Teratogenic effects in early chick embryos of solanine and glycoalkaloids from potatoes infected with late-blight, Phytophthora infestans.

Solanine or a preparation of mixed glycoalkaloids from potatoes naturally infected with the late-blight fungus, Phytophthora infestans, was injected into fertile chicken eggs between 0 and 26 h of incubation, before formation of the neural tube. The embryos were examined after a total of 72 h of incubation. Various abnormalities were found, the most conspicuous being absence of the tail or trunk below the wing bud (rumplessness). A statistically significant proportion of the abnormal embryos showed malformations that seemed to be related to this condition; these included fluid- or blood-filled vesicles in the lower trunk or tail region on one or both sides of the neural tube. Such abnormalities were not observed in control embryos.

CHIH-DFT determination of the molecular structure and infrared and ultraviolet spectra of gamma-solanine.

Glycoalkaloids are toxic secondary plant metabolites found in potatoes, tomatoes, and eggplants and they are considered potential precursors of steroids for applications in bionanotechnology. In this work, we make use of a new model chemistry within density functional theory, which is called CHIH-DFT, to calculate the molecular structure of gamma-solanine, as well to predict its infrared (IR) and ultraviolet (UV-vis) spectra, and some other electronic parameters.

Immunoaffinity sample purification and MALDI-TOF MS analysis of alpha-Solanine and alpha-chaconine in serum.

A sample purification technique was developed for the detection of potato glycoalkaloids (GAs) in blood serum by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). GAs were extracted from spiked serum (5 mL) using a C(18) solid-phase extraction cartridge. The GAs were then selectively captured on antibody-coated agarose beads. The agarose beads were washed with water and the GAs eluted with 25 microL of methanol. MALDI-TOF MS was used to detect the GAs in the methanol eluent. Immunoaffinity sample purification of the GAs effectively reduced the signal suppression observed during the analysis of unpurified samples. alpha-Chaconine and alpha-solanine were detected in serum spiked with 1 ng/mL of each GA.

Synthesis of gamma-chaconine and gamma-solanine are catalyzed in potato by two separate glycosyltransferases: UDP-glucose:solanidine glucosyltransferase and UDP-galactose:solanidine galactosyltransferase.

UDP-glucose:solanidine glucosyltransferase and UDP-galactose:solanidine galactosyltransferase from cytosol of potato sprouts were partially separated by Sephadex G-100 and Q-Sepharose chromatographies, proving the existence of different glycosylation systems in biosynthesis of alpha-chaconine and alpha-solanine.
