Studies on the phenazine methosulphate-tetrazolium salt capture reaction in NAD(P)+-dependent dehydrogenase cytochemistry. I. Localization artefacts caused by the escape of reduced co-enzyme during cytochemical reactions for NAD(P)+-dependent dehydrogenases.

The correct localization of oxidative enzymes using cytochemical tetrazolium methods, in which low molecular weight electron carriers such as NAD(P)H and reduced phenazine methosulphate (PMSH) are used, can be endangered by the escape of the reduced intermediates before they react to form the insoluble formazan at the true enzyme-containing sites. To investigate this phenomenon, the glucose-6-phosphate dehydrogenase reaction was studied in fixed erythrocytes which, because of their microscopic dimensions, are well-suited for studying the loss of intermediates. A mixture of active and heat-inactivated fixed erythrocytes was incubated in a PMS-supplemented medium for glucose-6-phosphate dehydrogenase. The cytophotometric histograms showed that the final formazan precipitate was equally distributed over both active and inactivated cells. When bovine serum albumin was added to the medium, all the formazan was found to be bound to this protein and the erythrocytes remained essentially unstained. The false localization in this system could be explained by an unfavourable balance between the capture of electrons carried by NADPH within the erythrocyte and the diffusion of NADPH out of the erythrocyte. The rate constant of NADPH oxidation was determined, as was also the diffusion constant of NADPH in a protein matrix. Substituting the data obtained into formulae derived from the enzyme cytochemical localization theory of Holt & O'Sullivan (1958), it was calculated that the capture reaction was highly deficient and, theoretically, less than 1% of the total amount of formazan produced was localized within the erythrocyte which explains the false localization observed. The importance of these findings for the cytochemical demonstration of NAD(P)+-dependent dehydrogenases in cells and electropherograms is briefly discussed.

The influence of a weak artificial magnetic 10 Hz field on the extinction of a conditioned flight reaction in rats.

36 three to four months old albino rats were conditioned to flee after the onset of an acoustic stimulus (1000 Hz). During the period of extinction of the flight reaction half of the animals were exposed to an alternating magnetic field (10 Hz, 0.03 mT peak to peak) for the time of investigation. These animals showed a significantly higher number of flight reactions than the control animals investigated without magnetic field, on the 4th, 5th, and 6th day of exposure (p less than 0.05).

Behavioral transition of crayfish avoidance reaction in response to uropod stimulation.

Unilateral mechanical stimulation of a uropod elicited an avoidance reaction in the crayfish Procambarus clarkii. Depending upon the animal's size, either one of two alternative behavioral acts emerged as the avoidance reaction to similar stimuli. A stationary resting posture of the crayfish and a passive extension of the abdomen were prerequisite conditions for inducing the avoidance reaction. In small animals (less than 10 cm in body length), the "dart" response was mainly elicited. Animals responded with a sudden closing of both uropods and rapid forward walking to scuttle away from the stimulus source. This act was regarded as one form of escape behavior. With increasing animal size, the "turn" response became more probable than the "dart" response. During the "turn" response, the uropod on the stimulated side opened and that on the contralateral side closed. This act was frequently followed by a rapid turning movement towards the stimulus source with an extension or flexion of the abdomen. This act was also regarded as a variant form of defensive behavior. In large animals with autotomized chelipeds, the response to the uropod stimulation reverted to that of the "dart" response after 2 to 3 days.

Oligopeptides in the regulation of feeding and avoidance behaviour in the land snail (Helix lucorum).

The effects of the oligopeptides pentagastrin, cholecystokinin octapeptide, substance P, and FMRF-amide on the feeding and avoidance behaviour of the land snail Helix lucorum were studied. Administration of 1 microM pentagastrin to fasted snails immediately before feeding led to a significant increase in the amount of food consumed in the first 20 min of the feeding period, although there was no change in the total amount of food consumed. Prefeeding injections of 0.05 microM pentagastrin or 0.05 microM or 1 microM solutions of cholecystokinin octapeptide, substance P, or FMRF-amide had no effect on feeding behaviour. Pentagastrin and cholecystokinin octapeptide (0.05 microM solutions) invoked a significant reversible decrease in the generalised avoidance reaction thresholds in fasted but not in prefed snails, whereas the same concentration of FMRF-amide reduced these thresholds only in prefed snails.

"Mind control" and the battering of women.

This paper describes one variation in the battering phenomenon which was initially observed among low-income women. The strategies of coercion and deception utilized by the abusive male in these relationships are described and compared with similar strategies of "mind control" utilized in more traditional "cultic" systems. The debilitating effects of these techniques on the battered female are described, as is the battering male's own separation reaction, and the probable dynamics of the men and women involved in this pathological family system. Some preliminary assessment and treatment guidelines are offered.

[The participation of the neostriatal cholinergic system in the differentiation of acoustic signals in dogs].

In chronic experiments on six dogs the influence was studied of micro-injections of choline agonist carbocholine (0.05-0.2 mkg) and of blocker of choline receptors atropine (40 mkg) in the caudate nucleus head of the left and right hemispheres on realization of instrumental defensive reflexes, connected with the maintenance of definite posture and on differentiation of signals in defensive situation. It has been shown that the cholinergic system of the neostriatum participated in realization of both the motor and sensory mechanisms in connection with the realization of motor responses to defensive and differentiation signals. Analysis of the obtained results also allowed to make a conclusion that the influence of carbocholine micro-injections into the neostriatum on differentiation depended on a number of factors: it did not take place when the signal was poorly distinguished (judging by the values of motor components to defensive and differentiation signals) or, on the other hand, against the background of stable differentiation reaction in other animals, i.e. in case of complete learning.

[Psychological mechanisms of refusal reactions in schizophrenic patients].

The psychological study is based on the clinical typology of refusal reactions (RR) in schizophrenic patients--"capitulation", "avoidance", "absenteism". Seventy patients with RR, mean age 27 years, 25 women, 45 men, were examined with the use of a number of psychological methods. The findings prove the hypothesis on the disturbances of motivation as a psychological mechanism of RR in schizophrenic patients. Deficiency is related to functions of the motivational process: incentive function, goal achievement, sense making and strengthened by trait anxiety and desize to avoid danger. The main factor promoting RR formation is basic motivation deficiency caused by endogenous process (schizophrenia). A mechanism of capitulation reaction formation is closely related to expectation of failure and refusal from acting as an opportunity to preserve self-estimation; avoidance reaction is characterized by significant disturbance of the motivation at the stage of incentive, goal achievement and sense making. Motivation deficiency in patients with absenteism reactions is predominantly associated with an specific individual's sense.

[The effect of a vasopressin analog on the reaction of the command neurons in the defensive behavior of the edible snail during nerve stimulation].

On the level of snail command neurones of the defensive behaviour an increase was revealed of the amplitude of summary EPSP recorded in command neurones in response to nerve stimulation, an increase of probability of appearance of action potential in the reaction and, under certain conditions, a significant decrease of habituation speed at rhythmic (0.1 Hz) nerve stimulation against the background of peptide. The latter effect was found during comparison of groups of neurones--control neurones and those to which the peptide was presented before the first series of stimulation in the test, i. e. without preliminary elaboration of habituation. The decrease of the speed of habituation concerned both the amplitude of summary EPSP and the probability of action potential appearance in the reaction. All these changes against the background of vasopressin analogue may be the basis of the increase of spike reactions of command neurones of the defensive behaviour and thus the basis of the initiation or the increase of behavioural defensive reactions. The obtained effects were not protracted and took place only in the presence of the peptide in the extracellular medium.

[Participation of the fear reaction in limiting the period of primary socialization in silver foxes].

Administration of chloditan, an inhibitor of adrenal cortex function, at the terminal stage of the sensitive period of socialization of foxes reduced the cortisol concentration in blood as compared with control and intact animals aged 45 and 60 days. Simultaneously, the weakening of fear reaction and an enhancement of exploratory and motor activities in an unknown surrounding were observed. However, administration of chloditan did not affect the duration of the sensitive period of socialization, since the aggressive defensive reactions changed neither at the age of 45 and 60 days, nor at the age of 7 months.

Oral administration of soybean lecithin transphosphatidylated phosphatidylserine improves memory impairment in aged rats.

Soybean lecithin transphosphatidylated phosphatidylserine (SB-tPS) was prepared from soybean lecithin and L-serine by a transphosphatidylation reaction, and its effect on age-related memory impairment was evaluated in rats by the Morris water maze test. Continuous oral administration of SB-tPS (60 mg x kg(-1) x d(-1) for 60 d) to male aged rats (24-25 mo) significantly improved performance in the water maze escape test (P < 0.01 vs. control aged rats) similar to bovine brain cortex-derived phosphatidylserine, which restores cognitive function in patients with senile dementia. SB-tPS also increased acetylcholine release and the Na(+), K(+)-ATPase activity of the synaptosomes prepared from these aged rats to the level in young rats. The nootropic actions of SB-tPS in the present study can be partly explained by the changes in these biochemical activities.

[Differences in the responses of identified neurons to chemical stimuli in satiated and hungry edible snails].

Reactions of command neurones for avoidance behaviour to food were investigated in hungry and satiated snails in "CNS-chemoreceptor" preparations, in which hemolymph was washed out by saline. Neuronal responses in hungry animal preparations differed significantly from responses in satiated animals preparations. Perfusion of hungry animal preparations with hemolymph of satiated animals changed significantly the responses of command neurones for avoidance behaviour. These responses resembled the reactions of the same neurones to food after aversive conditioning to food of hungry snails. The role of humoral factor in learning is discussed.

Components of predation defense behavior in chickens: evidence for endogenous rhythmicity.

The manifestation of diurnal periodicity and the extent of its control by the photoperiod was assessed in three predation defense reactions which constitute either components or outcomes of a predator-prey interaction sequence. Two-hundred White Leghorn chicks were reared from hatching for one week in either 24L or 12L and then tested at one of two clock hours previously demonstrated to define peak and trough response for one of the components. Putative evidence was found for an endogenous source of the periodicity manifested in all reactions. Maintenance schedule did not entrain the periodicity, but simple room entry and handling elicited anti-predator reactions, the extent of which varied as a function of clock hour. A general model of predation defense behavior was proposed.

Taming in wild rats following medial amygdaloid lesions.

Flight behavior, startle reactions, and defensiveness to nonpainful stimuli were examined before and after medial amygdaloid lesions in wild Rattus norvegicus and R. rattus. Lesions which included bilateral damage to the medial nucleus strongly reduced defensiveness but had no effect on flight behavior. In contrast, damage to nearby medial structures, which spared the medial nucleus, had no effect on defensiveness but reduced flight behavior. All lesions produced a transient decrease in activity but had no effect on startle reactions. The results suggest a differential organization of flight and defense behavior within the medial amygdala.

Amygdala kindling increased fear-response, but did not impair spatial memory in rats.

The behavioral effects of amygdala kindling, a model of experimental epilepsy in rats, are reported. The animals were stimulated twice a day until stage 5 (generalized clonic) seizures were obtained three times. Two weeks later the performance of the amygdala-kindled and sham-operated rats was tested in the open-field test, on the elevated plus maze, elevated bridges, and in the Morris water maze. The results show that amygdala kindling decreased exploratory and other motor activity in the open-field test, had anxiogenic effects on the elevated plus-maze, decreased boldness on the elevated bridges, but had a negligible affect in the spatial memory task. These results suggest that amygdala kindling affects the normal fear reaction of rats, a response that is known to be mediated through the amygdaloid pathways.

[A comparative neuropsychological approach to the analysis of hippocampal functions].

Formation of conditioned switching-over of heterogeneous instrumental reflexes in dogs was more successful than in rats, which testifies about significant differences of the functional organization of analytical-synthetic brain activity in rodents and predatory animals. Experiments with lesion of the hippocampus (in rats) and the records of its electrical activity (in dogs) allowed to conclude that the hippocampus of the both types of animals belongs to the system of structures, participating in the formation of conditioned switching-over. The data, obtained in the process of elaboration of the switching-over and probabilistic reinforcement of the alimentary conditioned stimulus, testify that providing for the reactions to signals with low probability of reinforcement in different species of animals is one of the functions of the hippocampus.

[Participation of the central noradrenergic system in the mechanisms of latent inhibition].

Male Wistar rats were learnt to conditioned reaction of passive avoidance 10 days after intraperitoneal administration of 50 mg/kg of specific neurotoxin DSP4. The character of conditioned reaction reproduction, duration of its conservation and its stability against amnestic processing were analyzed. It has been found that reduction of activity of noradrenergic coerulo-cortical system does not influence the conditioned reaction reproduction but inhibits its spontaneous extinction and prevents amnesia development. The obtained data are discussed in aspect of central noradrenaline participation in latent inhibition mechanisms.
