Allylated monosaccharides as precursors in triple reductive amination strategies: synthesis of castanospermine and swainsonine.

The feasibility of the triple-reductive amination reaction for the synthesis of complex indolizidine frameworks is illustrated by application to the potent glycosidase inhibitors castanospermine and swainsonine. The target compounds were obtained from known carbohydrate precursors in yields of 23 and 14%, over nine and 13 steps, respectively. The iodoetherification reaction of allylated monosaccharides was shown to be a practical reaction for the synthesis of the tricarbonyl precursors for the key triple reductive amination reactions.

Carbohydrate-based receptors with multiple thiourea binding sites. Multipoint hydrogen bond recognition of dicarboxylates and monosaccharides.

The binding properties of multitopic sugar thiourea receptors toward dicarboxylate and monosaccharide guests have been examined taking glutarate and octyl beta-D-glucopyranoside as model ligands. For the anionic hydrogen bond acceptor, both the complex stoichiometry and the association constants (K(as)) were found to be strongly dependent on the relative disposition of recognition elements in the host. In contrast, for the glucoside guest a 1:1 stoichiometry was observed in all cases, the K(as) values being largely independent of the unbound state provided that geometrically equivalent supramolecular topologies can be achieved.

Cyclic hexapeptides with free carboxylate groups as new receptors for monosaccharides.

[structure: see text]. Cyclic hexapeptides composed of alternating L-proline and 3-aminobenzoic acid subunits with substituents on the aromatic subunits that contain free carboxylate groups are able to bind monosaccharides in 4% CD3OD/CDCl3. The binding selectivity of these peptides depends on the structure of the substituents on the aromatic subunits.

Sulfo-protected hexosamine monosaccharides: potentially versatile building blocks for glycosaminoglycan synthesis.

2,2,2-Trifluorodiazoethane was investigated as a reagent for sulfo group protection on hexosamine monosaccharides. The synthesis of glucosamine and galactosamine building blocks fully differentiated for glycosaminoglycan synthesis and the synthesis of glycosyl donors are described. The compatibility of trifluoroethylsulfonate under a variety of reaction conditions has also been investigated. [structure: see text]

Monosaccharide compositions of Danaus plexippus (monarch butterfly) and Trichoplusia ni (cabbage looper) egg glycoproteins.

Monosaccharide compositions of eggs from Danaus plexippus (monarch butterfly) and Trichoplusia ni (cabbage looper) were analyzed. Analyses were performed mainly with high performance anion exchange chromatography (HPAEC) using crude extracts of eggs or SDS-PAGE separated and PVDF-blotted protein bands. Man and GlcN were the major components in all cases, but low levels of Gal and Fuc were possibly present in some samples. Some T. ni egg glycoproteins even contained GalN. Although a peak comigrating with Neu5Ac could be detected with HPAEC-PAD or RP-HPLC (fluorometry) after derivatization with 1,2-diamino-4,5-methylenedioxy-benzene, the quantities were too small to be significant as an integral part of the analyzed glycoproteins. These data suggests that most of glycans on the glycoproteins are pauci-Man type N-glycans, but a small portion of N-glycan may be either hybrid type or complex type.

[Transformation of glycyrrhizinic acid. XV. Synthesis of triterpene saponins with monosaccharide residues, attached through complex ester bonds].

Triterpene saponins, glycoside analogues of glycyrrhizic acid with a modified carbohydrate chain containing monosaccharide residues attached through ester bonds, were synthesized. To this end, peracetylated glycyrrhizic acid or its 30-methyl ester were glycosylated by 2,3,4,6-tetra-O-acetyl-alpha-D-gluco-or-alpha-D-galactopyranosyl bromide in dichloroethane in the presence of Ag2CO3. Glycerrhetinic acid saponin with D-Galp residues exhibited a higher antiulcer activity than glycyrrhizic acid in rats at a dose of 25 mg/kg. Selective inhibition of T suppressor-cell function by a monosaccharide.

Interactions between regulatory T lymphocytes and other cells are assumed to occur at the level of the cell surface. T cells which suppress the generation of specifically effector cells have been described as having antigenic, idiotypic, allotypic and I-region specificity. Other T suppressor cells generated by in vitro cultivation with or without mitogenic stimulation have suppressive activity for T and B cells but no specificity can be assigned to them. These T suppressor cells (Ts) inhibit various lymphoid functions-this either reflects their polyclonal origin or indicates that the structures recognized by the Ts receptors must be common for many cell types. Carbohydrates on cell membrane-inserted glycoproteins or glycolipids might function as specific ligands for recognition by cellular receptors or soluble factors. Almost all cell-surface proteins of mammalian cells are glycosylated. There is evidence for lectin-like carbohydrate binding proteins not only in plants but also in toxins, viruses, prokaryotic cells and even mammalian cells, including T cells. A functional role for these lectin-like proteins has been described for slime moulds and suggested for the selective association of embryonic cells. We report here that addition of a monosaccharide can counteract the effect of T suppressor cells during the generation of alloreactive cytotoxic T cells (CTLs) in vitro.

Spectral signatures and structural motifs in isolated and hydrated monosaccharides: phenyl alpha- and beta-l-fucopyranoside.

The conformation and structure of phenyl-alpha-l-fucopyranoside (alpha-PhFuc), phenyl-beta-L-fucopyranoside (beta-PhFuc) and their singly hydrated complexes (alpha,beta-PhFuc.H(2)O) isolated in a molecular beam, have been investigated by means of resonant two photon ionization (R2PI) spectroscopy and ultraviolet and infrared ion-dip spectroscopy. Conformational and structural assignments have been based on comparisons between their experimental and computed near IR spectra, calculated using density functional theory (DFT) and their relative energies, determined from ab initio (MP2) calculations. The near IR spectra of "free" and hydrated alpha- and beta-PhFuc, and many other mono- and di-saccharides, provide extremely sensitive probes of hydrogen-bonded interactions which can be finely tuned by small (or large) changes in the molecular conformation. They provide characteristic "signatures" which reflect anomeric, or axial vs. equatorial differences, both revealed through comparisons between alpha/beta-PhFuc and alpha/beta-PhXyl; or similarities, revealed through comparisons between fucose (6-deoxy galactose) and galactose; or binding motifs, for example, "insertion" vs. "addition" structures in hydrated complexes. At the monosaccharide level (the first step in the carbohydrate hierarchy), these trends appear to be general. In contrast to the monohydrates of galactose (beta-PhGal) and glucose (beta-PhGlc), the conformations of alpha- and beta-PhFuc are unaffected by the binding of a single water molecule though changes in the R2PI spectra of multiply hydrated alpha-PhFucW(n) however, may reflect a conformational transformation when n> or = 3.

Permeability of rat diaphragm muscle to some methyl glycosides and its relationship to monosaccharide transport.

1. Studies in vitro have shown that rat diaphragm muscle is freely permeable to alpha- and beta-methyl d-xylosides and beta-methyl l-arabinoside but not to alpha- or beta-methyl d-glucoside or alpha-methyl d-mannoside. 2. No competition for entry between methyl pentosides and d-xylose, l-arabinose, d-glucose or 3-O-methyl-d-glucose could be demonstrated and alpha-methyl xyloside entry was not inhibited by phlorrhizin. 3. It is concluded that the monosaccharide transport system is unlikely to contribute significantly to the entry of the methyl pentosides in this tissue.

The effect of membrane composition and alcohols on the insulin-sensitive reconstituted monosaccharide-transport system of rat adipocyte plasma membranes.

The monosaccharide transporter from the plasma membranes of rat adipocytes and insulin-stimulated adipocytes has been reconstituted in sonicated liposomes. The stereospecific D-glucose uptake by liposomes made from a range of phospholipids and incorporating fatty acids has been investigated. D-Glucose uptake is correlated with an increase in lipid fluidity as a consequence of the addition of fluidizing fatty acids, changes in phospholipid acyl chain length and temperature. Benzyl alcohol and ethyl alcohol, which are generally considered to increase bilayer fluidity, decrease stereo-specific D-glucose uptake in both whole adipocytes and reconstituted liposomes. It is suggested that, although these alcohols may affect D-glucose transport by lipid-mediated fluidity changes, they also interact directly with the transporter resulting in inhibition of transport.

Butyrate-stable monosaccharide derivatives induce maturation and apoptosis in human acute myeloid leukaemia cells.

The rapid degradation and subsequent lack of efficacy of n-butyric acid in vivo has been improved by the synthesis of monosaccharide stable pro-drugs of butyric acid. We studied the effects of D1 (O-n-butanoyl-2,3-O-isopropylidene-alpha-D-mannofuranoside), G1 (1-O-n-butanoyl-D,L-xylitol), and F1 (1-O-n-butanoyl 2,3-O-isopropylidene-D,L-xylitol) on the maturation and proliferation of AML cell lines HL 60 and FLG 29.1 and of purified blast cells from 10 cases of de novo acute myeloid leukaemia (AML). AML cell maturation was measured by surface antigen expression, morphology and cytochemistry. Toxicology in mice was also evaluated (DL50 1000 mg/kg). In HL 60 cells G1 and D1 increased the expression of CD15 and CD11a (presenting 62% of promyelo-metamyelocytes), and in 7/10 cases of primary AMLs that of CD11a, CD11b, CD15, and myeloperoxidase. D1, G1 and F1 induced a dose-dependent inhibition of tritiated thymidine uptake. Apoptosis (evaluated by flow cytometry and agarose gel electrophoresis) was induced in AML blasts by D1 and F1 (79% and 94% respectively for HL 60 cells) and, with less effect, by G1 (27%). The persistence of maturative and apoptotic activity in these new pro-drugs of butyric acid, hydrolysed only inside the tumour cell, suggests a possible use in differentiation therapy of myelodysplastic syndromes and AMLs.

A male gametophyte-specific monosaccharide transporter in Arabidopsis.

The AtSTP2 gene (sugar transport protein 2) of Arabidopsis thaliana encodes a high affinity, low specificity monosaccharide carrier that can transport a number of hexoses and pentoses at similar rates. AtSTP2 has 12 putative transmembrane helices and a molecular mass of 55.0 kDa. AtSTP2 expression was localized in AtSTP2 promoter-beta-glucuronidase (GUS) Arabidopsis plants showing AtSTP2-driven GUS activity during pollen maturation and also in germinating pollen. Immunohistochemical studies with anti-AtSTP2 antiserum as well as RNA in situ hybridization analyses modified these results and showed that AtSTP2 expression is confined to the early stages of gametophyte development. Both AtSTP2 mRNA and AtSTP2 protein are first seen at the time of beginning callose degradation and microspore release from the tetrades. AtSTP2 mRNA and AtSTP2 protein are no longer detected after the mitotic divisions and the formation of the trinucleate gametophyte. No AtSTP2 mRNA or AtSTP2 protein is seen in fully developed or germinating pollen. The putative role of AtSTP2 in the uptake of glucose units resulting from callose degradation during pollen maturation is discussed.

Apoptosis-inducing activity of galloyl monosaccharides in human histiocytic lymphoma U937 cells.

Three galloyl monosaccharides contained in medicinal plants were examined for apoptosis-inducing activity in human histiocytic lymphoma U937 cells. Tetragalloyl glucose (TgG) induced apoptosis as found by chromatin condensation, DNA ladder formation, and inhibition by a caspase inhibitor. Digalloyl hamamelose had moderate activity, while monogalloyl glucose was only marginally active. These findings suggest that the number and disposition of their phenolic groups are important for apoptosis induction. TgG induced apoptosis in human colon and stomach cancer cell lines as well, indicating it is potentially useful as an anti-cancer agent.
