Effect of guanosine on antitumor activity of fluorinated pyrimidines against P 388 leukemia.

The antitumor activity of the fluorinated pyrimidines 5-fluorouracil (FUra), 5-fluorouridine (FUrd), and 5-fluoro-2'-deoxyuridine (FdUrd) against P388 leukemia was markedly potentiated by the addition of guanosine (Guo), resulting in therapeutic synergism. Any combination of FUra at 1-20 mg/kg, FUrd at 0.3-1 mg/kg, or FdUrd at 1-100 mg/kg with Guo at 100 mg/kg significantly potentiated the activity of FUra, FUrd, or FdUrd, respectively. The potentiation of these fluorinated pyrimidines by guanosine was abolished by the simultaneous administration of cytidine or uridine, but not of thymidine. In particular, cytidine was the strongest inhibitor of antitumor activity of these fluorinated pyrimidines, alone and in combination with guanosine. To obtain more effective treatment with the combination of various fluorinated pyrimidines and Guo, the influence of the molar ratios of Guo to the fluorinated pyrimidines on the antitumor activity against P388 leukemia was investigated. The increase in life-span became more pronounced with increasing molar ratios. The optimal molar ratios of Guo/FUra, Guo/FUrd, and Guo/FdUrd were more than 5, 100, and 5, respectively.

Chain-fluorinated polyamines as tumor markers--IV. Comparison of 2-fluoroputrescine and 2,2-difluoroputrescine as substrates of spermidine synthase in vitro and in vivo.

1. 2-Fluoroputrescine has a high affinity for spermidine synthase (Km 12 microM) and obeys normal Michaelis-Menten kinetics. 2. The only product of the spermidine synthase-catalysed aminopropylation of 2-fluoroputrescine is 6-fluorospermidine. Formation of the isomeric 7-fluorospermidine could not be detected. 3. 2,2-Difluoroputrescine has even a higher affinity for spermidine synthase than putrescine and 2-fluoroputrescine; however, at concentrations greater than 25 microM one observes inhibition of the aminopropylation reaction. 4. Competition experiments between putrescine and 2,2-difluoroputrescine revealed mixed type inhibition. 5. HTC cells in suspension culture incorporated only small amounts of 2-fluoroputrescine, and even less in the case of 2,2-difluoroputrescine, if they were exposed to 10 microM concentrations of these diamines for up to 24 hr. However, in the presence of 0.5 mM DFMO, a concentration not sufficient to decrease cell growth significantly, but sufficient to decrease cellular putrescine and spermidine concentrations, the uptake of the chain-fluorinated diamines and their transformation into the fluorinated polyamine analogues was dramatically enhanced. In comparison with the difluoro analogues the accumulation rate of monofluoropolyamines was greater by a factor of about two. 6. 6-Fluorospermidine and 6-fluorospermine could be detected in significant quantities in nearly all tissues of mice 48 hr after a single dose (500 mg/kg) of 2-fluoroputrescine. In an analogous experiment with 2,2-difluoroputrescine, the formation of chain-fluorinated polyamines was considerably smaller. 7. Pretreatment of Lewis lung carcinoma bearing C57BL mice with alpha-difluoromethylornithine enhanced the incorporation of 2-fluoroputrescine into all organs, except the brain. Tumor and small intestines showed by far the highest accumulation of 6-fluoropolyamines. 8. Under identical experimental conditions the accumulation of chain-fluorinated polyamines in tumor tissue was more than twice as high with 2-fluoroputrescine as precursor than with the same dose of 2,2-difluoroputrescine. In normal tissues the difference between the uptake of 2-fluoroputrescine and 2,2-difluoroputrescine was usually even greater. 9. From the fact that the accumulation of 6-fluoropolyamines is less selective in tumors than that of 6,6-difluoropolyamines, and from the lower detection sensitivity due to its lower fluorine content, we conclude that 2,2-difluoroputrescine is more advantageous as a tumor marker than 2-fluoroputrescine for detection with 19F-NMR spectroscopy.(ABSTRACT TRUNCATED AT 400 WORDS)

The fluorinated anesthetic halothane as a potential NMR biologic probe.

Fluorinated anesthetics such as halothane preferentially partition into hydrophobic environments such as cell membranes. The 19F-NMR spectrum of halothane in a rat adenocarcinoma (with known altered lipid metabolism and membrane composition) shows an altered chemical shift pattern compared to the anesthetic in normal tissue. In eight tumor samples examined, the 19F-NMR spectra exhibit two distinct resonances, compared to a single resonance observed in normal tissues. This is explained by an enhanced or altered hydrophobic component in the tumor tissue giving rise to two discrete halothane environments. Another fluorinated anesthetic, isoflurane, shows similar behavior in distinguishing normal from diseased tissue. Given the large chemical shift range of fluorine and the inherent sensitivity of this nucleus, 19F-NMR spectra of fluorinated anesthetics can also be used to follow anesthetic degradation by the liver. The ability of fluorinated anesthetics to discriminate tissues and to monitor metabolic processes is potentially useful for in vivo 19F-NMR surface coil and imaging studies.

Fluorinated 2-nitroimidazole derivative hypoxic cell radiosensitizers: radiosensitizing activities and pharmacokinetics.

PURPOSE: To assess the effects of incorporation of a CF2 group into the side chain of a 2-nitroimidazole derivative, we evaluated the in vitro and in vivo radiosensitizing activities of KU-2285 (a 2-nitroimidazole derivative with an N1-substituent of -CH2CF2CONH(CH2)nOH, n = 2) and its related compounds in comparison with those of comparable nonfluorinated compounds. The pharmacokinetics of these compounds in murine tumors was also tested. METHODS AND MATERIALS: KU-2285, KU-3202 (n = 3) and KU-3207 (n = 4) are fluorinated 2-nitroimidazole derivative compounds with similar structures. Etanidazole (a 2-nitroimidazole derivative with an N1-substituent of -CH2CONH(CH2)nOH, n = 2) and its related compounds, KU-3205 (n = 3) and KU-3206 (n = 4) were also tested. The in vitro radiosensitizing activities of each compound for hypoxic cells was evaluated with a standard colony formation method. The in vivo radiosensitizing activities of these compounds were tested in female C3H/He mice bearing SCCVII tumors using an in vivo/in vitro clonogenic assay. The pharmacokinetic studies were performed in C3H/He mice bearing the SCCVII tumor. Samples were analyzed by reverse-phase high-performance liquid chromatography. RESULTS: The in vitro radiosensitizing activities of fluorinated 2-nitroimidazoles were higher than those of the nonfluorinated compounds. Although the in vivo radiosensitizing activity of KU-2285 was higher than that of etanidazole (p < 0.05), other fluorinated 2-nitroimidazoles showed less radiosensitizing activity than the comparable nonfluorinated compounds. The compound was eliminated from serum more rapidly with the increase in the number of CH2 group in the side chain of the compound in each series. CONCLUSION: Although the in vitro sensitizing activity of the fluorinated compounds was higher than that of the comparable nonfluorinated compounds, the in vivo radiosensitizing activity of all fluorinated compounds but KU-2285 was lower than that of comparable etanidazole group compounds, probably due to their lower molecular concentrations in tumor and rapid elimination.

Synthesis of carbon-11 labeled fluorinated 2-arylbenzothiazoles as novel potential PET cancer imaging agents.

Fluorinated 2-arylbenzothiazoles are new potential antitumor drugs, which show potent and selective inhibitory activity against breast, lung, and colon cancer cell lines. Carbon-11 labeled fluorinated 2-arylbenzothiazoles may serve as novel probes for positron emission tomography (PET) to image tyrosine kinase in cancers. The preparation of 4-fluorinated 2-arylbenzothiazoles 4-fluoro-2-(3-benzloxy-4-methoxyphenyl)benzothiazole (6a) and 4-fluoro-2-(3,4-dimethoxyphenyl)benzothiazole (6b) was achieved by a modification of Jacobson thioanilide radical cyclization chemistry. Hydrogenolytic cleavage of the benzyl ether group of compound 6a using H(2)/Pd-C provided the precursor 4-fluoro-2-(3-hydroxy-4-methoxyphenyl)benzothiazole (7) for radiolabeling. Synthesis of radiolabeling precursors and the reference standards 5- and 6-fluorinated arylbenzothiazoles (11c-n) was achieved via the reaction of o-aminothiophenol disulfides with substituted benzaldehydes under reducing conditions. The target radiotracers carbon-11 labeled 4-, 5-, and 6-fluorinated arylbenzothiazoles (3-[(11)C]6b, 4-[(11)C]11c, 3-[(11)C]11c, 5-[(11)C]11f, 4-[(11)C]11f, 4-[(11)C]11i, 3-[(11)C]11i, 5-[(11)C]11l, and 4-[(11)C]11l) were prepared by O-[(11)C]methylation of the phenolic hydroxyl precursors (7, 11d, 11e, 11g, 11h, 11j, 11k, 11m, and 11n) with [(11)C]methyl triflate and isolated by solid-phase extraction (SPE) purification in 30-55% radiochemical yields.

Thermodynamics of micellization for partially fluorinated cationic gemini surfactants and related single-chain surfactants in aqueous solution.

A series of partially fluorinated cationic gemini surfactants and their corresponding monomeric surfactants have been studied by isothermal titration microcalorimetry. The critical micelle concentration (CMC) and enthalpy of micellization (DeltaH(mic)) were obtained from calorimetric curves. The CMCs of the gemini surfactants are much lower than those of the corresponding monomeric surfactants and decrease with an increase in the number of fluorine atoms on the hydrophobic chain. The micellization of partially fluorinated cationic gemini surfactants is much more exothermic than that of the corresponding monomeric surfactants. Because of the incompatibility of hydrocarbon spacer and partially fluorinated chain, DeltaH(mic) values of the surfactants with a C6 spacer are more negative than those of the surfactants with a C12 spacer. The variations in the architecture of the fluorocarbon chain segments may be the reason of the irregularities in the change of DeltaH(mic) for the gemini surfactants. Moreover, the contribution of the enthalpy generally increases with an increase in the number of fluorine atoms.

Mixed fluorinated-hydrogenated surfactant-based system: preparation of ordered mesoporous materials.

We have investigated a mixed fluorinated-hydrogenated surfactant-based system [C8F17C2H4(OC2H4)9-C12H25(OC2H4)8] in water. The phase diagram exhibits that the micellar domain can be divided into three parts: above 80 wt% of water both hydrogenated and fluorinated surfactants are completely miscible and they formed mixed micelles in all proportion. When the water concentration is decreased from 80 to 60 wt% a gap of miscibility appears and two micellar zones, one fluorocarbon-rich micelles and one hydrocarbon-rich micelles are observed. The liquid crystal domain is composed of one fluorocarbon-rich (H(F)(1)) and one hydrocarbon-rich (H(H)(1)) hexagonal phase. The hydrophobic radius and the cross-sectional area remain constant in the H(H)(1) and in the H(F)(1) domains. Moreover, SAXS measurements proved that the hydrophobic chains in the liquid crystal phases adopt rather an extended conformation. Then the mixture of surfactants was used as template for the preparation of mesoporous materials. Mesostructured silicas with a well hexagonal array of their channels were prepared via a cooperative templating mechanism (CTM), if the loading of fluorinated surfactant is larger than 50%. Decreasing the proportion of the fluorinated amphiphile in the mixture leads to the formation of mesoporous silica with a disordered structure. In this case the channel arrangement is no longer governed by the fluorinated surfactant but by the hydrogenated one.

Phase behavior of fluorocarbon di-O-alkyl-glycerophosphocholines and glycerophosphoethanolamines and long-term shelf stability of fluorinated liposomes.

This paper is devoted to the morphological characterization, by freeze-fracture electron microscopy, and to the thermotropic phase behavior, by differential scanning calorimetry and X-ray diffraction of the aqueous dispersions of various fluorocarbon/fluorocarbon or mixed fluorocarbon/hydrocarbon 1,2- or 1,3-di-O-alkyl-glycerophosphocholines (PC) and 1,2-di-O-alkyl-glycerophosphoethanolamines (PE). The fluorinated PCs form classical lamellar phases and liposomes while an interdigitated lamellar phase has been evidenced for a hydrocarbon 1,3-analog. The fluorinated PEs display a lamellar to hexagonal phase transition which occurs almost simultaneously with the gel-to-fluid lamellar phase transition. The impact of each of the structural features [ether vs ester chemical junction connecting the hydrophobic chains on glycerol, their position (1,2- vs 1,3 isomers), number and length of the perfluoroalkylated chains, length of the fluorinated tail and hydrocarbon spacer, PC vs PE polar head] of the fluorinated phospholipids on the phase transition thermodynamic parameters (Tc, delta H, delta S) is discussed. Most of the liposomes formed from the fluorinated ether-PCs display a remarkable long-term shelf stability: they can be thermally sterilized and stored at room temperature for several months without any significant modification of their size and size distribution.

Structure and interactive properties of highly fluorinated phospholipid bilayers.

Because liposomes containing fluoroalkylated phospholipids are being developed for in vivo drug delivery, the structure and interactive properties of several fluoroalkylated glycerophosphocholines (PCs) were investigated by x-ray diffraction/osmotic stress, dipole potential, and hydrophobic ion binding measurements. The lipids included PCs with highly fluorinated tails on both alkyl chains and PCs with one hydrocarbon chain and one fluoroalkylated chain. Electron density profiles showed high electron density peaks in the center of the bilayer corresponding to the fluorine atoms. The height and width of these high density peaks varied systematically, depending on the number of fluorines and their position on the alkyl chains, and on whether the bilayer was in the gel or liquid crystalline phase. Wide-angle diffraction showed that in both gel and liquid crystalline bilayers the distance between adjacent alkyl chains was greater in fluoroalkylated PCs than in analogous hydrocarbon PCs. For interbilayer separations of less than about 8 A, pressure-distance relations for fluoroalkylated PCs were similar to those previously obtained from PC bilayers with hydrocarbon chains. However, for bilayer separations greater than 8A, the total repulsive pressure depended on whether the fluoroalkylated PC was in a gel or liquid-crystalline phase. We argue that these pressure-distance relations contain contributions from both hydration and entropic repulsive pressures. Dipole potentials ranged from -680 mV for PCs with both chains fluoroalkylated to -180 mV for PCs with one chain fluoroalkylated, compared to +415 mV for egg PC. The change in dipole potential as a function of subphase concentration of tetraphenyl-boron was much larger for egg PC than for fluorinated PC monolayers, indicating that the fluorine atoms modified the binding of this hydrophobic anion. Thus, compared to conventional liposomes, liposomes made from fluoroalkylated PCs have different binding properties, which may be relevant to their use as drug carriers.

Transmembrane pH-driven Na+ permeability of fluorinated phospholipid-based membranes.

The permeability to the H+/Na+ exchange of fluorinated phospholipid-based membranes has been evaluated by measuring the dissipation rate of a liposomal transmembrane pH gradient in the presence of Na+. The fluorinated liposomes are made from fluorocarbon/hydrocarbon or fluorocarbon/fluorocarbon double-chain ether-connected glycerophosphocholines or amido-connected phosphocholines deriving from diaminopropanol or serine. The fluorocarbon/hydrocarbon mixed-chain phospholipids, as compared to the fluorocarbon/fluorocarbon ones, form membranes that are substantially more able to maintain a transmembrane pH gradient in the presence of NA+ and display a lower Na+ permeability. However, these membranes are more permeable to the H+/Na/ exchange than conventional DSPC (1,2-distearoylphosphatidylcholine) ones. Our results indicate a detrimental impact of the membrane fluorination degree on H+/Na+ permeability: the lower the fluorination degree of the membrane, the lower its H+/Na+ permeability. Concerning structure/permeability relationships, it appears that the replacement of the ester connecting bond in their fluorinated phosphatidylcholine analogues for an ether or amide one lowers the transmembrane H+/Na+ exchange.

Improved stability of highly fluorinated phospholipid-based vesicles in the presence of bile salts.

The stability of fluorinated phospholipid-based vesicles in terms of detergent-induced release of encapsulated carboxyfluorescein has been evaluated. The fluorinated liposomes are substantially more resistant towards the lytic action of sodium taurocholate than conventional DSPC or even DSPC/CH 1/1 liposomes. Concerning structure/permeability relationships, the larger the fluorination degree of the membrane, the higher the resistance of the fluorinated liposomes to their destruction by the detergent. These results show that fluorinated liposomes have a promising potential as drug carrier and delivery systems for oral administration.

Dependence of selectivity on eluent composition and temperature in the HPLC separation of taxanes using fluorinated and hydrocarbon phases.

Optimized conditions of aqueous acetonitrile (ACN) eluent composition and temperature are established for the rapid separation of a standard mixture of 15 taxanes, on each of five different fluorinated phases and one C8 hydrocarbonaceous phase. On both types of stationary phase, the retention factors (k') of most of the taxanes decrease at the same rate with increasing ACN concentration. However, the taxanes containing a xylosyl group show a higher rate of decrease, which necessitates careful control of eluent composition to achieve separation of all the taxanes. Temperature can have a remarkable and counterintuitive effect on retention and selectivity. For the C8 phase with eluent compositions in the 40%-60% ACN range, the k' values of the xylosyl taxanes show an increase with increasing temperature over the range from 25 to 55 degrees C; the k' values for 10-deacetyl baccatin III and 10-deacetyl taxol go through a maximum over the same ranges. The other taxanes behave normally. The same pattern is observed on the propyl(perfluorophenyl) phase, although this and the other fluorinated phases are less retentive. This accounts for the common belief that fluorinated stationary phases offer resolution of taxanes superior to that on hydrocarbon phases. The higher retention on the latter requires eluent compositions near 50% ACN, where careful temperature optimization is required, which in practice is rarely performed. The lesser retention on the fluorinated phases allows use of lower ACN concentrations where the aberrant temperature effect is not found, so good separations without temperature optimization can be achieved. Further evidence of the lack of any fundamental difference in the selectivity of the two types of stationary phase is the similarity of the surface excess isotherms measured for ACN/H2O on both the fluorinated and hydrocarbon phases.

Recent highlights in electrophilic fluorination with 1-chloromethyl-4-fluoro- 1,4-diazoniabicyclo[2.2.2]octane bis(tetrafluoroborate).

Synthetic and structural aspects of organofluorine compounds continue to be the focal points of vigorous research activities, as evidenced by the appearance of a large number of publications. Among the various useful methodologies for the introduction of fluorine into organic molecules, electrophilic fluorination is a promising and exciting area of research. While a variety of electrophilic fluorinating reagents are available, currently 1-chloromethyl-4-fluoro-1,4-diazoniabicyclo[2.2.2]octane bis(tetrafluoroborate) (Selectfluor) provides a remarkably straightforward and effective route. The breadth of applications realizable from this reagent in its role as a key electrophilic fluorinating reagent is highlighted here. This Account covers the literature for electrophilic fluorination reactions that employ Selectfluor during the period January 1999-January 2003.

Proton and fluorine nuclear magnetic resonance spectroscopic observation of hemiacetal formation between N-acyl-p-fluorophenylalaninals and alpha-chymotrypsin.

Proton nuclear magnetic resonance (NMR) spectroscopy shows that the free aldehyde and not the hydrate of N-acetyl-DL-p-fluorophenylalaninal binds to alpha-chymotrypsin. A proton NMR cross-saturation experiment shows that the initial noncovalent complex is in equilibrium with a hemiacetal formed between the aldehyde and the active site serine residue. Fluorine NMR spectra of N-acetyl-DL- (and N-acetyl-L-) p-fluorophenylalaninal in the presence of alpha-chymotrypsin show separate signals for the hemiacetal complex, the bound aldehyde, the free aldehyde, and the free hydrate. N-Benzoyl-DL-p-fluorophenylalaninal fluorine NMR signals are also observed for all species except the bound aldehyde form in the presence of alpha-chymotrypsin. The D and L enantiomers of the hydrate of the N-acetyl aldehyde inhibitor give separate fluorine NMR signals, arising from chemical exchange between the L-aldehyde-alpha-chymotrypsin complex and the free L hydrate. The enzyme-bound inhibitor fluorine signals disappear upon proton decoupling due to a negative nuclear Overhauser effect. Upon gated decoupling of protons, the L hydrate and free aldehyde fluorine signals are reduced in intensity relative to that of the D hydrate signal in the racemate aldehyde complex. This is attributed to a saturation transfer of the heteronuclear nuclear Overhauser effect.

Proposed fluorination mechanism of CB(5)H(6)(-) and CB(9)H(10)(-) with HF. Evidence of kinetic control in the formation of 2-CB(5)H(5)F(-) and 6-CB(9)H(9)F(-).

Two pathways have been considered in the fluorination of CB(5)H(6)(-) and CB(9)H(10)(-) by HF. In the ionic HF fluorination pathway, the monocarborane anion cage is first protonated in a BBB face followed by H(2) elimination and fluoride anion addition. In the covalent HF fluorination pathway, HF is first coordinated through hydrogen to the BBB face. Next, the fluorine can add to either an axial or equatorial boron atom which opens the cage to a nido structure with an endo fluoride substituent. Endo to exo rearrangement occurs with a small activation barrier followed by H(2) elimination. In both pathways, fluorination at the equatorial boron position is predicted to have smaller activation barriers even though substitution at the axial position leads to the more stable products.

Crystal structure of ClF(4)(+)SbF(6)(-), normal coordinate analyses of ClF(4)(+), BrF(4)(+), IF(4)(+), SF(4), SeF(4), and TeF(4), and simple method for calculating the effects of fluorine bridging on the structure and vibrational spectra of ions in a strongly interacting ionic solid.

The crystal structure of the 1:1 adduct ClF(5).SbF(5) was determined and contains discrete ClF(4)(+) and SbF(6)(-) ions. The ClF(4)(+) cation has a pseudotrigonal bipyramidal structure with two longer and more ionic axial bonds and two shorter and more covalent equatorial bonds. The third equatorial position is occupied by a sterically active free valence electron pair of chlorine. The coordination about the chlorine atom is completed by two longer fluorine contacts in the equatorial plane, resulting in the formation of infinite zigzag chains of alternating ClF(4)(+) and cis-fluorine bridged SbF(6)(-) ions. Electronic structure calculations were carried out for the isoelectronic series ClF(4)(+), BrF(4)(+), IF(4)(+) and SF(4), SeF(4), TeF(4) at the B3LYP, MP2, and CCSD(T) levels of theory and used to revise the previous vibrational assignments and force fields. The discrepancies between the vibrational spectra observed for ClF(4)(+) in ClF(4)(+)SbF(6)(-) and those calculated for free ClF(4)(+) are largely due to the fluorine bridging that compresses the equatorial F-Cl-F bond angle and increases the barrier toward equatorial-axial fluorine exchange by the Berry mechanism. A computationally simple model, involving ClF(4)(+) and two fluorine-bridged HF molecules at a fixed distance as additional equatorial ligands, was used to simulate the bridging in the infinite chain structure and greatly improved the fit between observed and calculated spectra.

Hydration and dewetting near fluorinated superhydrophobic plates.

The water dynamics near nanoscale fluorinated (CF(3)(CF(2))(7)(CH(2))(2)SiH(3)) monolayers (plates) as well as possible dewetting transitions in-between two such plates have been studied with molecular dynamics simulations in this paper. A "weak water depletion" is found near the single fluorinated surface, with an average water density in the first solvation shells 6-8% lower than its hydrogenated counterpart. The fluorinated molecules are also found to be water impermeable, consistent with experimental findings. More surprisingly, a dewetting transition is found in the interplate region with a critical distance D(c) of 10 A (3-4 water diameters) for double plates with 8 x 8 molecules each (plate size approximately 4 nm x 4 nm). This transition, although occurring on a microscopic length scale, is reminiscent of a first-order phase transition from liquid to vapor. The unusual superhydrophobicity of fluorocarbons is found to be related to their larger size (or surface area) as compared to hydrocarbons, which "dilutes" their physical interactions with water. The water-plate interaction profile shows that the fluorinated carbons have a 10-12% weaker water-plate interaction than their hydrogenated counterparts in the nearest solvation shell, even though the fluorocarbons do have a stronger electrostatic interaction with water due to their larger partial charges. However, the van der Waals interactions dominate the water-plate interaction within the nearest shell, with up to 90% contributions to the total interaction energy, and fluorocarbons have a noticeably weaker (by 10-15%) van der Waals interaction with water in the nearest shell than do hydrocarbons. Both the slightly weaker water-plate interaction and larger surface area contribute to the stronger dewetting transition in the current fluorinated carbon plates.

Syntheses and adrenergic agonist properties of ring-fluorinated isoproterenols.

2-Fluoro-, 5-fluoro-, and 6-fluoroisoproterenol were synthesized by reduction of the Schiff base formed between the corresponding ring-fluorinated 3,4-bis(benzyloxy)phenethanolamine and acetone, followed by reductive debenzylation in the presence of oxalic acid to yield crystalline neutral oxalates. The apparent beta-adrenergic potencies were determined in the isolated guinea pig atria. 2-Fluoro- and 5-fluoroisoproterenol were equipotent with (+/-)-isoproterenol, while 6-fluoroisoprotenol was virtually inactive. No alpha-adrenergic agonist activity (guinea pig aorta) was shown by any of the fluoroisoproterenols. Displacement of alpha- and beta-specific radioligands from isolated membrane preparations from rat brain by the fluoroisoproterenols were in agreement with the responses of the organ preparations. Thus, the apparent fluorine-induced specificity is due to specificity at the receptor binding site. The effects of fluorine substitution are discussed with regard to the apparent negative influence of the 6-fluoro substituent on the beta-agonist properties of isoproterenol, the lack of any increase in potency due to the 2-fluoro substituent, and the possibility of fluorine-induced changes in the electron density of the aromatic ring as a possible rational for the fluorine-induced specificity of both the fluoroisoproterenols and the fluoronoirepinephrines.
