MTT: a software tool for quality control in sequence assembly.

A large-scale sequencing project requires a tool to control the quality of the input data because a sizable number of trace data may be of low quality. If these data are allowed to enter the sequence assembly pipeline, harm will be done. Hence, it is important to detect such data as soon as possible. MTT (Move-Track-Trim) is a software package analyzing the quality of the lanes. It subjects each lane to a series of tests, and if a lane does not pass all tests, it is flagged as a "bad" lane. The use has a chance to examine both the "good" and the "bad" lanes and reclassify a "bad" lane as "good," or vice versa. Alternatively, the user may decide to retrack the gel or get rid of some lanes altogether. As a by-product of the analysis, MTT performs other useful functions. It trims the lanes and compresses the lane files and moves them to the directories where assembly is carried out. It also generates some useful statistics describing the quality of the gel.

Quality control of mitigation methods for unusually high indoor radon concentrations.

The present study's objective was to control the quality of different mitigation methods for unusually high indoor radon (222Rn) concentrations of up to 274,000 Bq m(-3) in a village (Umhausen, 2,600 inhabitants) in western Tyrol, Austria. Five years after mitigation, five different remedial actions were examined on their quality by means of measuring indoor radon concentrations with charcoal liquid scintillation radon detectors and with a continuously recording AlphaGuard detector. Mitigation method in house 1--a mechanical intake and outlet ventilation system with heat exchanger in the basement, combined with a soil depressurization system--was characterized by long-term stability. With most favorable air pressure (+100 Pa) in the basement, mean basement radon concentrations in the winter were reduced from 200,000 Bq m(-3) to 3,000 Bq m(-3) by this method 5 y after mitigation. Acting against experts' instructions, the inhabitants had switched off the ventilation system most of the time to minimize power consumption although it had been proven that ventilation reduced mean basement radon concentration by a factor of about 3 in the winter and about 15 in the summer. Mitigation method in house 2-soil depressurization with two fans and loops of drainage tubes to withdraw radon from the region below the floor and outside the basement walls, and from soil below that part of the house with no basement-had been the most successful remedial measure until the winter of 1999 (i.e., 6 y after mitigation), when micro-cracks opened and consequently mean basement radon concentration increased from 250 Bq m(-3) to 1,500 Bq m(-3). Measures to block these microcracks and to minimize soil drying are being developed. Five years after mitigation, the remedial method used in house 3--a multilayer floor construction, where a fan was used to suck radon from a layer between bottom slab and floor-reduced winter mean radon concentration from 25,000 Bq m(-3) to 1,200 Bq m(-3), with the ventilation on and the basement door open. Mitigation method in house 4--a basement sealing technique--was unsuccessful with almost identical radon concentrations during all the five years since mitigation had started. Mitigation method in house 5--a waterproof basement technique especially for future homes--reduced mean basement radon concentration below 300 Bq m(-3) and mean ground floor radon concentration below 200 Bq m(-3), which is the Austrian action level for newly constructed buildings. These findings indicate that even in areas with extremely high radon concentrations, effective mitigation of indoor radon can be achieved provided that house-specific long-term, stable mitigation techniques are applied.

Quality control of gene expression in the nucleus.

Proteins are responsible for most cellular and extra-cellular functions. If altered, proteins can loose their normal activity and/or gain new properties. Either way the consequences may be deleterious for the cell and lead to disease at the organism level. Not surprisingly, eukaryotes have evolved mechanisms to recognize abnormal messenger RNAs and prevent them from producing faulty proteins. Protein-encoding genes are transcribed in the nucleus by RNA polymerase II as precursor RNAs that undergo extensive processing before being translocated to the cytoplasm for translation by the ribosomes. This spatial and temporal separation between RNA and protein synthesis offers an immense opportunity for control and regulation. Here we review recent studies that are beginning to unravel how the coupling between transcription, processing and transport of mRNAs contributes to control the quality of gene expression in the nucleus.

[Use of electronic data processing for quality control in perinatal medicine].

Pregnancy, delivery and neonatal period are influenced by many factors, which effect on perinatal and neonatal mortality and morbidity on the other-hand. Numerous parameters are to be recorded and analyzed for control of quality. With conventional technic of data processing this is only possible in large perinatological centers on a restricted scale. Therefore the electronic data processing project "perinatal medicine" was developed for recording important perinatological efficiency parameters. It was introduced in the practice of gynecological and infant hospital in Karl-Marx-Stadt.

[Blood-sugar self control. A means for the diabetic of controlling his metabolic management. Quality control of a battery-run pocket size reflectometer (glucose-meter)].

Home blood glucose monitoring by diabetic patients has recently been advocated as an effective means to improve metabolic control. The Glucocheck apparatus, a pocket-size battery-driven reflectance-meter (in Germany commercially available under the name Glucose-meter), has been evaluated for accuracy and practicability. In 450 blood glucose measurements, the variance between the values obtained using the Glucocheck apparatus and routine clinical laboratory procedures was +/- 11.7%. Especially in the low range of blood glucose concentrations, the Glucocheck method was very reliable. The quantitative precision of the Glucocheck method depends, however, quite considerably on the ability of the patient to use the apparatus correctly. In order to profit from Glucocheck in clinical practice, particular efforts to educate the patients in its use are necessary.

[Experimental studies on the quality control of xiaoluo pills].

Microscopic and physico-chemical methods have been used in the qualitative study of Xiaoluo Pills. The contents of peimine, peiminine and calcium carbonate in these pills have been determined. The proposed method can be used to control the quality of Xiaoluo Pills.

Development of quality control preparations for immunocytochemical assessment of urokinase-type plasminogen activator.

Quality control of immunochemical and/or immunocytochemical analyses warrants constant reproducibility and reliability of assay performance. In this respect, stable reference preparations containing known quantities of the components to be assessed may serve purposes in the quality assessment of antigen expression levels, including those of the plasminogen activation system. Quality control preparations for the immunocytochemical assessment of urokinase-type plasminogen activator (uPA) were developed using different combinations of cultured cell lines (BLM and IF6), each expressing immunochemically well-defined (by enzyme-linked immunosorbent assay[ELISA]) amounts of the respective component. Cytospins and frozen sections cut from sucrose/Tissue-Tek blocks containing these cell lines demonstrated stable and homogeneous expression of uPA. An excellent correlation was found between the immunocytochemical staining results and the data obtained by ELISA. Because these cell lines are available in practically unlimited quantities, large numbers of nearly identical quality control preparations can be made over a long period of time. Therefore, the incorporation of (combinations of) cell lines in cytospins or sucrose/Tissue-Tek blocks represents a simple model system in establishing quality control preparations for immunocytochemical assessment of components of the plasminogen activator system.

[Turning point in quality control in Japan--current status & problems and future issues].

Some authorities suggest that quality control in Japan is now at a turning point. During the 30 years since its introduction in the 1960s, quality control has improved dramatically, and a number of new quality control techniques have been designed and applied to routine examination. In actual practice, however, people tend to become insensitive to abnormalities or to ignore them, causing an increase in the number of abnormal results. This article will review a survey of quality control and describe the current status, problems and future issues in quality assurance.

[Results of a program of quality control in radiodiagnosis].

The results of a program of quality control on about 30 tomographic and fluoroscopic radiological equipments in the Radiological Departments of Spedali Civili (Brescia, Italy), are reported; the physical performances of the beams (kVp, emission, linearity and reproducibility, HVL, focal spot size) and the geometrical conditions (alignment and beam size, thickness of cut for tomography, resolution and homogeneity) have been verified. During one month we have also verified the working conditions for two automatic film processing systems. Three commercial quality control equipments have been employed and tested. Our results recommend a periodic control of physical and geometrical performances (particularly kVp, emission and beam alignment) and a daily control of the working conditions for the film processing systems. The three quality control systems have given results each other comparable.

[Quality control of care in a surgical department. Experience of the Fondation Berganié].

Activity in a surgical department can be evaluated quantitatively, but is should also be assessed qualitatively. One way to control the quality of care is to determine as accurately as possible the incidence of pre- peri- and post-operative complications and to analyse these date in a critical comparative study. This was accomplished in the surgical department of the Fondation Bergonié, Bordeaux during three test periods over the last five years--March 1987 (127 patients), June-July 1989 (276 patients), and June 1991 (147 patients). Results of this analysis cannot validly be compared with those of other departments with different patient recruitment and activity functions. However, this study within a department with regular, homogeneous activity did demonstrate, over a period of several years, a clear reduction in operative mortality, essentially by improved control of infectious complications by the extensive use of prophylactic antibiotic therapy for all surgery with a septic risk or for debilitated patients. Results also demonstrated the low cost effectiveness of routine pre-operative exploratory examinations.

Quality control of biochemical data in the Multiple Risk Factor Intervention Trial: Central Laboratory.

The acquisition of quality biochemical data in the MRFIT was enhanced by writing detailed protocols, instituting strict rules for collecting and shipping specimens, setting high goals for analytical performance, using reliable means to transfer data, employing dedicated and competent analysts, and having a director of the Central Laboratory who believed in and enforced an effective quality control system. A Laboratory Quality Control Subcommittee was formed to work with this director in solving laboratory problems, with the Coordinating Center in conducting appropriate monitoring, with the Quality Control Committee in documenting the quality of data and investigating any laboratory problems, and with other committees of the directors and Program Office in responding to any questions raised about laboratory aspects of the trial. All of these efforts culminated in reliable biochemical data.
