Effects of R-(+)-limonene on submerged cultures of the terpene transforming basidiomycete Pleurotus sapidus.

The biotransformation of limonene by the basidiomycete Pleurotus sapidus yielded cis/trans-carveol and carvone as the main products. The transformation period was extended from 4 days after direct addition to 12 days by gas phase addition of the substrate. After 2 days of transformation, 97% of the substrate had accumulated in the mycelium, while only 3% were present in the culture medium. Substrate toxicity led to a decrease of dry matter. Adaptation of the precultures with small amounts of substrate doubled the concentration of carveol and increased the concentration of carvone by a factor of 3-4. Total product concentrations of > 100 mg l-1 were reached.

Mechanisms of percutaneous absorption of tamoxifen by terpenes: eugenol, D-limonene and menthone.

The effect of chemical penetration enhancers (e.g., eugenol, d-limonene and menthone in combination with 50% ethanol) on the in vitro percutaneous absorption of tamoxifen through porcine epidermis has been investigated. The above enhancers significantly increased (p<0.05) the permeability coefficient of tamoxifen in comparison with the control (50% ethanol). Fourier transform infrared spectroscopy (FT-IR) was employed to investigate the biophysical changes in the stratum corneum (SC) lipids by the enhancer(s). FT-IR results showed that the treatment of the SC with enhancers did not produce a blue shift in the asymmetric and symmetric C-H stretching peak positions. However, all of the above enhancers showed a decrease in peak heights and areas for both asymmetric and symmetric C-H stretching absorbances in comparison with the untreated SC. A decrease in peak heights and areas is a measure of lipid extraction. Partitioning of tamoxifen to powdered SC from control and enhancer solutions was also determined. FT-IR and partitioning studies reveal that the enhancement in the permeability coefficient of tamoxifen by eugenol and d-limonene is due to lipid extraction and improvement in the partitioning of the drug to the SC. However, menthone enhanced the permeability of tamoxifen by increasing extraction of the SC lipids and not by improving the partitioning of the drug to the SC.

Essential oils analysis. II. Mass spectra identification of terpene and phenylpropane derivatives.

Mass spectra are widely used in order to identify the peaks resulting from a chromatographic separation. The most common approach to solve the problem for unknowns on whom very little other structural information is available is the use of a retrieval algorithm and a reference mass spectra database. The wide variety of mass spectra recorded with different instruments under various experimental conditions can lead to erroneous results. In order to improve the accuracy of the results, we proposed earlier an identification algorithm, which combines the information obtained from both GC and MS fingerprints. This paper presents a new algorithm based on the comparison of the unknown mass spectra with several libraries (including Wiley and NIST) by using reverse and direct search algorithms respectively. The results of the comparisons were quantified with respect to the match quality and the interference compounds. A global match index for the comparison using all the above information was computed and the results were presented as the match probability. This index expresses more accurately the matches between unknown and all the available libraries mass spectra. In order to verify our algorithm, we tried to identify the compounds separated by GC-MSD from different species of Acorus calamus L. (Araceae) essential oils. The probability of the matches increases compared with the quality of matches resulting from Wiley and NIST libraries.

Simultaneous determination of terpene lactones and flavonoid aglycones in Ginkgo biloba by high-performance liquid chromatography with evaporative light scattering detection.

A gradient high performance liquid chromatographic method with evaporative light scattering detection (ELSD) for the simultaneous determination of ginkgolide A, ginkgolide B, ginkgolide C, ginkgolide J, bilobalide, quercetin, kaempferol and isorhamnetin in Ginkgo biloba is described. Samples are analyzed by means of a reverse-phase column (Supelco Discovery C-18) using methanol (containing 0.05% TFA) and water (containing 5% methanol and 0.05% TFA) under gradient conditions as the mobile phase over 35 min. The evaporative light scattering detector (ELSD) used, is set at an evaporating temperature of 61 degrees C and compressed air pressure of 2.9 bar. The detection limits (S/N>3) of the compounds tested are 20-35 ng on the column. The exponential linear calibration curves are observed for all the compounds tested with r(2) more than 0.998. The reproducibility of the method was evaluated by analyzing three sets of controls on 3 consecutive days with RSD% and relative errors (RE%) less than 17.26 and 14.67%.

Secondary organic aerosol production from terpene ozonolysis. 2. Effect of NOx concentration.

We report secondary organic aerosol (SOA) yields from the ozonolysis of alpha-pinene in the presence of NO and NO2. Experimental conditions are characterized by the [VOC]0/ [NOx]0 ratio (ppbC/ppb), which varies from approximately 1 to approximately 300. SOA yield is constant for [VOC]0/[NOx]0 > approximately 15 and decreases dramatically (by more than a factor of 4) as [VOC]0/[NOx]0 decreases. Aerosol production is completely suppressed in the presence of NO for [VOC]0/[NOx]0 < or = 4.5. Fouriertransform IR analysis of filter samples reveals that nitrate-containing species contribute significantly to the total aerosol mass at low [VOC]0/[NOx]0. Yield reduction is a result of the formation of a more volatile product distribution as [VOC]0/[NOx]0 decreases; we propose that the change in the product distribution is driven by changes in the gas-phase chemistry as NOx concentration increases. We also present two-product model parameters to describe aerosol production from the alpha-pinene/0/NOx system under both high- and low-NOx conditions.

Terpene compounds as possible precursors of 1,8-cineole in red grapes and wines.

While the contribution of 1,8-cineole to the aroma of wine has been reported, it is a matter of controversy that the vineyards producing such wines are surrounded by Eucalyptus trees, which may contribute their essence to the grapes. However, experimental information presented in this paper suggests that 1,8-cineole can be produced by chemical transformation of limonene and alpha-terpineol, and this process may be responsible for the occurrence of Eucalyptus-like aroma in Tannat wines from vines not grown in the vicinity of Eucalyptus trees. A mechanism for the chemical transformation of these aroma compounds is proposed.
