An automated dosimetry data-acquisition and analysis system at the LAMPF pion therapy facility.

An automated data-acquisition and analysis system has been developed for dosimetry measurements on the pion therapy beam at the Clinton P. Anderson Meson Physics Facility Biomedical Channel in Los Alamos using a PDP-11/45 computer and CAMAC interface. Initialization, test, and monitor programs allow the user to set the physical limits of scanner travel, test the data lines, calibrate the analog signals for the scanner position, and monitor the analog versus digital values of the scanner position during operation. Data-acquisition programs scan beams in one, two, and three dimensions. Many options are available to the user in selecting the scan parameters and in changing some of these parameters during scanning. Data-analysis programs provide reproduction of stored data, comparison of linear scans, beam profiles along any line of a planar or volume scan, and isodose distributions from any planar scan or from any planar scan or from any plane of a volume scan. Other programs summarize stored data files and search for specific data according to the user's instruments.

A modified life-table analysis system for cohort studies.

A person-years at risk life-table analysis system of computer programs has been developed by the National Institute for Occupational Safety and Health (NIOSH) and is available with detailed documentation. The system was specifically designed to analyze occupational cohort mortality data. These programs require more computer core space and processing time than other available life-table programs. However, the NIOSH programs are advantageous because they include the following: (1) input data editing and modification, (2) mortality rates for 89 cause-of-death categories, (3) assignment of cumulative doses to specific person-years based on either personal or area exposure data, and (4) simultaneous examination of observed and expected deaths by duration of employment (or dose), latency, age, and calendar time.

SCANCANS--an interactive scanning cell analysis system.

This paper describes a computerized, general-purpose, interactive, scanning cell-analysis system developed at the Department of Clinical Cytology of the University Hospital in Uppsala. The system uses a Leitz MPV II scanning photometer, a PDP-8 computer with 24 k of core memory and a Tektronix 4010 graphic display terminal. A command interpreter and overlay structure which makes it simple to add new capabilities to the system is described. Some examples of command strings and the kind of operations the system can perform are included. Finally, the experience gained from using the system and some plans for future extensions are discussed.

Interactive image analysis system to determine the motility and velocity of cyanobacterial filaments.

An interactive image analysis system has been developed to analyse and quantify the percentage of motile filaments and the individual linear velocities of organisms. The technique is based on the "difference" image between two digitized images taken from a time-lapse video recording 80 s apart which is overlaid on the first image. The bright lines in the difference image represent the paths along which the filaments have moved and are measured using a crosshair cursor controlled by the mouse. Even short exposure to solar ultraviolet radiation strongly impairs the motility of the gliding cyanobacterium Phormidium uncinatum, while its velocity is not likewise affected. These effects are not due to either type I (free radical formation) or type II (singlet oxygen production) photodynamic reactions, since specific quenchers and scavengers, indicative of these reactions, failed to be effective.

Detecting differences in crown transparency assessments between countries using the image analysis system CROCO.

Estimates of crown transparency calculated using the semi-automatic image analysis system CROCO were compared with the visual estimates of survey teams from 12 European countries. For each of five European species (Norway spruce (Picea abies (L.) Karst.), silver fir (Abies alba Mill.), Scots pine (Pinus sylvestris L.), beech (Fagus sylvatica L.), oak (Quercus robur L.)) 15 trees were assessed. In 64% of the comparisons, CROCO estimates and the survey teams' scores differed significantly, while differences between countries were significant in 58% of all comparisons. We calculated Spearman's rank correlation and the standard deviation of the differences between CROCO and each country and between pairs of countries. We then computed the medians for CROCO and each country. CROCO and only a few countries had for all species a higher median correlation and lower median standard deviation than the median for the comparison between countries. In addition no country had a consistently higher correlation or lower standard deviation than CROCO. We conclude that CROCO can provide more consistent and less variable estimates of crown transparency than visual assessments and that it can serve as a reference to detect differences in visual transparency assessments between countries over time.

Quantification of human sperm morphology and motility by means of semi-automatic image analysis systems.

Sperm morphology and motility are determined using a semi-automatic image analysis system under opto-manual control. The system consists of a microscope equipped with a drawing-tube, a digitizer-tablet, and a cursor interfaced to a small microcomputer. A light-emitting diode is mounted on the cursor, visible as a bright red pilot-light in the microscopic field. Sperm head and midpiece as well as the pathway of motile spermatozoa are traced with the cursor's red pilot light. The microcomputer calculates area, circumference, and length simultaneously. Motility and sperm density are determined altogether using microchamber technique. In a selected group of male subjects with normal, doubtful, and pathological semen analyses biometrical analyses of sperm morphology and motility were performed. Sperm morphology is best described by the variation of the head size and by the average midpiece width. Motility is best described by % motile spermatozoa and mean velocity determined 1 hour after semen collection. Biometrical semen analyses are superior to subjective evaluations regarding degree of information, objectivity, and reproducibility.

[Systemic analysis of a material substrate for development of brain edema-swelling].

The biological system behavior was analysed by physical parameters: moisture content and density of brain tissue in biological modeling of compression brain edema-swelling (BES). The systemic analysis objectively assessed material substrate of the process pathogenetic mechanisms. Main shifts of such mechanisms exists in the intact brain and start to operate in response to the provoking factors. As a result, the system "brain tissue" rapidly and progressively loses its internal reserves. One can permanently trace the ambi-trends in the system by the factors interhemispheric and intrahemispheric "physical parameters".

Examination of accuracy of sleep stages by means of an automatic sleep analysis system 'Sleep Ukiha'.

We examined the differences between the results of an automatic sleep analysis system and inspection decision. Subjects were 10 males (average age 21.6 years). One section consists of 20 s records. The sections that deviated from the algorithm could not be decided. Each sleep stage decided by automatic analysis was compared with the inspection decision. The agreement ratio of stage 3 was 91.6% in the highest, and followed by stage 2, stage 4, stage W and stage 1. The lowest was 62.5% for movement time. The total agreement ratio was 85.8%. The agreement ratios of the apnea index (AI) and the apnea hypopnea index (AHI) were relatively high, but for types of sleep apnea, agreement ratios require improvement.

Constant topological organization of the coleopteran metamorphosing nervous system: analysis of persistent elements in the nervous system of Tenebrio molitor.

Evidence is provided that the topological organization of the larval neuropil is preserved during metamorphosis of Tenebrio molitor. Constancies in neuronal organization were due in part to persistence of individual neurons whose morphologies were individually followed. It appears that the phenomenologically static situation of the metamorphosing neuropil is achieved by stabilization and regulation due to cellular interactions. Comparisons are made with features of hemimetabolan postembryogenesis.

Cortical and cancellous bone in the human femoral neck: evaluation of an interactive image analysis system.

An interactive image analysis package was developed to examine whole cross-sections from the femoral neck. The package quantifies cortical width (Ct.Wi), cortical porosity (Ct.Po), and proportions of cortical, cancellous bone as a percentage of bone plus marrow area. Segmental analysis was used to quantify circumferential variations in bone distribution within the femoral cross-section. To evaluate reproducibility of data four independent operators analyzed previously prepared femoral neck sections from a 2000 BC population. Differences in total and circumferential distributions of cortical and cancellous bone with respect to gender and age of samples were demonstrated. Reproducibility was assessed using coefficients of variation (CV). Analysis of sections using a variable magnification, giving largest possible image size, rather than a set magnification reduced variation between operators for all measurements. Use of a calculated threshold significantly decreased variation between operators for the proportions of cortical and cancellous bone (p < or = 0.026). Dividing the image into 8 rather than 16 segments also improved reproducibility. There was little agreement between operators in the determination of cortical porosity. The mean CV for the other quantitative indices such as cortical width and proportions of cortical and cancellous bone ranged from 4.87% to 13.52%. The genders showed similar patterns in circumferential distribution of bone. Cortical width was significantly greater in the inferior region compared to the other areas, whereas percent cortical bone was lowest at the superior region. The center of mass (COM) for the younger age group was located anteriorly, whereas in the older samples the COM was located posteriorly of the center of area (p = 0.041). Basic data relating to cortical and cancellous bone of acceptable reproducibility in comparison with current standards in iliac histomorphometry can now be provided at modest cost in operator time and resources.

[Development of an analysis system for X-ray cephalometry by Windows platform].

Here in this paper introduced is an analysis system for X-ray cephalometry developed through Windows platform,which includes three parts: data input, cephalometry analysis & calculation, and display & printing of diagnostic results. It has been applied in orthodontic department of Stomatological Hospital of the Fourth Military Medical University, and it is quite convenient for doctors' cephalometry diagnosis.

[Application of automated image analysis system Allegro-MC for morphometric research].

Automated system of image analysis Allegro-MC is described, including its scheme and concise characteristic. System is used for automated macro- and micromorphometric studies. As the examples for the demonstration of system operation the following micropreparations were used: capillaries of rat uterus, demonstrated by Gomori alkaline phosphatase reaction, cerebral neurons, visualized using NADPH-diaphorase Hope and Vincent method, capillaries as demonstrated by electron microscopy. As a sample of macropreparation, carotid angiogram taken in direct projection, was used. The demonstration is supplemented by vital microscopic study of microvascular reaction in rat small intestinal mesenterium to irradiation with heliumneon laser.

Megakaryocyte ploidy in thrombocytosis: improved microdensitometric measurements with a new image analysis system.

Megakaryocyte (MK) nuclear deoxyribonucleic acid (DNA) content was measured on a new image analysis system. Feulgen-stained bone marrow aspirate smears were analysed from 9 patients with thrombocytosis. Average optical density (OD) of stained nuclei was evaluated by pixel discrimination over 128 grey levels. Projected nuclear area was delineated manually. The product of these two parameters gave an index of nuclear DNA content. Neutrophils were used as 2N reference cells. Reproducibility of OD, nuclear area and their product was excellent for individual cells (CV less than 2.0% for MKs, less than 4.0% for neutrophils). The average CV for DNA content of 18 groups of 10 neutrophils was 5.5% (range 3.0-9.7%). A significant linear regression existed between MK nuclear area and DNA content (p much less than 0.001) for 627 MKs in essential thrombocythaemia (ET) and 346 MKs in reactive thrombocytosis (RT). Compared with RT, more 8N and 64N MKs were seen in ET (p less than 0.05). 128N MKs were unique to ET. Image analysis of MK ploidy may assist the clinical discrimination between primary and secondary thrombocytosis.

[Development and clinical research of computer aided multivariate pattern analysis system (CAMPAS) OV-1 for diagnosis of ovarian carcinoma].

A computer aided multivariate pattern analysis system (CAMPAS) OV-1, which consisted of 10 discriminant functions based on eight tumor markers including CA125, IAP, TPA, LDH, CRP, CEA, amylase and alkaline phosphatase was developed to effectively diagnose patients with epithelial ovarian carcinoma. One hundred twenty-two patents with epithelial ovarian carcinoma and 215 patients with benign ovarian tumors were examined by using CAMPAS OV-1 retrospectively or prospectively. The clinical significance of CAMPAS OV-1 was compared with CA125 alone, and with a combined assay employing the eight tumor markers used in CAMPAS OV-1. The following results were obtained. 1. When CAMPAS OV-1 was applied to patients in which the value for each tumor marker was used to make the discriminant functions, the sensitivity, specificity and accuracy were 84.5%, 97.5% and 91.3%, respectively. The accuracy of CAMPAS OV-1 (91.3%) was significantly better than those of CA125 (80.0%) and combined assay (74.0%) [p < 0.001]. CAMPAS OV-1 showed relatively better sensitivity (63.3%) than CA125 (50.0%) in patients with stage I disease. Also CAMPAS OV-1 showed relatively better sensitivity (85.7%) than both CA125 (64.3%) and combined assay (78.6%) in patients with mucinous type tumors. Furthermore, the specificity of CAMPAS OV-1 (94.4%) was significantly better than those of CA125 (66.7%) and combined assay (55.6%) in patients with endometrial cysts [p < 0.05]. 2. When CAMPAS OV-1 was applied to the patients prospectively, whose values for each tumor marker were not used to make the discriminant functions, the sensitivity, specificity and accuracy were 88.2%, 83.8% and 85.0% respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Model TJ-IV computer-assisted vectorcardiogram analysis system.

During last 16 years we have successfully developed the computer-assisted vectorcardiogram analysis systems: model TJ-I, TJ-II, and TJ-III, but some technical problems remained unresolved, such as the recognition accuracy for vectorcardiograms, measurement of the parameters of complicated QRS waves, the ratio of T loop length to width, and the area of spatial vectors etc. A new system, model TJ-IV was designed to resolve these technical problems. The system was equipped with a 586 computer with a CPU of 120 MHz. Special new low-noise amplifier was employed and C language was used for programming. Three graph recognition techniques were used to enhance the accuracy of VCG recognition. 32 orthogonal electrocardiograms and vectorcardiograms were displayed and printed, and 566 parameters of vectorcardiograms were calculated. Our results with 150 cases showed that the system had high accuracy of graph recognition, and parameter calculation and the results were essentially consistent with those of manipulative methods. We were led to concluded when compared with TJ-III system, the new version has higher accuracy of processing and measurement for vectorcardiograms, is able to process more vectorcardiographic parameters, with higher processing speed.

A genetic analysis system of Burkholderia cepacia: construction of mobilizable transposons and a cloning vector.

A genetic analysis system of Burkholderia cepacia (Bc) was developed which included transposon mutagenesis and complementation of mutation with the cloned genes of interest. To deliver the transposon in this multidrug-resistant microorganism, two plasmids, pKN30 and pKN31, were constructed which contained Tn5 derivatives, Tn5-30Tp and Tn5-31Tp, respectively, carrying KmR and TpR genes. The plasmids have the origin of ColE1 replication and the mobilization gene of RP4. Tn5-31Tp was mobilized to Bc KF1, a strain isolated from a pneumonia patient, by the transfer system of RP4 integrated in the chromosome of Escherichia coli (Ec). Selection with trimethoprim resulted in generation of a number of transposants of Bc KF1. Fourteen protease-deficient mutants were isolated, all of which contained a single transposon marker in the chromosome. Thirteen protease-deficient mutants were also lipase deficient. An Ec-Bc shuttle plasmid, pTS1209, was constructed that consists of oriColE1, oripSa, ApR and CmR genes, and several unique restriction sites for cloning. Plasmid pTS1209 was successfully employed for cloning genes of Bc involved in protease production.
