[An experimental study on the anti-senility effects of shou xing bu zhi].

This paper presents the anti-senility effects of Shou Xing Bu Zhi (SXBZ. made of thirteen herbs: Polygonum multiflorum, Codonopsis silvestris, Astragalus membranaceus, Poria cocos, etc.) in mice. The mice were administered with the SXBZ orally at dosage of 10 g/kg daily for three months. The results showed that the lipofuscin of liver and brain tissues was significantly reduced in both young (1 month old) and adult (11 months) mice. Examination of lipid peroxidation of liver tissue revealed a marked decrease in adult mice. The lipid peroxidation of rat liver homogeneous was obviously inhibited after culturing with drug solutions (5.0 mg/0.1 ml), 37 degrees C for 90 minutes in vitro. In addition, the diminution of hydroxyproline of skin in both young and adult mice was observed. Results of this study indicated that the SXBZ was effective in slowing down aging.

Molluscicidal activity of Solanum aculeatum (family: Solanaceae) berries against Biomphalaria pfeifferi, Bulinus globosus and Lymnaea natalensis.

Aqueous suspensions of powder of sun- or freeze-dried berries of the plant Solanum aculeatum (Family Solanaceae), indigenous in Kenya, were tested for molluscicidal activity against Biomphalaria pfeifferi, Bulinus globosus and Lymnaea natalensis under laboratory conditions. One hundred or 50 mg powder L-1 of sun- or freeze-dried berries killed over 60% of the test B. pfeifferi, Bul. globosus and L. natalensis. Whereas 25 mg L-1 of the sun dried material killed less than 60% of the test snails, similar concentrations of the freeze dried molluscicide produced 60-80% mortality in the snails, under similar conditions. Using L. natalensis as the target snail, it was shown that the freeze dried material was more potent than the freeze-dried berries of S. incanum, S. nigrum or leaves of Polygonum senegalensis (Family Polygonaceae), all present in Kenya, and known to possess molluscicidal properties. The powdered material retained molluscicidal activity even after several months storage at room temperature. These findings suggest that S. aculeatum is a potent plant molluscicide, and has the potential for the control of vectors of schistosomiasis and fascioliasis in Kenya.

Effects of stilbenes on arachidonate metabolism in leukocytes.

The effects of various stilbenes (i.e, 3,4',5-trihydroxystilbene, 3,4',5-trihydroxystilbene 3-O-D-glucoside and 2,3,4',5-tetrahydroxystilbene 2-O-D-glucoside) isolated from the roots of Polygonum species on rat peritoneal polymorphonuclear leukocyte lipoxygenase and cyclooxygenase activities were studied. Resveratrol (3,4',5-trihydroxystilbene) was found to inhibit the 5-lipoxygenase product, 5-HETE, and cyclooxygenase products, HHT and thromboxane B2; its concentrations for 50% inhibition (IC50) were 2.72 +/- 0.262 microM for the leukocyte lipoxygenase product, 5-HETE, 0.683 +/- 0.163 microM for the formations of HHT and 0.810 +/- 0.274 microM for the formation of thromboxane B2. Piceid (3,4',5-trihydroxystilbene 3-O-D-glucoside) and 2,3,4',5-tetrahydroxystilbene 2-O-D-glucoside also inhibited the formation of 5-HETE, HHT and thromboxane B2, although less strongly. Their IC50 values were, respectively, 55.3 +/- 15.3 microM and greater than 1000 microM for the formation of 5-HETE, 196.7 +/- 48.0 microM and 300.0 +/- 10.4 microM for the formation of HHT and 251.7 +/- 24.9 microM and 366.7 +/- 37.1 microM for the formation of thromboxane B2.

Narthecium asiaticum Maxim. Poisoning of grazing cattle: observations on spontaneous and experimental cases.

A total of 39 Holstein cattle were grazed in tracts of wild grassland on account of shortage in pasture grass. Twenty-nine cattle were affected and 26 of them died during a 21-day period. The main signs were depression, anorexia, ascites, and oliguria. There was elevated serum urea nitrogen and sugar and protein in the urine. Pathological examination revealed turbid swelling of the kidney, an increase in the amount of fluid in the body cavity, edema in the perirenal adipose tissue and hemorrhage in various visceral organs and tissues. Histologically, acute tubular necrosis in the kidney, hypoplasia of the erythroblast series in the bone marrow, atrophy and degeneration of the lymphatic tissue and focal necrosis of the liver were observed in many of the cattle. Among cows experimentally fed Narthecium asiaticum Maxim., Polygonum sachalinense Fr. Schum., and Vitis coignetiae Pulliat which were presumed to have been ingested in large amounts by grazing cattle in the field, the cows fed N. asiaticum revealed the clinical, biochemical and pathological changes similar to those noticed in naturally affected cattle. Cows fed P. sachalinense and V. coignetiae showed no distinct systemic symptoms except transient anorexia and hypothermia.

Hypostase, embryonic sac and endosperm in Anethum graveolens L., and in various families.

The presence of pyriform hypostase tissue was investigated before double fertilization till the fusion of free endosperm nuclei in the embryonic sac of A. graveolens. Hypostase a glandular tissue, can secrete its cell contents into the embryonic sac during the development of free endosperm nuclei. Of 78 randomly selected families, 19 display different numbers of species with hypostase. In 15 of these families the hypostase is present together with the Polygonum type of embryonic sac and nuclear type of endosperm.

[Effect of guzhen recipe on glucocorticoid receptor in senile rate thymocyte].

The effects of Guzhen Recipe (GZ) which composed of Polygonum multiflorum, Cistanche deserticole, Rubus chiagii etc, on the number of thymocyte glucocorticoid receptor (GCR) sites, the GCR nuclear translocation rate, and the activity of serum thymic factors etc in senile rats were observed. The results showed: The number of thymocyte GCR sites, the contents of cytoplasmic protein, nuclear RNA and DNA, the thymus weight/body weight ratio and the serum thymic factors in senile rats were significantly decreased, while thymocyte GCR nuclear translocation rate increased obviously than that of young control rats. GZ, however, was capable of improving the above-mentioned changes of thymus in senile rats markedly, suggesting that GZ might weaken the inhibitory effect of glucocorticoid on the thymus by means of suppressing the translocation activity of GCR from cytoplasm to nucleus in senile rats, thus enhance thymus-dependent immune function in senile rats.

[The prognostic implication of determination of activation of leucocyte in severe burns].

Using NBT test, the relationship between the percentage change of activated neutrophils with the survival time of animals after severe burns was observed. The results showed that the NBT positive rate of neutrophils increased markedly after burn, and the NBT positive rate at 5 h postburn was negatively related to the survival time of burned animals. Continuous monitoring of neutrophil indicated that if the NBT positive rate after burn remained in the normal range, or increased initially to high level and then decreased to lower than normal, the survival time was longer. However, if the NBT positive rate increased continuously after burn, the survival time was shorter. Therefore, continuous assay of neutrophil activation with NBT is an important index for the prognosis of severe burns. Lowering of NBT positive rate was observed in rats treated with polygonum cuspidatum, and these rats lived much longer than those without the treatment.

[Research on the name and reality of zishen in ancient and modern].

The variety of Zishen was confused in the past records. Its original plant should be Polygonum bistorta. The crude drugs bearing the name of Zishen are varied presently. Their functions are completely different from what is recorded in "Benjing". Thus it is suggested that the name Zishen be discarded and replaced by "Quanshen".

[Changes in adhesion features of leukocytes in rats with severe burns].

Twenty-four rats were divided into 3 groups: normal controls (8), simple burned (8), burned plus Polygonum cuspidatum (p.c.)-treated (8). Three hours after burn, 3ml of whole blood was drawn to isolate polymorphonuclear neutrophilic leucocytes (PMNs). Confluent monolayer of rat microvessel endothelial cells was grown in a parallel plate of flow chamber. PMNs were driven through the flow chamber under different wall shear stresses. Leukocyte-endothelia adhesion was observed with an Olympus inverted microscope and recorded with a colour microscopic TV set. The adhesive characteristic curve of leukocyte was significantly different between the normal and burned groups. The number of PMNs adhesive to endothelium increased obviously 3 hours after burn. Under 0.196 Pa of wall shear stress, the adhesion rate of PMNs was 81.2 + 4.1% in the burned group and 58.8 + 4.6% in the normal group. The PNMs adhesion rate was decreased to near normal in p.c.-treated group. The average survival time in the burned group was 4.1 + 1.2 hours, much lower than that in the p.c. treated group (33.2 + 15.8 hours). However, in the simple burned group, one rat survived for 24 hours and its PMNs adhesive curve was similar to that of the normal or p.c.-treated groups. The findings indicate that PMNs adhesion features increase apparently after severe burn, which is closely related to the survival rate.

[The use of Chinese traditional medicines to improve impaired immune functions in scald mice].

For the purpose of searching for immunomodulators, this experiment studied the effects of 6 kinds of Chinese traditional herbs on the restoration of the suppressed immune functions in scald mice, including cell-mediated, humoral and non-specific immunity. All control non-treated scald mice showed definite depression of immune functions in various degrees. Polygonum cuspidatum, Taraxacum officinale and Oidenlandia diffusa (wild) roxb showed immunomodulating effects as measured with 3 immunological parameters. But the effects varied according to the dosage of drugs. Rehmannia glutinosa gave definite improving effects on cell-mediated and non-specific immunity, but no significant effect on humoral immunity, while Gui Ling Gao only showed some effect on humoral immunity.

[Effects of Chinese herbs on impaired lymphocyte functions after thermal injury in mice].

Four Chinese herbs were chosen for this study. They were Panax pseudoginseng wall (PPW), Loniconica japonica thunb (LJT), Polygonum cuspidatum (PC) and Jilin ginseng (JG). They were given from the 2nd to the 7th postburn day (PBD). The results showed that the response to antigen signal, the proliferative capacity, the interleukin II production function, and the antibody production ability by lymphocytes in control burned mice were more noticeably reduced than that in the normal mice. The PPW, LJT, PC might restore all the impaired functions in different degrees. The JG showed no effect at all.

Oncogene signal transduction inhibitors from medicinal plants.

Signal transduction is believed to be altered by cellular oncogenes or tumor suppressor genes during the transformation of normal cells into malignant cells. This proposition offers an attractive target for oncogene-based anticancer drug discovery from natural sources. Protein kinases encoded or modulated by oncogenes were used to prescreen the potential antitumor activity of medicinal plants. Protein-tyrosine kinase-directed fractionation and separation of the crude extracts of Polygonum cuspidatum and Koelreuteria henryi have led to the isolation of three different classes of protein-tyrosine kinase inhibitors, anthraquinone, stilbene and flavonoid. The anthraquinone inhibitor, emodin, displayed highly selective activities against src-Her-2/neu and ras-oncogenes.

[Relationship between disturbances of microcirculation and TNF during burn shock].

The changes in microcirculation and the level of plasma TNF in rat at the early stage of burn shock were measured, and the effects of Polygonum cuspidatum (PC) on above mentioned changes were observed. It was shown when the level of plasma TNF was increased, the number of adhesive leukocytes on venule wall was increased and the number of open capillaries were reduced significantly, and there were apparent aggregation of leukocytes and injury in lungs. The level of plasma TNF and the number of adhesive leukocytes were nearly normal, and the degree of aggregation of leukocytes and injury of lungs were attenuated by giving PC. The present study showed that the level of elevated plasma TNF in the early stage of burn shock was directly correlated with the adherence of leukocytes and the closing of capillaries. The administration of PC could apparently inhibit the increase of leukocytes, and at the meantime, attenuate the adherence of the neutrophils and injury of the lung. The results confirmed that the elevation of plasma TNF might be involved in the development of adherence of neutrophils and the disturbances of microcirculation in burn shock.

Uptake, translocation and metabolism of the herbicide florasulam in wheat and broadleaf weeds.

Florasulam is a triazolopyrimidine sulfonanilide post-emergence broadleaf herbicide for use in wheat (Triticum aestivum L.). The selectivity of florasulam to wheat has been determined to be related primarily to a differential rate of metabolism between wheat with a half-life of 2.4 h and broadleaf weeds with half-lives ranging from 19 to >48 h. To a lesser extent, selectivity, at least for the broadleaf weed cleavers (Galium aparine L.), involves uptake differences. Rate of metabolism data were generated using greenhouse-grown plants injected with radiolabelled florasulam and subsequent extraction and processing by high-performance liquid chromatography (HPLC). Structures of metabolites were determined by isolation for nuclear magnetic resonance and liquid chromatography/mass spectrometry. Wheat plants metabolised florasulam by hydroxylation of the aniline ring para to the nitrogen, followed by conjugation to glucose. Metabolism by broadleaf weeds was so slow that isolation of metabolite was not possible, but comparison of HPLC data suggested hydroxylation as the major pathway.

Naturally occurring antifungal agents against Zygosaccharomyces bailii and their synergism.

Polygodial was found to exhibit a fungicidal activity against a food spoilage yeast, Zygosaccharomyces bailii, with the minimum fungicidal concentration (MFC) of 50 microg/mL (0.17 mM). The time-kill curve study showed that polygodial was fungicidal at any growth stage. The primary action of polygodial comes from its ability to disrupt the native membrane-associated function of integral proteins as nonionic surface active agents (surfactants) followed by a decrease in plasma membrane fluidity. The fungicidal activity of polygodial was increased 128-fold in combination with a sublethal amount (equivalent of 1/2 MFC) of anethole and vice versa relative to the fungicidal activity of anethole. The fungicidal activity of sorbic acid was enhanced 512-fold in combination with 1/2 MFC of polygodial. Conversely, the fungicidal activity of polygodial was enhanced 128-fold in combination with 1/2 MFC of sorbic acid.
