The ascidian Aplidium conicum collected along Sardinia coasts (Italy) contained two novel prenylated benzoquinones, designated thiaplidiaquinone A (1) and thiaplidiaquinone B (2). These compounds showed an unprecedented tetracyclic structure. We have studied the pro-apototic mechanisms of both prenylated benzoquinones in the Jurkat cell line that is derived from a human T lymphoma, and we show that both compounds induce a strong production of intracellular reactive oxygen species (ROS) in this cell line. Moreover, kinetic experiments, comparing the timing of ROS induction with the collapse of the mitochondria potential (DeltaPsi(m)), clearly showed that ROS preceded the disruption of the mitochondrial potential, and the later one paralleled the appearance of apoptotic cells. Thus, thiaplidiaquinones A and B can enter into the cells and induce cell death by apoptosis.

Biogenesis of the enantiomeric sesquiterpenes 1 and 5 of the marine nudibranch Doriopsilla areolata was investigated by feeding of [1-(13)C]glucose, [1,2-(13)C(2)]glucose, and [1,2-(13)C(2)]acetate. Evidence is presented that supports de novo origin of both compounds via mevalonic acid.
Fused tetracyclic oxetanes 4, highly substituted cyclobutenes 6, and the pentacyclic derivatives 7 and 8 were obtained by irradiation of derivatives 3 that were prepared from commercial R-(+)-sclareolide (1) in three steps. Compounds 4 are formed through a Paterno-Büchi reaction, while tricyclic derivatives 6 are the fragmentation products of the first formed oxetanes. In clear contrast, cyclopentenyl and 3-furyl derivatives, 3e and 3f, gave the [2+2] adducts, namely pentacyclic derivatives 7 and 8. All the reported reactions are totally regio- and stereoselective, with the exception of the cyclization of furyl derivative 3f, which gave the mixture of both the crossed (7b) and the right (8) isomers.
Lipophilic extracts of the stembark of Buddleja globosa were found to have antifungal activity at 125 microg/mL against three dermatophytic fungal species but had no activity at 1000 microg/mL against four other fungal species or two yeast species. Bioassay-guided fractionation of Si gel column eluates using the sensitive fungal species resulted in active fractions from which were isolated five compounds that were characterized by spectroscopic methods as one novel and four known compounds. The known compounds were the diterpene buddlejone (1), the bisditerpene maytenone, and the two sesquiterpenes buddledin A and buddledin B, while the novel compound was characterized as the diterpene deoxybuddlejone (2). The minimum inhibitory concentration of all the compounds was determined against all the microorganisms under test, and buddledins A and B were shown to exhibit the greatest antifungal activity, with values of 43 microM and 51 microM, respectively, against the sensitive fungi Trichophyton rubrum, Tricophyton interdigitale, and Epidermophyton floccosum.
Two terpenoid biosynthetic pathways, the mevalonate and non-mevalonate (glyceraldehyde phosphate-pyruvate) routes, were examined by feeding (13)C-labeled serines ([1-(13)C]- and [3-(13)C]-) to the cultured cells of the liverwort, Heteroscyphus planus. The labeling patterns observed in the isoprenoid unit of the biosynthetically (13)C-labeled stigmasterol corresponded to those expected from the mevalonate pathway, while those of the phytyl side chain corresponded to those from the non-mevalonate pathway. Thus, serine is a potential probe to determine the origin of terpenoid biosynthesis, in either the mevalonate or non-mevalonate pathway.
Seven new nitrogenous terpenoids, (1R,6R,7S,10S)-10-isothiocyanatocadin-4-ene (1), (1S,2S,5S,6S,7R,8S)-13-isothiocyanatocubebane (2), (1R,3S,4R,7S,8S,12S,13S)-7-isocyanoamphilecta-10,14-diene (3), (1S,3S,4R,7S,8S,12S,13S)-8-isocyanoamphilecta-11(20),14-diene (4), (3S,4R,7S,8S,11S,13S)-8-isocyanoamphilecta-1(12),14-diene (5), 8-isocyanatocycloamphilect-10-ene (6), and 8-isothiocyanatocycloamphilect-10-ene (7), were isolated from the Okinawan sponge Stylissasp., along with 12 known related compounds. Structural determinations of these compounds were made by spectroscopic analysis, and assessment was made of their cytotoxicity toward HeLa cells.
