Antioxidant enzymes of larvae of the cabbage looper moth, Trichoplusia ni: subcellular distribution and activities of superoxide dismutase, catalase and glutathione reductase.

In the mid-fifth instar larvae of the cabbage looper moth, Trichoplusia ni, the subcellular distribution of total superoxide dismutase was as follows: 3.05 units (70.0%), 0.97 units (22.3%), and 0.33 units (7.6%) mg-1 protein in the mitochondrial, cytosolic and nuclear fractions, respectively. No superoxide dismutase activity was detected in the microsomal fraction. Catalase activity was unusually high and as follows: 283.4 units (47.3%), 150.1 units (25.1%), 142.3 units (23.8%), and 22.9 units (3.8%) mg-1 protein in the mitochondrial, cytosolic, microsomal (containing peroxisomes), and nuclear fractions. No glutathione peroxidase activity was found, but appreciable glutathione reductase activity was detected with broad subcellular distribution as follows: 3.86 units (36.1%), 3.68 units (34.0%), 2.46 units (23.0%), and 0.70 units (6.5%) mg-1 protein in the nuclear, mitochondrial, and cytosolic fractions, respectively. The unusually wide intracellular distribution of catalase in this phytophagous insect is apparently an evolutionary adaptation to the absence of glutathione peroxidase; hence, lack of a glutathione peroxidase-glutathione reductase role in alleviating stress from lipid peroxidation. Catalase working sequentially to superoxide dismutase, may nearly completely prevent the formation of the lipid peroxidizing .OH radical from all intracellular compartments by the destruction of H2O2 which together with O2- is a precursor of .OH.

Effects of cabbage leaf protein concentrate on the serum and liver lipid concentrations in rats.

The effects of cabbage leaf protein concentrate (CLPC) on serum and liver lipid concentrations were determined in rats fed cholesterol-enriched and cholesterol-free diets. In rats fed the cholesterol-enriched diet with CLPC, total cholesterol, triacylglycerol and phospholipid concentrations in both the serum and liver, as well as the atherogenic index diet were significantly lower than those of the rats fed a casein diet. A supplement of methionine to the CLPC diet raised serum HDL-cholesterol and body weight gain, indicating that the addition of methionine to the CLPC diet is not only available to improve the nutritive value of CLPC but also to lower the atherogenic index. In rats fed the cholesterol-free diet, the liver total cholesterol and triacylglycerol concentrations of the CLPC-fed rats also showed lower values than those of the casein-fed rats, however, the serum total cholesterol concentration of the CLPC-fed rats did not differ from that of the casein-fed rats.

Effect of dietary cabbage fermentation extract and young barley leaf powder on immune function of Sprague-Dawley rats.

We investigated dietary effects of cabbage fermentation extract (CFE) and young barley leaf powder (YBLP) on rat immune functions. Male Sprague-Dawley rats of 4 wk age were fed for 3 wk diets containing these samples at 0.1 or 1% level. After the feeding period, serum IgG level was significantly higher in the rats fed 1% CFE. IgG productivity of spleen lymphocytes was enhanced dose-dependently in both groups of CFE and YBLP. Furthermore, IgG productivity of mesenteric lymph node (MLN) lymphocytes was approximately 2 times higher in the rats fed 1% CFE diet than in the control ones. IgA productivity of MLN lymphocytes tended to increase in both of CFE and YBLP groups. From these results, it was suggested that dietary CFE and YBLP reinforced Ig productivity in both systemic and intestinal immune systems. Moreover, CFE feeding tended to enhance the production of TNF-alpha by spleen lymphocytes. In spleen phospholipids, the level of arachidonic acid, a substrate for inflammatory lipid mediators, was not affected by CFE or YBLP feeding.

Antimicrobial activity of sulfur compounds derived from cabbage.

Selected sulfur compounds found in cabbage and its fermentation product, sauerkraut, were tested for minimum inhibitory concentration (MIC) against growth of 15 species of bacteria and 4 species of yeasts. S-Methyl-L-cysteine sulfoxide, sinigrin, and dimethyl sulfide at 500 ppm were not inhibitory to any of the bacteria and yeasts tested. Dimethyl disulfide at 500 ppm retarded some, but did not prevent growth of any of the test microorganisms. Dimethyl trisulfide had an MIC to bacteria of 200 ppm and to yeast of 20 ppm. Methyl methanethiosulfinate had an MIC between 50 and 200 ppm for all bacteria, and between 6 and 10 ppm for all yeasts tested. Methyl methanethiosulfonate had an MIC between 20 and 100 ppm for bacteria and between 50 and 500 ppm for yeasts. Allyl isothiocyanate had an MIC between 50 and 500 ppm for bacteria and between 1 and 4 ppm for yeasts. Methyl methanethiosulfinate was 10 to 100 times more inhibitory against Listeria monocytogenes at pH values of 5, 6, and 7 and was much less influenced by pH than was sodium benzoate.

Antimicrobial resistance of Listeria monocytogenes isolated from various cabbage farms and packing sheds in Texas.

Twenty-one isolates of Listeria monocytogenes from cabbage, environmental, and water samples were evaluated for antimicrobial resistance by the disk diffusion method. Ninety-five percent (20 of 21) of the isolates tested were resistant to two or more antimicrobial agents. This finding is significant, since multiresistant strains of Listeria spp. are not commonly found in nature. Eighty-five percent (17 of 20) of the multiresistant strains were resistant to penicillin, and the remaining multiresistant isolates were somewhat sensitive to penicillin. A multiresistant strain showing intermediate sensitivity to penicillin was resistant to gentamicin. One isolate was susceptible to all antimicrobial agents except penicillin. Penicillin- and gentamicin-resistant L. monocytogenes have not previously been reported from human, food, or environmental samples. This study provides evidence of the emergence of multiresistant L. monocytogenes strains, pointing to an increase in the potential threat to human health posed by this pathogen.

Efficacy of acidified sodium chlorite treatments in reducing Escherichia coli O157:H7 on Chinese cabbage.

Efficacy of acidified sodium chlorite for reducing the population of Escherichia coli O157:H7 pathogens on Chinese cabbage leaves was evaluated. Washing leaves with distilled water could reduce the population of E. coli O157:H7 by approximately 1.0 log CFU/g, whereas treating with acidified chlorite solution could reduce the population by 3.0 log CFU/g without changing the leaf color. A similar level of reduction was achieved by washing with sodium chlorite solution containing various organic acids. However, acidified sodium chlorite in combination with a mild heat treatment reduced the population by approximately 4.0 log CFU/g without affecting the color, but it softened the leaves. Moreover, the efficacy of the washing treatment was similar at low (4 degrees C) and room (25 degrees C) temperatures, indicating that acidified sodium chloride solution could be useful as a sanitizer for surface washing of fresh produce.

[Morphology of hemolymph cells in cabbage butterfly catterpillars].

Using morphological and cytochemical methods, a comparative analysis of the haemocytes isolated from 5-aged caterpillars of cabbage butterfly was made. The following groups of haemocytes were found in haemolymph: proleucocytes--7%, macronucleocytes--25, micronucleocytes--48, enocytoides--5, phagocytes--15%, and single eosinophil-like cells. The number of haemocytes in each group is closely related to their functional activity. Micronucleocytes, phagocytes and macronucleocytes are most numerous in 5-aged larvae before their pupation. This phenomenon is in connection with pinocytotic activity of micronucleocytes, and with phagocytotic function of phagocytes and macronucleocytes. The liquid media of haemolymph is rich in proteins and polysaccharides.

Cabbage root fly control using non-organophosphorus insecticides.

The need to find non-organophosphorus insecticides to control the cabbage root fly has never been so urgent. Of the six non-OP insecticides tested, fipronil was the most effective but spinosad, diflubenzuron and cyromazine also showed considerable promise. As expected, the transplant drenches (34 mg active ingredient (a.i./plant) were more effective than the module drenches (5 mg a.i./plant), which in turn were more effective than the film-coated seed treatments (0.001 mg a.i./plant), simply because of the different amounts of insecticide applied per plant. One remaining major problem is that, even if effective non-OP insecticides can be found, the manufacturers may still not support their insecticides being applied to minor crops such as field vegetables.

[Effects of IAA and stimulated microgravity on formation of adventitious roots of Chinese cabbage].

Under the induction of indole-3-acetic acid (IAA), adventitious roots were differentiated on hypocotyl segments derived from seedlings of Chinese cabbage (Brassica campestris spp. pekinensis). IAA at concentration of 0.4-1.0 mg/L in solid MS medium incited many adventitious roots on hypocotyl segments. The earliest anatomic changes were observed on cut surface of hypocotyl segments under optical microscope 24 hours after IAA treatment: cytoplasmic and nuclear density became higher in a few of parenchytmatous cells adjacent to phloem in tissue of pericycle, followed by cell divisions. Lately, the dividing cells expanded and developed into root primordium from which root cap was differentiated. After five days, most roots protruded through hypocotyl cortex and appeared just below the cut surface. The rooting capacity of the segments derived from three regions of each hypocotyl was different. High level of IAA modified the polarity of root formation on segment inserted upside down and sucrose increased the function of IAA. Additionally, microgravity did not significantly change the rooting polarity under the condition of stimulated microgravity, but it increased the competence of explants to IAA treatment. The results presented here provided an experimental system for further investigation of molecular events associated with adventitious root initiation.

[Prokaryote expression and western analysis of BcpLH gene of Chinese cabbage].

BcpLH gene preferentially expressed in folding leaf of Chinese cabbage contains dsRNA-binding domains. The cDNA of BcpLH gene was cloned into a His-fusion expression vector pET-28a (+) and was induced to express in E. coli strain BL21 (DE3). Then, the specific protein was partially purified and the rabbit was immunized to prepare the anti-serum. Meanwhile BcpLH cDNA was cloned into the pMAL-c2 containing the solubizing partner, and then the soluble protein generated. It was demonstrated from Western dot assay that the BcpLH protein was specific. The BcpLH active protein and its anti-serum made it possible to study RNA-binding activity and regulation mechanism in plant development.

[Freezing injury of plant and oxidative stress--superoxide generation in freeze-thaw cycles of cabbage leaf tissues].

To test the hypothesis that oxidative stress is involved in inducing freezing injury of plants, nitroblue tetrazolium (NBT) was infiltrated into cabbage leaf tissues before they were frozen. NBT was reduced to formazans in freeze-thaw cycles. The ethanol-extractable mono-formazan was measured by a spectrophotometer, and used as a quantitative indicator of oxidative stress that developed during a freeze-thaw cycle. NBT per se as an oxidant increased the freezing injury slightly. When Me2SO as a cryoprotectant was infiltrated into leaf tissue, it apparently enhanced the freezing tolerance, and this is correlated to its ability in ameliorating oxidative stress, as indicated by the partial inhibition of NBT reduction. The results supported the hypothesis that freezing injury of plants is related to oxidative stress. The experiments indicated that it was possible that the reductant of NBT in the freeze-thaw cycle was superoxide anion free radical.

Evaluation of genetic diversity and uniformity of head cabbage DH lines by the use of RAPD markers.

DH lines derived from cabbage cvs. Kamienna Głowa, Sława z Enkhuizen and Langendijker, representing R1 generation, were analysed by the use of RAPD markers for their diversity and uniformity. For the evaluation of genetic diversity, eight primers yielding informative bands were used. Of the total of 83 RAPD bands scored in this study, 16.9% were polymorphic between a set of 13 DH lines. The similarity of the DH lines, estimated by Jaccard's coefficient, was depicted in the UPGMA dendrogram. Fourteen generated informative RAPD bands allowed the identification of DH lines developed from each cultivar. The evaluation of the uniformity for six closely related DH lines was possible by the use of three primers which generate one or two polymorphic bands. The lack of differences among ten plants of the five investigated DH lines manifested their uniformity. One line showed intraline polymorphism with two RAPD primers. The occurrence of the differences at the molecular level among ten plants indicated that their parental R0 plant was probably obtained from somatic cells, not by androgenesis.

[Field efficacy of emulsifiable suspensions of Beauveria bassiana conidia for control of Myzus persicae population on cabbage].

The conidial powder of Beauveria bassiana SG8702 produced using a diphasic fermentation technology was formulated into emulsifiable suspension I (EI-I, > 10(10) conidia.ml-1) and ES-II, which resulted from adding 1% (W/V) of imidacloprid 10% WP to the ES-I. Three dilutions (10(7), 10(6), and 10(5) conidia.ml-1 of each were sprayed onto cabbage plants for control of the green peach aphid Myzus persicae population in field plots in Kunming, Yunnan during July 2001. Each dilution and a water spray as control were replicated three times. During a 28 d period of sampling at 3 or 4 d intervals, spray of the ES-II at 10(7) conidia.ml-1 controlled the increase of the aphid population by > 90% consistently from day 7 after spray. The efficacy for the ES-I at the same concentration reached 85% on day 7 after spray, exceeded 70% in the following two weeks, but declined to 64.4% on day 24 and 52.6% on day 28. At the lower concentrations sprayed, both suspensions significantly suppressed the increase of M. persicae population, but the ES-II resulted in higher efficacy than the ES-I. Local summer weather with mild temperature and frequent rainy days was favorable to the use of the formulations for aphid control.

An evaluation of some morphological traits in doubled haploid lines and their F1 hybrids of head cabbage Kamienna Głowa in the vegetative phase.

The internal stump length, head mass and head shape of doubled haploid (DH) lines and their F1 hybrids of head cabbage Kamienna Głowa were compared. It was found that the range of variation in the investigated traits of DH lines was higher than that of their F1 hybrids. The head mass of the DH lines indicated some level of inbreeding depression, but their F1 hybrids showed a significant effect of heterosis. Genes responsible for flattened head shape were dominant over rounded shape genes. The longer internal stump trait was dominant over the shorter one.

[LDL-cholesterol-lowering effect of a mixed green vegetable and fruit beverage containing broccoli and cabbage in hypercholesterolemic subjects].

The serum LDL-cholesterol (LDL-C)-lowering effects of two types of canned beverage containing mixed green vegetables and fruits, with or without broccoli and cabbage (B&C), were examined in a randomized double-blind study design. Seventy-seven adults subjects with mild to moderate hypercholesterolemia participated in this study after giving their informed consent. The subjects were randomly divided into two groups. One group(test group) was allocated a test sample, containing B&C as the main materials. Another group(control group) was allocated a placebo sample made from the same materials but without B&C. The subjects were administered 2 cans of the assigned sample (160 g contents/can) per day for 12 weeks. Forty-nine out of 77 subjects, whose LDL-C levels were greater than or equal to 140 mg/dl and less than 180 mg/dl, were analyzed for the effectiveness. Serum LDL-C levels in the test group were significantly(p < 0.05) reduced at 3, 6 and 9 weeks after administration in comparison to the baseline levels (155.7 mg/dl in average). The average LDL-C value at 9 weeks was 142.5 mg/dl and the reduction rate was 8.5%. But serum LDL-C levels in the control group were not significantly reduced. Significant differences(p < 0.05) between the groups were observed in the LDL-C levels at 6 and 9 weeks and also in the total cholesterol levels at 9 week. Thus daily intake of the beverage tested containing B&C are useful for lowering serum LDL-C levels in hypercholesterolemic subjects.

Molecular cloning and characterization of nucleoside diphosphate (NDP) kinases from Chinese cabbage (Brassica campestris).

Nucleoside diphosphate kinases (NDPKs) are key metabolic enzymes that catalyze the synthesis of non-adenosine nucleoside triphosphates (NTP) by transfer of the terminal phosphate between NDP and NTP. Recently we isolated three NDPK cDNAs from Chinese cabbage cDNA library. BcNDK1 has 636 bp and encodes a putative 17.4 kDa protein, BcNDK2 has 854 bp and encodes a putative 25.5 kDa protein, and BcNDK3 is 986 bp long and encodes a putative 25.7 kDa protein. The precursor proteins of BcNDK2 and BcNDK3 have long N-terminal extensions containing putative chloroplast or mitochondrial targeting sequences. A phylogenic tree showed that the 3 BcNDKs are highly homologous to other plant NDPK genes, especially those of Arabidopsis. Expression of the BcNDK genes depended on the developmental stage and the conditions of seed germination. Most notably, expression of BcNDK2 increased dramatically in seedlings transferred to the light after germinating in the dark. In addition, BcNDK3 differed from BcNDK1 in being highly expressed in the hooks and cotyledons of seedlings. Although all BcNDKs were highly expressed in petals, BcNDK1 was also expressed in pistils. Expression of each of the BcNDKs increased as the flower bud matured. These results indicate that NDPKs are involved in physiological pathways activated by a variety of environmental conditions and at different developmental stages.
