Fate of radioactive exocrine pancreatic proteins injected into the blood circulation of the rat. Tissue uptake and transepithelial excretion.

[35S]methionine or [35S]methionine-labeled exocrine pancreatic proteins were injected into the bloodstream of conscious rats. Samples of blood, urine, bile, and pancreatic juice were collected at varying intervals through 7 h. Injection of [35S]methionine resulted in the appearance of trichloroacetic acid--soluble radioactivity [( 35S]methionine) in bile and urine within 4 min and trichloroacetic acid-insoluble radioactivity in blood, bile, and pancreatic juice after 20 min. Analysis of these body fluids by two-dimensional isoelectric focusing/sodium dodecyl sulfate gel electrophoresis and fluorography indicated that rat serum, biliary, and pancreatic proteins were labeled, respectively. After the injection of [35S]methionine-labeled pancreatic proteins, half of the trichloroacetic acid-insoluble radioactivity disappeared from the serum in 10-15 min. Radioactive proteins appeared after 5 min in urine and bile, and, over the course of the experiment, accounted for 1%-2% and 0.3%-0.5% of the injected radioactivity, respectively. Analysis of individual radioactive proteins excreted into bile by two-dimensional isoelectric focusing/sodium dodecyl sulfate gel electrophoresis indicated preferential transhepatic transport of negatively charged pancreatic proteins. The majority of pancreatic proteins (approximately 97%) were taken up by a variety of body tissues, particularly kidney, liver, spleen, and lung. Trichloroacetic acid-soluble radioactivity, largely representing [35S]methionine, appeared sequentially in serum, urine, and bile within 2-12 min. At later experimental time points (greater than 60-90 min), radioactive rat serum, biliary, and pancreatic proteins appeared in blood, bile, and pancreatic juice, respectively. After the injection of 35S-labeled guinea pig pancreatic proteins into the blood circulation of the rat, trichloroacetic acid-insoluble radioactivity, observed in pancreatic juice after 60-90 min, exclusively represented rat exocrine pancreatic proteins as judged by the two-dimensional gel procedure. These studies indicate that pancreatic proteins are removed from the blood circulation by at least three separate pathways: (a) uptake and degradation by a variety of tissues in the body (approximately 97% of injected radioactivity), (b) excretion of intact proteins into urine (1%-2%), and (c) transport of intact proteins into bile (0.3%-0.5%). Transport of exocrine pancreatic proteins from the blood circulation to pancreatic juice could not be demonstrated.

[Effect of TCM therapy by principle for nourishing Qi, activating blood circulation and resolving phlegm on quality of life of patients with chronic obstructive pulmonary disease].

OBJECTIVE: To observe the effect of TCM therapy, by principle for nourishing Qi, activating blood circulation and resolving phlegm, on quality of life of patients with chronic obstructive pulmonary disease (COPD). METHODS: Seventy-two COPD patients with Qi-deficiency, blood stasis and phlegm retention syndrome were randomly divided into two groups. Thirty-five patients in the treated group were treated with the integrative traditional and Western medicine, that is, conventional Western medicine combined with Chinese compound recipe prescribed based on the principle of nourishing Qi, activating blood circulation and resolving phlegm, or with Feikang granule and Bailing capsule, while 37 in the control group was treated with conventional Western medicine alone. The clinical efficacy was evaluated by assessment software of COPD quality of life after patients were treated for 3 months. RESULTS: The clinical efficacy in the treated group was superior to that in the control group, showing significant difference (P < 0.01). CONCLUSION: Compound recipe prescribed according to the therapeutic principle for nourishing Qi, activating blood circulation and resolving phlegm can improve the symptoms of patients with COPD at stable stage, elevate their quality of life.

[Supplementing Qi and activating blood circulation in gene regulation of vascular smooth muscle cells].

According to the theory of traditional Chinese medicine, blood stasis is a main pathological mechanism in the development of vascular diseases. Supplementing Qi and activating blood circulation, as the therapeutic principle for the blood stasis, has been adapted. Studies demonstrated that the supplementing Qi and activating blood circulation recipe could regulate the expression of vasoactive peptides in vascular smooth muscle cells (VSMCs). The recipe inhibited the formation of neointima following arterial intimal lesions through down-regulating expression of proliferation-related genes and phenotypic modulation of VSMCs. The recipe also markedly inhibited the adhesion and migration of VSMCs and matrix remodelling by means of a mechanism that balances extracellular matrix turnover rate. The purpose of this review is to summarize the recent advances made in our understanding of new functions for the recipe in regulating VSMCs behaviours and their microenvironment relevant to vascular diseases and maintaining proper homeostasis.

[Gaseous signaling molecule and study on activating blood circulation and removing stasis].

The finding of gaseous signaling molecule NO, CO and H2S, and the advocacy of "gas biology" concept have provided a novel thinking for the study of Huoxue Huayu (HXHY, a traditional Chinese medicine method of promoting blood circulation to remove blood stasis). The concept "Qi" in TCM and gaseous signaling molecule are correlated in respects of their source and mesomeric function. Study of HXHY could only be deepened through an overall cognition on blood stasis syndrome with the view of Qi-blood correlation.

Is big endothelin converted to endothelin-1 in circulating blood?

Although evidence has been accumulating to support an intracellular processing of big endothelin-1 (big ET-1) to ET-1, molecular conversion in the circulating blood remains to be elucidated. The present study was undertaken to investigate whether big ET-1 was converted to ET-1 in human blood. In the first experiment, normal serum with synthetic big ET-1 exogenously added or serum from patients with chronic renal failure was incubated in vitro at 37 degrees C for 1 h. In the second experiment, synthetic big ET-1 was incubated in the whole blood at 37 degrees C for 1 h. In the third experiment, synthetic big ET-1 was administered intravenously in anesthetized rat and a plasma sample was obtained before and after 15 min and 1 h. After respective incubation, molecular forms of ET were determined by a combination of reverse-phase high-performance liquid chromatography and radioimmunoassay. There was no significant conversion of big ET-1 to ET-1 in the serum obtained from normal and CRF patients. However, there was a slight but significant increase of ET-1 after incubation of big ET-1 in the whole blood or after administration of big ET-1 in anesthetized rat. The conversion in the whole blood was inhibited by 5 mM EDTA. These results suggest that circulating blood may not be a major site of molecular conversion from big ET-1 to ET-1, although conversion does occur in the circulation by blood cell-mediated process.

[Effect of different drug dosage to activate blood circulation and to nourish qi on cardiac function and structure of congestive heart failure rats after acute myocardial infarction].

OBJECTIVE: To observe the effect of different drug dosage to activate blood circulation and to nourish Qi on cardiac function and structure of heart failure rats after acute myocardial infarction. METHOD: The congestive heart failure post cardiac infarction rats models subjected to left coronary occlusion were made and given different dosage of drug of Huoxue Yiqi and captopril (positive control) from 4 weeks to 8 weeks after operation. The cardiac function before and after using drugs were observed by impedance methods. RESULT: Treated by different dosage of drug of Huoxue Yiqi and captopil, cardiac function of heart failure rats after acute myocardial infarction improved, SV, CO, CI(P < 0.01); at the same time, prescription of Huoxue Yiqi and Qixue could improve the model rats' heart coefficient, VS SHAM(P > 0.05), but prescription of Yiqi Huoxue and captopril could not improve it. CONCLUSION: Drug to activate blood circulation and to increase qi can treat heart failure rats after acute myocardial infarction, through improving their function and structure. At the same time, more dosage of drug to activate blood circulation and to increase qi(Huoxue/Yiqi drug) may improve the model rats' heart coefficient.

Effects of CO2 inhalation on cochlear blood circulation.

The cochlear blood flow was measured with the microsphere method before, during and after CO2 inhalation in anesthetized rabbits. Different gas mixtures and exposure times were used. The greatest increase of cochlear blood flow was measured after 5 min inhalation of 7% CO2 in air (115%) and the least increase (44%) in animals breathing 7% CO2 in oxygen. The difference is explained as due to a vasoconstriction caused by a high oxygen tension counteracting the vasodilating effect of CO2. The treatment with CO2 of supposed vascular diseases in the cochlea is discussed and the negative effects due to respiratory acidosis are emphasized.

[Physiological characteristics of the smooth muscles vessels of the small circle of blood circulation].

Now sireos problem of pulmonology there are the diseases connected with infringement of coordinated regulation of a tone of smooth muscles of vessels and airways of ways that conducts to dissociation of parameters haemodinamyc and ventilation of lungs and as consequence, to infringement airwave-perfusion attitudes. In the review features humoral regulation contractile activity of smooth muscles of vessels of a small circle of blood circulation, a role of endocellular alarm systems in these mechanisms, and endothelium, as the local modulator endocrine functions are considered. Disgusting muscles of a small circle are distinguished from the main vessels of the big circle of blood circulation with predisposition to the raised mechanical pressure. In spite of the fact that endothelium renders modulating relaxe influence on contractile answers of smooth muscles of vessels of a venous and arterial small circle of blood circulation at action corresponding vasoconstriction, pulmonary veins are capable to endothelium-dependent dilatation to a lesser degree, in comparison with pulmonary arteries. And, on the contrary, in absence endothelium, they are characterized with high sensitivity to vasopression to substances--serotonin, histamine, phenylephrine. Features of regulation smooth muscle pressure pulmonary an artery are shown in contractile reactions of its isolated segments in reply to influence beta-adreno agonist--isoprotherenol and phosphoesterase inhibitors. Though, increase in endocellular concentration cyclic nucleotides (cAMP and\or cGMP), on the standard representations, cannot explain growth of a mechanical pressure of smooth muscles, apparently, in contractile reactions of a pulmonary artery to influence biologically and physiologically active substances interfere more complex mechanisms in which basis processes of interaction of smooth muscles cells lay, endothelium and cells of a microenvironment. Finding-out of the contribution cyclic nucleotides in these processes demands the further researches.

Artificial blood circulation: stabilization, physiological control, and optimization.

The requirements for creating an efficient Artificial Blood Circulation System (ABCS) have been determined. A hierarchical three-level adaptive control system is suggested for ABCS to solve the following problems: stabilization of the circulation conditions, left and right pump coordination, physiological control for maintaining a proper relation between the cardiac output and the level of gas exchange required for metabolism, and optimization of the system behavior. The adaptations to varying load and body parameters will be accomplished using the signals which characterize the real-time computer-processed values of correlations between the changes in hydraulic resistance of blood vessels, or the changes in aortic pressure, and the oxygen (or carbon dioxide) concentration.

Cochlear blood circulation studied with microspheres.

The cochlear blood flow was studied with the microsphere method in guinea pigs, rats and cats. Results from experiments using different sizes of microspheres were compared and the effect of middle ear infection or perforation of the otic capsule on the cochlear microcirculation was investigated. Neither the opening of the cochlear bony wall nor the middle ear infection influenced the blood flow in the cochlea. 9- and 15-micrometer spheres were given simultaneously and the results were equivalent, indicating no more shunting of the smaller ones than of the bigger ones. Thus, the smaller spheres are to be preferred, since the smaller ones can be used in a larger number without negative effects on the general circulation. In cats, the cochlear blood circulation was determined before and during cervical sympathetic stimulation. Sympathetic stimulation caused a reduction of 25% of the cochlear blood flow.
