[Informative value of immunologic analysis of blood and ejaculate in diagnosing chronic prostatitis].

Ejaculate and blood serum from 40 patients with chronic prostatitis were studied immunologically, IgG, IgA, sIgA, IgM concentrations in ejaculate may be greatly influenced by multicomponent composition of ejaculate. This means that it is impossible to diagnose chronic prostatitis only by changes of immunoglobulin concentrations in ejaculate. Data on IgA, sIgA concentrations plus on leukocyte count in ejaculate give more information for precise diagnosis of chronic prostatitis.

[Fructose--its importance to quality of human ejaculate (author's transl)].

No statistically significant differences regarding fructose in one millilitre and ejaculated volume were established from a comparison between 95 ejaculates, among them 14 normospermatic, 47 asthenospermatic, 16 oligospermatic with more than ten million of spermia in one millilitre, six oligospermatic with less than ten million, and twelve azoospermatic. Very low and statistically insignificant correlations, also, were found to exist between the fructose level in a given ejaculate and other qualitative parameters, such as volume, sperm count, as well as morphology and motility of spermia one hour and five hours after ejaculation. The correlation coefficient usually was close to zero. These results seem to cast doubt at the diagnostic value of fructose determination in spermatological routine tests. However, studies into the correlations between fructose, glucose and, perhaps, other sugars in both clinically intact males and patients with metabolic disorders might produce new insights into the dynamics of glucose metabolism in ejaculated semen.

[Relationships between spermiological, biochemical, endocrinological and fertilizing characteristics of boar ejaculates].

Ejaculate volume, sperm concentration, total number of abnormal spermatozoa, number of spermatozoa with protoplasmic droplet, and the content of chlorides, magnesium, calcium and testosterone in semen plasma were evaluated in 155 ejaculates obtained from 77 boars. Further, the evaluation included the fertilizing ability of the ejaculates, expressed by conception rate after the first insemination, average number of all piglets and live-born piglets in litter after inseminations with the studied semen. The following significant (P less than 0.01) relationships were found after the evaluation of the mutual correlations of the studied characteristics: ejaculate volume to sperm concentration (r = -0.496), chlorides to calcium (r = -0.240), chlorides to magnesium (r = -0.492), magnesium to calcium (r = +0.261), testosterone to conception rate after the first insemination (r = -0.222), boar age to ejaculate volume (r = +0.285), boar age to sperm concentration (r = -0.375), boar age to magnesium (r = -0.281), length of sexual rest to total percentage of abnormal spermatozoa (r = +0.323) and to the percentage of spermatozoa with protoplasmic droplet (r = +0.367). The importance of these findings is treated in discussion.

[Concentration of ions of selected trace elements in the ejaculates of boars after interruption of the flow of the secretion from the testes, epididymis and accessory sexual glands].

The investigations were carried out on 6 boars aged from 10 to 12 months. Before operation, 5 ejaculates were collected from each animal. In the second stage of the investigations, the boars were subjected to the following procedures: the deferent ducts were cut and ligated, and the bulbo-urethral glands and vesicular glands were removed. Four to 28 ejaculates were collected after each operation. The ejaculates obtained at the individual stages of the experiments were subjected to the following tests: introductory assessment, concentration and morphology of spermatozoa, and biochemical investigations concerning the concentration of Mn, Co, Cd, Fe, Cr, Ni, Sr, Ag and Li. The authors proved that the secretions from the individual segments of the reproductive organ of the boar are different in respect of their biochemical composition. The secretion of the testicles and epididymides determines the concentration of Mn, Cd and Cr in the ejaculate of the animals. The level of Mn and Fe seems to be largely influenced by the secretion from the vesicular glands and deferent ducts. The secretion of the prostate and urethral glands is responsible for the level of Cr and Ni in semen plasma, and also for the concentration of Mn, Ca and Fe. The concentration of Fe in the semen of boars distinctly depends on the presence of spermatozoa and of the secretion of the bulbo-urethral glands. The AAS method failed to detect traces of Ag and Li in the ejaculates of these boars; Co was present in vestigial amounts only.

[Effect of the standard of hygiene in the care for breeding boars and of the technic of semen collection on the number of pathogens in the boar ejaculate].

The microbiological examination of 48 ejaculates obtained by an artificial vagina revealed an average number of 178 000 germs per 1 ml of semen (1900 - 758 000). When the technique was changed, the manual method being employed, 23 ejaculates were obtained from the same boars and the microbiological examination revealed 44 200 germs per 1 ml of semen, on an average. In the second part of the trial 85 ejaculates obtained by an artificial vagina from 17 breeding boars were subject to microbiological examination. The average number of germs found in this case was 99 377 (10 000 - 400 000). When boar body was washed with water or with water and soap and rubbed with a sterile cloth, prior to each collection, the number of germs per 1 ml of semen decreased to 6071 (0 - 40 000). The reduction of the number of germs in the ejaculate of washed boars is statistically highly significant, as compared with semen collection from boars without washing. The number of fungus-bearing ejaculates decreased from the original 39.9% to 9.3%.

Five meters of H(2)O: the pressure at the urinary bladder neck during human ejaculation.

BACKGROUND: There are no data in the literature on pressure changes in the prostatic urethra during ejaculation. In healthy men, it has always been postulated that there must be a pressure gradient in order to prevent retrograde ejaculation, but scientific proof for that is pending. METHODS: In five healthy male volunteers, the pressure profile in the prostatic urethra was registered during ejaculation, using a 10 French balloon catheter with 16 pressure channels. The channels were arranged in pairs at 5-mm intervals, beginning just below the balloon at the bladder neck and extending down to the external urethral sphincter. RESULTS: In the proximal part of the prostatic urethra, a pressure of up to 500 cm of H(2)O was measured in all subjects. Contrary to that, pressures did not exceed 400 cm of H(2)O distally to the verumontanum. CONCLUSIONS: A novel method to register the pressure profile in the lower urinary tract during ejaculation (ejaculomanometry) is presented. This study adds to the knowledge of the normal physiology of reproductive function and may be useful in the evaluation of male sexual and reproductive disorders.

Ejaculate of sneaker males is pheromonally inconspicuous in the black goby, Gobius niger (Teleostei, Gobiidae).

The black goby, Gobius niger, shows alternative male mating tactics, i.e., parental and sneaker males. Males release a sexual pheromone that attracts females and stimulates aggressive displays in males. This pheromone is produced by the mesorchial gland, a structure well developed in parental males but markedly undeveloped in sneakers. We measured the behavioral response of parental males to the ejaculates of males performing different reproductive tactics. Parental males reacted to the ejaculate of other parental males, with stereotypic aggressive behaviors, but not to the ejaculate of sneakers; consequently sneaker male ejaculate appears to be pheromonally inconspicuous.

Ejaculate-hormonal traits in the leopard cat (Felis bengalensis) and sperm function as measured by in vitro penetration of zona-free hamster ova and zona-intact domestic cat oocytes.

Electroejaculate traits and circulating follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone concentrations were analyzed in adult leopard cats (Felis bengalensis), a rare felid species indigenous to east Asia. The ability of leopard cat sperm to bind and penetrate zona-free hamster ova and zona-intact domestic cat oocytes in vitro was examined as a means of testing sperm function. The influence of culture media [Biggers, Whitten, Whittingham (BWW) vs. modified Krebs Ringer bicarbonate (mKRB)], seminal plasma removal, and swim-up separation on sperm motility, sperm morphology, and oocyte penetration also were assessed. Sperm treatments included dilution of raw semen (DR), ejaculate centrifugation, and either resuspension (NS) or swim-up processing (SU). The percentage of oocytes penetrated (penetration rate) and the number of penetrated sperm/oocyte (penetration index) were determined. Ejaculates from each male consisted of at least a 50% sperm motility rating, and hormone concentrations in individual males were unrelated to any ejaculate trait measured concurrently on the same day. The SU technique improved (P less than 0.05) percent sperm motility and the proportion of structurally normal sperm compared to DR and NS treatments. Leopard cat spermatozoa were capable of binding to and penetrating hamster ova and domestic cat oocytes; however, penetration was influenced by culture medium and seminal processing. In the hamster assay, a higher (P less than 0.05) penetration rate and penetration index were achieved when mKRB was used for gamete incubation instead of BWW. NS processing also increased (P less than 0.05) overall penetration compared to DR and SU. In the cat oocyte assay, zona penetration rate was similar (P greater than 0.05) in the DR, NS, and SU aliquots; however, the zona penetration index was increased (P less than 0.05) by the NS compared to the DR and SU treatments. This study 1) provides baseline ejaculate and endocrine norms for the leopard cat, 2) demonstrates that leopard cat sperm undergo nuclear decondensation in hamster ova and penetrate zona-intact domestic cat oocytes, 3) indicates that seminal plasma removal enhances leopard cat sperm fertilizing ability and ovum penetration, and 4) suggests that heterologous oocyte penetration is effective for assessing factors influencing fertilization and sperm function in this nondomestic felid.

Prolongation of the ejaculation latency in the male rat by thioridazine and chlorimipramine.

Thioridazine (3 mg/kg) and chlorimipramine (1.5-6.0 mg/kg) prolonged the ejaculation latency and increased the number of mounts but did not change the number of intromissions preceding ejaculation. Blockade of peripheral and central noradrenaline receptors by phentolamine and phenoxybenzamine respectively resulted in a suppression of all aspects of the sexual behavior with increasing doses. DL-5-HTP (25-100 mg/kg) in combination with an inhibitory of peripheral 5-HTP decarboxylase (benserazide, 25 mg/kg) produced, like chlorimipramine and thioridazine, a prolongation of ejactulation latency and an increase in the number of mounts preceding ejaculation. Selective inhibition of 5-HT reuptake however, by zimelidine (0-20 mg/kg) or alaproclate (0-20 mg/kg) did not affect the mating behavior. At higher doses of these drugs some animals failed to initiate sexual activities. There was an increase in the postejaculatory interval but not change in the ejaculatory latency. It is concluded tha the prolonged ejaculation latencies observed following treatment with thioridazine or chlorimipramine is not due to a blockade of central or peripheral adrenergic alpha-receptors.

The role of anxiety in premature ejaculation: a psychophysiological model.

Despite the prevalence of premature ejaculation and the success in its treatment, researchers have yet to offer a compelling, empirically based theory regarding its nature or etiology. This study explored a model that argues that anxiety may not be necessary for the existence of this dysfunction. Fifteen premature ejaculators (PEs) and 17 nonpremature ejaculators (NPEs) engaged in self-stimulation to orgasm both in the laboratory and at home. The following specific hypotheses were tested: Compared to NPEs, PEs would demonstrate (i) shorter orgasmic latencies, both in the lab and at home, and (ii) equally accurate estimates of these latencies. Results offered strong support for both hypotheses. These findings, and those derived from a questionnaire completed by subjects, were seen as consistent with a psychophysiologic model of premature ejaculation. According to this model, the role of anxiety is seen as variable, interacting with the somatic vulnerability of the individual to determine orgasmic latency.

Multiple ejaculations during prolonged sexual tests and lack of resultant serum testosterone increases in Male Stumptail Macaques (M. arctoides).

The sexual behavior of four male and four female adult stumptail macaques was observed in standardized 3-hr heterosexual pair tests. The males achieved from 11 to 19 ejaculations during a single test, thus apparently exceeding any other primate studied under laboratory conditions for the ability to display multiple ejaculatory patterns in relatively short periods of time. The stage of the menstrual cycle was not found to be related to the performance of the male or female, although this variable was not analyzed in depth. Progressive increases in the interejaculatory interval (IEI) occurred for all four males for the initial two to four ejaculations of the series, and then either a plateau or a transient decrease in this measure occurred. Since the data varied considerably for IEI as well as for other quantitative measures of sexual behavior for a given pair, individual data rather than group averages are presented. Blood samples drawn immediately before and after behavioral testing as well as at comparable intervals 1 week prior to behavioral testing were analyzed for testosterone concentration by radioimmunoassay for each male. Serum testosterone values were not found to be influenced by multiple ejaculations. Instead, decreased levels of serum testosterone were encountered on the second blood sample of the day for most males, regardless of whether sexual behavior occurred. The possibility that the decreases were related to stress effects of handling is discussed. Because of this complication, it could be only tentatively concluded that repeated copulation to ejaculation over a 3-hr period did not result in appreciable changes in testosterone levels.

Retrograde ejaculation after retroperitoneal lower lumbar interbody fusion.

We have studied the incidence and functional outcome of retrograde ejaculation as a postoperative complication of anterior lumbar interbody fusion. A questionnaire, specifically designed to analyse this problem, has been used over a 6 to 13 year follow-up. Out of 50 men, 41 completed the questionnaire; 2 complained they had permanent retrograde ejaculation after the operation; one stopped ejaculating for 6 months, and thereafter had a 50% reduction. The Dallas pain questionnaire showed that retrograde ejaculation did not have a negative effect on the functional outcome, but male genital dysfunction was a complication of anterior spinal fusion in 8% of cases.

Guidelines for the pharmacotherapy of premature ejaculation.

In the 1960s, premature ejaculation was considered a learned behaviour that had to be treated by behavioural therapy, such as the so-called stop-start technique. The efficacy of this method was questioned. Physicians have always looked for medication that could delay ejaculation. Many studies, controlled and open label, have now clearly shown that rapid ejaculation can be treated quickly and effectively with clomipramine and SSRIs. This article describes the guidelines for the pharmacological treatment of premature ejaculation. These guidelines were developed to ensure that medication are used when clear indications are present.

Cholecystokinin facilitates ejaculation in male rats: blockade with proglumide and apomorphine.

Systemic administration of the active, sulfated form of cholecystokinin-octapeptide (CCK-8S), at a dose known to inhibit dopamine (DA) release, significantly reduced the latency to ejaculate and number of intromissions preceding ejaculation in sexually active male rats. This effect was identical to that reported for low doses of the DA receptor agonist apomorphine, which bind preferentially to presynaptic DA autoreceptors. The facilitatory effect of CCK-8S on ejaculation was blocked by the putative CCK receptor antagonist proglumide and by a presynaptic dose of apomorphine. Blockade of the effect of CCK-8S with a presynaptic dose of apomorphine is consistent with CCK induction of depolarization block in DA neurons. These data suggest that CCK-8S and apomorphine may reduce the ejaculation threshold in sexually active male rats by inhibition of DA release via two different mechanisms of action.

Clinical use of immunoelectrophoresis of split ejaculates. I. Variation of patterns due to antisera.

As a preliminary investigation for the clinical application of immunoelectrophoresis of ejaculate in the diagnosis of male reproductive organ diseases, variations of immunoelectrophoretic patterns in normal subjects due to the difference of antigenicity between split ejaculates were demonstrated with rabbit antisera. Fresh samples and antisera against them gave larger numbers of and more intense precipitin lines than in stored samples. The precipitin lines developed by acid phosphatase were most remarkable between first portion of ejaculate (F1) and anti-F1, whereas the line caused by protein from the seminal vesicle was most prominent between the remainder of the ejaculate following F1 (F2) and anti-F2.

PCA: effects on ejaculation, thermoregulation, salivation, and irritability in rats.

The short term monoamine releaser p-chloroamphetamine (PCA) was injected intraperitoneally in male rats housed at 20 degrees C. Within 2 hr of PCA injections (2.5, 5.0, 8.0 or 10.0 mg/kg), rats showed ejaculation, decreased colonic temperature, increased salivation, and increased irritability. Ejaculation and salivation scores were considerably lower in the 2.5 mg/kg than in the higher dose groups, but otherwise were not dose dependent at the doses used. Hypothermia was of similar magnitude in all groups, but lasted longer in the higher dose groups. Irritability increased with dose size. In order to study the role of ambient temperature in PCA-induced behavioral changes, observations were made on an additional group or rats housed at the higher ambient temperature of 25 degrees C. In these rats an increase, rather than a decrease, in mean colonic temperature was observed following PCA injection (5 mg/kg). Ejaculation and irritability scores were similar to those observed at the lower ambient temperature, but salivation was enhanced. It is suggested that PCA induces ejaculation, salivation, irritability and, depending on the ambient temperature, either hypothermia or hyperthermia.

Stimulation of dopamine autoreceptors elicits "premature ejaculation" in rats.

Following treatment with dopamine (DA) receptor agonists, such as apomorphine, N-n-propyl-norapomorphine, lisuride and 3-(3-hydroxyphenyl)-N-n-propyl-piperidine (3-PPP) (50, 2.5, 400 and 5000 micrograms/kg, respectively), male rats attain ejaculation with receptive females sooner and after fewer penile intromissions than controls. Since doses of DA agonists needed to produce "premature ejaculation" are within the low dose range needed to stimulate DA autoreceptors, it is suggested that "premature ejaculation" in rats results from inhibition of DA neurotransmission. This hypothesis is supported by the finding that 6 hr after haloperidol (1 mg/kg), rats achieve ejaculation after fewer intromissions than normal.

Imipramine-induced erection, masturbation, and ejaculation in male horses.

Imipramine hydrochloride was administered to five male horses (400-500 kg b.wt.): one experienced young stallion, two mature normal breeding stallions, one 5-year-old stallion with erection and ejaculatory dysfunction, and one long-term castrated male horse. Oral imipramine treatment (100 to 600 mg, twice daily) led to frequent erection and masturbation while at rest in the stall in a nonsexual context. Intravenous imipramine treatment over a range of doses (50 to 1000 mg) similarly induced erection and masturbation in all animals. Erection typically occurred within 10 minutes after injection, and the erection and masturbation continued intermittently for 1 to 2 hours. These erections proceeded as during sexual excitement to a normal firmness and eventual engorgement of the glans penis. Two stallions ejaculated while masturbating. Mild ataxia and drowsiness appeared at the higher doses, but the animals remained responsive to auditory, visual, and tactile stimuli. Erection and masturbation were often interrupted by activities about the barn or the approach of the handler, suggesting cortical inhibitory control of the erection. When tested in a sexual context immediately following IV treatment (500 mg), the two mature breeding stallions bred normally. The 5-year-old stallion, which had not ejaculated over several months of breeding attempts, spontaneously ejaculated following IV imipramine treatment. Subsequently, this stallion has ejaculated during copulation while on low dose oral (100 mg. twice daily) imipramine treatment. Plasma total androgens increased during treatment in these stallions. The long-term castrate showed erection and masturbation following IV imipramine treatment, suggesting that the effect of imipramine is not testosterone dependent.
