Glutathione and related enzymes in fascioliasis before and after treatment with bithionol.

The present work is an update evaluation of the glutathione status in patients with established fascioliasis before and after treatment with bithionol. Blood glutathione (GSH), erythrocyte glutathione S-transferase (GST) and serum gamma-glutamyl transferase (GGT) activities were studied. After treatment, the variations observed in these parameters were restored to the corresponding normal control values confirming the toxic features resulting from fascioliasis and suggesting no adverse effect of bithionol on the parameters studied. We recommend the use of serum GGT, blood GSH and erythrocyte GST for the early detection of therapeutic response in fascioliasis.

Human fascioliasis: ultrastructural study on the liver before and after bithionol treatment.

The pathology of human fascioliasis was studied before and after bithionol treatment using light and transmission electron microscopy. Fine needle biopsies were taken from five patients, with established fascioliasis, before and after drug administration. By light microscope the pathology of human fascioliasis was similar to that reported in experimental fascioliasis. The ultrastructural picture revealed bile ductular hyperplasia, fibrosis of portal tracts, widening of the interhepatic spaces by many microvilli and dilated Disse space with collagen fibres. Bile ductular hyperplasia may be the initial factor to fibrinogenesis, which subsequently enhance the development of the microvilli on the surface of the hepatocytes. Both light and electron microscopic studies revealed regression of the picture of fascioliasis to normal after bithionol treatment with no sign of toxicity on the liver.

Bithionol (Bitin) treatment in established fascioliasis in Egyptians.

The present study is an evaluation of bithionol treatment in established fascioliasis. Eight patients were enrolled in the study. All were passing eggs of Fasciola spp. and seven had concomitant Schistosoma mansoni infection. The clinical and haematological findings conformed with the known picture of fascioliasis. Bitin tablets (200 mg) were administered in the dose of 30 mg kg-1 body wt every other day for five doses. Eggs were counted during and for 4 months after the end of therapy. Fasciola ova disappeared after the second or third dose giving a cure rate of 100%. Ova of S. mansoni did not show any change in number. As regards the liver function tests; bilirubin, SGOT and SGPT values were within the normal range and were not affected by treatment. A raised SAP level was observed before therapy which was significantly higher after bithionol. Creatinine phosphokinase and lactate dehydrogenase enzymes and kidney function tests showed almost normal values and were not altered by therapy. It was concluded that Bitin was a potent fasciolicidal drug in the dose used. No side effects were observed.

[The prevention and therapy of fascioloidiasis using bithionol sulfoxide].

The elimination of the sources of invasion, i. e. hay and forage coming from the infested biotopes, combined with the administration of bithionol sulphoxide, resulted in a reduction of the invasion of cattle by Fascioloides magna from 21.1 to 3.2% within two years. The Czechoslovak-produced bithionol sulphoxide, administered at a rate of 40 to 50 mg per kg body weight as medicated feed, showed 100% effectiveness. At doses higher than 30 mg per kg body weight, the protective period should be prolonged to three or four weeks.

[Autoradiographic study of the distribution of 14C bithionol sulfoxide in mice (author's transl)].

The distribution of a single oral dose of 14C bithionol sulfoxide in mice was determined by whole body autoradiography. The bithionol sulfoxide was found in the whole organism mainly in richly vascularized organs, liver, kidney, lung. It did not pass through the blood-brain barrier and did not accumulate in any particular organ but persisted in the blood. Observations were performed in 18-day pregnant mice in order to investigate an eventual placental transfer. Six hours after the administration, radioactivity was detected in blood and fetal vascularized tissues.

[Treatment Of Taeniasis With Bithionol]

Ten cases infected with Taenia saginata were treated with Bithionol, 2, 2' thiobis (4,6-dichlorophenol) which is well known as an excellent anthelminthhic against paragonimiasis and their follow-up studies were carried on within 3 to 12 months after treatment. Bithionol, 50 mg dose per kg of body weight was divided into two equal dose and administered orally in 30 minutes intervals. Three hours later, this was followed with a sodium sulfate or magnesium sulfate purge. The strobilas without scolices were found in the stools after the medication and the follow-up studies have shown the complete deworming in all cases. It is well tolerated, toxic effects being uncommon and when present, limited to mild gastro-intestinal disturbances.

Successful praziquantel treatment of paragonimiasis following bithionol failure. A case report.

A patient from Southeast Asia with cough and hemoptysis was documented to have pulmonary paragonimiasis. Initial treatment with bithionol failed. The patient was then treated with praziquantel 75 mg/kg body weight a day for 2 days under an investigational protocol. Subsequent follow-up studies showed clinical improvement and indicated parasitologic cure. A concurrent asymptomatic Clonorchis infection was also cured following praziquantel treatment. Side effects were minor.

[Study of the effect of tegalide and bithionol on biological membranes using fluorescent probes].

Fluorescent probes were used to study structural changes in different membranes affected by gamma-radiation, protonophores and radioprotective agents, tegalide and bithionol. The preparations of the defined concentrations decreased the microviscosity of membranes, lowered the peaks and changed the temperature of phase transitions in liposomes from dipalmitoyl lecithin, and induced the output of Ca2+ from mitochondria. The effects depended on the radiation dose, the structure, concentration and lipophilicity of the preparation; protonophores produced a specific effect.

Compartive studies on the effect of bithionol, praziqantel and triclabendazole in rabbit's fascioliasis. 1. Parasitological studies.

Rabbits were infected with 30 metacercariae each. They were then divided into four batches each of two groups. One batch served as controls and the others were treated each with a drug. The first group from each batch was treated when worms were still immature, the other group was treated after maturity. The effect of the drugs was monitored by: (1) Stool examination for eggs and studying the percent of egg reduction. (2) Worm recovery at autopsy and calculating the percent of the drug efficacy. (3) Culturing some eggs and following their embryonation and hatching. Results pointed out that Triclabendazole was the best fasciolicide followed by Bithionol. Praziquantel had a negligible effect.

[A Study On The Chemotherapy In Clonorchiasis: Report 1. An Experimental Study On Chemotherapy With Dithiazanine Iodide And Bithionol Sulfoxide In Clonorchiasis]

Authors carried out experimental study on chemotherapy with oral administration of Dithiazanine iodide (D.I.) and Bithionol sulfoxide(B.S.) in rabbit clonorchiasis. And the following result was obtained. 1)In change of the E.P.G (eggs per gram feces) by D.I. administration, it was rather increased in early stage of the administration than prior to administration, and thereafter decreased gradually. 2)In the change of the C.S. worm body by D.I. administration, there was not only prominent change of supporting tissue but also the change of reproductive organ was found. 3)In considering the wormicidal effect of D.I. from detecting rate of survival worms, the effect was slight in group of 100 mg(80 mg/kg) per day dosage, but the effects were very excellent and almost complete by proper times of administration in groups of 200 mg (130 mg/kg) per day or higher dosage. 4)But the side effect and intoxication sign of D.I. were appearent in groups of 200 mg or higher dosage. 5)By B.S. administration, E.P.G. was decreased gradually. 6)In changes of the C.S. worm body by B.S. administration, prominent inhibitory chnnge was seen in egg formation ability. 7)Slight wormicidal effect of B.S. was observed in groups of 140 mg (100 mg/kg) per day or higher dosage. 8)Side effect and intoxication sign of B.S. were found little in groups of 140 mg or lesser dosage.

The effect of bithionol sulphoxide on Echinococcus granulosus and Taenia hydatigena infections in dogs.

Trials with bithionol sulphoxide involving 130 dogs, infected with either Echinococcus granulosus or Taenia hydatigena, indicated that a single treatment at a dose rate of 400 mg/kg or two treatments at 50 mg/kg eliminated the former, but a single treatment at 50 mg/kg was sufficient to eliminate the latter. Dose rates 200 mg/kg or more were lethal to some dogs. With two doses of 50 mg/kg, there was only minor discomfort from vomiting or diarrhoea in some dogs.
