Bitterness suppression of BCAA solutions by L-ornithine.

The purpose of the present study was to evaluate the bitterness-suppressing effect of L-ornithine (L-Orn) on single or mixed solutions of branched-chain amino acids (BCAAs) using human gustatory sensation tests and an artificial taste sensor. The BCAAs tested (L-isoleucine (L-Ile), L-leucine (L-Leu), and L-valine (L-Val)) are the main components of various enteral nutrients or supplements. The bitterness-suppression effect of L-Orn was also compared with the effect of L-Arg. L-Orn was effective in suppressing the bitterness of single or mixed solutions of BCAAs in human gustatory sensation tests, the effect being similar to or greater than that of L-Arg. The artificial taste sensor was able to predict the bitterness-suppressing effects of L-Orn and L-Arg. The response electric potential patterns of L-Val, L-Leu and L-Ile solutions to which 100 mM L-Arg had been added were quite similar to the sensor response patterns of the 100 mM L-Arg solutions alone. The relative response electric potential patterns of L-Val, L-Leu or L-Ile solutions containing 100 mM L-Orn in channels 5-8 (positively charged) are similar to that of single solution of 100 mM L-Orn.

[Metabolic effects of GFX+BCAA solution in the old aged rats after operation].

Effects of total parenteral nutrition (TPN) on the perioperative metabolism in old aged rats were evaluated. Extremely old rats (over 100 weeks old) and young rats undergoing laparotomy were treated with TPN (200 ml/kg/days, 182 cal/kg/day, 148 nonprotein cal/N ratio) consisting of carbohydrate mixtures (combination of glucose, fructose, and xylitol, 4:2:1 GFX solution) and 10% amino acids solution with 30% of branched chain amino acids (BCAA). Blood, liver, and urine samples were analyzed on 1, 3 postoperative days. Increased body weight, positive balance in daily nitrogen excretion, and decreased water content of the liver were observed in old rats. Protein, glycogen, and triglyceride contents of the liver tissue increased in old rats. However the young rats showed negative nitrogen balance and stronger catabolic effect than the old rats. It suggested negative nutritional balance in young rats with this decreased dosage of GFX+BCAA solution suitable for the old rats. In conclusion, decreased amounts of GFX+BCAA solution improved perioperative metabolism in the extremely old.

Amino acid (GAS:BCAA) ratios in plasma and gut contents of short-term protein depleted rats.

The ratio of total concentrations or molar ratios (moles/1,000 amino acid residues) of three non-essential amino acids (glycine, alanine, serine-GAS) and three essential-branched chain amino acids (valine, isoleucine, leucine-BCAA) were investigated in rat systemic and portal vein plasma and jejunal and ileal gut contents after feeding normoprotein (NP) or protein-free (PF) diets for 7 days. Amino acid analysis of gut content showed that the GAS:BCAA ratio was not significantly altered by the PF diet either in the jejunum or in the ileum. On the contrary, the PF diet, caused a three and four-fold increase in this ratio in the portal and systemic plasma, respectively. The situation was produced by the higher concentrations of GAS, which remained near control levels (portal plasma) or exceeded these values (systemic plasma), in contrast to the decreasing levels of BCAA found in both plasmas of the PF group.

Administration of granulated BCAA and quality of life.

The PEM (protein energy malnutrition) is an important factor for improvement of prognosis and QOL in the patients with liver cirrhosis. In 453 chronic liver disease patients, QOL decreased significantly according to the progression of disease assessed by SF-36 (Kruskal-Wallis test: p<0.05) Moreover, the QOL of patients with chronic liver diseases was improved in BCAA administration group (n=13) compared with non-administration group (n=12) after 6 months period. In conclusion, BCAA administration showed improvement of PEM and QOL of patient with chronic liver diseases.

Overtraining and the BCAA hypothesis.

The purpose of this review was to give an answer to the question whether there are convincing data to support the hypothesis of an amino acid imbalance as one possible mechanism to explain overtraining syndrome. Animal studies point to an enhanced synthesis of the neurotransmitter 5-hydroxytryptamine through an amino acid imbalance at the blood-brain barrier with a preferable tryptophan uptake into the brain, resulting in premature fatigue. Human studies, however, show contradictory results, mainly because of nonstandardized methodology, so that a final conclusion cannot be made at present. BCAA supplementation in addition to standard carbohydrate ingestion during sustained exercise seems to be of no eminent advantage to delay fatigue. The overall results concerning the BCAA hypothesis to explain overtraining are inconclusive and require more controlled experimental research.

Suppression of the bitterness of enteral nutrients using increased particle sizes of branched-chain amino acids (BCAAs) and various flavours: a taste sensor study.

An improved formulation of the enteral nutrient Aminoleban EN (Otsuka Pharmaceutical Co., Ltd., Tokyo, Japan), has been commercially available since Spring 2004. Like the previous formulation, the improved product contains branched-chain amino acids (BCAAs) L-isoleucine (L-Ile), L-leucine (L-Leu), and L-valine (L-Val), but the average particle size of these amino acids has been increased to 180 to 250 microm in the improved formulation, compared with 40 to 90 microm in the old product. The improved formulation has a significantly lower bitterness intensity score than the older formulation, as evaluated both in human gustatory tests and using the artificial taste sensor. We propose that this improved taste masking is due to the larger particle size of the BCAA crystals, due to which their release rates are reduced. The addition of improved flavours has also helped to reduce the bitterness of the improved Aminoleban EN formulation significantly. Analysis of the taste sensor data suggests that the sourness and sweetness of the added flavours were critical in diminishing the bitterness of Aminoleban EN.

[Usefulness of branched-chain amino acid (BCAA)-enriched nutrient mixture for nutritional treatment undergoing endoscopic treatment for esophageal varices].

We investigated the alteration of nutritional status in 144 patients who were treated for the first time with endoscopic sclerotherapy or endoscopic variceal ligation during their therapies. The serum levels of albumin, cholinesterase and total cholesterol were compared before and after treatment. The serum level of cholinesterase declined significantly. To investigate the impact of aging on the changes of nutritional status we divided all patients into two groups: (1) under 65 years, and (2) over 65 years. The decline of serum albumin of elderly patients (n=65) was significantly greater than that of younger patients (n=79). A branched-chain amino acid (BCAA)-enriched nutrient mixture for nutritional treatment significantly suppressed the decline of serum albumin in elderly patients. Nutritional treatment with a BCAA-enriched nutrient mixture should be considered during endoscopic therapy for esophageal varices, especially in elderly patients.

The utilization of the Antarctic environmental specimen bank (BCAA) in monitoring Cd and Hg in an Antarctic coastal area in Terra Nova Bay (Ross Sea--Northern Victoria Land).

The first projects relating to levels of Cd and Hg on marine biota and sediments from Terra Nova Bay (Ross Sea--Antarctica) and their bioaccumulation and biomagnification in this trophic web have been carried out by research programmes pertaining to the Italian Antarctic Research Program (PNRA) since 1989. Making use of this data, and checking the same metals after 10 years thanks to the samples stored in the BCAA, we have looked for the levels of Cd and Hg in a coastal marine ecosystem of Terra Nova Bay, and have proposed using some organisms to monitor the levels of these two heavy metals in this environment where the Italian Base is located, using the data determinate in this work as background levels. In our work, the amount of Hg and Cd concentrations have been determined in biota from the inner shelf of Terra Nova Bay (Adamussium colbecki, Laternula elliptica, Odontaster validus, Sterechinus neumayeri, Trematomus bernacchii, Iridaea cordata, Phyllophora antarctica, Parborlasia corrugatus), and in two different size fractions of sieved marine sediments (<2000 microm and <63 microm). To widen the distribution of Cd and Hg in this ecosystem we have also investigated the fraction of these metals bound to the labile phase of the marine sediments, and their presence in the particulate matter found in pack-ice cores, recent snow, water column and sea microlayer.

Nocturnal energy and BCAA supplementation in patients with liver cirrhosis.

Patients with liver cirrhosis have protein-energy malnutrition (PEM) and run short of proteins and energy. Protein deficiency leads to hypoalbuminemia, which induces peritoneal effusion and edema. Energy deficiency decreases fat and muscle mass and causes muscle weakness, which decreases the QOL of these patients. A decrease in triceps skinfold thickness (TSF) and arm muscle circumference (AMC), which are indicators of hypoalbuminemia and energy deficiency, and a decrease in respiratory quotient (RQ) affect the prognosis of these patients. Therefore, appropriate nutritional assessment should be performed in order to detect PEM in these patients, and necessary proteins or energy should be supplied. TSF, AMC and RQ are improved by increasing the energy supply to correct its deficiency and by dividing daily food intake (in some cases, adding nocturnal supplementation). Serum albumin levels increase when branched-chain amino acids (BCAAs) are administered after every meal or at bedtime. Consequently, nutritional assessment on PEM, and nutritional therapy based on this assessment are essential for improving the QOL and prognosis of patients with liver cirrhosis.

Controlled overexpression of BCKD kinase expression: metabolic engineering applied to BCAA metabolism in a mammalian system.

A common metabolic complication of human disease is uncontrolled muscle protein breakdown or cachexia, which occurs in patients with chronic diseases such as cancer, AIDS, renal failure, and diabetes. Increased branched-chain amino acid catabolism is implicated as causal and has stimulated the investigation of methods to regulate the metabolism of these amino acids. Here we demonstrate doxycycline-controlled overexpression of a branched-chain alpha-ketoacid dehydrogenase (BCKD) kinase transgene in mammalian cell culture. This kinase functions to inactivate the BCKD complex by phosphorylation, thus preventing the catabolism of these essential, regulatory metabolites. In this study, doxycycline treatment leads to a 10-fold increase in BCKD kinase protein. The transgene-generated kinase is rapidly incorporated within mitochondria and functions correctly to inactivate the BCKD complex. The maximum reduction in basal BCKD activity achieved was 94%. Unexpectedly, total BCKD activity was also decreased by kinase overexpression despite no observable change in expression of the BCKD catalytic proteins. These results demonstrate that artificial regulation of branched-chain amino acid metabolism is possible through the controlled overexpression of a single endogenous enzyme and suggest the feasibility of clinical applications.

[The behavior of visceral proteins in digestive surgery patients undergoing total peripheral parenteral nutrition enriched with branched-chain amino acids (BCAA)].

The objective of this study, is to confirm a clear benefit of protein synthesis after administration of branched amino acids in parenteral nutrition solutions. Particularly, on a parenteral total hypocaloric nutrition including lipids. Eighty one digestive surgical patients, were included in this study. Fifty with neoplastic disease and thirty one without. Patients in both study groups received a similar peripheral vein nutritional support regimen one week after surgical treatment. To evaluate the effects of the branched chain amino acid enrichment on total parenteral nutrition, we observed the behavior of short middle life proteins such as transferrin, prealbumin, and retinol binding protein, for their reliability as indexes of protein synthesis.
