Hormone profile of T3, T4 at the end of pregnancy and during puerperium.

The Authors have studied the T3, T4 and TBG behaviour at the end of pregnancy and after 96 h and 144 h from delivery in 25 pregnant women. The patients have been randomly divided into two groups: the first was studied in basal conditions and the second after administration of enantate testosterone and valerianate estradiol. In the first group T3 and T4 values increased slightly. A higher increase was noticed from the 96th to the 144th h from delivery. In the second group T3 and T4 values were discording.

[Induction of lactation in cattle after short-term hormonal treatment].

In wintertime 7 nonpregnant cows (3 nonlactating and 4 milk secreting less than 3 liter/day) and 3 heifers were treated:day 1 cloprostenol (530 microgram); day 2 progesterone capronate (0,25/Kg); days 3 to 5 progesterone (0,25 mg/Kg)+ estradiol valerianate (0,01 mg/Kg) and reserpine (0,01 mg/Kg); days 6 to 8 progesterone (0,25 mg/Kg) + estradiol (0,2 mg/Kg) and reserpine (0,01 mg/Kg); day 9 betametasone acetate (0,2 mg/Kg) and phosphate (0,55 mg/Kg); days 9 to 16 estradiol (0,0075 mg/Kg). 9 days later the treatment started again. In all the period (5/12-25/2) the animals where machine milked. 2 lactating nonpregnant cows where kept as control. The milk secretion started in nonlactating animals but the peak daily milk yield was 3 liter about only. In lactating animals the yield no increased. The second treatment (and the first in lactating cows) induced secreting less milk with more fat and protein. The low milk yields was associated to a season's negative effect.

HGH behaviour at term of pregnancy and during puerperium.

Human growth hormone (HGH) has been measured in the plasma of 30 subjects at term of pregnancy, at 96 and at 144 h after delivery. The subjects were then selected into three groups: 10 were studied in basal conditions, 10 were given pyridoxine, 10 were given enantate testosterone and valerianate estradiol. In the first group the correlation index (t of Student) was not significant showing the lack of correlation among the tested averages. IN the second group the index of Student was weakly significant (t = 2.36, p less than 0.05). In the third group the Authors found a high representative correlation between the term of pregnancy and the 144th hour after delivery (t = 3.81, p less than 0.01) and between the 96th and 144th hour after delivery (t = 2.95, p less than 0.01).

Transport of various substances through human enamel and dentine.

The penetration of 14C-labelled alcohols (methanol, ethanol, n-butanol), 14C-labelled carbonic acids (formic, acidic, propionic, valerianic, octanoic, malonic, succinic and lactic acid), 14C-drugs (procain, barbital), and 14C-sugars (saccharose, xylose) into about 800 human deciduous or permanent teeth, both healthy and carious, was investigated. Dental enamel up to the cemento-enamel junction was incubated at a pH-value of 5.0 or 6.8 for 1 or 24 hours. For measurement of radioactivity, the dentine of the root was obtained by trepanation. Between intact and carious permanent teeth only slight differences were observed in case of the diffusion of methanol and ethanol (2% of the incubation medium), while n-butanol penetrated the dentine to an extent of 4.2% at a pH of 5.0. The monocarbonic acids penetrated the enamel of healthy teeth within 24 hours to an extent of 6.6-19.2% of the content of the incubation medium, while the dicarbonic (succinic and malonic) acids reached amounts of 3.6 and 9.2%, and the percentage of lactid acid which penetrated the enamel reached 2.9%, respectively. Under all conditions tested, saccharose penetration was higher in carious than in healthy teeth (3.8 vs 6.5%). The highest uptake was found in experiments with barbital; it was more pronounced in deciduous than permanent teeth (16.2 vs 12.4%). The data could be of interest in the therapy of inflammatory and other processes of the pulp.

In vitro study on the interaction of Valeriana officinalis L. extracts and their amino acids on GABAA receptor in rat brain.

This work studied in vitro the interaction of aqueous and hydroalcoholic extracts of Valeriana officinalis L. and compounds that are present in the extracts (amino acids and valerenic acid) with the GABAA (gamma-aminobutyric acid) receptor, using the [3H] muscimol binding technique to crude synaptic membranes from rat brain cortices. Both extracts displaced [3H]muscimol bound and this effect is probably due only to their amino acid content, specially GABA. This fact explains the in vitro effect of valerian extracts on GABAA receptor but not their sedative effect.

NCDEU update. Natural product formulations available in europe for psychotropic indications.

Until the middle of this century, development of medical treatment for human disease was intimately connected with the plant kingdom. Despite advances of the last three decades in utilizing chemical synthetic approaches to drug design and sophisticated structure-activity studies, there is still a great need for novel compounds with unique mechanisms of action in the field of medicine. While many thousands of structural analogs have been synthesized and tested, numerous gaps remain in the therapeutic armamentarium for psychiatric illnesses. Most new drugs marketed for psychotherapeutic indications in recent years have been only incremental improvements on existing medications. Major breakthroughs have resulted primarily from the study of natural products. Some of our most valuable drugs have been isolated from plant and animal sources, including aspirin, morphine, reserpine (the first antipsychotic), almost all of our antibiotics, digitalis, and such anti-cancer agents as vincristine, vinblastine, and taxol. Recent political and social events suggest that new emphasis will be placed on natural products research in the years to come. This article highlights therapeutic applications of Ginkgo biloba, Hypericum perforatum, Valerian officinalis, and Panex ginseng.

[Medicinal plants--concentrators of chromium. The role of chromium in alkaloid metabolism].

Mass screening of medicinal plants of the flora of Russia (196 species) was performed for chromium content. A total of 124 species-chromium concentrators were found, in which the chromium content markedly exceeded the mean values, this excess being 4- to 6-fold in 54 species, 7- to 3-fold in 62 species, and 37- to 114-fold in seven species. The greatest capacity of chromium accumulation was shown for the sand immortelle, foxglove, Alexandrian laurel, Greek valerian, marsh cudweed, adenostilis, and lobelia. These species are considered as potential sources of chromium for correction of its deficiency in humans. Some mechanisms underlying the effect of chromium on metabolism of alkaloids derivative of quinolizidine, tropane, isoquinoline, and indole, were deciphered.

[Comparison of vaginal cytologic effects and blood elimination curves of different oestrogen drugs (author's transl)].

With different oral oestrogens it was studied, if relations are existing between pharmacokinetic and vaginal effect. By means of Karyopycnotic Index and Dynamic Oestrogenicity Index the cytological effect was quantified from a single dose of oestradiol valerianate, mestranol, ethinyloestradiol and 3 depot oestrogens proved on postmenopause women and compared with adequate pharmacocinetical investigations. The courses of oestrogen levels did not correlate with indices. Both methods of investigation do not compensate, but complete one another. Depotoestrogens and short acting oestrogens are distinguishable with regard to proliferation maxium and steepness of index decline.

[Additive treatment of metastasizing breast cancer with special reference to postmenopausal age (results of a randomized study)].

Two therapy groups were formed at random out of 82 patients with progressive carcinoma of the breast. One was treated with estrogens, namely 17-ethinyl 1,3,5-estratriene 3,17 diol (SH 8.1083). The second group was treated with depot preparations consisting of a combination of estrogens and gestagens; these patients received 90 mg estradiol valerianate and 300 mg 17-hydroxy-19-norprogesterone caproate (SH 8.0834) weekly. The estrogen therapy resulted in a remission rate of 48% (complete remission 21.1%, partial remission 26.9%). The remission rate of the combination treatment was 46.6% (complete remission 23.3%, partial remission 23.3%). There was no considerable difference between the remission rates produced by the two therapeutik methods. It is remarkable that much more osseous remissions were found under the combination therapy of estrogens and gestagens. There were no substantial differences between the remission rates of the soft-tissue metastases and the pleuropulmonary metastases. In the estrogen group the remissions took 7.7 months, whereas an average remission period of 8.3 months was observed in the estrogen-gestagen group.

Incorporation of isobutyrate and valerate into cellular plasmalogen by Bacteroides succinogenes.

Wegner, G. H. (University of Wisconsin, Madison) and E. M. Foster. Incorporation of isobutyrate and valerate into cellular plasmalogen by Bacteroides succinogenes. J. Bacteriol. 85:53-61. 1963.-Bacteroides succinogenes was found to require both a branched-chain volatile fatty acid (e.g., isobutyric) and a straight-chain acid (e.g., valeric) for growth. The organism used the acids as precursors for the synthesis of long-chain fatty acids and fatty aldehydes, which in turn were employed in the synthesis of phospholipid, mainly ethanolamine plasmalogen. Isobutyric acid was incorporated primarily into branched-chain C(14) and C(16) acids (tentatively identified as 12-methyl tridecanoic and 14-methyl pentadecanoic acids, respectively), and into fatty aldehydes. Valeric acid was used mainly for the synthesis of n-C(13) and n-C(15) fatty acids and fatty aldehydes. Apparently the two short-chain fatty acids were built up by the addition of two-carbon units to form the long-chain acids and aldehydes of the plasmalogen.
