Fowl cholera immunization in turkeys. 3. Significance of market quality in the evaluation of fowl cholera vaccines.

Turkeys vaccinated with various experimental vaccines and a commerical bacterin for fowl cholera and surviving an artificially induced epornitic were killed, and their carcasses were examined for wholesomeness. It was evident that, if the "fitness for human consumption" judgement was considered in addition to mortality, efficacy ratings of the various vaccines changed. This suggests that the "fitness for human consumption" factor be considered in future evaluation of biologicals for use in meat-producing birds.

Scanning electron microscopy of fowl oviducal cells and HeLa and BHK-21 cells incubated with or without fowl spermatozoa in vitro.

The morphology of cells cultured at 41 C with and without fowl spermatozoa was observed with the scanning electron microscope. At the beginning of incubation, abnormal spermatozoa were comparatively few, and spermatozoa were found on the surface of the cultured cells. Morphological alteration of spermatozoa increased with increasing incubation interval and included head and tail coiling with occasional separation of heads from flagella.

[Interaction of the M protein of the wild-type fowl pest virus and its ts-mutant with the ribonucleoprotein of the fowl pest virus in vitro].

The extent of inhibition of transcription realized in vitro by fowl plague virus (FPV) ribonucleoprotein (RNP) upon the addition of M protein isolated from FPV virions does not depend on nonionic detergent concentration in the reaction medium but does depend greatly on NaCl concentration. The highest inhibition of transcription is observed at a low ionic strength (0.02 M NaCl); inhibition is completely eliminated by increasing NaCl concentration to 0.3 M. When M protein isolated from FPV virions with ts mutation of M protein is added to RNP, addition to the system of 0.3 M NaCl decreases transcription inhibition but does not eliminate it completely.

Evaluation of a commercial modified live virus fowl pox vaccine for the control of "variant" fowl poxvirus infections.

Three-week-old specific-pathogen-free chickens were vaccinated with either a commercial modified live virus fowl pox vaccine or five "variant" poxvirus field isolates. Immunity engendered by the commercial modified vaccine or field isolates was challenged with either the variant isolates or commercial modified vaccine virus. The commercial modified vaccine did not adequately protect vaccinates against challenge with the variant isolates. The percentages of vaccinated chickens protected following challenge with each of the variant isolates were 70%, 20%, 30%, 20%, and 25%. However, when the isolates were applied as vaccines, 100% of the vaccinates were protected against challenge from the modified vaccine virus. Furthermore, the variant poxvirus isolates offered excellent protection from challenge with homologous variant isolates. The modified live virus vaccine was expected to offer significant protection against challenge from the variant pox isolates, but in this experiment it did not. The variant isolates tested may be good vaccine candidates to prevent the vaccine breaks currently encountered in previously pox-vaccinated flocks.

Polymorphism in fowl serum albumin. VI. Changes in in vitro protein synthesizing activity in developing embryonic fowl liver.

Cell-free protein synthesizing systems were prepared from the livers of chick embryos at selected ages and the characteristics of individual fractions were compared. While polysomes showed decreasing size with older embryos, isolated polysomes did not differ significantly in amino acid incorporating activity when assayed with standard cell sap. When assayed with standard polysomes, cell sap activity decreased with increasing developmental age whether incorporation was measured using (3H)lysine, (3H)leucine, or [3H]aminoacyl-tRNA. Free amino acid concentrations in the cell sap showed reproducidble independent variation during development which was taken into consideration in calculating net amino acid incorporation. A larger increase in ribonuclease activity was observed during development; however, nuclease inhibitor activity was absent before day 15 but increased thereafter. Aminoacyl-tRNA sythetase activity did not vary significantly. It is proposed that the observed changes in the rate of cell-free protein synthesis result not only from increasing ribonuclease activity with increasing developmental age but also from changes in the activity of other soluble factors.

Polymorphism in fowl serum albumin. VII. Distribution and activity of free and membrane-bound polysomes in developing fowl liver.

The quantity and activities of membrane-bound and free polysomes in livers from chick embryos at successive stages of development were compared in cell-free protein-synthesizing systems. Membrane-bound polysomes increased 2-fold between 8 and 18 days of development, while total ribosome content remained constant. Free polysome activity also remained constant during this period, while that of membrane-bound (total--free) polysomes decreased, possibly because of an increase in ribonuclease activity in this fraction. Serum albumin biosynthesis occurred primarily on membrane-bound polysomes. With liver development, increased secretion of serum proteins may be correlated with synthesis of serum albumin on increasing numbers of membrane bound polyribosomes.
