I. A bibliographic database for quantitative analysis of phosphorus flow in ruminants.

The present study describes a quantitative analysis of phosphorus metabolism in ruminants. The database compiles 100 sources either published (95) or unpublished (5) involving studies on true or apparent phosphorus balances carried out before 1999. The database reports 652 distinct experimental conditions (lines) described with experimental variables (column) involving a total of 2982 animals: 414 lambs (33 references), 1078 sheep (37 references), 64 pregnant ewes (3 references), 212 lactating ewes (5 references), 528 calves (20 references), 4 maintenance cows (1 reference), 519 lactating cows (6 references), 42 kids (1 reference) and 121 lactating goats (3 references). The reliability of the database was tested using some general relationships.

Quantitative analysis for computed tomography findings of various diffuse lung diseases using volume histogram analysis.

OBJECTIVES: : The aim of this study was to achieve the quantitative analysis of the characteristic computed tomography (CT) findings and course of interstitial pneumonia using the volume histogram method. METHODS: : Contrast (CNT), variance (VAR), and entropy (EPY) values from whole-lung volume data were compared between normal lungs and 5 diseases that have characteristic CT findings. Thirteen cases with nonspecific interstitial pneumonia (NSIP) were evaluated before and after treatment. RESULTS: : In cases with thickening of the bronchovascular bundles and interlobular thickening, ground-glass attenuation, airspace consolidation, and honeycombing, the values of VAR and EPY were greater than those in the normal cases (P < 0.05). In the cases with NSIP, the CNT value after treatment was significantly greater and the values of VAR and EPY after treatment were significantly lower than those before treatment (P < 0.05). CONCLUSIONS: : Volume histogram analysis is a promising method for the evaluation of diffuse lung diseases and the effectiveness of treatment.

Quantitative analysis of bone metastasis in prostate cancer.

A new method of quantitative analysis of bone metastasis in prostate cancer is described. By using a variance-dependent filter and a smoothing filter, the increased uptake lesions were distinctly demonstrated on the bone scintigraphy. The images were divided into several areas and analyzed. These procedures resulted in a clear identification of the metastatic lesions. Repeated measurements of the metastatic areas from the same scintigrams disclosed a high reproducibility. This method appears to be highly consistent and clinically applicable.

Computerized quantitative analysis of left ventricular wall motion by two-dimensional echocardiography.

Quantitative detection of left ventricular segmental wall motion abnormalities by any modality depends on the reference system used because of the dynamic geometry of contraction and cardiac motion within the thorax. To assess the feasibility and accuracy of quantitative analysis of left ventricular wall motion by two-dimensional echocardiography, we studied 61 subjects with the use of 44 different reference methods in each of three echocardiographic views: the parasternal short-axis view at the levels of the mitral valve and of the papillary muscles and an apical four-chamber view. The three major groups of reference systems used were those with a fixed external reference, a floating reference correcting for translation, and systems correcting for both translation and rotation. In the first part of this study the end-diastolic and end-systolic outlines of 20 normal subjects were stored in a computer and composite data of these 20 subjects were plotted to obtain a 95% confidence interval for measured normal fractional change for each reference method. In the second part of the project an additional prospective group of 10 normal subjects and a group of 31 "abnormal" patients had their left ventricular wall motion analyzed by similar methods and the results were compared with all the confidence intervals. One reference method was selected for each two-dimensional echocardiographic view based on the highest sensitivity and specificity found by statistical analysis; a floating-reference system including translation was found to be optimal for the apical four-chamber and parasternal short-axis views at the level of the mitral valve and a fixed external reference system was optimal for the short-axis view at the papillary level. The percent fractional shortening of radial dimensions (radial methods) and the percent fractional change in area measurements (area methods) during the cardiac cycle were also calculated at 5, 10, 20, 30 and 45 degree intervals around the outline perimeter for each subject according to each of the 44 different methods. Area methods yielded the same specificity and sensitivity as radial dimension analysis methods at 5 to 45 degree intervals. Ten normal subjects underwent repeat echocardiography within 2 days of their first study to examine day-to-day variation. Average change in mean contraction from day to day was 7% to 9% for radial methods and 9% to 13% for area methods. In conclusion, we present a computerized system for unbiased selection of optimal methods of analysis of left ventricular wall motion by two-dimensional echocardiography.(ABSTRACT TRUNCATED AT 400 WORDS)

A quantitative analysis of head-shaking nystagmus of peripheral vestibular origin.

A quantitative analysis of horizontal head-shaking nystagmus (HSN) was made on 48 patients with unilateral peripheral vestibular lesions in conjunction with stimulus intensity. Each patient underwent three head-shaking tests with 10, 30 and 50 horizontal head-excursions at a frequency of approximately 2 Hz, and HSN was recorded on ENG with eyes open in total darkness. i) HSN appeared in a biphasic or monophasic pattern. ii) The maximal slow-phase eye velocity (MSV) of the 1st phase (PI) of biphasic HSN increased significantly in proportion to stimulus intensity, and was significantly greater than that of monophasic HSN. iii) The duration of HSN was greater in the 2nd phase (PII) of biphasic HSN than in PI and increased markedly in proportion to stimulus intensity. iv) As the stimulus intensity rose to a high level, the interval between PI and PII (2nd phase latency) shortened, and the PII tended to appear more quickly after head-shaking. It was especially noteworthy that in response to an increase in stimulus intensity, both the MSV in PI and the duration of PII of biphasic HSN increased, but the duration of PI was reversely suppressed by the PII.

[A quantitative analysis of pulmonary arterioles in 402 cases of cor pulmonale].

A quantitative analysis of pulmonary arterioles were made in 402 cases of cor pulmonale. The results showed that arteriole muscularization and medial hypertrophy, intimal fibrosis of pulmonary small arteries were important lesions. The authors found that right ventricular hypertrophy was bound to follow intimal fibrosis of pulmonary small arteries but was not along with medial hypertrophy and arteriole muscularization, and that the lesion of pulmonary arteries was not balanced with the course of disease. The authors considered that these results were related to vessel dilatation and medial atrophy because of intimal pressure in late stage of the disease.

[Application of MALT-CLS method to FTIR quantitative analysis of atmospheric trace gas].

The MALT-CLS method for quantitative analysis of atmosphere trace gas by FTIR spectrometry was studied. Some experiments are described, such as long-path White cell, and passive remote sensing of aircraft. The characteristic of this method is that the calibration spectra are calculated from a database of absorption line parameters HITRAN using MALT program, including environmental and instrumental effects in the calculation. It is particularly useful in long open-path and solar FTIR spectroscopy, or passive remote sensing by FTIR.

Quantitative analysis of anti-emetic principle in the tubers of Pinellia ternata by enzyme immunoassay.

A crude globulin fraction was isolated from the tubers of Pinellia ternata (Thunb.) Breit. Tubers of P. ternata are one of the well known Chinese traditional medicines. It is used on account of its anti-emetic effect. 6KDP was separated by means of gel filtration and chromatography on CM-Toyopearl 650 M from the crude globulin fraction. 6KDP is a glycoprotein of M.W. 6 kDa. 6KDP has an anti-emetic activity and hemagglutination activity. Both activities dropped during heat treatment. For the purpose to evaluate the quality of the powdered tubers of P. ternata, the quantitative analysis by means of EIA was established. The contents of 6KDP varied from 5.75 to 8.30%. The assay demonstrated a low cross-reaction with the crude globulin fraction of Colocasia antiquorum, a member of the same family as P. ternata. 6KDP is one of major proteins in the tubers of P. ternata. Thus 6KDP is a marker for quantitative analysis of the tubers of P. ternata.

Quantitative analysis of antibiotics by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.

Comparative studies of the matrix-assisted laser desorption/ionization (MALDI) of 30 antibiotics were made using alpha-cyano-4-hydroxycinnamic acid (HCCA), 2,5-dihydroxybenzoic acid (DHB), 5,10,15,20-tetrakis(4-hydroxyphenyl)-21H,23H-porphyrin and meso-tetra(N-methyl-4-pyridyl)porphyrin matrices. Most antibiotics generated intense protonated molecules in HCCA and DHB matrices, and sodium or potassium adduct ions in porphyrin matrices. Using sulfonamide antibiotics as model compounds, DHB was found to be the most suitable matrix for quantitative analysis. Detection limits were about 1 picomole. Linear correlation (R2 > 0.97), between the sample quantity over the range 1 to 100 picomole and the signal response, was obtained when ratios of the sum of peak areas of protonated molecules and sodium adduct ions, with reference to that of a structurally analogous internal standard (acetaminophen), were measured. The precision was found to be in the range of 4 to 32% relative standard deviation, dependent on the type and concentration of the analyte. A simple acylation derivatization process was developed to confirm the presence of suspected antibiotic residues. It is demonstrated that MALDI is a viable technique for the quantitative analysis of low molecular weight antibiotics.

Quantitative analysis of constitutive heterochromatin in couples with fetal wastage.

PROBLEM: Heteromorphism of constitutive heterochromatin is a stable evolutionary feature that is thought to cause no phenotypic alterations. Nevertheless, the role of constitutive heterochromatin is still unknown. The instability of constitutive heterochromatin was generally restricted to T-lymphocytes and was associated with variable immunodeficiency. The heterochromatin regions of chromosomes 1, 9, 16, and Y have been postulated to play a role in the immune response and during early embryo development. METHOD OF STUDY: To investigate a possible influence of constitutive heterochromatin in human reproductive ability, quantitative analysis of constitutive heterochromatin in human chromosomes 1, 9, 16 and Y was done. Thirty couples were divided into two groups, owing to the clinical heterogeneity of their reproductive disorders. The first group included couples with two or more spontaneous abortions as the only pregnancy outcomes, and the second group included couples with a stillborn child with or without malformations. In the control group were couples with one or more healthy children without a history of fetal wastage. All of the persons in this study had normal karyotypes. The amount of constitutive heterochromatin was expressed by relative value using the simple transformation [q/(p + q)]. This value, obtained on GTG-banded metaphase chromosomes, represented an indirect measure of heterochromatin content. The Y/F index was used to express the relative amount of heterochromatin in chromosome Y. RESULTS: There was a significant increase in the heterochromatin content of the chromosomes 16 homologue pair in males and females with a stillborn or a stillborn malformed child (P < 0.01) and an increase in total heterochromatin cell content compared to controls (P = 0.005). The same couples had significantly increased mean maximal heterochromatin content in the potential zygotes (P < 0.02). The couples who experienced spontaneous abortions only had a minimal total heterochromatin content in the potential zygotes (P < 0.05). The Y/F index was significantly lower in the males in both groups compared to controls (P1 < 0.02; P2 < 0.02). CONCLUSION: The quantitative analysis of constitutive heterochromatin could be valuable in predicting pregnancy outcome.

Improving the linearity of infrared diffuse reflection spectroscopy data for quantitative analysis: an application in quantifying organophosphorus contamination in soil.

Diffuse reflection data are presented for ethyl methylphosphonate in a fine Utah dirt sample as a model system for organophosphate-contaminated soil. The data revealed a chemometric artifact when the spectra were represented in Kubelka-Munk units that manifests as a linear dependence of spectral peak height on variations in the observed baseline position (i.e., the position of the observed transmission intensity where no absorption features occur in the sample spectrum). We believe that this artifact is the result of the mathematical process by which the raw data are converted into Kubelka-Munk units, and we developed a numerical strategy for compensating for the observed effect and restoring chemometric precision to the diffuse reflection data for quantitative analysis while retaining the benefits of linear calibration afforded by the Kubelka-Munk approach. We validated our Kubelka-Munk correction strategy by repeating the experiment using a simpler system--pure caffeine in potassium bromide. The numerical preprocessing includes conventional multiplicative scatter correction coupled with a baseline offset correction that facilitates the use of quantitative diffuse reflection data in the Kubelka-Munk formalism for the quantitation of contaminants in a complex soil matrix, but is also applicable to more fundamental diffuse reflection quantitative analysis experiments.

Quantitative analysis of ductus proliferation, proliferative activity, Kupffer cell proliferation and angiogenesis in differential diagnosis of biliary atresia and neonatal hepatitis.

BACKGROUND/AIMS: The clinical presentation of cholestasis in infancy caused by neonatal hepatitis and biliary atresia are very similar. Differential diagnosis is sometimes very difficult. The diagnostic accuracy is very important. The surgical treatment of biliary atresia should be performed as early as possible. If cases of biliary atresia are misdiagnosed, they will become progressive cirrhosis and if cases of neonatal hepatitis are misdiagnosed, they will result in unnecessary laparotomy. The aim of this study is to determine the role of quantitative analysis of ductus proliferation, proliferative activity, Kupffer cell proliferation and angiogenesis in the differential diagnosis of biliary atresia and neonatal hepatitis. METHODOLOGY: This study included 60 infants, 30 with neonatal hepatitis and 30 with biliary atresia. Differential diagnosis was done by ultrasonography, cholescintigraphy and liver biopsy. The parafifin-embedded tissue sections of liver biopsies underwent immunohistochemistry with Ki67 to mark proliferation activity, cytokeratin to mark the proliferating ductuli, vimentin to mark Kupffer cells, and CD34 to mark capillary vessels. Number of ductuli per high power field, number of Ki-67 positive cells per ductus, number of Kupffer cells per high power field and number of capillary vessels per high power field were calculated. Independent T test was used for statistical evaluation. RESULTS: Independent sample T test indicated that there is a significant difference for proliferating ductuli and proliferation activity between neonatal hepatitis and biliary atresia. Biliary atresia cases represent more proliferating ductuli and proliferation activity in ductal epithelial cells than neonatal hepatitis cases. There is no statistical significance for Kupffer cell proliferation and vascularization. CONCLUSIONS: Our results indicate that, quantitative analysis of proliferating ductuli and proliferation activity of ductal epithelial cells may be helpful in differential diagnosis of neonatal hepatitis and biliary atresia. Besides neither Kupffer cell proliferation nor vascularization are found to be useful in differential diagnosis.

Expression of telomerase reverse transcriptase mRNA and its quantitative analysis in human endometrial cancer.

Three major components of telomerase, i.e., human telomerase RNA (hTERC), telomerase-associated protein (TEP1), and the human telomerase reverse transcriptase (hTERT), have been identified. Among them, TERT expression is very closely related to telomerase activity. The purpose of this study was to evaluate the implications of TERT expression and telomerase activity in endometrial cancer. Fresh surgical specimens of 36 endometrial carcinomas (CA group) and 9 samples of postmenopausal endometrial tissue without malignancy (NP group) were obtained at operation in our hospital. These specimens were analyzed for telomerase activity and TERT expression by TRAP assay and RT-PCR, respectively, and the detection and quantitative analysis were made. The results for endometrial cancer were compared with those for normal endometrium and with the clinical data. In the CA group, TERT expression was detected in 35/36 subjects (97.2%), whereas in 1/9 subject (11.1%) from the NP group. Relative TERT mRNA expression was 0.50 in the CA group, and this was significantly higher compared with the level of 0.10 in the NP group (p<0.05). Telomerase activity was detected in 34/36 subjects (94.4%) from the CA group and in 3/9 subjects (33.3%) from the NP group (p<0.05), while the RTA was 30.9 and 0.2, respectively (p<0.05). There was a significant correlation between the relative TERT expression and RTA (n=45, R=0.413, p<0.05). RTA was significantly higher at an advanced surgical stage (FIGO II, III or IV) than at an early stage (FIGO 0 or I) (52.4 vs. 20.4, p<0.05), but other clinical factors showed no relationship with TERT and RTA values. The detection and quantitative analysis of telomerase activity and TERT expression is helpful for distinguishing malignant from normal endometrium when the patient is postmenopausal, even if the tumor is very small or of low malignancy.

Automated quantitative analysis of activator protein-2alpha subcellular expression in melanoma tissue microarrays correlates with survival prediction.

The activator protein-2alpha (AP-2) transcription factor plays a key role in regulating expression of genes involved in tumor growth and metastasis of human melanoma. We sought to assess the prognostic significance of AP-2 expression and its role in the transition of nevi to metastatic melanoma. Two cohorts were analyzed. One was a "progression" microarray containing melanoma specimens from M.D. Anderson Cancer Center representing 84 cases and the other was a retrospective cohort from Yale University representing 214 primary melanomas and 293 metastases. Analysis of total AP-2 expression using two quantitative systems [automated quantitative analysis (AQUA) and laser scanning cytometry (LSC)] revealed no correlation with diagnosis group. LSC analysis of the M.D. Anderson Cancer Center array showed that the number of cells expressing nuclear AP-2 was highest in the benign nevi group (11.85%) and significantly decreased in each phase of melanoma progression to 0.39% in the metastatic group. Both LSC and AQUA showed decreased nuclear AP-2 levels and increased cytoplasmic AP-2 that is directly proportional to progression. Neither nuclear nor cytoplasmic expression levels correlated with outcome. Intriguingly, the ratio of cytoplasmic to nuclear AP-2 predicted outcome in the entire population and in the primary tumors alone, demonstrating the power of the ratio to normalize for variations. Furthermore, the AP-2 ratio directly correlated with other clinicopathologic factors, including Breslow depth (R = 0.334, P < 0.001). We show that a high level of AP-2 expression in the cytoplasm relative to the nucleus correlates with poor prognosis and the loss of nuclear AP-2 expression is associated with malignant transformation and progression of melanoma.

[A new computed tomography method for quantitative analysis of abdominal aortic atherosclerosis].

Clinical evaluation of aortic atherosclerosis has been most commonly performed by visual analysis of aortic calcification on conventional radiographs or X-ray CT. However, precise evaluation of the degree of calcification or mild changes with increasing age can be difficult by these methods of visual analysis. A new quantitative method of evaluating the abdominal aortic atherosclerosis using CT is reported. Target CT scans of the abdominal aorta were performed at the level of the 1st, 3rd and 4th lumbar vertebrae. Two circular regions of interest (ROI) were selected along the outer margin and inner margin of the abdominal aortic wall on CT images, since it was difficult to precisely trace the aortic wall. Histograms of CT value for each pixel were made from two ROIs. A histogram at the ROI of the outer margin was depicted using class values over the maximum CT value in histogram at the ROI of inner margin which indicated flowing blood. A sum of products of the value in each class by the number in that class was divided by the aortic diameter. The value thus obtained was defined as the atherosclerotic index (S.I.). Forty-five cases were studied with this method. Six cases were excluded because of artifacts. The remaining 39 cases (16 males, 23 females) were analyzed. S.I. increased with aging and was higher in men than in women. However, it increased more rapidly in women (8.68/year) than in men (2.73/year). Because this new method employed the CT value, it is more objectively quantitative than previous methods of visual analysis in evaluating abdominal aortic atherosclerosis.(ABSTRACT TRUNCATED AT 250 WORDS)

Myocardial contrast echocardiography yields best accuracy using quantitative analysis of digital data from pulse inversion technique: comparison with second harmonic imaging and harmonic power Doppler during simultaneous dipyridamole stress SPECT studies.

AIMS: This prospective study assesses the (1) feasibility of quantifying ultrasound myocardial perfusion studies based on the densitometric analysis of digital data and the (2) comparison of pulse inversion, second harmonic and harmonic power Doppler modalities with SPECT. METHODS AND RESULTS: Twenty-three patients with suspected ischaemic heart disease had i.v. injections of Tc-Sestamibi and Optison during a dipyridamole stress test for echocardiography in pulse inversion, second harmonic and harmonic power Doppler mode. Analysis was (a) visual by scoring and (b) quantitative by densitometry of digital data for background subtracted myocardial opacification (a.u.) and normalized contrast effect (%). In the nine control patients, myocardial opacification at stress was greater (P< or =0.002) than in the pathologic group (5. +/- 3.3 vs 2.6 +/- 2.5 a.u. in pulse inversion, 5.4 +/- 2.1 vs 2.4 +/- 1.8 in second harmonic and 7.1 +/- 3.7 vs 4.9 +/- 3.7 a.u. in harmonic power Doppler). In the pathologic group, normalized contrast effect decreased significantly during stress (23.7 +/- 18.8 to 11.3 +/- 10.8%, P<0.003) only in pulse inversion. Kappa values for patient based diagnostic agreement with SPECT were 0.75 by pulse inversion, 0.62 by second harmonic and 0.52 by harmonic power Doppler for quantitative analysis, and 0.51, 0.37 and 0.35 respectively, for visual assessment. CONCLUSION: Myocardial contrast echocardiography should be analysed using densitometry of digital data. The new technique pulse inversion demonstrates best agreement with SPECT data.

Quantitative analysis of types I and II collagen in the disc annulus in adolescent idiopathic scoliosis.

OBJECTIVE: To find the possible roles of collagen in the pathogenesis of AIS through studies of the distribution of the collagen I and II in the disc annulus fibrous in adolescent idiopathic scoliosis (AIS). METHODS: The disc annulus fibrous of apex curve were harvested during anterior surgery and divided into concave and convex samples. 25 AIS cases were covered in this study including 6 males and 19 females, with an average age of 14.6 years (range 12-18 years). 11 specimens were intervertebral disc from the thoracic region (from T8 to T11) and 14 sample were from lumbar discs (from L1 to L2). RT-PCR was employed to investigate the distribution and content of collagen I (220 bp) and collagen II (359 bp) in the intervertebral disc specimens. 1% agarose gel electrophoresis and the Gelwork image analysis system was used in the semi-quantitative analysis of the product. RESULTS: For the AIS group, type I collagen and type II collagen significantly increased on convex side compared with that of the concave side (p<0.01). The same trend was observed in both thoracic and lumber disc annulus fibrosis, but there was no statistical difference expect for the expression of type II collagen in convex side of disc annulus fibrous (p<0.05). CONCLUSION: The asymmetric expression of collagen I and II collagen showing that there was degeneration in the intervertebral disc of AIS. The abnormal collagen metabolism may be one of reasons in the development of AIS and probably an important factor in the progression of AIS.

[Assessment of myocardial viability by quantitative analysis of stress Tl-SPECT--comparison with FDG-PET].

Stress Tl-201 tomography (SPECT) is widely used for evaluating myocardial viability. To assess its value, redistribution (RD) on SPECT was compared with metabolic imaging using FDG. Thirty patients with coronary artery disease underwent stress-3 hour Tl-201 SPECT and PET using N-13 ammonia and FDG. RD was classified into 4 grading, including complete RD (CR), incomplete RD (IR), persistent defect (PD) and additional minimal RD (MR) defined as no definite RD on visual analysis but faint RD with Bull's eye quantitative analysis (QNT). All but one segment with CR or IR were viable regions (normal or ischemic regions) by PET. Of 74 segments without RD on visual analysis, 31 segments (42%) had RD by QNT (MR). All of them were viable regions by PET. Thus, QNT identified 31 segments (63%) of the metabolically viable segments which the visual Tl-201 analysis did not show RD and classified as myocardial scar. However, even such QNT cannot detect ischemic myocardium in 18 segments (42%) containing metabolic activity on PET. These data indicate that QNT of RD on Tl-201 SPECT is considered as a valuable means for assessing myocardial ischemia.
