Detection of aromatic catabolic gene expression in heterogeneous organic matter used for reduction of volatile organic compounds (VOC) by biofiltration.

A qualitative procedure of purified DNA/RNA co-extraction from complex organic matter, used as biofilter support for removing volatile organic compounds, was set up and applied to detect xylene monooxygenase gene expression by RT-PCR. A DNA/RNA extraction protocol based on a combination of sample lyophilization pre-treatment and CTAB--phenol/chloroform extraction procedure was optimized for the recovery of purified nucleic acids [100-500 ng DNA (10 kb) and 0.5-2 microg of rRNA 16S from 100 mg matrix]. PCR and RT-PCR protocols were established to detect xylene monooxygenase gene expression starting from differentially induced organic matrices obtained by biofiltration technology. This work allowed the microbial degradation activities in heterogeneous organic solid media to be studied and suggests a rapid method to follow specific biological activities during solid and/or semisolid organic substrates biotransformation.

Angiotensin-converting enzyme is present in the subfornical organ and other circumventricular organs of the rat.

Angiotensin-converting enzyme (ACE, kininase II, EC 3.4.15.1) activity was measured by a radiochemical assay in isolated circumventricular organs of the rat. ACE activity was unevenly distributed, with a 100-fold difference between the lowest (subcommisural) and the highest (subfornical organ) activities. Our results suggest that angiotensin II could be locally formed in circumventricular organs and especially in the subfornical organ. The high angiotensin-converting enzyme activity in the subfornical organ could indicate a physiological role of endogenous angiotensin II in this structure.

Olfactory mucosa-expressed organic anion transporter, Oat6, manifests high affinity interactions with odorant organic anions.

We have characterized the expression of organic anion transporter 6, Oat6 (slc22a20), in olfactory mucosa, as well as its interaction with several odorant organic anions. In situ hybridization reveals diffuse Oat6 expression throughout olfactory epithelium, yet olfactory neurons laser-capture microdissected from either the main olfactory epithelium (MOE) or the vomeronasal organ (VNO) did not express Oat6 mRNA. These data suggest that Oat6 is expressed in non-neuronal cells of olfactory tissue, such as epithelial and/or other supporting cells. We next investigated interaction of Oat6 with several small organic anions that have previously been identified as odortype components in mouse urine. We find that each of these compounds, propionate, 2- and 3-methylbutyrate, benzoate, heptanoate, and 2-ethylhexanoate, inhibits Oat6-mediated uptake of a labeled tracer, estrone sulfate, consistent with their being Oat6 substrates. Previously, we noted defects in the renal elimination of odortype and odortype-like molecules in Oat1 knockout mice. The finding that such molecules interact with Oat6 raises the possibility that odorants secreted into the urine through one OAT-mediated mechanism (Eraly et al., JBC 2006) are transported through the olfactory mucosa through another OAT-mediated mechanism. Oat6 might play a direct or indirect role in olfaction, such as modulation of the availability of odorant organic anions at the mucosal surface for presentation to olfactory neurons or facilitation of delivery to a distal site of chemosensation, among other possibilities that we discuss.

Critical organ dosimetry in permanent seed prostate brachytherapy: defining the organs at risk.

PURPOSE: Although permanent seed prostate brachytherapy is associated with a low risk of serious morbidity, proctitis and prolonged irritative and obstructive urinary symptoms may occur. Data are accumulating to help establish thresholds or guidelines for minimizing toxicity, however, no uniform method of defining and calculating the dose to critical organs currently exists. We set out to examine the existing data and propose a uniform method of reporting such that results from different centers can more easily be compared. METHODS AND MATERIALS: In preparation for a panel discussion at the American Brachytherapy Society 2004 Annual Meeting, four members with expertise in prostate dosimetry and critical organ assessment performed a literature search and, supplemented with their clinical experience, formulated a proposal for defining and reporting dose in a standardized fashion to the critical organs for permanent seed prostate brachytherapy. RESULTS: As previously recommended by the American Brachytherapy Society, postimplant dosimetry should be performed on all patients undergoing permanent prostate brachytherapy. The standard imaging for postplan assessment is the CT scan. The interval between seed implantation and postplan assessment should be reported. For rectal and urinary morbidities, the critical organs are considered to be the anterior rectum and the prostatic urethra, respectively. For erectile dysfunction, both the neurovascular bundle and penile bulb have been implicated. The rectum should be contoured on all CT scan slices where radioactive seeds are visible. Both the inner and outer walls should be contoured. The dose should be reported as RV100 and RV150, the volumes in cubic centimeters of the rectal wall receiving 100% and 150% of the prescribed dose, respectively. The urethra should be contoured as a structure on each slice where seeds can be seen. The urethra should be identified by either catheterization or fusion with transrectal ultrasound. The dose should be reported as UrD5 and UrD30, which are, respectively, the dose to 5% and 30% of the urethra in Gray. As well, a UrV150 should be reported, which is the volume in cubic centimeters of the urethra receiving 150% of the prescribed dose. No recommendations can be made at this time for reporting neurovascular bundle or penile bulb doses. CONCLUSIONS: It is essential that toxicity data be collected and reported in a uniform fashion. Thus, the critical organs for toxicity must be defined and the corresponding dosimetry reported in a standard fashion such that guidelines can be established in the future based on data from a cross-section of centers.

Regulation by protein kinase C of organic anion transport driven by rat organic anion transporter 3 (rOAT3).

The organic anion transporter 3 (rOAT3) is a multispecific OAT localized at the basolateral membrane of the proximal tubule. The purpose of this study was to elucidate the role of protein kinase C (PKC) in the regulation of organic anion transport driven by rOAT3 and its mechanism of action. For this purpose, we established and utilized cells derived from the second segment of proximal tubule from mice stably expressing rOAT3 (S2 rOAT3). Phorbol 12-myristate 13-acetate (PMA), a PKC stimulator, attenuated the cellular uptake of estrone sulfate (ES), a prototype organic anion for rOAT3, in a dose- and time-dependent manner. PMA treatment resulted in a decrease in the Vmax, but not the Km of uptake of ES in S2 rOAT3. Treatment of S2 rOAT3 with other PKC stimulators or diacylglycerols also inhibited the uptake of ES, whereas that with an inactive phorbol ester did not. Chelerythrine chloride, a PKC inhibitor, reversed the PMA-induced decrease in uptake of ES in S2 rOAT3. These results suggest that PKC activation downregulates rOAT3-mediated organic anion transport. This down-regulation may be due to the inhibition of translocation or internalization of the rOAT3 protein, resulting in the decrease in the Vmax of rOAT3-mediated organic anion transport.

Organic reactions without organic solvents and oils and fats as renewable raw materials for the chemical industry.

The use of organic solvents should be minimized as far as possible to reduce atmospheric volatile organic compounds (VOCs). Examples of solvent-free organic syntheses are described. The increasing usage of renewable feedstocks taking advantage of the synthetic potential of nature is another way to avoid organic solvents. Most important is the development of chemical products, i.e., coatings that can be processed without organic solvents.

Characterisation of organic matter from anaerobic digestion of organic waste by aerobic microbial activity.

The aim of this study was to examine whether the characterisation of organic matter on the basis of an oxygen uptake rate (OUR) could be applied to organic waste from an anaerobic waste treatment process. Three anaerobic digestion experiments were carried out in a bioreactor. Volatile fatty acids (VFA) and dissolved organic carbon (DOC) were monitored. OUR-experiments were carried out with diluted samples from the process. The graphs of the OUR-experiments showed a clear lag-phase, which was due to the slow adaptation of aerobic microorganisms. Model simulations of the OUR versus time curve showed sufficient agreement, if based on one fraction of readily biodegradable and two fractions of less easily biodegradable organic matter. The shape of the simulated graphs was affected considerably by the value of the maintenance energy requirement rate qm and could be improved by reducing the standard value qm = 1 d(-1) to qm = 0.1 d(-1). Only little agreement was achieved when comparing the results of the OUR-experiments with the VFA- and DOC-concentrations. Experiments with additional trace elements and vitamins led to an increase in the OUR and proved that the oxygen consumption was not exclusively determined by the availability of organic matter.

Quality of life. Organ transplantation and artificial organs.

Changes in quality of life for patients who require transplantation or permanent artificial organ support begin with the onset of serious organ dysfunction. These changes, which initially are almost universally negative, greatly affect close family members as well as the patient. To the extent that alternate therapies are available, transplantation appears preferable to chronic artificial organ support in allowing the patient to return to work, to school, or to similar self-selected activities. There is some evidence that transplant and artificial organ patients may describe their subjective well-being more positively than would be anticipated from objective data. There is also evidence, at least for hemodialysis patients, that this satisfaction is time-limited. The right to choose death over life dependent on a machine is being exercised by some alert patients as well as by family members and attending physicians when the patient is not competent. Little data is available regarding the long-range impact of transplantation or artificial organ support on members of the patients' families, but in the author's experience one characteristic predominates. To the extent that the life-extending therapy can be forgotten, or is non-intrusive, for days or weeks at a time, as can be the case with transplantation, family functioning returns to normal. The adjustments needed to cope successfully with the stresses of the acute phase are completed. To the extent that reminders and uncertainties of the therapy are chronically present, family interaction continues to center upon (or is designed to avoid) the patient. The family's resilience and capacity to maintain its integrity diminish with each new stressful event.

Hydration of natural organic matter: effect on sorption of organic compounds by humin and humic acid fractions vs original peat material.

Natural organic matter (NOM) hydration is found to change activity-based sorption of test organic compounds by as much as 2-3 orders of magnitude, depending on the compound and the specific NOM sorbent. This is demonstrated for sorption on humin, humic acid, and the NOM source material. Hydration assistance in organic compound sorption correlates with the ability of the sorbate to interact strongly with hydrated sorbents, demonstrating the important role of noncovalent polar links in organizing the sorbent structure. Differences in hydration effect between the sorbents are caused mainly by differences in compound-sorbent interactions in the dry state. For a given compound, hydration of the sorbent tends to equalize the sorption capability of the three sorbents. No correlation was found between the strength of sorbate-sorbent interactions or the type of sorbate functional groups and the extent of sorption nonlinearity. Sorption nonlinearity compared over the same sorbed concentration range is greater on the original NOM than on either of the two extracted fractions. In elucidating sorption mechanisms on hydrated NOM, it is important to explicitly consider the participation of water molecules in organic compound interactions in the NOM phase.

Polyparameter linear free energy relationships for estimating the equilibrium partition of organic compounds between water and the natural organic matter in soils and sediments.

Values of the organic-carbon-based partition coefficient (Koc) have often been estimated using one-parameter linear free energy relationships (op-LFERs), which include both correlations between log Koc and log Kow, where Kow is the octanol-water partition coefficient, and op-LFERs that are based on first-order molecular connectivity indices. For chemicals with tendencies toward strong hydrogen-bonding or other specific interactions with the organic phase, however, these methods are notsufficientlyaccurate. Polyparameter LFERs (pp-LFERs) address these shortcomings by explicitly considering contributions toward free energy change from multiple kinds of molecular interactions with both water and bulk organic phases. This paper reviews pp-LFER theory and presents the development of a new pp-LFER for organic chemical partitioning with soil/sediment organic matter (SOM) using a data set of 356 carefully selected experimental values of log Koc for 75 chemicals, including apolar, monopolar, and bipolar compounds. The paradigm of representing SOM by a single pp-LFER is qualitatively supported by our results, and the set of coefficients for the regression log Koc= (1.10+/-0.10)E - (0.72+/-0.14)S + (0.15+/-0.15)A - (1.98+/-0.14)B + (2.28+/-0.14)V + (0.14+/-0.10) represents a proposed set of water-SOM-specific properties for estimating log Koc. The developed correlation outperformed other currently recommended approaches with the given Koc data set and also compared favorably against the use of new multiple class-specific op-LFER regressions. Overall, the pp-LFER approach is recommended over other current methods for the purpose of Koc estimation and especially for polar chemicals.

Impaired organic anion transport in kidney and choroid plexus of organic anion transporter 3 (Oat3 (Slc22a8)) knockout mice.

To begin to develop in vivo model systems for the assessment of the contributions of specific organic anion transporter (OAT) family members to detoxification, development, and disease, we carried out a targeted disruption of the murine organic anion transporter 3 (Oat3) gene. Surviving Oat3(-/-) animals appear healthy, are fertile, and do not exhibit any gross morphological tissue abnormalities. No Oat3 mRNA expression was detected in kidney, liver, or choroid plexus (CP) of Oat3(-/-) mice. A distinct phenotype manifested by a substantial loss of organic anion transport capacity in kidney and CP was identified. Uptake sensitive to inhibition by bromosulfophthalein or probenecid was observed for taurocholate, estrone sulfate, and para-aminohippurate in renal slices from wild-type mice, whereas in Oat3(-/-) animals transport of these substances was greatly reduced. No discernable differences in uptake were observed between hepatic slices from wild-type and Oat3(-/-) littermates, suggesting Oat3 does not play a major role in hepatic organic anion uptake. Cellular accumulation of fluorescein was reduced by approximately 75% in CP from Oat3(-/-) mice. However, capillary accumulation of fluorescein-methotrexate was unchanged, indicating the effects of Oat3 loss are restricted to the entry step and that Oat3 is localized to the apical membrane of CP. These data indicate a key role for Oat3 in systemic detoxification and in control of the organic anion distribution in cerebrospinal fluid.

Development and organization of a multiple organ transplantation program.

Multiple organ transplantation has come of age. Indications are that it will continue to grow, if not flourish. The complexity of modern surgical care, its multiperson dependency, and the need for the surgeon to retain knowledge and involvement with his patient's care and problems are nowhere more evident than in multiple organ transplantation. Each organ presents its own associated challenges, the prime solution of which lies with a skillful and dedicated surgeon; but with all organs there are challenges of expectantly waiting patients, housing during the wait and for postoperative observation, procurement, erratic scheduling of the operating room, nursing, social service, immunosuppression, immunopathology, interest of hospital public relations and of the news media, and perennial care. This report concerns the growth and development of the multiple organ transplant program at the University Health Center of Pittsburgh and describes some answers to the challenges presented.

Analysis of organ procurement failure at an urban trauma center and the impact of HIV on organ procurement at a regional transplantation center.

A 42-month experience with 100 patients with fatal head injuries was analyzed to identify areas of organ procurement failure. Thirty-six patients were ineligible for organ donation. Reasons for exclusion included advanced age (7), sepsis (16), hepatitis (1), systemic illnesses (3), and HIV infection or risk (9). Resuscitation failure (17 patients) and late deaths from failed support (16 patients) left 31 potential donors. Of the 30 families asked to donate, 17 consented (56.7%). Annual consent rates were 25%, 71%, 75%, and 67%. Efforts to improve organ procurement should focus on resuscitation and physiologic support of potential donors. To assess the impact of HIV infection or risk on organ procurement, a 3-year experience of the regional transplantation center (RTP) was reviewed. Of 1,714 referrals to the RTP from 102 hospitals, 1,120 were from trauma centers. The incidence of rejection because of HIV risk or infection was significantly higher in the trauma center group than in the group from non-trauma centers, 17.2% versus 10.2% (p less than 0.004). A similar difference was noted between metropolitan and suburban hospitals (p less than 0.0001). Hepatitis risk was comparable, 3.9% vs. 3.2%. The risk of HIV infection is emerging as a factor limiting organ donation at urban trauma centers.

Using a standardized donor ratio to assess the performance of organ procurement organizations.

OBJECTIVE: To develop a Standardized Donor Ratio (SDR) as an outcome measure for evaluating the effectiveness of organ procurement organizations (OPOs). DATA SOURCES/STUDY SETTING: All deaths by cause in the United States during 1993-1994 as reported in the Vital Mortality Statistics, Multiple Cause of Death files. The OPO-specific data were provided by the United Network for Organ Sharing (UNOS). STUDY DESIGN: Each OPO's expected number of donors was calculated by applying national donation rates to deaths with potential for donation in 24 age, sex, and race cells. The SDR was calculated by dividing the observed number of donors by the expected number. The chi2 tests of the hypothesis that the OPO's performance differed from the national norm of 1.0 were performed. The SDR was compared to the existing performance standard based on the unadjusted number of donors per million live population in the OPO's service area. An ordinary least squares (OLS) regression assessed predictors of the SDR. PRINCIPAL FINDINGS: The SDRs ranged from 0.41 to 1.99. Twenty-nine of 64 OPOs had SDRs significantly different than 1.0. The SDRs were positively associated with the percent of white living population and the number of organ types transplanted per transplant center served by the OPO. CONCLUSIONS: The SDRs can be used by Centers for Medicare and Medicaid Services (CMS), UNOS, and OPOs to target quality improvement initiatives, present more accurate comparisons of OPO performance, and develop public policy on the evaluation of the effectiveness of organ procurement efforts.

Role of intermolecular disulfide bonds of the organic solvent-stable PST-01 protease in its organic solvent stability.

The PST-01 protease is secreted by the organic solvent-tolerant microorganism Pseudomonas aeruginosa PST-01 and is stable in the presence of various organic solvents. Therefore, the PST-01 strain and the PST-01 protease are very useful for fermentation and reactions in the presence of organic solvents, respectively. The organic solvent-stable PST-01 protease has two disulfide bonds (between Cys-30 and Cys-58 and between Cys-270 and Cys-297) in its molecule. Mutant PST-01 proteases in which one or both of the disulfide bonds were deleted were constructed by site-directed mutagenesis, and the effect of the disulfide bonds on the activity and the various stabilities was investigated. The disulfide bond between Cys-270 and Cys-297 in the PST-01 protease was found to be essential for its activity. The disulfide bond between Cys-30 and Cys-58 played an important role in the organic solvent stability of the PST-01 protease.

Is it ethical to prioritize patients for organ allocation according to their values about organ donation?

Because the supply of deceased donor organs fails to meet demand, patients needing a transplant frequently have lengthy waits or die while waiting. In an effort to reduce waiting times, the concept of "preferred status" has emerged. In the United States, preferred status has taken the form of a community of individuals called LifeSharers. Using directed donation, this group aims to facilitate priority organ allocation to its members-people who have agreed to be organ donors. Such preferred status programs increase societal awareness about organ donation and transplantation, but they are not without ethical controversy, as some term them "clubs." In the case of LifeSharers, the potential to increase the pool of deceased donor organs is a worthy goal that would benefit the community of patients awaiting transplantation, not just LifeSharers members.

Influence of lacquer thinner and some organic solvents on reproductive and accessory reproductive organs in the male rat.

The effects of thinner and its main components, toluene, xylene, methanol, and ethyl acetate, on reproductive and accessory reproductive organs in male rats were studied. The vapour from these organic solvents was inhaled twice a day for 7 d. Following inhalation of thinner vapour for 7 d, the weights of the testes and prostate fell and acid phosphatase activity in the prostate and plasma testosterone levels were significantly decreased compared with the control group. Both ethyl acetate and xylene caused a decrease in the weight of the testes and accessory reproductive organs, as well as reducing acid phosphatase activity in the prostate and plasma testosterone levels. In contrast, toluene and methanol had no effect on organ weights, circulating testosterone levels, or on enzyme activity. Body weight was decreased by inhalation of thinner or ethyl acetate vapour. Spermatozoa levels in the epididymis were decreased by inhalation of ethyl acetate and xylene vapour. These results suggest that thinner, particularly the components ethyl acetate and xylene, interfere with the functions of the testes and accessory reproductive organ; toluene has no effect on these functions.

Distinctive sorption mechanisms of soil organic matter and mineral components as elucidated by organic vapor uptake kinetics.

Sorption kinetics and capacities of volatile organic compounds (VOCs) affect the remediation and fate of these pollutants in soils. The soil organic-mineral compositional heterogeneity complicates the transport and fate of VOCs in soils. The sorption kinetics of toluene vapor with two common soil components, kaolinite and humic acid, shows two distinct sorption patterns. Results with kaolinite are characteristic of surface adsorption, whereas results with humic acid are characteristic of solvation and partition effects. On soils, the kinetics of toluene vapor sorption show a two-stage sorption phenomenon. The first stage is reflective of surface adsorption (1-4 h to completion) and the second stage of much slower partitioning into soil organic matter, which was preceded by a lag phase (approximately 4 h) and took as long as 15 h for completion. The relative contributions of these two stages to soil uptake are quantifiable by two independent parameters, the soil organic fraction and the surface area. A better understanding of the effect of soil compositional heterogeneity on sorption kinetics and capacities facilitates our understanding of the prediction for the fate of organic contaminants in the environment.
