Phosphorylation of H2AX histones in response to double-strand breaks and induction of premature chromatin condensation in hydroxyurea-treated root meristem cells of Raphanus sativus, Vicia faba, and Allium porrum.

Histone H2A variant H2AX is rapidly phosphorylated on the induction of DNA double-strand breaks by ionizing radiation and hydroxyurea-mediated replication arrest, resulting in the formation of gamma-H2AX foci along megabase chromatin domains nearby the sites of incurred DNA damage. In an attempt to establish a relationship between species-specific nuclear architecture and H2AX phosphorylation in S/G(2) phase-arrested root meristem cells, immunocytochemical comparisons using an antibody raised against human gamma-H2AX were made among three plants differing with respect to DNA contents: Allium porrum, representing a reticulate type of DNA package, Vicia faba, having semireticulate cell nuclei, and Raphanus sativus, characterised by a chromocentric type of chromatin. Another approach was aimed at determining possible correlations between the extent of hydroxyurea-induced phosphorylation of H2AX histones and the quantities of root meristem cells induced by caffeine to enter aberrant mitotic division (premature chromosome condensation). It was concluded that the higher-order structure of chromatin may contribute to the accessibility of molecular factors engaged in the recognition and repair of genetic lesions. Consequently, in contrast to A. porrum and V. faba, a diffuse chromatin in chromocentric cell nuclei of R. sativus may become more vulnerable both to generate DNA double-strand breaks and to recruit molecular elements needed to arrange the cell cycle checkpoint functions, and thus, more resistant to factors which allow the cells to enter premature chromosome condensation spontaneously. On the other hand, however, caffeine-mediated overriding of the S-M checkpoint control system resulted in the typical appearance of premature chromosome condensation, irrespective of the genomic content of DNA.

Sesquiterpene lactone dehydroleucodine selectively induces transient arrest in G2 in Allium cepa root meristematic cells.

Dehydroleucodine is a sesquiterpene lactone recently isolated from aerial parts of a medicinal herb, Artemisia douglasiana Besser. We have previously shown that 25 and 100 microM dehydroleucodine slowed down onion root growth by 30 and 70%, respectively, affecting neither cell viability nor cell elongation. In the present study we analyze the effect of dehydroleucodine on cell cycle phases in onion (Allium cepa L.) root meristematic cells synchronized with caffeine or caffeine and hydroxyurea. Synchronized root cells treated with 100 and 200 microM dehydroleucodine present an interphase lengthening of 5.2 h and 8.2 h, respectively. The S-phase length, estimated by [3H]thymidine incorporation assay, was 6 h for both control roots and roots that had been immersed in dehydroleucodine. The peak of [3H]leucine incorporation was observed 6 h after release from synchronization in controls and in dehydroleucodine-treated roots, indicating that protein synthesis in G2 was not affected. Thus, these results show that dose-dependently dehydroleucodine selectively induces a transient arrest of meristematic cell in G2 and that dehydroleucodine can be used experimentally as a cell cycle suppressor.

Effect of low storage temperature on some of the flavour precursors in garlic (Allium sativum).

Garlic (Allium sativum) cloves were stored at ambient temperature and 4 degrees C for periods up to six months to establish the effect of position of the individual clove within the bulb and of low storage temperature on the composition of several flavours precursors and other organic sulphur compounds, measured by gradient High Pressure Liquid Chromatography. Levels of alliin, gamma glutamyl allyl cysteine sulphoxide and gamma glutamyl isoallyl cysteine sulphoxide were statistically significantly higher in outer than in inner cloves. There was no statistically significant change in levels of alliin, the major flavour precursor, in cloves stored at 4 degrees C, remaining in the average range 17.5+/-3.8-39.1+/-7.5 mM. However, isoalliin increased significantly during storage at 4 degrees C, rising from an average 0.6+/-0.2 mM (outer cloves) -- 0.7+/-0.4 mM (inner cloves) to 7.1+/-1.7 mM (outer cloves) -- 4.1+/-0.7 mM (inner cloves). A decline in other sulphur-containing compounds, most likely to be the peptides gamma-glutamyl allylcysteine sulphoxide and gamma-glutamyl isoallylcysteine sulphoxide, occurred at the same time and possibly contributed to the increase in the flavour precursor compounds. The degree of chemical changes during storage will be of interest to the food and pharmaceutical industries.

Two different Bacillus thuringiensis toxin genes confer resistance to beet armyworm (Spodoptera exigua Hübner) in transgenic Bt-shallots (Allium cepa L.).

Agrobacterium-mediated genetic transformation was applied to produce beet armyworm (Spodoptera exigua Hübner) resistant tropical shallots (Allium cepa L. group Aggregatum). A cry1Ca or a H04 hybrid gene from Bacillus thuringiensis, driven by the chrysanthemum ribulose-1,5-bisphosphate carboxylase/oxygenase small subunit (Rubisco SSU) promoter, along with the hygromycin phosphotransferase gene (hpt) driven by the CaMV 35S promoter, was employed for genetic transformation. An average transformation frequency of 3.68% was obtained from two shallot cultivars, Tropix and Kuning. After transfer of the in vitro plants to the greenhouse 69% of the cry1Ca and 39% of the H04 transgenic shallots survived the first half year. After one year of cultivation in the greenhouse the remaining cry1Ca and H04 transgenic plants grew vigorously and had a normal bulb formation, although the cry1Ca transgenic plants (and controls) had darker green leaves compared to their H04 counterparts. Standard PCR, adaptor ligation PCR and Southern analyses confirmed the integration of T-DNA into the shallot genome. Northern blot and ELISA analyses revealed expression of the cry1Ca or H04 gene in the transgenic plants. The amount of Cry1Ca expressed in transgenic plants was higher than the expression levels of H04 (0.39 vs. 0.16% of the total soluble leaf proteins, respectively). There was a good correlation between protein expression and beet armyworm resistance. Cry1Ca or H04 gene expression of at least 0.22 or 0.08% of the total soluble protein in shallot leaves was sufficient to give a complete resistance against beet armyworm. This confirms earlier observations that the H04 toxin is more toxic to S. exigua than the Cry1Ca toxin. The results from this study suggest that the cry1Ca and H04 transgenic shallots developed could be used for introducing resistance to beet armyworm in (sub) tropical shallot.

The C-S lyases of higher plants: preparation and properties of homogeneous alliin lyase from garlic (Allium sativum).

Alliin lyase from garlic (Allium sativum) has been purified to homogeneity. The purification procedure involves the use of affinity chromatography on concanavalin A-Sepharose 4B. Addition of polyvinylpolypyrrolidone to the homogenizing medium greatly improves the specific activity of the extract. The enzyme is a glycoprotein as seen by its ability to bind to concanavalin A-Sepharose 4B and by its positive periodic acid-Schiff base stain. It has a carbohydrate content of 5.5%. Km values for this enzyme were estimated to be 5.7 mM for S-ethyl-L-cysteine sulfoxide and 3.3 mM for S-allyl-L-cysteine sulfoxide. The molecular weight of this garlic enzyme, as determined by gel filtration, was found to be 85,000; the molecule consists of two equal subunits of Mr 42,000. The amino acid content was found to be similar to that reported previously for onion alliin lyase, although there is twice as much tryptophan in the garlic alliin lyase as in the onion enzyme. By both chemical and spectral methods the enzyme was found to have two molecules of pyridoxal 5-phosphate per enzyme molecule, suggesting one per subunit. There are significant differences in the nature of these findings from those previously reported from this laboratory for the onion enzyme. Studies are in progress to compare further the alliin lyases from garlic and onion.

Structural studies on Allium cepa L. chromatin: enhanced stability of internucleosome interactions in plant chromatin.

The pattern of micrococcal nuclease digestion of chromatin from different organs of Allium cepa has been studied. The DNA from small oligonucleosomes appears to be highly degraded and heterogeneous. In solutions of intermediate ionic strength (0.15 M NaCl) histones H2A and H2B form dimers, however at high salt concentrations (2 M NaCl) they tend to form complexes of higher order, such as tetramers. It is proposed that a correlation exists between the ability of these proteins to form tetramers and the particular stability of internucleosome interactions.

High-resolution analysis of Allium cepa chromosome replication by an FPG technique.

In Allium cepa meristematic cells, BrdU incorporation into late-replicating DNA and its detection by FPG technique has shown to allow the visualization of characteristic late replicated segments (LRS) in metaphase chromosomes not detected till now. Since these LRS have very similar characteristics with the 'dot-like' exchanges reported previously in this system, they seem to represent a new support for the idea of a regular organization of chromatin in metaphase.

The flavonoids of Allium ursinum.

From wild garlic Allium ursinum three new flavonoid glycosides were identified as kaempferol 3-O-beta-neohesperidoside-7-O-[2-O-(trans-p-coumaroyl)]-beta -D- glucopyranoside, kaempferol 3-O-beta-neohesperidoside-7-O-[2-O-(trans-feruloyl)]-beta-D- glucopyranoside, kaempferol 3-O-beta-neohesperidoside-7-O-[2-O-(trans-p-coumaroyl)-3-O-b eta-D- glucopyranosyl]-beta-D-glucopyranoside and characterized as the peracetates. Additionally, two known flavonoid glycosides kaempferol 3-O-beta-glucopyranoside and kaempferol 3-O-beta-neohesperidoside were isolated. The isolated compounds showed an inhibition of human platelet aggregation.

Mitodepressive effect of (-)-cathinone, from Catha edulis (khat), on the meristematic region of Allium cepa root tips.

(-)-Cathinone, a psychostimulant alkaloid from khat, showed a significant mitodepressive effect on the dividing cells of Allium cepa root tips. This effect was dose-dependent and irreversible. Exposure of root cell to cathinone for 48 hr produced significant condensation and clumping of chromosomes, sticky metaphases and anaphase bridges. Our study suggests that the teratogenic and mutagenic effects of khat extracts as reported by earlier workers might be partially or totally due to their (-)-cathinone content.

Isolation and identification of two isomeric trihydroxy octadecenoic acids with prostaglandin E-like activity from onion bulbs (Allium cepa).

Two fractions with prostaglandin E-like activity were isolated from onion (Allium cepa) by using XAD-2 adsorption, silicic acid column chromatography and thin layer chromatography. The fractions were analyzed by gas chromatography/mass spectrometry and were characterized as isomeric mixtures of 9,10,13-trihydroxy-11-octadecenoic and 9,12,13-trihydroxy-10-octadecenoic acid, which are lipoxygenase metabolites of linoleic acid. Bio-assay, for which cascade superfusion was used and the rabbit coeliac and mesenteric arteries and the rat fundus strip were employed as assay organs, was utilized to monitor the bio-active profile throughout the isolation procedures. The activity of 1 microgram of the pharmacologically active fractions T1 and T2 was found to be equivalent to that of respectively 1.33 and 0.63 ng of prostaglandin E2.

Effects of onion (Allium cepa) extract on platelet aggregation and thromboxane synthesis.

Oral administration of onion and garlic reportedly decreases platelet aggregation in both human and animal subjects. An oily chloroform extract of onion (Allium Cepa) was prepared and separated by column chromatography on silicic acid into six fractions by elution with solvents of increasing polarity. The least polar fraction contained most of the inhibitory activity towards platelet aggregation induced by either ADP or arachidonic acid. Further purification was afforded by thin-layer chromatography. The specific activity of this major active fraction (I50 per ml of PRP) was approximately 7 units per milligram. Platelets incubated in the presence of onion inhibitor and (1-14C)-arachidonic acid showed striking changes in the pattern of arachidonic acid metabolites formed. Thromboxane B2 synthesis was almost completely suppressed without significant decreases in total hydroxy fatty acid formation. It was concluded that the observed antiplatelet activity of onion relates to the presence of a non-polar, heat stable inhibitor of thromboxane synthesis. This appears to be the first demonstration of this type of inhibitor present in significant quantities in a common foodstuff.

Effects of certain food dyes on chromosomes of Allium cepa.

The effects of 4 permitted food dyes, i.e., fast green FCF, indigo carmine, orange G and tartrazine, and the non-permitted dye metanil yellow on chromosomes of Allium cepa are reported. A significant increase in polyploid cells was observed in all cases. High doses of these dyes induced chromosome breaks and micronucleus formation. Although all dyes produced mitotic aberrations, metanil yellow and fast green FCF showed comparatively stronger clastogenic activity.

Effects of lead on root growth, cell division, and nucleolus of Allium cepa.

The effects of different concentrations of lead nitrate on root growth, cell division, chromosome morphology and nucleolus in root tip cells of Allium cepa were studied. The concentrations of lead nitrate (Pb(NO(3))(2)) were 10(-7), 10(-6), 10(-5), 10(-4), 10(-3) and 10(-2) m. It was observed that lead reduces root growth and causes mitotic irregularities, including c-mitosis, anaphase bridges, and chromosome stickiness. Also, interphase cells with micronuclei, irregularly shaped nuclei and nuclei with decomposed nuclear material were noticed. The c-mitotic effect reached its maximum in the meristem at above 10(-4) m Pb, when practically almost all the anomalous dividing cells are of this type. After treatment with Pb, there were many similar silver-stained particulate materials scattered in the nucleus in short, rounded meristem cells and in long, oblong root cap cells.

Allium sativum (garlic)--a natural antibiotic.

Allium sativum (garlic) has been recognized not only as a spice but also as a substance which exerts a control on microorganisms. Recent publications indicate that garlic extract has broad-spectrum antimicrobial activity against many genera of bacteria and fungi. The active component (allicin) has been isolated and characterized. Because many of the microorganisms susceptible to garlic extract are medically significant, garlic holds a promising position as a broad-spectrum therapeutic agent. Furthermore, garlic plants may also play an important role in the ecological control of pathogenic microorganisms in nature.

Short duration of the mitotic and endomitotic cell cycle in the heterochromatin-rich monocot Allium carinatum.

Fraction labeled mitoses curves and caffeine-induced binucleate cell populations, respectively, were used to determine the duration of the mitotic and endomitotic cycle in roots of Allium carinatum, a leak with a 2C DNA content of 22.4 pg and a high proportion of heterochromatin. The mitotic cycle lasts, on an average, 8.1 h, the endomitotic one 7.5 h. These relatively short times (with respect to the nuclear DNA content) are discussed in the view of a possible role of heterochromatin in shortening cell cycle duration and possible advantage of the endomitotic cycle for rapid cell and tissue growth.

Inhibition of protein synthesis during G2 induces irregular metaphase and anaphase in mitosis of root cells of Allium sativum L.

When partially synchronized root tip cells of Allium sativum L. (Garlic) were treated with cycloheximide during G2, some cells showed condensed chromosomes in arrested prophase nuclei. Other cells that were able to proceed into metaphase exhibited abnormal alignments of chromosomes at the metaphase plate, resulting in irregular anaphase and telophase movements of the chromosomes. Electron microscopy showed that kinetochores were poorly developed with only few kinetochore-to-pole microtubules attached. Other microtubules, probably mostly pole-to-pole microtubules, exhibited irregular pathways making the spindle appear disorganized. These observations seem to suggest that a protein (or proteins) necessary for kinetochore and spindle function are synthesized at a time in G2 similar to the synthesis of the mitotic proteins that are necessary for chromosome condensation and nuclear membrane breakdown. It is suggested that the protein(s) needed for normal spindle function are newly synthesized during each cell cycle and therefore belong functionally to the mitotic proteins.

Inhibitory effect of garlic (Allium sativum) on sodium transport in isolated toad skin.

The effect of an aqueous fraction from the bulbs of Allium sativum (GE) was investigated in toad skin. When added to the inner (serosal) solution, GE caused a maximal reversible reduction of the transepithelial potential difference and short circuit current of 38% and 45%, respectively. When added to the outer (mucosal) solution, the effect was only partially reversible. Isaacson's amiloride test showed that GE decreased sodium potential (ENa.) and sodium conductance (GNa.). The net Na+ flux decreased due principally to a fall in Na+ flux in the active direction. GE decreased Na(+)-K+ ATPase activity in vitro. Partial replacement of sodium by choline in the outer solution reduced the effect of GE on the skin and substitution of normal Ringer's solution with isethionate Ringer's solution in the outer solution significantly enhanced the effect of GE on the skin. These results indicate that GE decreases active Na transport in the toad skin.

Subcellular localization of copper in the root cells of Allium sativum by electron energy loss spectroscopy (EELS).

Ultrastructural investigation of the root cells of Allium sativum L. exposed to three different concentrations of Cu (1, 10 and 100 microM) for 9 days was carried out using transmission electron microscopy (TEM) and electron energy loss spectroscopy (EELS). The results presented here indicate that excess Cu induces ultrastructural changes such as strong vacuolization, condensed nuclear chromatin, decreased endoplasmic reticulum (ER) and ribosome and serious plasmolysis. EELS analysis indicated that electron dense granules containing Cu appeared in the cells after Cu treatment. The vacuoles of the root tip cells were the main Cu-accumulation site. Small amounts of copper were also localized to cytoplasmic vesicles or cell walls of cortical cells. The results of the present investigation have significant importance in further understanding the mechanisms of absorption, transportation and accumulation of heavy metals in plants grown in polluted soil.
