Comparison of a Modified Atmosphere Stunning-Killing System to conventional electrical stunning and killing on selected broiler breast muscle rigor development and meat quality attributes.

Two experiments were conducted to compare the effects of a 70:30 argon:carbon dioxide Modified Atmosphere Stun-Kill (MASK) system to both a low voltage (LV) and a high current (HC) electrical stunning and conventional killing system on broiler breast rigor development and meat quality. In Experiment 1, the effects on breast muscle pH and meat tenderness of the MASK system and the LV system were compared. In Experiment 2, the MASK system was compared to both HC and LV stunning for pH, R-value, C.I.E. L*, a*, b*, cooked yield, and tenderness of broiler breast meat. In Experiment 1, there were no significant differences in breast muscle pH between LV and MASK, except at 24 h. Only at 5 h post-mortem did the MASK system result in significantly tougher breast meat. In Experiment 2, there were no significant differences between treatments in breast muscle color or cooked yield at any sampling time. The breast muscle from birds subjected to the HC stun had consistently higher pH values until 5 h post-mortem, at which time the breast meat from birds subjected to the MASK system had higher pH. R-value data showed a similar trend, with the HC stunned birds having the lower R-values until 5 h post-mortem. Breast meat tenderness values for the three treatments were significantly different only at 3 and 5 h post-mortem, when birds stunned with HC had the highest shear values. The MASK system did not exhibit any rigor accelerating benefits when compared to LV stunning and conventional killing, and only minimal rigor acceleration when compared to HC killing.

Metabolic disturbances in male broilers of different strains. 2. Relationship between the thyroid and somatotropic axes with growth rate and mortality.

Seven male broiler strains (Arbor Acres, Avian Farms, Cobb-500, Hubbard-Peterson, ISA, Naked Neck, and Ross) were compared for their growth rate, feed efficiency, and mortality due to sudden death and ascites. In addition, weekly plasma levels of thyroid hormones [3,3',5-triiodothyronine (T3) thyroxine (T4), T3: T4 ratio, growth hormone (GH), and insulin-like growth factor-I (IGF-I)] were determined. The highly productive, commercial strains were very similar in their endocrine profiles but differed markedly from the Naked Neck chickens. Naked Neck chickens were characterized by higher plasma T3 and lower T4 levels at similar ages as well as when compared on the same body weight basis. The present findings support the hypothesis that the slightly hypothyroid state of high productive broilers renders them more sensitive to metabolic disorders. Naked Neck chickens also had higher plasma GH levels than those of their age-matched commercial broilers. The coefficient of variation for GH was highest for Naked Neck chickens, which is indicative for an amplified GH burst amplitude. It may be stated that changes in plasma thyroid hormone concentration in indirect response to selection for low feed conversion and fast growth may be causatively linked to susceptibility for metabolic disturbances such as sudden death syndrome and ascites.

Research and development in 2000: directions and priorities for the world's poultry science community.

The challenges and targets facing the world's poultry science community in the immediate future are reviewed in the context of meeting the dietary needs for animal protein of the world population. The prior need to provide for the increasing demand for cereals, oil seeds, and grain legumes for human consumption is assessed at having a reasonable chance of success. If this need is met, the requirement for extra feed resources for increased poultry production targets is also assessed as having a reasonable chance of success. A major component of this equation is the prediction of improved efficiency of poultry production of a similar order to that of the last 50 yr arising from 1) extension of the 20th century revolution in poultry technology to over 50% of the world population compared with the present 20 to 25%; 2) recent advances in genetics, nutrition, health, housing, and husbandry still awaiting application in industry; 3) future applications from current and future research in molecular biotechnology, nutrition, health, and reproduction; and 4) the development of efficient, small-scale, extensive poultry production systems especially in countries where over 25% of the world population will still not be able to afford the products of a modern, intensive poultry industry, even in 50 yr. These challenges, targets, and predictions simply cannot be met unless the world's poultry science community increases its own efficiency, its professional initiatives to deal with the real challenges, and its social initiatives to influence socio-economic decisions on national and world stages.

Roaster breast meat condemned for cyanosis: a dark firm dry-like condition?

A case-control study (n = 68) of roaster chickens condemned for cyanosis was conducted. Color (CIE L*a*b*) and pH were measured at slaughter, and after 24 h aging on ice, at four predetermined sites of the Pectoralis major. Cyanotic carcasses (dark) had a higher pH than controls at the time of slaughter and at 24 h postmortem (P < 0.01). Perimortem pH was significantly correlated with pH at 24 h postmortem (r = 0.64) and also was correlated with lightness (L*) perimortem and postmortem (24 h; r = -0.36 and -0.50, respectively). Perimortem pH was not correlated with meat redness (a*) at slaughter time and after 24 h. Ultimate pH and lightness at 24 h postmortem were also correlated. Tests based on pH, L*, and a* of the P. major were assessed: the sensitivity and specificity at various cut-off points were, respectively, pH(6.3) = 76.47 and 88.24%, L*(41) = 91.18 and 79.41%, and a*(3) = 76.47 and 97.06%. The repeatability (p) of pH and color measurements was excellent and ranged from 0.87 to 0.98. Breast meat from roasters condemned for cyanosis had dark, firm, and dry (DFD)-like traits, and accurate tests based on color and pH could be described as a means of identifying chickens condemned for cyanosis.

Avian T helper one/two immune response balance can be shifted toward inflammation by antigen delivery to scavenger receptors.

Whether immune responses are dominated by inflammation or antibody production is often key to surviving infections. Therefore, differential control of these immune pathways determined by CD4 T cells is of fundamental interest for vaccine design. Little is known about how inflammatory [T helper cell (Th) type 1 (Th1)] versus antibody-inducing (Th2) choices are controlled in domestic fowl. To address this, MHC-matched chickens were immunized to test whether antibody-dominated Th2 or inflammatory Th1 responses could be preferentially activated, and our findings subsequently extended to outbred broiler breeders. Strategies used were known to shift the response in mice from Th2 to Th1 by delivering the injected antigen preferentially to macrophages. The model antigen, BSA, was maleylated to allow binding to scavenger receptors (SR) present on mammalian macrophages. Maleyl-BSA bound well in receptor-specific fashion to a chicken macrophage cell line. Compared with native BSA, immunization with SR-binding, maleyl-BSA modulated the immune response toward the Th1 pathway, as evident by increases in the magnitude of in vivo inflammatory reactions and declines in antibody-making responses. Initiation of a maleyl-BSA Th1 pathway is further supported by the enhanced ability of splenocytes to express mRNA for interferon-gamma in response to antigens. Together, these data establish the presence and functional relevance of SR in domestic fowl as well as provide a system for investigating the mechanisms controlling Th1/Th2 pathways in chickens. Moreover, the ability to direct immune responses toward either pathway by antigen maleylation will contribute significantly to the development of better vaccines for poultry diseases.

Evaluation of the efficacy of a feed additive to ameliorate the toxic effects of 4,15-diacetoxiscirpenol in growing chicks.

The possible protective effect of a feed additive (Mycofix) against the toxic effects of 4,15-diacetoxiscirpenol (DAS) in growing broiler chickens was investigated in a 21-d fully randomized trial consisting of seven dietary treatments (control with no DAS or Mycofix added, 1 ppm DAS alone, 1 ppm DAS supplemented with 0.75 g/kg Mycofix, 1 ppm DAS supplemented with 1.5 g/kg Mycofix, 2 ppm DAS alone, 2 ppm DAS supplemented with 0.75 g/kg Mycofix, and 2 ppm DAS supplemented with 1.5 g/kg Mycofix). When no feed additive was included, both levels of dietary DAS significantly decreased BW and feed intake and caused oral lesions, with the effect of 2 ppm DAS being more severe. When 1 ppm DAS was added to the diet, supplementation of Mycofix protected against the adverse effects of DAS on feed intake and BW at both levels of inclusion (0.75 and 1.5 g/kg); however, no protection against oral lesions was obtained by Mycofix supplementation. This finding suggests that the adverse effect of DAS on performance is not due to the oral lesions per se but it is likely the result of the systemic absorption of the mycotoxin. When 2 ppm dietary DAS was present in the diet, only partial protection on BW and feed intake was obtained by Mycofix supplementation. More studies are required to determine if a higher dose of Mycofix could be capable of counteracting the adverse effects of 2 ppm dietary DAS on chicken performance.

Antibiotic residues in poultry tissues and eggs: human health concerns?

Antibiotics are used by the poultry industry to enhance the health and productivity of flocks. The use of antimicrobials is strictly regulated by the Food and Drug Administration (FDA) and the USDA to warrant their safety and efficacy. Prior to regulatory approval, the pharmacokinetics and tissue tolerances of an antimicrobial are determined to set the proper dosage. To ensure proper use, both the FDA and USDA have research, surveillance, and compliance programs to develop detection methods and monitor poultry tissues for antimicrobials. Unfortunately, there is the perception among many consumers that our food supply contains high concentrations of drug or hormone residues causing significant health concerns or problems. In fact, foods produced in this country (including poultry) are very safe and meet the highest standards to exclude chemical contaminants. An overview will be presented on the federal oversight and monitoring of antimicrobial residues in poultry tissues.

Effect of selection for growth rate on muscle damage during turkey breast muscle development.

Damage to the turkey pectoralis major muscle was studied in a randombred control line (RBC2), a subline (F) from the RBC2 line selected long-term for increased 16-wk BW only, and a commercial sire line (B) at 25 d of incubation and at 1, 4, 8, 16, and 20 wk posthatch. Pectoralis major muscle samples were obtained from three females and three males from each line in a manner to avoid contraction. After being fixed and sectioned, the muscle samples were stained with hematoxylin and eosin to view muscle fiber and muscle fiber bundle morphology. Beginning at 8 wk posthatch, differences in muscle fiber morphology were observed among the different lines. The RBC2 line throughout the duration of the study maintained well-organized muscle fibers and muscle fiber bundles with large capillary networks. In contrast, the growth-selected F line began to show muscle fiber degeneration at 8 wk posthatch, and limited capillary beds were observed as development proceeded. The B line had intermediate muscle morphology between the RBC2 and F lines, but by 20 wk posthatch significant muscle fiber degeneration was present with limited capillary supply. The degenerative muscle fiber changes were predominant in the growth-selected F-line, suggesting that growth selection for just BW may be associated with posthatch muscle damage.

Selection strategies for body weight and reduced ascites susceptibility in broilers.

Ascites syndrome is a metabolic disorder in broilers. Mortality due to ascites results in significant economic losses and has a negative impact on animal welfare. It has been shown that genetic factors play a considerable role in susceptibility of birds to ascites, which offers perspectives for selection against this syndrome. The aim of the present study was to evaluate the consequences of alternative selection strategies for BW and resistance to ascites syndrome using deterministic simulation. In addition to the consequences of current selection (i.e., selection for increased BW only) alternative selection strategies including information on different ascites-related traits measured under normal or cold conditions and the consequences of having information on the underlying genes (i.e., MAS) were quantified. Five different breeding schemes were compared based on the selection response for BW, ascites susceptibility, and the rate of inbreeding. Traits investigated in the index as indicators for ascites were hematocrit value (HCT) and ratio of right ventricle to the total ventricular weight of the heart (RV:TV). The results indicated that by ignoring ascites susceptibility in the breeding goal, the gain for BW is 130 g and the birds will become more susceptible to ascites. Testing 50% of the birds under cold temperature conditions and including information of ascites related traits (HCT and RV:TV) measured under normal and cold conditions makes it possible to achieve a relatively high gain for BW (111.4 g) while controlling the genetic level for ascites susceptibility (selection response was 0). The results of scenarios including QTL information of ascites susceptibility showed that QTL information could be used very effectively in controlling ascites susceptibility.

Comparison of three lines of broiler breeders differing in ascites susceptibility or growth rate. 1. Relationship between acoustic resonance data and embryonic or hatching parameters.

Ascites is a prevalent cardiovascular disease among modern broilers with negative impacts on production and animal welfare. The peak of mortality due to ascites occurs at the end of the growing period, but the etiology of this problem may start during embryonic development. A few recent reports have demonstrated that the signs of ascites susceptibility are manifested during the late stages of incubation. In the current study, we used a nondestructive method based on egg acoustic resonance parameters [resonant frequency (RF) and damping] to establish a relationship between embryo physiological events during early development in broiler eggs and susceptibility to ascites. The hatching eggs of 3 broiler lines differing in ascites susceptibility were used for this study: ascites-resistant dam line (DAR), ascites-sensitive dam line (DAS), and ascites-sensitive sire line (SASL). These lines were selected on the basis of fast growth, high breast meat yield, and ascites induction at low temperatures such that the order of ascites susceptibility in terms of mortality was SASL >> DAS > DAR. Eggs were incubated under standard conditions in forced-draft incubators. We measured egg weights at setting, albumen pH, Haugh units (HU) at setting, and embryo weights at d 11 and 18, at internal pipping (IP), and at hatch. The durations of IP, external pipping (EP), and hatching were also determined. At 2 hourly periods during incubation, egg RF and damping were also measured. There were differences in egg weights between DAR and SASL vs. DAS, but albumen HU, albumen pH, and the ratio of yolk weight to egg weight were similar. There were differences in RF, damping, embryonic growth rates, and hatching events. Changes in resonant frequency and damping, which certainly suggest eggshell differences among lines, were not totally related to variations in physiological events during early and late embryonic development. A comparison between DAR and DAS, between DAS and SASL, or DAR and SASL indicates that sensitivity to ascites and selection for rapid growth rate in ascites-sensitive lines have different effects on embryonic parameters. We concluded that the sensitivity of broiler breeders to ascites does not influence egg internal quality, but the occurrence of ascites sensitivity in broilers could not be reliably predicted by early in ovo acoustic resonance parameters and hatching events.

The occurrence of deep pectoral myopathy in roaster chickens.

A study was conducted to determine the incidence of deep pectoral myopathy (DPM) in male roaster chickens reared under intensive conditions, processed at different ages (from 47 to 65 d of age), and belonging to 2 commercial genotypes (Ross 508 and Cobb 500). The study was carried out in a major Italian processing plant on a total of 120,700 male roaster chickens chosen at random from 151 flocks during a 6-mo period. The evaluation of DPM was performed on pectoralis minor muscles and consisted of a visual assessment of the presence or absence of the myopathy as well as the scoring of muscle damage level as being in an "early" (hemorrhagic appearance) or "old" (gray or green discoloration) developing stage. The average incidence of carcasses affected by DPM was found to be 0.84% (0.62 and 0.22% in early and old stages, respectively). The range in the incidence of total DPM was fairly large and varied from 0 to 16.7%. Considerable variations were also observed for early (range: 0 to 12.0%) and old (range: 0 to 5.6%) developing stages. Considering the effect of genotype, Ross 508 exhibited a higher incidence of DPM in respect to Cobb 500 (1.27 vs. 0.35%; P < or = 0.01). This result was due to the higher incidence of carcasses affected by both early (0.94 vs. 0.26%; P < or = 0.01) and old (0.33 vs. 0.09%; P < or = 0.01) developing stages of DPM in Ross 508 birds.

Pathological features in dead on arrival broilers with special reference to heart disorders.

A gross postmortem investigation was done on 302 broilers that died between catching and slaughter to establish predisposing factors for dying in this period. Special attention was paid to heart disorders, which were established by determining the ratio of the right ventricle mass to the total ventricle mass (RV:TV) and to postmortem changes in hearts and lungs of broilers that were dead on arrival (DOA). Macroscopic pathologic lesions were found in 89.4% of DOA broilers. Signs of infectious diseases appeared to be most frequent (64.9%), followed by heart and circulation disorders (42.4%), and trauma (29.5%). The RV:TV was significantly higher for DOA broilers in comparison with slaughtered broilers. The prevalence of hearts with an abnormal RV:TV in DOA broilers was 34.4 vs. 4.1% in slaughtered broilers. The DOA broilers with an abnormal heart ratio more frequently showed ascites and hydropericardium. Postmortem changes in lungs depend on the position of the carcass the first several hours after death. Broilers, which remain in dorsal recumbency for several hours after death, develop engorged lungs. A good health status as well as more attention for the catching and crating process is crucial in decreasing the percentage of DOA broilers. Prevention of an increased heart ratio and of ascites will improve the livability in the broiler house and also decrease the DOA rate enormously.

Maternal antibody transfer from dams to their egg yolks, egg whites, and chicks in meat lines of chickens.

Maternal antibodies are transferred from hens to the chicks via the egg. To gain insight into maternal antibody transfer and endogenous production of antibodies in broiler chicks, total IgY, IgA, IgM, as well as anti-Newcastle disease virus (NDV) and anti-infectious bronchitis (IBV) antibody levels were examined in the dams' plasma, egg yolks, egg whites, and chicks' plasma on d 3, 7, 14, and 21. Blood was collected from 39-wk-old breeder hens (line 1, n = 17; line 2, n = 21). Fertile eggs were used for antibody extraction from the egg yolks and egg whites (4 to 5 eggs/dam) and for hatching. Unvaccinated chicks (4 to 5 chicks/dam) were reared in a HEPA-filtered room and were bled on d 3, 7, 14 and 21. Based on ELISA methods, plasma levels of IgY and IgM were higher (P < 0.0001), and those of IgA were similar (P = 0.31), in line 2 compared with line 1. Egg yolk IgY and IgA, as well as egg white IgY, IgA, and IgM levels were higher in line 2 compared with line 1 (P < 0.0001). Independent of line of chicken, the percentage dam-to-chick (3 d) plasma transfer of IgY was estimated to be approximately 30%, with that for IgM and IgA less than 1%. Chicks synthesized IgM first, followed by IgA and IgY. Anti-NDV and anti-IBV antibodies were detected in the dams' plasma, egg yolks, and in the chicks' plasma on d 3 and 7, with line 2 having higher anti-IBV and lower anti-NDV levels than line 1 in all samples (P < 0.0001). In summary, IgY levels, total or antigen-specific, in the dams' plasma or eggs were found to be a direct indicator of maternal antibody transfer to the chicks' circulation, with an expected percentage transfer of approximately 30%. This knowledge, together with the observed time course of endogenous antibody production in broiler chicks, may find direct application in formulating strategies for protecting chicks, especially during the first few weeks of age when their immune system is not yet fully functional.

Peripheral neuropathy of dietary riboflavin deficiency in chickens.

A strain of rapidly growing meat-type chickens was fed a diet deficient in riboflavin from 1-40 days of age. Diminished growth rate, progressive gait abnormality and reluctance to move were noted beginning on day 8. Neurologic abnormalities were related to peripheral neuropathy characterized by Schwann cell hypertrophy and degeneration with cytoplasmic lipid droplets' and segmental demyelination. Lesions were initially detected on day 10, and in concert with clinical signs became more profound between days 14 and 21. Sequestration of myelin debris within Schwann cells was common. Other features of the neuropathy included the presence of endoneurial edema and axonal degeneration involving small numbers of fibers. Remyelination of peripheral nerve fibers in birds on the deficient diet was occasionally seen on day 10, became progressively more prominent, and was marked by day 37. There was an associated, variable but incomplete, clinical improvement evident in later stages of the study. Liver concentrations of riboflavin in deficient birds were significantly reduced on day 13 but not on day 26. This neuropathy may be related to diminished tissue levels of the riboflavin-based coenzymes flavin-adenine dinucleotide (FAD) and flavin mononucleotide (FMN) leading to reduced cellular energy levels and profoundly affecting Schwann cells at some critical point in growth.

Drug plasma levels following administration of trimethoprim and sulphonamide combinations to broilers.

Trimethoprim (TMP) was administered in combination with either sulphadiazine or sulphadimidine to broilers, and plasma concentrations were determined simultaneously by newly developed thin-layer and/or high-performance liquid-chromatographic procedures, which also allowed quantification of the N4-acetyl metabolites of the sulphonamides. After i.v. injection of TMP (20 mg/kg body wt) and sulphadiazine (100 mg/kg body wt), both compounds were rapidly eliminated from plasma with half-lives of 1 and 2.7 h, respectively. Apparent volumes of distribution (3.3 and 0.96 l/kg, respectively) indicated that the tissue distribution of TMP was more extensive than that of the sulphonamide. After oral administration of the same dosages, elimination appeared to be slower compared to the i.v. injection, but this was obviously related to delayed absorption. Bioavailability after oral administration was approximately 100% of sulphadiazine, but only about 60% for TMP. Oral dosing of TMP in combination with sulphadimidine yielded similar maximum plasma concentrations of both compounds to those obtained with the combination of TMP with sulphadiazine, but the plasma concentration decline of sulphadimidine appeared to be more rapid than that of sulphadiazine after oral administration. During prolonged administration of different dosages of TMP-sulphadiazine combinations via drinking water, only low plasma concentrations were attained by the recommended dosage of the combination. Up to 10-fold higher dosages were tolerated by the animals without side-effects. In view of the fact that the sensitivity of bacterial strains to TMP-sulphonamide combinations differs widely, the plasma concentrations determined in the present study during prolonged drinking-water medication with different dosages of a TMP-sulphadiazine combination can be used to select effective doses for treatment of different poultry diseases.

Seroprevalence of Ornithobacterium rhinotracheale infection in broiler and broiler breeder chickens in West Azerbaijan Province, Iran.

Ornithobacterium rhinotracheale is a pleomorphic Gram-negative rod shaped bacterium of the rRNA superfamily V that is associated with respiratory disease in poultry. This study was conducted to determine the seroprevalence of O. rhinotracheale infection in broiler and broiler breeder chickens in West Azerbaijan (Urmia lake region) by using a commercial enzyme-linked immunosorbent assay. In this study, 463 serum samples were obtained from 50 broiler flocks and 472 blood sera from 42 broiler breeder flocks. Results showed that 41 broiler flocks (82%) and 39 broiler breeder flocks (92.8%) were positive. Ornithobacterium rhinotracheale antibodies were detected in 205 (44.2%) of the 463 broiler serum samples. Of the 472 blood sera examined from broiler breeder, 340 (72%) were positive. The results of this study indicated that the prevalence of O. rhinotracheale antibodies is high in the broiler and broiler breeder flocks in West Azerbaijan.
