cases].

The authors present a series of 45 plastic operations for bicornuate uterus. Three women were operated on for functional symptoms, eight for sterility of which seven were primary, and thirty four for infertility. They are sorry that three women were lost to follow up. They used the Bret-Palmer technique with modifications that they themselves describe. They justify this choice as against other techniques of metroplasty. The anatomical results are, in the main, good when controlled by hysterosalpingography 3 to 6 months after the operation. The functional results, by which is meant the pregnancies that have been obtained, are medium as far as sterility is concerned. Four out of the eight women who were operated on became pregnant. But they are very good in cases of infertility, because thirty out of the thirty four cases that were operated on had one or two pregnancies. The authors justify their indications for metroplasty after a first spontaneous abortion by the good results they have had, and because bicornuate uteri give rise to many obstetrical complications. Furthermore in unoperated cases there is often difficulty in attempting a new pregnancy, and quite often spontaneous late abortions or premature labours. In certain cases they carried out the operation after first spontaneous abortions, but they are more conservative in operating in cases of primary sterility. The authors do not think it is necessary always to carry out a cerclage procedure nor a caesarean operation automatically in cases that have become pregnant after metroplasty.

16 alpha-[77Br]bromoestradiol-17 beta: a high specific-activity, gamma-emitting tracer with uptake in rat uterus and uterus and induced mammary tumors.

16 alpha-[77Br]Bromoestradiol-17 beta (Compound 1) has been synthesized by radiobromination of estrone enoldiacetate. Tissue uptake studies performed 1 hr after administration of Compound 1 to immature or mature female rats showed uterus-to-blood ratios of 13, with nontarget issue-to-blood ratios ranging from 0.6 to 2. Co-administration of unlabelled estradiol caused a selective depression in the uterine uptake with no effect on nontarget tissue uptake. In adult animals bearing adenocarcinomas induced by DMBA (7,12-dimethylbenz(a)anthracene), tumor-to-blood ratios of 6.3 were obtained, this uptake also being depressed in animals treated with unlabeled estradiol. The studies demonstrate that Compound 1 has suitable binding properties and sufficiently high specific activity so that its uptake in estrogen target tissues in vivo is mediated primarily by the estrogen receptor. Furthermore, they suggest that this compound may be suitable for imaging human breast tumors that contain estrogen receptors.
