Two new antifungal saponins from the Tibetan herbal medicine Clematis tangutica.

Bioassay-guided fractionation of the ethanol extract of the aerial parts of Clematis tangutica led to the isolation of two new antifungal triterpene saponins. Their structures were determined to be 3- O-alpha- L-arabinopyranosyl hederagenin 28- O-alpha- L-rhamnopyranosyl ester ( 1) and 3- O-beta- D-glucopyranosyl-(1--> 4)-alpha- L-arabinopyranosyl hederagenin 28- O-alpha- L-rhamnopyranosyl ester ( 2) on the basis of spectral data and chemical evidence. Inhibitory activities of the two saponins against seven fungal strains were evaluated. Compounds 1 and 2 showed evident antifungal activity (MIA approximately 2.5 micrograms/disc) against Saccharomyces cerevisiae, similar to the positive control amphotericin B and ordinary activities (MIA approximately 10 micrograms/disc) against Penicillium avellaneum UC-4376, Candida glabrata, Trichosporon beigelii and Pyricularia oryzae. Compound 2 is a better antifungal agent than compound 1 against most of the fungal strains that were tested.

Clematis huchouensis Tamura: a traditional Chinese herbal medicine and its quality control using a high performance liquid chromatography technique.

A simple, specific and reliable high performance liquid chromatography (HPLC) method has been developed and validated for study of fingerprint chromatograms of extracts from the whole plant of Clematis huchouensis Tamura (CHT) for quality control of a traditional Chinese medicinal (TCM) herb. An Agilent C18 column was used to separate extracts in this protocol and detection was made by ultraviolet absorbance at 340 nm. The column temperature was maintained at 25 degrees C. A mobile phase consisting of (a) water (with 30 mM KH2PO4) and (b) CH3CN, (c) CH3OH was found to be suitable for this separation at a flow rate of 0.8 microl min(-1) with gradient elution. Under the described conditions, a fingerprint profile of 8 compounds was collected within 35 min, which made the HPLC method unique and interesting. The fingerprint chromatograms had good stability, precision and reproducibility. Moreover, eco-climatic (habitat) effects were studied by comparison of fingerprint chromatograms obtained from extracts of CHT collected from two habitats, with rutin as a reference marker peak. The protocol developed is quite suitable for differentiation of extracts of CHT and can be used as a quality control method for this herb and a model for other herbal drugs.

Bumblebee-pollination and temporal change of the calyx tube length in Clematis stans(Ranunculaceae).

Clematis stans is dioecious semi-arboreal, with pale purple-blue, nodding, tubulous flowers in a paniculate inflorescence. Both male and female flowers produce nectar from the base of the calyx tube during a flowering period of 3 or 4 days, and are pollinated by two bumblebee species, Bombus diversus and B. honshuensis, with different proboscis lengths. When the flowers open, four sepals constructing a calyx tube separate at the top and their respective tips gradually curl up, so that a tubular part shortens. Observations at two field sites showed that B. diversus (with a longer proboscis) most often visits the flowers with a longer calyx tube, and B. honshuensis (with a shorter proboscis) the flowers with a shorter calyx tube, i.e., later in the flowering period. By changing the calyx tube length, the flowers of C. stans accept the two bumblebee species with different proboscis length as pollinators and thus increase the chance of pollination for each flower. It was also found that the two bumblebee species prefer the male flowers to the female flowers, although the female flowers secrete more nectar as a reward than male flowers. This is likely because they visit the male flowers to collect pollen grains in addition to nectar.

The hypotensive action of Desmodium styracifolium and Clematis chinensis.

The cardiovascular pharmacology of aqueous extracts of Desmodium styracifolium (DSE) and Clematis chinensis (CCE) were studied in rats both in vivo and in vitro. DSE produced two successive hypotensive actions: the first one via cholinergic receptor stimulation, while the second one potentiated by blockades of autonomic ganglion and alpha-adrenoceptor. In contrast to DSE, CCE produced only one hypotensive response which was mediated through histaminergic activity. Furthermore, both extracts relaxed isolated methoxamine preconstricted helical tail artery strips. CCE also produced both negative chronotropic and inotropic effects on isolated atria, while DSE was positive chronotropic without apparent effect on the contractile force.

[Identification of Tougucao derived from Clematis species].

Comparative studies were carried out on the morphology and histology of 5 Clematis species, namely, C. intricata, C. aethusifolia, C. glauca, C. tangutica and C. hexapetala, allegedly used as "Tougucao". New characteristics, a sclerenchymatous ring composed of primary phloem fibers and lignified parenchyma cells, and a parenchymatous tertiary ring were discovered. A parameter representing the ratio of the diameter of cambium ring to the diameter of the stem (RCR) is created. A key for identification of the stems of the above five species is made according to the presence or absence of the sclerenchymatous ring, the diameter and numbers of hair cells, the thickness of fiber wall and the value of RCR, etc.
