We investigated the influence of butyl 3-(1H-tetrazol-5-yl) oxanilate (MTB) on the release of histamine and slow reacting substance of anaphylaxis (SRS-A) in vitro. MTB dose-dependently inhibited the release of not only histamine but also SRS-A from passively sensitized guinea-pig lung, while disodium cromoglycate (DSCG) hardly affected either release. MTB also resulted in a dose-dependent inhibition of the release of these mediators from the passively sensitized cynomolgus and rhesus monkey and that from human lung at concentrations similar to those inhibiting the release in the guinea-pig. Relatively lower inhibitory activities on the releases of both mediators from the cynomolgus monkey and human lung, but no effects on those from the rhesus monkey were observed with DSCG. MTB dose-dependently inhibited only SRS release from guinea-pig lung induced by phospholipase A2, although the compound did not show any inhibitory activity on the release of those from the calcium ionophore (A23187)-stimulated one. On the other hand, the release of SRS, but not that of histamine from the lung stimulated with A23187 as well as phospholipase A2 was inhibited by N-(3,4-dimethoxycinnamoyl)-anthranilic acid (tranilast, N-5'). From these results, MTB was a potent inhibitor of the anaphylactic release of the mediators, particularly SRS-A. It was suggested that the inhibitory mechanism of MTB is different from those of DSCG and N-5'.
INTRODUCTION: The available clinical guidelines have been successful in improving awareness of the inflammatory nature of asthma and have promoted the use of inhaled corticosteroids (ICSs) to achieve long-term control of symptoms. Because of lingering concerns over the possible adverse consequences of ICS use, an expert panel was convened with a mandate to identify the critical questions that impact decisions regarding the use of ICSs and to evaluate the available evidence with respect to risk. METHODS: A university librarian retrieved citations and abstracts from the MEDLINE and EMBASE databases using a list of National Library of Medicine search terms and key words. Reviewers were asked to systematically abstract relevant information from each of their assigned articles and to list their own clinical or scientific conclusions based on the study results. A predefined grading algorithm was used to calculate a summary quality rating score for the relevant evidence. RESULTS: The results are presented as a series of key questions followed by a summary of the relevant evidence. An evidence grade is assigned, followed by a summary statement reflecting the panel's consensus opinion following review. CONCLUSIONS: The preponderance of evidence supports a conclusion that the proven clinical effectiveness of ICS treatment decidedly outweighs the proven risks.

Cells in spleens from tumor bearing animals were found to inhibit in vitro reactivity of lymphocytes to a variety of stimulants. These suppressor cells inhibited the stimulation of normal lymphocytes by mitogens : P.H.A.-Con A-L.P.S. They were adherent cells, bore immunoglobulins at their surface and did not bear the 0 marker. They were also demonstrated in T deprived tumor bearing animals. They might be B cells, or, more probably, macrophages. Other activities that are inhibited by the suppressor cells are: -reactivity to allogenic cells in the mixed lymphocyte culture, -in vitro generation of secondary anti tumor cytotoxic effector cells, -macrophage migration inhibition in the presence of immune lymphocytes and tumor extracts, -reactivity against syngeneic tumor cells or tumor extracts. Suppressor cells may play a role in the general immunodepression of tumor bearing hosts, and in the inability of tumor bearing hosts to reject the tumor. However, the biological role of these non T suppressor cells is not clear, as they also inhibit tumor cell growth, and in vivo experiments favour suppressor cells of thymic origin.
In the quest for the elusive cure of human neoplasms, considerable effort has been expended over the course of the past 10 years to define the immune system as it relates to the development and evolution of cancer in man. At this time thera is ample evidence for, but incomplete of, the existence of a multifactorial immunosurveillance system that is important in the production and control of neoplasms. The framework for this system has been defined and its modification, with the development and progression of various neoplasms, has been studied with great detail. These studies have been undertaken with the hope of harnessing immune mechanisms and altering their response in favor of the host's defenses related to the evolving tumor. This paper will review the major principles and concepts that have evolved.
