Resveratrol as a novel agent for treatment of multiple myeloma with matrix metalloproteinase inhibitory activity.

AIM: To examine the in vitro antitumor activity of resveratrol against multiple myeloma (MM) cell lines (RPMI 8226, U266, and KM3), and the mechanisms involved. METHODS: The growth inhibition of resveratrol was determined by 3-(4, 5-dimethyl-2-thiazyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. The effect of resveratrol on the apoptosis was investigated by combined annexin V-propidium iodide staining. The effect of resveratrol on the invasion through Matrigel matrix was detected by transwell invasion analyses. The activity of matrix metalloproteinase (MMP)-2 and -9 proteins were determined by gelatin zymography analysis. The expression of MMP-2, MMP-9, Bcl-2, Bcl-x(L), XIAP and Bax protein were detected using Western blotting analysis. RESULTS: Resveratrol inhibited proliferation of MM cells in a dose- and time-dependent manner. Incubation of MM cells with resveratrol resulted in apoptotic cell death. Resveratrol down-regulated the expression of the antiapoptotic proteins Bcl-2, Bcl-x(L) and XIAP and up-regulated the expression of the proapoptotic protein Bax. Furthermore, resveratrol inhibited invasion of RPMI 8226, U266, and KM3 cells with IC50 values of 64+/-8 micromol/L, 93+/-11 micromol/L, and 153+/-11 micromol/L, respectively. Resveratrol inhibited the constitutive expression of MMP-2 and -9 proteins of MM cells and suppressed its gelatinolytic activity. CONCLUSION: Resveratrol inhibits the proliferation of MM cells by inducing apoptotic cell death. Resveratrol also inhibits MM cell invasion. The inhibition of invasion may be associated with the attenuation of the enzymatic activities of MMP-2 and -9.

Transient population dynamics in periodic matrix models: methodology and effects of cyclic permutations.

Many biological populations are subject to periodically changing environments such as years with or without fire, or rotation of crop types. The dynamics and management options for such populations are frequently investigated using periodic matrix models. However the analysis is usually limited to long-term results (asymptotic population growth rate and its sensitivity to perturbations of vital rates). In non-periodic matrix models it has been shown that long-term results may be misleading as populations are rarely in their stable structure. We therefore develop methods to analyze transient dynamics of periodic matrix models. In particular, we show how to calculate the effects of perturbations on population size within and at the end of environmental cycles. Using a model of a weed population subject to a crop rotation, we show that different cyclic permutations produce different patterns of sensitivity of population size and different population sizes. By examining how the starting environment interacts with the initial conditions, we explain how different patterns arise. Such understanding is critical to developing effective management and monitoring strategies for populations subject to periodically recurring environments.

Bond durability of resin cements to Au-Pd-Ag alloy under cyclic impact load.

The bond durability of resin cements to a 12% Au-Pd-Ag alloy was studied through cyclic impact tests with different loads. A piece of casting alloy was bonded to a cast block with two types of resin cements, Super Bond C&B and Bistite II. A shear load was applied onto a small piece of alloy until debonding of the specimen, using different weights of plungers, 130 g, 230 g, 330 g and 430 g. The specimen bonded with Super Bond exhibited a higher resistance than that with Bistite II. The fracture modes of the debonded cements were completely different from each other. That is, Bistite II showed a bulk fracture of cement by the crack penetrating through the cement layer. On the other hand, Super Bond showed damages limited to the surface and no bulk fracture. The mode of fracture was dependent not on the loading weight but the types of resin cements used.

[Determination of stilbene in radix Polygoni multiflori by HPLC and its stability study].

OBJECTIVE: To develop a HPLC method for determining stilbene in Radix Polygoni Multiflori and studying its stability. METHOD: HPLC conditions: An Econosphere C18 column, a mixture of methanol-water(30:70) as mobile phase, and UV detection at 310 nm. In addition, the stability of stilbene was studied at high temperature and in acidic solution. RESULT: The linear range of calibration curve was 2.24 x 10(-2)-1.12 x 10(-1) micrograms(n = 6) with a correlation coefficient of 0.99998, the average recovery with RSD being 99.96% +/- 1.54%. CONCLUSION: The method is sensitive, accurate and speedy. Stilbene in aqueous solution is stable, but high temperature(80 degrees C) tends to affect the stability. In acidic solution no stability can be found.

[Antiviral action of combined use of rhizoma Polygoni cuspidati and radix Astragali on HSV-1 strain].

OBJECTIVE: The clinical action of combined use of Rhizoma Polygoni Cuspidati and Radix Astragali on HSV-1 was investigated with a view to developing a new antiviral drug. METHOD: The action was analyzed by way of plaque reduction assay and median-effect principle. RESULTS: In the HEp-2 cell system, if the combination ratio of Rhizoma Polygoni Cuspidati and Radix Astragali was 1(ED50):1(ED50) then, (1) In directly annihilating HSV-1 F strain, when the plaque reduction rate was 20%-80%, and the combination index was < 1.0, there was synergism. (2) In inhibiting the multiplication of HSV-1 F strain, when the plaque reduction rate was 20%-60%, and the combination index was < 1.0, there was also synergism. (3) In blockading HSV-1 F strain infection, when the plaque reduction rate was 20%-90% and the combination index was < 1.0, there was synergism. So this ratio of 1(ED50):1(ED50) should be the first choice for combination. CONCLUSION: The treatment index of the above two Chinese medicinal herbs equals 10(3), and the cytotoxicity is not potentiated, indicating that the combination is helpful as a virucide for HSV-1 F strain.
