Early detection of radiation-induced glomerular injury by albumin permeability assay.

Renal irradiation leads predictably to glomerular vascular injury, cell lysis, matrix accumulation, sclerosis and loss of renal function. The immediate effects of renal irradiation that may be associated with glomerular pathology and proteinuria are not clear in the human disease or its rat model. We hypothesized that radiation-induced injury causes immediate and subtle alterations in glomerular physiology independent of the neurohumoral and hemodynamic regulatory mechanisms. We employed a sensitive in vitro functional assay of glomerular albumin permeability (P(alb)) to demonstrate radiation-induced damage to the glomerular filtration barrier immediately after total-body irradiation of rats. In blinded experiments, control rats were sham-treated, and experimental rats received 9.5 Gy X rays. Rats were killed humanely at 1 h to 9 weeks after irradiation and glomeruli were isolated. In parallel experiments, glomeruli were isolated from normal rats and irradiated in vitro. The change in glomerular capillary permeability due to an experimental oncotic gradient was determined using videomicroscopy and P(alb) was calculated. Results show that in vivo or in vitro irradiation of glomeruli caused an increased P(alb) at 1 h. Increased P(alb) was observed up to 3 weeks after irradiation. Glomeruli from mice irradiated with 9.5 or 19.0 Gy X rays did not show increased P(alb) at 1 h postirradiation. We conclude that glomerular protein permeability of irradiated rats increases in a dose-dependent manner immediately after irradiation and that it appears to be independent of hemodynamic or systemic influences.

Different responses of albumin-containing and alpha-fetoprotein-containing cells to estriol in the adult mouse liver.

A single intraperitoneal injection of 10 mg estriol (E3) rapidly induced a transient decrease in the serum albumin (ALB) level in 3 to 5 days in adult mice. At a dose of 5 mg, E3 showed a remarkable threshold effect on the decline of serum ALB. By immunohistochemistry, two types of ALB-positive cells were found in the liver. Type 1 cells were stained intensely and almost homogeneously throughout all or nearly all their entire cytoplasm, and type 2 cells showed only granular or vesicular cytoplasmic deposits. After administration of 10 mg E3, the type 1 cells diminished rapidly and markedly in direct proportion to the fall in serum ALB from the centro- and mediolobular zones, where they had been normally localized either singly or in small groups. Simultaneously, the granular or vesicular deposits that had been normally stained very weakly in the type 2 cells in the centro- and mediolobular zones became intensely stained along with cells in the perilobular zone. Alpha-fetoprotein (AFP)-containing cells in the liver appeared in maximal numbers at 5 to 7 days after E3 administration. These serological and cytological changes after E3 administration are here discussed on the basis of a possible inverse relationship between ALB and AFP gene expression.

Modulation of incipient glomerular lesions in experimental diabetic nephropathy by hypotensive and subhypotensive dosages of an ACE inhibitor.

A glomerular permeability defect occurs early in the course of type 1 diabetes and precedes the onset of microalbuminuria and renal morphological changes. Recently, ACE inhibitors have been shown to prevent loss of glomerular membrane permselective function, but the mechanism of this nephroprotective effect is still being debated. The objective of the present study was to evaluate the effects of hypotensive and subhypotensive dosages of the ACE inhibitor quinapril ex vivo and of its active metabolite quinaprilat in vitro on the glomerular albumin permeability (P(alb)) defect in the early phases of experimental diabetes. For the ex vivo study, six groups of male Wistar rats were evaluated for 4 weeks. One group served as a nondiabetic control (C); the other five groups were rendered diabetic and included untreated diabetic rats (D) and diabetic rats receiving quinapril at the dosages of 5 (DQ1), 2.5 (DQ2), 1.25 (DQ3), and 0.625 (DQ4) mg. kg(-1). day(-1). Dosage-dependent effects of quinapril on systolic blood pressure and the glomerular filtration rate were observed. In contrast, control of P(alb) in isolated glomeruli exposed to oncotic gradients, proteinuria, and glomerular and tubular hypertrophy was obtained with subhypotensive dosages (DQ3 and DQ4 groups) of the ACE inhibitor. In the in vitro study, quinaprilat reduced P(alb) significantly in concentration ranges from 10(-6) to 10(-14) mol/l compared with results in control glomeruli. The effect on P(alb) may have occurred by mechanisms different from kidney ACE inhibitor. These study results indicated that ACE inhibitor treatment prevents the early onset of the P(alb) defect in experimental diabetes. This effect seemed to occur independently of systemic or glomerular hemodynamic changes and, at least partially, from kidney ACE inhibition.

Detection of microalbuminuria. Receiver operating characteristic curve analysis favors albumin-to-creatinine ratio over albumin concentration.

OBJECTIVE: To compare albumin-to-creatinine ratio (ACR) and albumin concentration (ALB) for detection of microalbuminuria using precise data on CIs for sensitivity and specificity and to determine sex- and age-specific discriminator values for ACR. RESEARCH DESIGN AND METHODS: All consecutive patients investigated for the presence of microalbuminuria with timed overnight urine collection were included in the study. Results from 1.171 men and 1,223 women were eligible for evaluation. Albumin excretion rate (the reference procedure), ALB, and ACR were compared by receiver operating characteristic (ROC) curve analysis. Age- and sex-specific discriminator values were established. Sensitivity and specificity were determined, and their CIs were compared with those calculated from data presented in the literature. RESULTS: Sensitivity and specificity were 89% (range, 85-92) and 90% (88-92), respectively, for women, and 90% (87-93) and 89% (86-91), respectively, for men, at a discriminator value of 15 mg/l for the ALB. This discriminator value did not show any relationship to sex or age. For the ACR, which depends on sex, the sensitivity and specificity were 94% (92-96) and 92% (91-94), respectively, at 2.5 g/mol for women and 94% (92-97) and 93% (91-95), respectively, at 1.8 g/mol for men. The discriminator value for the ACR appears to depend on age in both men and women. CONCLUSIONS: ROC curve analysis clearly shows that the ACR performs better than the ALB in screening for microalbuminuria. However, in contrast to the ALB, the ACR needs sex- and age-specific discriminator values.

[Effect of alpha and beta-adrenergic agonists on methacholine-induced nasal secretion in man].

To examine the roles of alpha-adrenergic, beta-adrenergic and cholinergic agonists on the nasal secretion mechanism, the author repeated nasal methacholine (Mch, 1 time 20 mg) challenges ten times at 10-min intervals in 13 healthy male subjects. As it was difficult to obtain a sufficient amount of nasal secretion for analysis using simple nasal provocation tests with alpha-and beta-adrenergic agonists before repeated Mch challenges, alpha-adrenergic (phenylephrine, Phe, 2 mg) and beta-adrenergic (isoproterenol, Iso, 0.2 mg) agonists were administered into both nostrils. The effects of these agents on the volume of nasal secretions (Vol), the fucose volume (Fu, an index of glandular secretion) and the albumin volume (Alb, an index of vascular permeability) in the Mch induced nasal secretions were analyzed each time. Repeated Mch challenges revealed that the initial challenge produced the largest response as reflected by the Vol, Fu and Alb of the secretions, and by Vol and Fu production being almost constant during the 10 repetitions of Mch challenge at 10-min intervals. However, Alb demonstrated the greatest decrements. Phe administration before Mch challenges gave rise to Fu increasing more markedly while Alb was unchanged. On the other hand, that of Iso before, produced greater decreases in Vol, Fu and Alb levels. Therefore, it is suggested that repeated Mch challenge causes the maintenance of glandular secretion while stopping albumin permeability, that with pre-administration of alpha-agonists glandular secretion increases, and that pre-administration of beta-agonists inhibits glandular secretion. When both alpha- and beta-agonists are given before the challenge, however, there is no increase in vascular permeability.

[Observations of bone and incisor dentin of rabbits injected with rhodamine-labeled albumin by confocal laser scanning microscope].

The confocal laser scanning microscope (CLSM) is the most recently developed microscope using the confocal system and laser light source. This microscope is possible to create optical sectioning images from any depth of the specimen. In this study, the incorporation of rhodamine-labeled albumin (R-ALB) into the bone and incisor dentin was observed by CLSM. Male Japanese white rabbits were given an intravenous injection of R-ALB every two days for 10 days and sacrificed 3 days after the last injection. Transverse ground section of the tibia about 80 microns thickness showed the incorporation of orange-red fluorescence labeling into the endosteal surface as well as the Haversian system of bone. Each labeling line of R-ALB was clearly visible by CLSM compared with the normal fluorescence image. Three-dimensional reconstruction of the sectioning images showed the process of the incorporation of R-ALB into the bone and the complexity of the labeled pattern. R-ALB was incorporated into the incisor dentin and showed the periodic labeling pattern. The high resolution image of the periodic labeling pattern was observed by CLSM. These results indicated that CLSM is a valuable apparatus for studying the fluorescence labeling images of hard tissues.

[An intervention trial to postpone aging in competent elderly. Trial of nutritional improvement in the retirement home].

The purpose of this study was to evaluate the effectiveness of a comprehensive intervention program in changing dietary habits and nutritional status, for postponing aging in competent elderly. The intervention group (n = 44, 11 men, 33 women, mean age: 74.3 +/- 5.4 years) comprised residents of a retirement home in a suburb of Tokyo. The intervention program, which consisted of 82 lectures and practice sessions focusing mainly on improvement of life styles, was performed between May 1993 and May 1995. In addition, individualized dietary consultations were provided every week during the intervention period. The age and sex matched control group (n = 133, 33 men, 100 women) was randomly selected from participants of the community-based health examination in Tokyo. Changes in dietary habits were measured by a 15-food frequency questionnaire method. As parameters of nutritional status, serum albumin (ALB), total cholesterol (TC), HDL cholesterol (HDL-C) and body mass index (BMI) were simultaneously examined at pre- and post-intervention. The results obtained were as follows: 1) The mean score of Tokyo Metropolitan Institute of Gerontology Index of Competence (TMIG Index of Competence) was 11.3 +/- 1.6 in the intervention group, 12.0 +/- 1.6 in the control group, the competent level of functional capacity was equally high in both groups. 2) Food frequencies of meat, fruit, and fat and oil significantly increased in the intervention group. A significant increase in the food frequency of meat was seen in the control group. 3) Significant increases in ALB, HDL-C and BMI were consequently observed in the intervention group, whereas levels of ALB and BMI significantly decreased in the control group. 4) The number of times of participation in the comprehensive intervention program was significantly positively related to the change in ALB between pre- and post-intervention, after adjusted for age, sex, education, TMIG Index of Competence score and ALB level at baseline. These results indicate that the comprehensive intervention program is effective for postponing aging in the competent elderly.

The biological activity of ABA-1-like protein from Ascaris lumbricoides.

The elevation of non-specific IgE (total IgE) in Ascaris infection can be seen one week after infection, and reaches a peak after approximately two weeks. It has been reported that ABA-1 protein is the main constituent in the pseudocoelomic fluid of Ascaris suum. To investigate the effect of the ABA-1-like protein from Ascaris lumbricoides (ALB), the cDNA was cloned by reverse transcriptase polymerase chain reaction, using original primers based on the consensus sequences of ABA-1 and TBA-1, that is an ABA-1-like protein from Toxocara canis. The clone was sequenced, we constructed the recombinant polyprotein of ALB (rALB14 and rALB7) based on the ALB sequence, and rALB was administrated to BALB/c mice. Fourteen days after inoculation with rALB14 which is the full length of ALB, the elevation of total IgE which we supposed to contain non-specific IgE was observed, and the results were as we expected. Furthermore, in an in-vitro experiment, we confirmed that the spleen cells proliferated when stimulated by rALB14 and concanavalin A. Therefore, the whole conformation of ALB is considered to be involved in the elevation of non-specific IgE, and is involved in the activation of T cells.

[Detection of aflatoxin-albumin adducts in human serum and its application].

Exposure to aflatoxin B1 (AFB1) is one of the major risk factors in the etiology of human hepatocellular carcinoma (HCC). The development of highly sensitive and specific methods for detecting AFB1 and its macromolecular biomarkers are critical for identifying individuals at risk for hepatocellular carcinoma. The detection of aflatoxin-albumin adducts (AFB-ALB) in serum has been used to investigate the relationship between aflatoxin exposure and HCC. A more sensitive and reliable method to detect AFB-ALB in serum was modified by separating serum albumin with Microcon-50 microconcentrators in stead of using precipitation. The method could separate serum albumin quickly and make protein digested in the same container. Thus, the step for transferring samples was reduced and recovery rate was improved. The AFB-ALB adducts were then determined by radioimmunoassay. The AFB-ALB adducts of human serum samples from Qidong, Jiangsu Province, and Fusui, Guangxi Zhuang Autonomous Region of the People's Republic of China were detected. The results indicated that the exposure of AFB1 is common and the positive rate for AFB-ALB adducts in serum of these populations is high.

Validation and evaluation of biomarkers in workers exposed to benzene in China.

This study was conducted to validate biomarkers for early detection of benzene exposure and effect in 2 phases. The main purpose of phase 1 was to determine whether these biomarkers could reliably detect differences between workers with high exposure levels and unexposed subjects, which is the minimal screening criterion for a biomarker assay. Phase 2 of the study mainly focused on evaluating the exposure-response relation, confounding factors, and sensitivities of biomarkers for low benzene exposures. The Chinese occupational population studied had a broad range of benzene exposures. On the day of biological sample collection, exposures ranged from 0.06 to 122 ppm with a median exposure of 3.2 ppm. The median of the 4-week mean benzene exposures was 3.8 ppm, and the median lifetime cumulative exposure was 51.1 ppm-years. Compared with benzene levels in collected samples, toluene levels were relatively high, with a median of 12.6 ppm (mean, 26.3 ppm), but xylene levels were low, with a median of 0.30 ppm (mean, 0.40 ppm). The biomarkers evaluated were urinary metabolites S-phenylmercapturic acid (S-PMA*), trans,trans-muconic acid (t,t-MA), hydroquinone (HQ), catechol (CAT), and phenol; albumin adducts of benzene oxide and 1,4-benzoquinone (BO-Alb and 1,4-BQ-Alb, respectively) in blood; blood cell counts; and chromosomal aberrations. Blood cell counts in this population, including red blood cells (RBCs), white blood cells (WBCs), and neutrophils, decreased significantly with increased exposures but remained in normal ranges. Chromosomal aberration data showed significant increases of chromatid breaks and total chromosomal aberrations in exposed subjects compared with unexposed subjects. Among the urinary metabolites, the levels of S-PMA and t,t-MA were significantly elevated after benzene exposures. Both markers showed significant exposure-response trends even over the exposure range from 0 to 1 ppm. However, HQ, CAT, and phenol showed significant increases only for benzene exposure levels above 5 ppm. Multiple regression analyses of these urinary metabolites on benzene exposure indicated that toluene exposure, smoking status, and cotinine levels had no significant effects on urinary metabolite levels. A time-course study estimated the half-lives of S-PMA, t,t-MA, HQ, CAT, and phenol to be 12.8, 13.7, 12.7, 15.0, and 16.3 hours, respectively. Both BO-Alb and 1,4-BQ-Alb showed strong exposure-response associations with benzene. Regression analyses showed that after adjustment for potential confounding by smoking, there was still a strong association between benzene exposure and these markers. Furthermore, the analyses for correlations among biomarkers revealed that the urinary metabolites correlated substantially with each other. The albumin adducts also correlated well with the urinary biomarkers, especially with S-PMA. BO-Alb and 1,4-BQ adducts also correlated well with each other (r = 0.74). For benzene exposure monitoring, both S-PMA and t,t-MA were judged to be good and sensitive markers, which detected benzene exposures at around 0.1 ppm and 1 ppm, respectively. But S-PMA was clearly superior to t,t-MA as a biomarker for low levels of benzene exposure.

Distribution of albumin and alpha-fetoprotein mRNAs in normal, hyperplastic, and preneoplastic rat liver.

The nature of bile duct-like (oval) cells proliferating during chemical hepatocarcinogenesis has been controversial. To investigate this issue further, the authors compared the hepatic distribution of albumin (ALB) and alpha-fetoprotein (AFP) mRNAs in rats in which oval cell proliferation was induced by feeding a choline-devoid diet containing 0.1% ethionine (CDE, a hepatocarcinogenic diet) with that in normal rats and in rats in which biliary epithelial cell hyperplasia was induced by either bile duct ligation or feeding alpha-naphthylisothiocyanate (ANIT). Northern blot analysis in parenchymal and nonparenchymal liver cells isolated from these animals demonstrated that ALB mRNA was present in the hepatocytes of both control and experimental animals, whereas this transcript was detected in nonparenchymal epithelial cells only in CDE-fed rats. Alpha-fetoprotein mRNA was not seen in either parenchymal or nonparenchymal cells isolated from normal or hyperplastic livers induced by bile duct ligation or ANIT feeding. In CDE-fed rats, however, both parenchymal and nonparenchymal cell populations displayed AFP message. In situ hybridization directly demonstrated nonparenchymal cell expression of both ALB and AFP transcripts in CDE-fed rats. Most surprisingly, ALB and AFP mRNAs were also detected by in situ hybridization in occasional nonparenchymal cells located in portal tracts near the limiting plate in normal liver, as well as under conditions associated with bile duct hyperplasia. Immunohistochemical studies of intermediate filament proteins, cytokeratin 19 (a marker of glandular epithelia), vimentin (a marker of mesenchymal lineage), and desmin (a marker of muscle cell differentiation) demonstrated that oval cells, as well as normal and hyperplastic bile duct cells, were positive for cytokeratin 19 and negative for both vimentin and desmin. Cytokeratin-positive oval cells formed duct profiles and were connected to preexisting ductules and ducts. These results are construed to suggest that oval cells proliferating during CDE hepatocarcinogenesis are derived from epithelial cells within the biliary tree. The presence of cells with similar morphologic appearance, periportal location, and AFP and ALB expression in normal liver suggests that these cells may be the progenitors of oval cells induced by some carcinogenic regimens.

Hepatic differentiation from embryonic stem cells in vitro.

OBJECTIVE: To investigate an method for hepatic differentiation from embryonic stem cells (ES cells) in vitro and the resulting differentiation ratio, in order to develop a procedure for producing a new type of hepatocyte for hepatocyte replacement therapy in the treatment of liver failure. METHODS: ES cells from Balb/C mice were cultured and maintained in an undifferentiated state in gelatin-coated dishes using Dulbecco's modified Eagle's medium (DMEM) containing 1000 U/ml leukemia inhibitory factor (LIF). Then, LIF was withdrawn from the DMEM to allow the ES cells to develop into embryonic bodies (EBs). EBs were plated onto tissue culture dishes, and growth factors such as acidicfibroblast growth factor (aFGF) and hepatocyte growth factor (HGF) were added to the medium to promote directional differentiation. The course of development and differentiation was observed dynamically using an inversion microscope. The expression of hepatic proteins, such as alpha-fetoprotein (AFP), albumin (ALB), cytokeratin 8 (CK8), cytokeratin 18 (CK18), in cytoplasm was analyzed by immunocytochemistry (ICC). The concentration of ALB in the medium was determined dynamically by radioimmunoassay (RIA). RESULTS: ES cells replicated as clones, without differentiating, in DMEM containing LIF. They developed into EBs in medium without LIF. Our ICC assay showed that differentiating cells did not express hepatic proteins, such as AFP, ALB, CK8, and CK18 until day 7, day 9, day 11, and day 11, respectively (up to 2 days later when growth factors are not present). The concentration of AFP in the medium was first detected on day 8, at a concentration of 3.4 ng/ml, and increased to 22.8 ng/ml by day 15. The concentration of ALB in the medium was 0.2 micro g/ml on day 11, and increased to 2.2 micro g/ml by day 15. ALB-positive cells under ICC manifest morphological structures were consistent with normal mouse hepatocytes. The differentiation ratio of hepatocytes in the ES cell differentiation system was 30% on day 15 (significantly lower without the presence of growth factors). CONCLUSIONS: ES cells can differentiate into mature hepatocytes. Growth factors, such as aFGF and HGF, can enhance this differentiation and produce sufficient numbers of functional hepatocytes. This method may be a reliable new way of differentiating ES cells into hepatocytes for use in replacement therapy in the treatment of liver failure.

A novel method to synthesize polyaluminum chloride with a membrane reactor.

Al13 or Alb is usually regarded as the most efficient species of polyaluminum chloride (PAC), the performance flocculant for water treatment. This paper was intended to report a new method to synthesize PAC with high content Alb, by using the membrane reactor. NaOH solutions were managed to permeate slowly through the micropores of ultrafiltration membrane into AICl3 solutions under the suitable transmembrane pressure(TMP). Meanwhile NaOH drops size was limited to nano-scale, resulting in dramatical reduction of the characteristic diffusion time and great increment of contact interface between the strong base and Al ions in solution to favor the formation of Al(OH)4-, the precursor of Al13, so few precipitates and much Alb are produced. When the initial concentration of AlCl3/NaOH is 0.40/2.0 (mol/L), MWCO = 10000, TMP = 0.0085 MPa, T = 305 K and B (molar ratio of OH-/Al3+) = 2.25, the quantity of Alb attains about 80%. The results of 27Al-NMR determination showed that the Al13 content is equal to Alb content. And our PAC product has shown better flocculation effects than the commercial product.

[Extraction and characterization of soluble protein fractions from Phaseolus lunatus L seeds].

Legume proteins as a potential source of valuable nutrients, are the object of several studies in order to obtain the best use. A basic knowledge becomes more important for those proteins from species not wholly utilized, before using them as food ingredients. The objective of this work was to determine several structural and nutritional characteristics of the protein fractions from Phaseolus lunatus, separated in different solvents. The relative amount of extraction for the albumins (ALB), globulins (GLB), prolamines (PRL), and glutelins (GLT) was 62.3, 34.8, 1.4 and 1.5%, respectively. The SDS-PAGE electrophoretic profile of both ALB and GLB, showed seven common bands in intervals from 10 to 95 kDa, and 14 to 99 kDa, respectively; the amino acids profile showed that PRL was the rich fraction in sulfurated amino acids (11.5 g/100 g protein); the content of lysine in the fraction of ALB was smaller than expected but the requirement of the FAO in the fractions of GLB and GLT was covered. In general, the fraction of GLB had the best balance of amino acids and digestibility (80%); however, it had a relationship of calculated protein efficiency ratio (C-PER) of 0.11, smaller than the ratio in ALB (0.97). The calorimetric analysis showed denatured temperatures around 90 degrees C for the ALB, GLB, and GLU fractions. The PRL fraction probably did not present a thermal transition because the proteins were denaturalized by the extraction conditions.

[Role of clinical data in NST: from the viewpoint of the clinical laboratory].

Serum albumin (Alb) sampling is used for patients who object to screening by NST. As Alb is decreased by malnutrition, it is available for nutritional assessment, reflecting the long-term nutritive condition. Recently, Alb was correlated with nutritional assessment proteins with RTP, like RBP, TTR and Tf. We recently set up a screening line of Alb < or =3.0g/dl; however, we will gradually elevate it to < or =3.5g/dl. The role of the medical technologist is to estimate the number of patients with < or =3.5g/dl, and investigate correlations between the Alb concentration and other markers, including ChE, TTR, TP and CRP.

[Hepatocyte growth factor and fibroblast growth factor-4-induced differentiation of human bone marrow mesenchymal stem cells into hepatocyte-like cells in vitro].

OBJECTIVE: To induce the differentiation of human bone marrow mesenchymal stem cells (HMSCs) into hepatocyte-like cells with hepatocyte growth factor (HGF) and fibroblast growth factor-4 (FGF-4) in vitro. METHODS: HMSCs were induced to differentiate into hepatocyte-like cells by HGF (group B), FGF-4 (group C) and HGF+FGF-4 (group D) in vitro. Undifferentiated HMSCs and L-02 cells were used as the negative (group A) and positive (group E) controls, respectively. The changes of cell morphology were observed microscopically. The expressions of hepatic markers, alpha fetoprotein (AFP) and CK-18, were detected by immunocytochemical staining at different times after induction, and the differentiation ratios of the various groups of HMSCs were calculated on the basis of image analysis. The expressions of AFP and ALB were detected by immunofluorescence assay in each group at different times after induction, and the expressions of AFP and ALB mRNA by RT-PCR. RESULTS: HMSCs gradually transformed into spindle-shaped, round, polygonal or irregular cells after induction. Immunocytochemical staining revealed positive AFP and CK18 expressions in groups B, C, and D after induction as well as in group E. The positive units (PU) of AFP and CK18 in group D calculated according to image analysis were significantly higher than that of groups A, B, and C. The expressions of AFP and ALB detected by immunofluorescence were both positive after induction in all groups except group A, similar to the findings of the expressions of AFP and ALB mRNA by RT-PCR. CONCLUSION: HMSCs can be induced to differentiate into hepatocyte-like cells by HGF, FGF-4 and their combination at certain concentrations, and the hepatocyte-like cells can express some hepatic markers such as AFP, ALB, CK18, etc. HGF+FGF-4 may achieve more effective induction of HMSC differentiation into hepatocyte-like cells, and the efficiency of HGF is greater than that of FGF-4.

[Investigation on renal function of workers exposed to toluene and xylene].

beta 2-microglobulin (beta 2-M), albumin (Alb), IgG, Protein in urine were measured in 59 workers exposed to toluene, and xylene, and 70 workers not so exposed as control. The results showed that the urine levels of beta 2-M, Alb in exposed groups were significantly higher and increased progressively with increasing work time. Urinary beta 2-M showed significantly positive correlation with Alb, protein. Conformability of beta 2-M, Alb with other renal damage indexes seems better. It is suggested that toluene and xylene might cause renal damage, changes of urinary beta 2-M and Alb concentrations might be used as sensitive indexes for early detection of renal dysfunction in toluene and xylene workers.

[Pharmacokinetics of four benzimidazoles administered intragastrically to mice infected with secondary cysts of Echinococcus granulosus].

Mice infected with secondary cysts of Echinococcus granulosus were treated ig with mebendazole (Meb), albendazole (Alb), albendazole sulfoxide (AlbSO) or albendazole sulphone (AlbSP) at equal effective dosage. The blood concentrations-time courses of the four drugs were adequately fitted to a liner one-compartment open model analysed with a modified NONLIN program, and the major pharmaco- kinetic parameters were then calculated (Table 1). When Alb was given ig to the infected mice, 76.2-84.5% of the drug absorbed was metabolized to AlbSO. There fore, the pharmacokinetics of AlbSO in Alb group was determined. After ig administration of Meb to the infected mice, the ratio of the drug contents in cyst wall to corresponding plasma concentrations of Meb was 10.6-26.9%, but much less in cyst fluid. In Alb and AlbSO groups, similar AlbSO contents in cyst wall and cyst fluid with higher ratio to plasma concentrations were observed (Fig 3). The results suggest that E granulosus cysts were more susceptible to Meb than to Alb or AlbSO, and improvement of Meb absorption would be favourable for enhancement of efficacy in the treatment of hydatid disease.
