Plasma testosterone in Klinefelter's syndrome: diurnal variation and response to ACTH and dexamethasone.

The mean basal plasma testosterone level in 28 patients with chromatin positive Klinefelter's syndrome was significantly lower than in 58 healthy male controls. In 12 of the patients the hormone levels were in the normal range. In both the eugonadal males and the Klinefelter patients the plasma testosterone levels spontaneously decreased throughout the day, the relative decrease in both groups being of the same order of magnitude. Short term ACTH infusion and the administration of dexamethasone did not significantly influence the plasma testosterone concentration in the Klinefelter patients. These findings do not support the view that the adrenal cortex plays a major role in contributing to the circulating plasma testosterone levels in this syndrome.

[Clinical biological possibilities of the early detection of the side effects of antibiotics].

The study of the dynamics of biogenic amines in the blood of patients provided to a certain extent determination of the possible thread of the drug complications. In this respect the serotonin level was of special significance. As for the reaction of leucocyte agglomeration it may be used before the treatment if there is a doubt in the tolerance of the antibacterial drug prescribed. Increased serotonin blood levels in this case also serve a confirmation of the thread of the possible allergic or toxic reaction in the patient.

[Sex hormone excretion in stomach cancer patients].

Study on the balance of sex hormones in 43 males and 45 females with gastric cancer indicated the presence of some deviations from normal level of androgens and estrogens excretion in gastric cancer patients. The character and intensity of the deviations observed were dependent on patients' sex and stage of the disease. In males the leading factor in sex hormones excretion disorders is a regular decrease both of 17-KS and estrogens, and only in stage UV hyperestrogenization phenomena are observed on account of active estrogens being predominant over androsterone. In females relative hyperestrogenization was observed already in stage III on acount of androsterone decrease, while in stage IV--due to reduction of total 17-KS in normal level of total enstrogens.

Laboratory medicine. Series on clinical testing. 2. Fractionation of urinary ketosteroids. Procedure and clinical significance.

Ketosteroids, the excretory metabolities of adrenal and gonadal steroids, can be analyzed individually in urine by a simple extraction procedure followed by separation and quantitation by the use of gas/liquid chromatography. The ketosteroids quantitatively detected by the technique are androsterone, etiocholanolone, dehydroepiandrosterone, 11-hydroxyandrosterone, 11-hydroxyetiocholanolone, 11-ketandrosterone, and 11-ketoetiocholanolone. Other steroid metabolities detected are pregnanediol, pregnanetriol, delta5-pregnenetriol, and 11-ketopregnanetriol. In comparison with concentrations of these steroids observed in urine specimens collected from healthy individuals, abnormal results occur in specimens from patients with testicular disease, Cushing's syndrome, adrenal hyperplasia, and several types of female hirsutism. Characteristic profiles for each of these diseases are presented.

[Studies on gamma-oryzanol. III. Influence of gamma-oryzanol on circadian rhythms of serum gastrin, 11-OHCS and gastric secretion in rats (author's transl)].

In the course of investigations of gamma-oryzanol in rats, circadian rhythms in antiulcerogenic action on gamma-oryzanol were observed. Circadian rhythms in several other parameters were investigated and correlated with the antiulcerogenic action. Pretreatment of rats with gamma-oryzanol 100 mg/kg, s.c., for 5 days tended to decrease serum gastrin levels and gastric secretion, and to increase serum 11-OHCS. Suppresive patterns of serum gastrin and gastric secretion following the pretreatment with gamma-oryzanol corresponded to the circadian rhythms in antiulcerogenic action of gamma-oryzanol. A complication of our results suggests that increment of catecholamines in rat brain after gamma-oryzanol plays a role in the above mentioned actions.

Tryptophan metabolism in baboons: effect of riboflavin and pyridoxine deficiency.

Seven urinary metabolites of tryptophan-kynurenine pathway were measured in riboflavin-deficient, pyridoxine-deficient, pair-fed and natural diet baboons. The most significant changes in the pyridoxine-deficient baboons was a mean sevenfold increase in the excretion of xanthurenic acid and a threefold decrease in 3-hydroxy anthranilic acid. The riboflavin-deficient baboons showed a twelvefold increase in the excretion of anthranilic acid, and a tenfold decrease in 3-hydroxy kynurenine. Red blood-cell pyridine nucleotides decreased only in the pyridoxine-deficient baboons, while plasma 11-hydroxy corticosteroids increased only in the riboflavin-deficient baboons. These results are discussed in relation to enzymatic changes that may be expected during these deficiencies.

[Structuro-metabolic mechanisms for integrating several endocrine organs].

Mechanisms of integration in the neuroendocrine system in the regimens of physiological rhythm (oestral cycle) and of isolated effect upon one link of the system (metapyrone administration) were studied with the aid of qualitative histoenzymological analysis of adenohypohysis and adrenal cortex, estimating the comoplex reorganization of various structural components of adenohypophysis and adrenal cortex in response to specific depression of glycocorticoid synthesis. The role of the so called unspecific reorganization in the maintenance of functional response to isolated effects upon certain endocrine functions is discussed along with the necessity of further studies on the principles of interaction in the endocrine system using morphofunctional analysis.

[Effect of sealed chamber conditions on the functional state of the human adrenal cortex (based on data from a study of the 11-hydroxycortico-steroid content in the blood plasma)].

The effect of a changed atmosphere, hypoxia, hypercapnia, their combinations and different motor activities on the adrenocortical function was studied in 36 test subjects kept in an 8 m3 altitude chamber. Human adaptation to the environmental changes developed with an active involvement of the adrenal cortex. The level and direction of the changes depended on both the force of the influences and on the initial state of the test subjects.

[Hormone and mediator content in the blood of the adrenals and in the peripheral blood in hypertension].

The concentration of the fractions of corticosteroids, aldosterone, catecholamines, and cyclic nucleotides (cAMP and cGMP) in blood of the adrenal veins and the activity of renin in blood of the renal veins were studied in 22 patients with stages IIA and IIB hypertensive disease. At the same time the content of these substances in the peripheral blood was determined and compared with the level of steroid and catecholamine excretion in the daily urine. An increase in the content of free 11 OCS and F fractions in the peripheral blood and blood of the adrenal veins was revealed in all patients examined.

[The character of functional connections in the endocrine system of female white rats and their transformation with age].

The authors studied the functional interrelations between the adrenal glands and the thyroid gland of female albino rats of various age groups. Blood content of 11-oxycorticosteroids and protein-bound iodine was determined. Analogous determinations were carried out in partial exclusion of the adrenal gland function (metapyron block) and of the thyroid gland function (6-methyluracil block). It is supposed that in the young sexually-mature female albino rats the functions of the adrenal glands and the thyroid gland were in direct relationship irrespective of the cyclic variations in the sex steroid secretion; with the advance of age this association became deranged.

[Activity of adrenal cytoplasmic dehydrogenase following prolonged ACTH administration].

The activity of cytoplasmic dehydrogenases of the adrenal cells was examined during the prolonged injections of ACTH to Wistar rats. The indices of specific steroid synthesis function of the glands remained relatively high in the course of the whole experiment. Changes in the rats of corticosterone synthesis and dehydrogenase activities were phasic in character; they included an initial synchronic activation the first two days with its subsequent decrease (7 days of ACTH injections). The prevalence of activities of NADP-dependent dehydrogenases in combination with reactivation of steroidogenesis was marked during the concluding phase (13 days) of the experiment. A possibility of the adaptive role of selective activation of NADP-dependent enzymes in the maintenance of a high level of hormone biosynthesis under conditions of prolonged ACTH stimulation is discussed.

[Effect of long-lasting noise on the plasma concentration of catecholamines, glucocorticosteroids and PBI in pigs].

Exposure of pigs with a body mass of 25-30 kg to long-lasting motor noise (108 dB, 60-8000 Hz, 72 hours) results in a considerable increase of catecholamine concentration in the plasma. Coincidently there is a sharp continuous decrease of the 11-OHKS in the plasma during the whole period of exposure to noise. Immediately after stopping the acoustic irritation the concentration of plasma corticosteroids increases again. The electroencephalogram (EEG) giving evidence of inhibition signs during exposure to noise an adaptation effect of the information processing system is concluded. The increase of the PBJ during the period of acoustic stress points to an increased function of the thyroid gland but simultaneously it is considered to be an expression of the elevated adrenergic situation. By reason of the strong deviation of the three observed regulatory systems must be concluded that longer-lasting exposure to noise with the applied quality and intensity represents a stress situation for pigs having noise character as established for men. A possible influence on the productivity of the pork type pig by permanent acoustic stress is indicated.

[Indices of lipid metabolism, thyroid gland and adrenal cortical function in prolonged stress of the central nervous system in chronic stimulation of the adrenals].

The experiments were conducted in 14 male rabbits dynamically on the 5th, 10th, 20th, 30th, 40th, 60th and 80th day of the central nervous system tension or at rest, including the studies of lipid metabolism (total cholesterol and beta-lipoproteins content), the function of the thyroid gland (protein bound iodine content) and the function of the adrenal cortex (11-OCS content). Such lasting stimulation of the adrenal cortex was found to be accompanied by hypercholesterolemia, hyper-beta-lipoproteinemia, elevated 11-OCS level and lack of thyroid function changes. A lasting tension of the central nervous system against the background of chronic stimulation of the adrenals delays the development of hypercholesterolemia and hyper-beta-lipoproteinemia, reduces the function of the thyroid gland, and increases the function of the adrenal cortex. The observed shifts in the lipid metabolism may be of atherogenic importance. The obtained data may be useful for the study of the pathogenesis of hormonal shifts in atherosclerosis.

[Glucocorticosteroid, catecholamine and protein-bound iodine (PBI) plasma levels in swine following electrocutaneous stimulation].

Application of electrocutaneous stimuli for short-time excitation of swine, accompanied by high locomotor activity, led in animals weighing between 30 kg and 45 kg to steep rises of catecholamine and glucocorticosteroid plasma concentrations, while little change was recordable from protein-linked iodine. Increase in plasma glucose and RQ in animals with higher rises in plasma-borne catecholamines and glucocorticoids was higher than that in animals with lower catecholamine deflection. Both the results reported in this paper and findings obtained from other studies into swine are likely to suggest major involvement of the sympathico-adrenomedullary system in the buildup of transport stress. Interdependence was found to exist between the intensity of the adrenergic reaction, on the one hand, and rises in plasma glucose and RQ as well as drop of pH, on the other. Those findings from various tests seem to support the conclusion that under conditions of severe excitation and high locomotor activity the differentiated resistance of swine to stress is atributable to different intensities of sympathico-adrenergic reaction of the animal concerned.
