Mechanisms of the therapeutic effect of astragalus membranaceus on sodium and water retention in experimental heart failure.

OBJECTIVE: To investigate the therapeutic effect of Astragali on sodium and water retention in aortocaval fistula-caused experimental congestive heart failure and its involved mechanisms. METHODS: In aortocaval fistula-caused chronic (5 wk), heart failure rats treated with and without Astragali 1.0 g/day intraperitoneally, changes of cardiac and renal function, renal response to atrial natriuretic peptide (ANP) were examined. Dot blot analysis was used to determine the effect of Astragali on hypothalamic arginine vasopresin (AVP) mRNA expression, and mRNA expressions of aortic and renal AVP V1a receptor, renal AVP V2 receptor and aquaporin-2 (AQP2) were simultaneously detected by RT-PCR method. RESULTS: Rats with aortocaval fistula impaired cardiac and renal functions evidenced by higher right atrial pressure (RAP), left ventricular end-diastolic pressure (LVEDP), lower + dP/dtmax of left ventricle, glomerular filtration rate (GFR), renal plasma flow (RPF), urine volume (UV), urinary sodium excretion (UNaV) and free water clearance (CH2O) compared with sham-operated control (P < 0.05). There was no change in serum sodium, hematocrit and plasma osmolality. Astragali could remarkably improve the cardiac and renal functions. Dot blot analysis demonstrated upregulated hypothalamic AVP mRNA expression in this experimental heart failure. The AVP V1a receptor mRNA level of aortic arch and renal medulla were reduced, while in renal cortex it was elevated. The mRNA expressions of AVP V2 receptor and AQP2 were increased in renal cortex while decreased in medulla. Astragali could partially or completely correct those abnormal mRNA expressions. Analysis on plasma atrial natriuretic peptide (ANP), urinary cyclic guanidino monophosphate excretion (UcGMP V), urinary cyclic guanidino monophosphate excretion/plasma atrial natriuretic peptide (UcGMP V/pANP), and further correlation and linear regression analysis between UcGMP V and plasma ANP showed that there was blunted renal response to ANP in heart failure rat, and astragali could improve the renal reaction to ANP significantly. CONCLUSION: Chinese herb, astragali have therapeutic effects on sodium and water retention in aortocaval fistula-induced heart failure, the mechanisms of which might be the improvement of cardiac and renal functions, partly correction of abnormal mRNA expressions of AVP system and AQP2, and amelioration of blunted renal response to ANP.

Identification of a gene associated with astragalus and angelica's renal protective effects by silver staining mRNA differential display.

OBJECTIVE: To identify genes associated with the chronic progression of renal disease and a stragalus and angelica (A&A)'s renal protective effects. METHODS: The technique of silver staining mRNA differential display (DD) was used to investigate changes of gene expression in normal, sclerotic and A&A treated sclerotic kidneys. We isolated genes differentially expressed during the progression of renal disease which could be normalized by A&A. RESULTS: Several genes related to A&A's protective effects were isolated and one of them was confirmed by Northern blot. CONCLUSION: Silver staining mRNA differential display is a simple and effective technique for isolating differentially expressed genes. The isolated new gene may be related to the progression of chronic renal disease and contribute to A&A's protective effects.

[Influence of leaf-picking on effective constituents of the Astragalus mongholicus roots].

After the application of the orthogonal design, experiments show that the proportion and time of leaf-picking have a great influence not only on the total content of 17 kinds of amino acids but also on that of Astragaloside. If the proportion is large and the time is delated, both contents of 17 kinds of amino acids and Astragaloside will be reduced accordingly compared to the control group. There is less influence on the contents if the time of leaf-picking is set on June 15 each year and the picking proportion is 1 out 3.

[Change of superficial shape and cell microstructure of milk-vetch (Astragalus L.) root under simulated Cu pollution in a red soil].

This study carried out experiments to investigate changes of shape, inter-structures and cell microstructures of milk-vetch root under simulated Cu stress at non- or contaminated levels in a red soil using observation and bio-microscopic technique. It resulted that when Cu concentration ranged from 0 to 40 mg.kg-1 soil, the milk-vetch grew well and it had a whole root which worked normally. When Cu concentration reached 50 mg.kg-1 soil, the growth of milk-vetch began to get influence with decline of biomass, the taproot crooked and was less branched, root became short and hazel and had fewer shorter hairs, tubby appeared, epidermis began to shrink and cell wall cockled slightly and unevenly, the boundary between plasmalemma and organelle blurred as well. When treatment concentration reaching to 200 mg.kg-1 soil, milk-vetch roots became rotted and black, the cell wall broke and cytoplasm shrank so severely that plasmolysis happened and the plant died. So the critical Cu concentration in experimented soil was 50 mg.kg-1 soil, and the resistance of milk-vetch root to Cu contamination buildup with the growth of aboveground part, and cell wall was the main part to Cu tolerance.
