[NK cell activity in patients with kidney insufficiency].

Natural killer cell (NKC) plays an important role in anti-cancer, anti-virus and the regulation of immune reaction in human body. NK cell activities from the peripheral blood were determined with 51Cr release method from the human erythroleukemia cell line K562 in 61 patients, who were selected in the light of the standard of deficiency syndrome in TCM with insufficiency of the Kidney in which 12 kinds of diseases in western medicine were included, and 60 healthy subjects as control group. The patients with insufficiency of the Kidney showed significantly decrease in NK cell activity compared to the control group (25.29 +/- 10.20 vs 56.27 +/- 16.72, P less than 0.001). NK cell activities in 20 out of 60 patients, with deficiency of Kidney-Yang, were lower than 21 with deficiency or Kidney-Yin (19.38 +/- 7.44 vs 26.60 +/- 8.95, P less than 0.01) and 20 with deficiency of Kidney-Qi (19.38 +/- 7.44 vs 29.38 +/- 11.30, P less than 0.01), respectively, while there was no difference between the deficiency of Kidney-Qi (P greater than 0.05). The authors also found that NK cell activities of healthy subjects were strongly inhibited by plasma from 43 out of 61 patients. These results suggested that decreased NK cell activity is one of the common characters in patients with insufficiency of the Kidney in TCM and there are some inhibitory substances against NK cell activity in plasma of ones. The fact that NK cell activity of patients with deficiency of Kidney-Yang was the lowest among three kinds patients with insufficiency of the Kidney.(ABSTRACT TRUNCATED AT 250 WORDS)

[Research on the mechanism of Yin-enriching and Qi-reinforcing, strengthening body and tonification deficiency syndrome action of radix Panacis quinquefolii].

Radix Panacis Quinquefolii could significantly promote 3H-TdR infiltrating of spontaneously and ConA-stimulated splenic lymphocytes of mice; Promote the splenic lymphocytes capacity of mice in interleukin2 (IL-2) production; Significantly enhance erythropoitin (EPO) level of mice in serum; Markedly promote production of colony stimulating factors (CSFs) in murine spleen-conditioned medium (SCM); And promote production of CSFs in lung-conditioned medium (LCM) of mice. The results suggest that yin-enriching and qi-reinforcing, strengthening body and tonification deficiency syndrome action of Radix Panacis Quinquefolii is in association with its lymphocytes activation-stimulating, IL-2 secrection-promoting, EPO secrection, and CSTs production enhancing effectes.

Studies on the luminescence of channels in rats and its law of changes with "syndromes" and treatment of acupuncture and moxibustion.

We previously reported the discovery of 14 channels in the human body, which possessed the biophysical property of high emission of light. In this study we found the same property on the Ren and Du channels in healthy rats. Additionally, we discovered that the luminance of the related channels in rat models with different "syndromes" varied greatly. For instance, a markedly low luminance appeared on the Du channel in animals with experimental syndrome of Yang deficiency induced by hydrocortisone; while in animals with experimental syndrome of blood deficiency caused by bleeding, an apparently low luminance occurred on the Ren channel. The intensity of the emitted light on Du and Ren channels increased after acupuncture treatment, but not significantly. This phenomenon conforms to the theory in traditional Chinese medicine that the Du channel is the "sea of Yang channels", while the Ren channel is the "sea of Yin channels".

Clinical observation on treatment of non-parvicellular carcinoma of the lung with jin fu kang oral liquid.

Jin Fu Kang Oral Liquid ([symbol: see text]), made of traditional Chinese drugs for supplementing qi and nourishing yin, was developed according to the common symptoms in lung carcinoma with deficiency of both qi and yin. Of the 96 cases in the Jin Fu Kang group, 1 case got complete remission (CR) after treatment, 8 cases partial remission (PR), 52 cases no change (NC), PR + NC covering 63.5%. Of the 52 cases in the group of Jin Fu Kang plus chemotherapy, 11 cases got PR after treatment, 26 cases NC, PR + NC covering 71.2%. Of the 25 cases in the chemotherapy group, 4 cases got PR after treatment, 11 cases NC, PR + NC covering 60.0%. The results show that the therapeutic effectiveness in the Jin Fu Kang group and the group of Jin Fu Kang plus chemotherapy was better than that in the chemotherapy group. The one-year survival rate and the two-year survival rate after treatment in the Jin Fu Kang group were 67.3% and 67.3% respectively; 66.7% and 66.7% in the group of Jin Fu Kang plus chemotherapy; and 40.3% and 0.0% in the chemotherapy group. The improvement of clinical symptoms, increase of body weight and improvement of health situation (KPS marks) after treatment in both the Jin Fu Kang group and the group of Jin Fu Kang plus chemotherapy were better than that in the chemotherapy group. Some indicators of immunology and hemogram after treatment were greatly improved in the Jin Fu Kang group, worse in the chemotherapy group, but no obvious improvement in the group of Jin Fu Kang plus chemotherapy.

[17-ketosteroid sulfates and its application to kampo-medicine].

Traditional oriental medicine tells us that everything is interrelated and consists of combinations of opposing factors, "Yin-Yang". So we need to evaluate the normal state and pathologic condition from the perspective of dynamic hormone balance between cortisol and DHEA which are opposing and physiologically very important. Cortisol has a more functional side and is considered Yang, while DHEA has a more material side and is considered Yin. We tried to explain the concepts of KAMPO which include SHO (Yin-Yang. Excess-Deficiency), the 6 Stages of Disease and Deficiency of the Kidney, by measuring urinary 17-OHCS and 17-KS-S, metabolites of cortisol and DHEA. We think our effort may develop a good objective indicator of KAMPO's diagnosis and be useful for selecting of treatment.

Therapeutic approaches for chronic gastralgia based on differentiation of symptoms and signs.

OBJECTIVE: To report our clinical experience with the treatment of chronic gastralgia on the basis of the differentiation of symptoms and signs. METHOD: A total of 160 cases of chronic gastralgia we treated in recent years were reviewed. RESULTS: Among the 160 cases, 120 falls in the category characterized by the predominant clinical symptoms such as depression of liver-energy and spleen-asthenia. For the other 40 cases, their clinical symptoms are of deficiency of stomach-yin, and retention of damp-heat in the interior or accumulation of blood stasis constituted the major clinical presentations, but more or less featured by depression of liver-energy and spleen-asthenia. Through analysis by differentiation of the symptoms and signs, we were convinced that the major symptoms of chronic gastralgia and some accompanying minor symptoms were all the representation and extension of the underlying cause, disorder of stomach-energy. The treatment was therefore targeted at dispersing the depression of liver-energy and invigorating the spleen, smoothing and regulating the stomach-energy. The treatment adopts primary prescription and good effects were achieved. CONCLUSION: The pathogenesis of chronic gastralgia predominantly lies in the depression of liver-energy and spleen-asthenia, together with disordered stomach-energy. The treatment should be implemented to disperse the depression of liver-energy and invigorate the spleen, with attention to smoothing and regulating the stomach-energy throughout the whole treatment course.

[Comparison between two diagnostic methods of computer's mathematic model and clinical diagnosis on TCM syndromes of rheumatoid arthritis].

Based on the test result of 14 trace elements in hair of 163 cases of rheumatoid arthritis, we used one of the methods of the computer's mathematic model, DYNAMIC to assign the patients to five groups among multidimensional space. Then, another method, M-DEC was used to diagnose it back on one plane. The result was compared with that by the usual clinical diagnosis, also five groups were obtained, the group of Deficiency of both Liver and Kidney, syndrome of intermingled Cold with Heat, Deficiency of Qi and Yin, syndrome of Dampness-Heat stagnating in collaterals and syndrome of Phlegm-Dampness stagnating in collaterals. And Kappa was 0.77, greater than 0.6. It displayed the consistency in observation of these two methods, which was shown reliable. After that, we have detected the sensitivity of the syndrome diagnosis by computer's mathematic model method, the result was 96.67% and the specificity of that was 95.15%. So we consider this method could give us an objective judgment on TCM syndromes of rheumatoid arthritis.

Dietary potassium restriction stimulates endocytosis of ROMK channel in rat cortical collecting duct.

ROMK potassium channels are present in the cortical collecting ducts (CCDs) of the kidney and serve as the exit pathways for K+ secretion in this nephron segment. Dietary K+ restriction reduces the abundance of ROMK in the kidney. We have previously shown that ROMK undergoes endocytosis via clathrin-coated vesicles in Xenopus laevis oocytes and in cultured cells. Here, we examined the effect of dietary K+ restriction on endocytosis of ROMK in CCDs using double-labeling immunofluorescent staining and confocal microscopic imaging in whole kidney sections as well as in individually isolated tubules. We found that ROMK abundance in kidney cortex and CCDs was reduced in rats fed a K+-restricted diet compared with rats fed the control K+ diet. In the control animals, ROMK staining was preferentially localized to the apical membrane of CCDs. Compared with control tubules, ROMK staining in CCDs was markedly shifted toward intracellular locations in animals fed a K+-deficient diet for 48 h. Some of the intracellular distribution of ROMK colocalized with an early endosomal marker, early endosomal antigen-1 or with a late endosomal/lysosomal marker, lysosomal membrane glycoprotein-120. These results suggest that K+ restriction reduces the abundance of ROMK in CCDs by increasing endocytosis and degradation of the channel protein. This decrease in the abundance of ROMK is likely important for maintaining K+ homeostasis during K+ deficiency.

Diet-induced DNA damage and altered nucleotide metabolism in lymphocytes from methyl-donor-deficient rats.

Tumor induction with chronic feeding of methyl-donor-deficient diets has been well established; however, the biochemical and molecular mechanisms which predipose to tumorigenesis in this model are still not well understood. The purpose of the present investigation was to assess DNA damage and altered nucleotide metabolism in lymphocytes from Fischer 344 rats fed one of four semi-purified diets: (i) deficient in methionine and choline; (ii) deficient in folic acid; (iii) deficient in methionine, choline and folic acid; or (iv) a supplemented control diet. The accumulation of DNA-strand breaks, as assessed by DNA unwinding in alkali, was increased in lymphocytes from both the methionine/choline-deficient and folate-deficient groups, but was most severe in the group deficient in all three methyl donors. Lymphocyte DNA damage was consistently associated with alterations in folate-dependent thymidylate synthesis, and a decrease in intracellular levels of the DNA-repair-associated pyridine nucleotide, nicotinamide adenine dinucleotide. In the liver, a synergistic lipotropic interaction between folate deficiency and methionine/choline deficiency was observed, confirming the metabolic inter-relationship between these nutrients. Taken together, the results suggest that folate deficiency interacts with methionine/choline deficiency to potentiate symptoms of methyl-donor deficiency and that alterations in folate-dependent thymidylate synthesis are related to DNA damage in lymphocytes. These metabolic aberrations may contribute to immune dysfunction with chronic feeding of methyl-donor-deficient diets.

DNA strand break accumulation, thymidylate synthesis and NAD levels in lymphocytes from methyl donor-deficient rats.

Although tumor development with methyl donor-deficient diets has been well established, the biochemical and molecular mechanisms which predispose to tumor development are not yet understood. In the present study, DNA damage and nucleotide metabolism associated with DNA synthesis and repair were evaluated in splenic lymphocytes from rats fed a basal diet low in methionine and lacking in choline and folate or a supplemented control diet for a period of 3 wk. The accumulation of DNA strand breaks, as assessed by DNA unwinding in alkali, was found to be significantly elevated in lymphocytes from rats fed the methyl donor-deficient diet and was associated with an increase in mitogen-stimulated incorporation of [3H]-thymidine via the salvage pathway for thymidylate synthesis. In addition, a significant decrease in the DNA repair-associated pyridine nucleotide, NAD, was observed in lymphocytes from the deficient group and was associated with a decrease in total spleen cell numbers. These results suggest that alterations in nucleotide metabolism and DNA damage are induced when methyl donor pools are stressed by dietary deficiency.

Involvement of rapid nucleotide synthesis in recovery from phosphate starvation of Catharanthus roseus cells.

Growth of suspension-cultured Catharanthus roseus cells ceased during phosphate starvation, but the cells grew again upon addition of Pi even after long-term starvation. The metabolic fate of [(33)P]Pi was studied in 1-week-old stationary phase cells in ordinary culture and in 1- or 2-week-old Pi-starved cells. Immediately after administration, the most heavily labelled organic compounds are nucleotides, followed by sugar phosphates. Two weeks Pi starvation slowed down the speed of incorporation of (33)P into nucleotides. The RNA, protein, and free nucleotide content all decreased gradually during Pi starvation; however, these compounds, especially nucleotides, increased markedly in the 24 h after addition of Pi. These responses are found in all cells examined, although the total amounts of these compounds were lower in the long-term Pi-deficient cells. Of the nucleotides, a marked increase was observed in nucleoside triphosphates and UDP-glucose. The transcript level of phosphate transporter and the activities of acid phosphatase, 5'- and 3'-nucleotidase, and adenosine nucleosidase were all reduced by the addition of Pi. In contrast, the activities of adenine phosphoribosyltransferase, nicotinate phosphoribosyltransferase, and nicotinamidase, which are salvage enzymes of purine and pyridine nucleotides, were markedly increased in the Pi-fed cells. Little or no increase was observed in adenosine kinase. In the light of these results, the possible involvement of net nucleotide synthesis in the initial metabolic events of recovery from Pi deficiency are discussed.
