Electric birefringence and circular dichroism measurements have been made on solutions of two po!y (L-lysine) homologs. The specific Kerr constant and the molar ellipticity at 222 nm of poly (L-alpha, gamma-diaminobutyric acid hydrochloride) in methanol/water mixtures underwent an abrupt change between 75 and 80 vol% methanol at 25 degrees C, corresponding to a solvent-induced helix-coil transition. On the helix side of the transition region, i.e., between 78 and 80 vol% methanol, anomalous birefringence transients indicative of field-induced helix-to-coil transition were observed at high fields. In the case of poly (L-ornithine hydrobromide) in methanol/water mixtures, a helix-coil transition was induced between 93 and 98 vol% methanol and anomalous birefringence transients were observed between 96 and 98 vol% methanol. The double logarithmic plots of the steady-state specific birefringence versus the square of field strength for various solvent compositions and polymer concentrations could be superimposed on one another by horizontal and vertical shifts, except for the range where anomalous birefringence transients were observed. This enabled us to estimate the threshold field strength.
In order to test the diffuse ion atmosphere polarization model recently developed by us, the effects of ionic strength, titrating with Mg2+ and Co(NH3)3+6, and coion charge on the electric polarizability of short fragments of DNA are investigated. The results are consistent with the predictions of the theory and show that the diffuse ion atmosphere polarization contributes significantly to the overall orientation of DNA. At low ionic strengths, we attempt to separate the total dipole moment into two components: one that agrees well with the Debye-Hückel ion atmosphere calculations, while the other, presumably due to condensed counterion polarization, appears to be substantially independent of the ionic strength. At higher salt concentrations, however, a simple separation into dipole components is not possible, perhaps due to a significant coupling of ion flows between the diffuse atmosphere and the condensed counterion layer.
Lac repressor and its tryptic core have been investigated by electro-optical methods. The reduced dichroism measured as a function of the electric field strength is not consistent with an induced dipole, but indicates the existence of a strong permanent dipole moment (approximately 4 X 10(-27) C m) for the holo-repressor, which is almost independent of ion concentration and pH. A dominant contribution of a permanent dipole is also demonstrated by the shape of the dichroism rise curve. The experimental data are not consistent with a counterion polarization phenomenon and also do not indicate a major contribution from proton fluctuations. Probably the nature of the dipole is similar to that found for compounds with a tetrahedral substitution by angular residues. Other potential models involve large conformational fluctuations or inherent asymmetry of the lac repressor. Rotation time constants obtained from the dichroism decay are not consistent with a spherical shape, for either the holo- or core repressor. A simple interpretation of the data by prolate ellipsoids suggests a short diameter of 6 nm for both holo- and core repressor and long diameters of 14 and 12 nm for holo- and core repressor, respectively. Addition of the inducer isopropyl-beta-D-thiogalactopyranoside leads to a change of the limit dichroism, but does not affect the rotation time constants within experimental accuracy.
The impedence of sheep Purkinje strands, measured to 3-5 kHz, is interpreted with circuit models based on morphology. The strand is described as a one-dimensional electrical cable. Clefts between myocytes of the strand allow radial current to flow in parallel with current across the outer membrane. A lumped model of the clefts, in which all the cleft membrane is in series with 100 omega-cm2, fits only below 20 Hz. Two distributed models, pie and disk, fit at all frequencies with somewhat different (31%) luminal resistivities, but with similar membrane parameters. Series resistance representing the endothelial sheath is small. Simulations of voltage clamp experiments include measured linear parameters and nonlinear membrane channels, as well as radial variation of cleft concentration, membrane flux, voltage, and current. Cleft potential is drastically nonuniform when sodium current flows. Cleft potential is reasonably uniform when calcium and potassium currents flow, but the calcium and potassium concentrations change markedly, enough to turn off the calcium current, even if the calcium channel did not inactivate. We conclude that physiological current flows produce significant nonuniformities in electrochemical potentials in the clefts of this cardiac preparation.
The structure and charge distribution of T-vimentin, which differs from the intact intermediate filament protein vimentin through the absence of the first 70 amino acids, has been studied by transient electric birefringence measurements. It is found that in 0.7 mM phosphate, pH 7.5 buffer, exclusively single dimers (with a hydrodynamic length of 40 to 43 nm) are present, which are considerably bent and/or flexible and which have a relatively large permanent dipole moment. This indicates a parallel alignment of two protein chains. In 0.2 mM phosphate, 0.5 mM MgCl2, pH 7.5, predominantly tetrameric T-vimentin is found with a rigid structure, no permanent dipole moment, and a length of 63 to 68 nm. Tetramer formation is likely to be induced by binding of Mg2+ to the protein. The observed length is in agreement with that of intact vimentin tetramers in which the 1B regions of the rod domains of the dimers overlap (A11 configuration). A minor part of the tetramers may be in a flexible or bent A22 form. The loss of the permanent dipole moment when tetramers are formed is, apart from charge compensation, presumably due to the antiparallel alignment of the constituting dimers in which their dipoles cancel.
Twenty-four patients with self-reported "sensitivity to electricity" were divided into two groups and tested in a double-blind provocation study. These patients, who reported increased skin symptoms when exposed to electromagnetic fields, were compared with 12 age- and sex-matched controls. Both groups were exposed to 30-minute periods of high or low stress situations, with and without simultaneous exposure to electromagnetic fields from a visual display unit. The matched controls were tested twice and given the same exposure as the patients but had the fields turned on every time. Stress was induced by requiring the participants to act in accordance with a random sequence of flashing lights while simultaneously solving complicated mathematical problems. Blood samples were analyzed for levels of the stress-related hormones melatonin, prolactin, adrenocorticotrophic hormone, neuropeptide Y, and growth hormone, and the expression of different peptides, cellular markers, and cytokines (somatostatin, CD1, factor XIIIa, and tumor necrosis factor-alpha). Skin biopsies were also analyzed for the occurrence of mast cells. Stress provocation resulted in feelings of more intense mental stress and elevated heart rate. The patients reported increased skin symptoms when they knew or believed that the electromagnetic field was turned on. With the blind conditions there were no differences between "on" or "off." Inflammatory mediators and mast cells in the skin were not affected by the stress exposure or by exposure to electromagnetic fields. The main conclusion was that the patients did not react to the fields.
Fast electron generation and propagation were studied in the interaction of a green laser with solids. The experiment, carried out with the LULI TW laser (350 fs, 15 J), used K(alpha) emission from buried fluorescent layers to measure electron transport. Results for conductors (Al) and insulators (plastic) are compared with simulations: in plastic, inhibition in the propagation of fast electrons is observed, due to electric fields which become the dominant factor in electron transport.
A study based on electrical impedance analysis of tissue properties was conducted in order to understand the relationship between impedance components and ethylene biosynthesis induced by direct current in cucumber (Cucumis sativus L.) fruit. Impedances were measured at a range of alternating current frequencies from 500 Hz to 1 MHz. We calculated capacitances representing the plasma membranes, C1, and organelle membranes, C2, and resistances representing extracellular space, R1, cytoplasm, R2, and organelle interior, R3. Direct current of 1 to 3 mA induced ethylene synthesis with a sharp peak at 1 h. The rate of production was greater with a stronger current. This abrupt induction of ethylene synthesis was accompanied by an equally abrupt activation of 1-aminocyclopropane-1-carboxylic acid (ACC) synthase within 1 h, but not that of ACC oxidase, which was activated only at the later stages of the treatment at a time when ethylene production and ACC synthase activity were declining. Using direct current of 0 to 3 mA, C2, R1, and R2 increased abruptly, and C1 increased gradually after 3 h. The rates of increases were greater with currents of larger magnitude, R3 was not affected during passage of the current. Diazocyclopentadiene, an inhibitor of ethylene action, eliminated the direct current induction of R1 but had no effect on the increases in C2 and R2. Diazocyclopentadiene counteracted the stimulative effects of exogenously applied ethylene with respect to respiration and activities of ACC oxidase and phenylalanine ammonia-lyase. These results indicate that an externally applied current may generate signal(s) by altering the functions of organelle membranes and/or cytoplasmic pH to induce ACC synthase.
Although electroacupuncture (EA) has been widely used to treat pain, the optimal frequency of EA therapy remains unclear. The study sought to determine the effect of different EA frequencies in a Sprague-Dawley (SD) rat model of pain. Electric stimulation (ES) at frequencies of 2 Hz, 15 Hz or 100 Hz was applied to the ipsilateral or contralateral sciatic nerve of the injected hindpaw of SD rats. Formalin (50 microl, 5%) was subcutaneously injected into the plantar surface of the left hindpaw to induce a nociceptive response. Behavior, including licking and biting, was observed to have two distinct periods, an early phase during the first 5 mins and a late phase from 21-35 mins after injection. The total biting or licking count served as an Indicator of nociceptive response. Our results indicate that ES of the ipsilateral sciatic nerve at a frequency of 2 Hz or 15 Hz reduced the nociceptive responses in both the early and the late phases of the formalin test, whereas ES at 2 Hz had greater antinociceptive effect than ES at 15 Hz in the early phase. No similar analgesic effect in the early phase was observed for ES at 100 Hz. Both pretreatment with ES at 2 Hz and naloxone (3 mg/kg, s.c.) produced a greater antinociceptive response in the late phase than when ES at 2 Hz was delivered immediately after formalin administration. In addition, ES of the neck muscle or contralateral sciatic nerve at a frequency of 2 Hz also decreased licking and biting activity in both phases. The results of this study indicate that different analgesic mechanisms are involved in the response to ES at frequencies of 2 Hz, 15 Hz and 100 Hz, and that ES at 2 Hz has a greater analgesic effect on formalin-induced nociceptive response, especially when it is delivered prior to the onset of pain. The analgesic effect of ES may be mediated via a central origin in the supraspinal level. These findings suggest that 2 Hz may be a good frequency selection for clinical EA applications in analgesia, and that pretreatment with EA at 2 Hz may be an effective method to treat post-operative pain.
Key-pressing responses in the cat were maintained under conditions in which brief electric shock was first postponed by responses (avoidance), then periodically presented independently of responses, and finally produced by responses on a fixed-interval schedule of 15 min (FI 15-min). A steady rate of responding occurred under shock avoidance and under response-independent shock; positively accelerated responding was engendered by the FI 15-min schedule. A second experiment studied responding under second-order schedules composed of three FI 5-min components. Responding was suppressed when a stimulus was presented briefly at completion of each FI 5-min component and a shock followed the brief stimulus at completion of the third component. Responding was maintained when each of the first two components was completed either with or without presentation of a brief stimulus and a shock alone was presented at completion of the third FI 5-min component.
The trace of Henri Moissan's pioneer work 100 years ago is clearly evidenced by an overview of achievements in high temperature devices; 1987: "Le four électrique" by Henri Moissan; 1948-1952: "High temperature heating in a cavity rotary kiln using focusing of solar radiation" by Félix Trombe; 1962: "The cavity rotary kiln using focused solar radiation jointly with a plasma gun" by Marc Foëx; 1970: "The rotary kiln with two plasma guns and arc transfer" by Marc Foëx; 1984: "The plasma furnace" by Electricité de France (EDF) at Renardières; 1997: "The plasma furnace" by the Atomic Energy Center (CEA) at Cadarache, the VULCANO program. The first part of this contribution is devoted to Henri Moissan. Re-reading his early book on the electric furnace, especially the first chapter and the sections on silica, carbon vapor and experiments performed in casting molten metal--the conclusions are outstanding--provides modern readers with an amazing insight into future developments. The last two parts are devoted to Félix Trombe and Marc Foëx, tracing the evolution of high temperature cavity processus leading to the solar furnace and the present day plasma furnace at the CEA. Focus is placed on research conducted by the French National Center for Scientific Research (CNRS) with the solar and plasma furnaces at Odeillo. The relationships with Henri Moissan's early work are amazing, offering a well deserved homage to this pioneer researcher.
We studied the slow electrical activity of the brain in people subjected to irradiation as a result of the Chernobyl accident and in healthy donors. The mean constant potential, which is an index of energy metabolism in different regions of the cerebral cortex, is reliably lower in eliminators, as compared to the donors, thus suggesting a postradiation energy deficiency of the brain. The structure of correlations between the constant potential indices is different: the number of significant correlations in the group of eliminators is lower by 30% than in the group of donors, thus suggesting disturbed intercentral relations in the CNS under the conditions of postradiation energy deficiency of the brain.
In 6 patients with medicamentously therapy-refractory tachycardias (3 times auricular flutter/auricular fibrillation with very rapid atrioventricular conduction, once focal atrial tachycardia, once paroxysmal atrioventricular reentry tachycardia, once recurrent ventricular tachycardia on the basis of an interventricular reentry) percutaneous transvenous catheter ablations of the atrioventricular conduction region (bundle of His ablation, 5 cases) and the right Tawara branch (1 case), respectively, were performed. During a follow-up period of 2-26 months (on an average 10.5 +/- 8.0 months) a permanent total block in 4 cases could be obtained (3 times AV-block III. once complete right bundle branch block). In two other patients the total AV block receded to the AV block I. and II. respectively, within 2 and 8 days, respectively; in the first case the AV node reentry tachycardias were no more to be evoked, in the second case a tolerable ventricular frequency was the result during the auricular flutter recidivations. In all patients treated by means of catheter ablation after the intervention an impressive improvement of the clinical symptomatology developed. Due to possible complication and the dependence upon the pacemaker of the patients concerned which is to be expected the indication to the ablation should be made only after the exhaustion of all medicamentous possibilities and taking into consideration other electric therapy methods.
