Cerebral vasodilation and spasmolytic activity of diltiazem in anesthetized animals.

The effects of diltiazem on the cerebral blood flow and cerebrovascular spasm were studied in pentobarbital anesthetized animals. In Rhesus monkeys, the common carotid and internal carotid blood flow were measured by an electromagnetic flowmeter. Diltiazem (10-300 micrograms/kg, i.v.) dose-dependently increased both the common carotid and internal carotid blood flow, and the increase in internal carotid blood flow persisted for a longer period than that in the common carotid blood flow. In dogs, regional blood flow in the cerebral cortex (rCBF) was measured by means of the hydrogen gas clearance method. Diltiazem (20 micrograms/kg/min, i.v.) increased rCBF by about 20% of the control during the infusion, and the increase in rCBF was still continued 40 min after the infusion was stopped. In cats, the basilar artery was exposed by craniotomy through the transcervico-transclival approach, and vasospasm was induced by topical administration of 5-HT, PGF2 alpha and incubated blood. Diltiazem, either applied topically to the artery (100 micrograms/ml) or infused continuously into the femoral vein (20 or 40 micrograms/kg/min), suppressed the vasoconstriction evoked by the spasmogen.

Rociverine citrate: a new spasmolytic agent, potentially useful in the treatment of urinary bladder hyperreflexia.

Rociverine citrate was evaluated for its ability to affect the motility of rat urinary bladder, in vitro and in vivo, in comparison with flavoxate hydrochloride. Rociverine counteracted both methacholine- and high K+-induced tonic contractions of bladder strips. In anaesthetized rats, intravenous rociverine inhibited dose-dependently frequency and amplitude of the distension-induced rhythmic contractions (DIRCs) of urinary bladder and counteracted the topical high K+-induced pressure increase in the same organ. Orally administered rociverine produced a dose-related reversal of the reserpine-induced detrusor hyperreflexia in anaesthetized rats. In each of these experimental models rociverine was more effective than flavoxate. These results point to the usefulness of rociverine in the treatment of urinary bladder motility disorders.

[Influence of spasmolytic agents on the cumulative calcium dose-effect curve in isolation-perfused peripheral vascular beds].

The influence of verapamil, theophylline, and papaverine on the cumulative calcium dose-effect curve was studied at the isolated perfused rat tail. The results indicate different changes of the contraction and relexation phase respectively. The findings are discussed in connection with the action mechanism of the drugs and the processes of excitation-contraction coupling.

Trefoil factor-2, human spasmolytic polypeptide, promotes branching morphogenesis in MCF-7 cells.

Members of the trefoil factor (TFF) family are highly expressed in endodermal ulcerative wound healing and selectively in neoplastic proliferation of various glandular epithelia. There is some evidence that TFF1 and TFF3 affect cell motility, are indirectly involved in growth suppression, and are associated with mucin expression. TFF2 is co-expressed with TFF1 in gastric surface epithelial cells, but its potential role in vivo is unclear. We analyzed potential effects on cell proliferation and morphogenesis of TFF2 on a panel of epithelial and mesenchymal cell lines. TFF2 had no measurable effect on the proliferation of any of the cell lines tested. In type 1 collagen lattices, TFF2 at a low concentration (25-100 nM) induced the formation of highly complex branched structures in the breast carcinoma cell line MCF-7 over a period of 14 to 42 days. No significant effect was shown with other cell lines. This morphogenic effect was abolished by monoclonal antibodies specific for either TFF2 or TFF1. TFF2 did not affect cell motility in MCF-7 cells as measured by videomicroscopy, in contrast to previous studies using TFF1. TFF2-treated MCF-7 colonies showed a 30% reduction in the number of apoptotic bodies, corroborated by trypan blue exclusion and DNA fragmentation ELISA, indicating TFF2 promotes cell survival via inhibition of apoptosis and can act as a morphogen in the presence of TFF1. These properties may complement the actions of TFF1 as a motogen and may explain differential expression in endodermal wound healing.

[Spasmolytic effect of histamine in the isolated intestine of the hamster (Cricetus auratus)].

A comparative study of the effects produced by histamine on 15 different sections of the small intestine of hamsters (Cricetus auratus) from the stomach to the ileocecal valve was performed to find a substitute for that of guinea pigs and to ascertain which of the sections was the most sensitive. Changes in tone, amplitude and frequency of contractions with respect to control (spontaneous motility, were significant (P less than 0.01); the variance analysis showed no significant variations in the sensitivity or the motility of the different sections.
