Topical nylidrin and aqueous humor dynamics in rabbits and monkeys.

Topical administration of nylidrin hydrochloride lowered intraocular pressure in both albino rabbits and cynomologus monkeys. The maximum decrease occurred two hours following drug application and was unilateral. The response lasted four hours in monkeys and at least six hours in rabbits. Outflow facility, episcleral venous pressure, and aqueous humor flow measured by fluorophotometry were unaltered in both species. In rabbits in the treated eyes, ocular blood flow was increased, and anterior and posterior chamber ascorbate concentrations were unchanged. The mechanism of IOP reduction appears to be increased aqueous humor outflow via uveoscleral outflow routes that are not pressure dependent.

Effects of buphenine (nylidrin) on the perfused mammalian eye.

Buphenine (nylidrin), a beta-adrenergic agonist, is used therapeutically for its vasodilating effect on the peripheral circulation and possibly on the cerebral circulation as well. In spite of its ophthalmic usage in degenerative retinal disease and glaucoma, buphenine's ocular effects and their mechanisms are not sufficiently established. Consequently, we studied the action of 4.5-120 microM buphenine in isolated, arterially perfused cat eyes, and then used light-evoked electrical signals, perfusion flow rates and the diameters of the retinal vessels as parameters for the drug's effect. Our findings showed that buphenine induced a marked, dose-related, reversible increase in the amplitude of the electroretinogram (ERG) b-wave, a decrease in the c-wave, but no significant changes in the standing potential and light peak of the DC-ERG. The compound action potential of the optic nerve revealed dose-dependent and reversible changes in configuration. However, the flow of perfusate was not affected by the drug, and the diameter of retinal vessels did not change significantly. Our studies suggest that the interaction of buphenine with retinal adrenergic receptors is not related to the vasculature present but to elements involved in information processing. It is likely that these receptors are linked to neurons, since the beta-agonist affected the ERG b-waves as well as the compound action potential of the optic nerve.

[Action of nylidrin (Dilatol) on utero-placental blood supply (author's transl)].

The effects of one single intravenous injection of 5 mg of nylidrin, following administration of 113m-indium, on utero-placental blood supply were studied in 23 women in advanced pregnancy, all of them afflicted with hypertension. Pulse rates over the placenta rose in 16 cases, primarily in patients from which prolonged half-life was measured.

[Prevention of premature birth with nylidrin and verapamil (author's transl)].

108 cases of threatened premature birth were treated with i.v. drip-infusion, followed by oral treatment with Dilatol and Isoptin. This was successful in 81% of cases. A birth-weight of over 2500 g was considered a positive result. In 14% of cases there were side effects. The results are compared with earlier investigations.
