Effects of guanidine derivatives on mitochondrial function. II. Reversal of guanidine-derivative inhibiton by free fatty acids.

Long chain free fatty acids interfere with the inhibitory action of phenethylbiguanide and related compounds on mitochondrial respiration in vitro. This interference depends on binding of fatty acids to mitochondria and diminishes with decreasing chain length. Reversal of guanidine-derivative inhibition by fatty acids differs from that caused by dinitrophenol in that the effect of fatty acid is achieved without alteration in coupling or respiratory control. The binding of phenethylbiguanide to mitochondria is inhibited by both fatty acid and dinitrophenol. Serum albumin potentiates the inhibitory potency of guanidine derivatives, probably by removing endogenous mitochondrial free fatty acids.

Modified guanidines as potential chiral superbases. 1. Preparation Of 1,3-disubstituted 2-iminoimidazolidines and the related guanidines through chloroamidine derivatives

Modified guanidines were explored as potential chiral superbases. Thus, chiral 1,3-dimethyl-2-iminoimidazolidines with or without 4, 5-diphenyl groups, their guanidinium salts, and the 2-iminoimidazolidines with (S)-1-phenylethyl groups on the ring nitrogens were prepared by treatment of 2-chloroimidazolinium chlorides with appropriate amines. Bicyclic guanidines were also prepared from a prolinamide using a similar procedure.

Modified guanidines as potential chiral superbases. 2. Preparation Of 1,3-unsubstituted and 1-substituted 2-iminoimidazolidine derivatives and a related guanidine by the 2-chloro-1, 3-dimethylimidazolinium chloride-induced cyclization of thioureas

Simple preparation methods for modified guanidines were explored for new chiral superbases. Thus, (4S,5S)-4,5-diphenyl- and diastereomeric cyclohexane-fused 2-iminoimidazolidines were prepared from (1S,2S)-1,2-diphenylethylenediamine and (1R,2R)- or (1S,2S)-1, 2-diaminocyclohexanes through cyclization of protected thiourea intermediates with 2-chloro-1,3-dimethylimidazolinium chloride (DMC) as a key reaction. In the (4S,5S)-4,5-diphenyl series 1-methyl-2-iminoimidazolidines and 2-diethylaminoimidazoline were also prepared as related guanidines.

Structural evidence for guanidine-protein side chain interactions: crystal structure of CutA from Pyrococcus horikoshii in 3 M guanidine hydrochloride.

This study was carried out to investigate the structural perturbation of the protein's local structure by the denaturants under non-denaturing conditions. Crystal structure of CutA from an archaeon Pyrococcus horikosii (PhoCutA), a heavy-metal binding protein, was determined at 1.6-angstroms resolution in the presence of 3 M guanidine HCl (GdnHCl). Native PhoCutA has a large number of short intramolecular hydrogen bonds and salt bridges on the protein surface, of which greater than 90% of hydrogen bonds and all salt bridges were retained in 3 M GdnHCl. Hydrogen bonds that disappeared in the GdnHCl crystal structure were mainly located on the protein surface, especially around the structurally perturbed loop, suggesting interactions between peptide groups and GdnHCl. Only a few GdnH+ ions were observed in the crystal structure, although none at the surface, of the protein. Two GdnH+ ions were observed in the center of the trimeric structure, replacing water molecules, and were hydrogen bonded with Asp84 and Asp86 of each chain. The exterior loop from Tyr39 to Lys44, including Trp40-Trp41, was perturbed structurally. Decreases in temperature factors were observed in beta strand 5 and the N terminus of helix 3. These results suggest the specific bindings of GdnH+ with some acidic residues and the non-specific bindings around Trp residues and peptide groups on the protein surface and that binding of GdnHCl to the native protein is limited, resulting in local structural perturbation.

Catalytic construction and reconstruction of guanidines: Ti-mediated guanylation of amines and transamination of guanidines.

Bis(guanidinate) titanium imido complexes [{(Me2N)C(NiPr)2}2TiNAr'] (Ar' = 2,6-Me2C6H3 (1a); C6F5 (1b)) are competent catalysts for the guanylation of a variety of arylamines with carbodiimide. The reversible [2 + 2] addition of iPrN=C=NiPr to 1b is demonstrated and is proposed to be part of the catalytic cycle. Compounds 1a and 1b are also effective precatalysts for the transamination of trialkylguanidines with arylamines to yield aryldialkylguanidines.

The insertion of carbodiimides into Al and Ga amido linkages. Guanidinates and mixed amido guanidinates of aluminum and gallium.

The insertion of carbodiimides into existing metal-heteroatom bonds is an important preparative route for the synthesis of useful ligand systems such as amidinates and guanidinates. Our interest lies in multiple insertions at one metal center and the mechanisms of insertion and rearrangement. We have synthesized and characterized [Me(2)NC(N(i)Pr)(2)](n)M(NMe(2))(3)(-)(n) (n = 1, 2, 3; M = Al, Ga). We have investigated the mechanism of synthesis and discovered a ligand transfer step that is important for the formation of the final products.

Transition Metal Complexes of Guanidinate Dianions: Reactions between Guanidines and M(NMe(2))(5) (M = Ta, Nb).

The protonation of two metal-amido groups of M(NMe(2))(5) with trialkylguanidines yielded a series of novel complexes with formulas [RNC(NR)NR]M(NMe(2))(3) (1-4) (M = Ta, Nb; R = isopropyl, cyclohexyl). These complexes contained dianionic N,N',N' '-trialkylguanidinate ligands which were coordinated in a chelating bidentate mode. A single-crystal X-ray study of [CyNC(NCy)NCy]Ta(NMe(2))(3) (3) (C(25)H(51)N(6)Ta, triclinic, P&onemacr;, a = 9.4155(2) Å, b = 13.3188(2) Å, c = 13.5215(2) Å, alpha = 117.075(1) degrees, beta = 101.744(1) degrees, gamma = 98.507(1) degrees, Z = 2) confirmed the connectivity of these species. These guanidinate ligands exhibited both planarity of the central CN(3) group and the correct orientation of the three NR substituents to allow for pi conjugation within the ligand core.
