Effects of captopril chronic intake on the aerobic performance and muscle strength of normotensive trained subjects.

Fifteen normotensive athletes specializing in dynamic sports took part in a randomized double-blind and cross-over study: captopril (50 mg/24 h) vs placebo. Each treatment lasted one month. Maximal exercise tests on cycle ergometer were performed at the end of each period. No significant differences were observed in the maximal values of oxygen uptake, power, heart rate or blood lactate value. The anaerobic threshold, defined as the exercise intensity which corresponded to a 4 mmol.l-1 blood lactate level was unchanged. With captopril, the end-of-exercise systolic and diastolic BP were slightly altered (NS). Lower limb muscle strength, as explored with a Cybex isokinetic system, was not modified by captopril intake. The results indicate that maximal aerobic performance and isokinetic strength of the lower limbs are not altered by captopril chronic administration (50 mg/day) in normotensive trained subjects.

A novel model-free approach for reconstruction of time-delayed gene regulatory networks.

Reconstruction of genetic networks is one of the key scientific challenges in functional genomics. This paper describes a novel approach for addressing the regulatory dependencies between genes whose activities can be delayed by multiple units of time. The aim of the proposed approach termed TdGRN (time-delayed gene regulatory networking) is to reversely engineer the dynamic mechanisms of gene regulations, which is realized by identifying the time-delayed gene regulations through supervised decision-tree analysis of the newly designed time-delayed gene expression matrix, derived from the original time-series microarray data. A permutation technique is used to determine the statistical classification threshold of a tree, from which a gene regulatory rule(s) is extracted. The proposed TdGRN is a model-free approach that attempts to learn the underlying regulatory rules without relying on any model assumptions. Compared with model-based approaches, it has several significant advantages: it requires neither any arbitrary threshold for discretization of gene transcriptional values nor the definition of the number of regulators (k). We have applied this novel method to the publicly available data for budding yeast cell cycling. The numerical results demonstrate that most of the identified time-delayed gene regulations have current biological knowledge supports.

pH and H2O2 concentration of hydrogen peroxide disinfection systems.

Hydrogen peroxide disinfection has caused discomfort for some soft contact lens wearers even though the solution was neutralized properly. The discomfort could be due to the endpoint pH of the system used or the residual H2O2 concentration. Using a digital pH meter and H2O2 concentration detection strips, we measured the pH and H2O2 concentration of seven different hydrogen peroxide disinfection systems over their recommended disinfection and neutralization cycles to determine their time courses and endpoint values. The final pH of the seven systems varied from 6.15 to 7.74 pH units. All systems resulted in residual H2O2 concentrations below threshold values. These findings indicated that the final pH may be the cause of patient discomfort after hydrogen peroxide disinfection and neutralization.

Delayed pattern discrimination in patients with unilateral temporal lobe damage.

Behavioral and neurophysiological studies in macaque monkeys suggest a role of the inferior temporal cortex in pattern discrimination and visual memory. To determine whether this cortical area is also involved in human short-term visual memory, we measured spatial frequency discrimination thresholds for sequentially presented stimuli in 17 patients with unilateral, postoperative focal damage to the temporal cortex (11 left, 6 right hemisphere). These results are compared to those of 17 age-matched control subjects. Contrast detection thresholds and difference thresholds for spatial frequency were determined for spatially truncated sine wave gratings presented in the left and right visual fields. Detection thresholds were measured for sine wave gratings in a spatial two-alternative forced-choice procedure for three spatial frequencies [2.5, 5, and 10 cycles (c)/degree] for each hemifield. Discrimination thresholds were determined for two gratings sequentially presented either 4 degrees to the left or right of fixation. Grating contrast was five times the value of detection threshold and reference frequency was 5 c/degree. Within each trial, the gratings were separated in time by 1, 3, and 10 sec interstimulus intervals (ISIs), and subjects signaled which grating had the higher spatial frequency. The results indicate that (1) contrast detection thresholds overall did not differ between patient and control groups; (2) spatial frequency discrimination thresholds were, however, significantly elevated in patients and this elevation was significantly more pronounced in the visual field contralateral to the damaged hemisphere; and (3) patients with inferotemporal damage exhibited higher discrimination thresholds for the longest ISI, whereas patients with medial/superior temporal lobe damage did not show this effect. The results suggest that visual areas in human temporal cortex are involved in the higher visual processes underlying delayed pattern discrimination.

Validity of the heart rate deflection point as a predictor of lactate threshold concepts during cycling.

This paper examines the validity of the heart rate deflection point (HRDP) obtained with the "updated" Conconi test. Eleven male road cyclists performed 2 progressive incremental cycling tests and a 30-minute prolonged exercise test (PET). From the data obtained, comparisons were made and correlation coefficients were calculated between HRDP, the lactate threshold (LT), and the 3 mmol.L(-1) threshold (AT3). The PET at HRDP demonstrated whether or not a steady state in blood lactate concentration (BLaSS) could be maintained. Significantly lower values for power output (p < 0.01) and heart rate (HR) (p < 0.01) were found at LT compared with HRDP. No differences were found between HRDP and AT3. Only a moderate correlation for power output between HRDP and AT3 (rs = 0.69; p < 0.05) could be observed. During the PET, only 6 out of 11 cyclists reached the target time of 30 minutes, and only 4 cyclists maintained a BLaSS. We conclude that the updated Conconi test is not a valid method for assessing LT or AT3. Therefore, this method seems not suitable to evaluate endurance performance and prescribe exercise intensities in road cycling.

Menstrual cycle-dependent changes in visual field analysis of healthy women.

PURPOSE: To determine the effect of menstrual cycle phases on the visual field analysis of healthy females. MATERIAL AND METHODS: One randomly selected eye each of 59 healthy normally menstruating women, and of 54 men with no systemic and ocular problems, other than refractive error, were included in the study. Subjects underwent complete ocular examination, and standard achromatic perimetric (SAP) and short-wavelength automated perimetric (SWAP) analysis in both follicular (7th to 10th day of the cycle) and luteal phases (days 3-7 before the menstrual bleeding) of the menstrual cycle. Visual field analysis was performed using a Model 750 Humphrey Field Analyzer II (Humphrey Instruments Inc., San Leandro, Calif., USA) with full-threshold, central 30-2 program. Visual fields were divided into four regions as superior temporal, inferior temporal, superior nasal and inferior nasal, respectively. RESULTS: The mean age of female (n = 59) and the male subjects (n = 54) were 34.6 +/- 2.9 and 35.0 +/- 2.7 years, respectively (p = 0.49). SWAP tests demonstrated a significantly decreased mean MS value in the luteal phase (p < 0.05). However, it did not change significantly with SAP tests. Regional MS values of both SAP and SWAP tests were not different in both phases of the menstrual cycle (all p values >0.05). Mean perimetric test durations obtained with both SAP and SWAP were not different throughout the menstrual cycle (both p values >0.05). CONCLUSION: Clinicians should verify menstrual status when evaluating a suspected loss of visual field sensitivity in menstruating women. The findings of the present study suggest that the SWAP test may be more sensitive to determine subtle sex hormone-dependent changes in visual field analysis of healthy women.

Effects of dobutamine on critical capillary PO(2) and lactic acidosis threshold in patients with cardiovascular disease.

BACKGROUND: Muscle capillary PO(2) has been found to reach a minimal value, ie, a critical capillary PO(2), in the midrange of work capacity in patients with cardiovascular disease. However, it is not known if the critical capillary PO(2) can be influenced by a change in blood flow response to exercise. This study was carried out to determine the effect of changing the blood flow response to exercise, using low-dose infusion of dobutamine, on muscle end-capillary PO(2) (as approximated by femoral vein PO(2)), lactate concentration, oxygen uptake (O(2)), and the relation among these variables. METHODS: Eleven male patients with coronary artery disease performed an incremental exercise test on a cycle ergometer with and without continuous infusion of dobutamine, 6 microg/kg/min. Respiratory gas analysis was performed on a breath-by-breath basis; femoral vein blood was sampled every minute through a percutaneous catheter. RESULTS: Dobutamine increased resting O(2) and O(2) at the lactic acidosis threshold (LAT) but not peak O(2). The femoral vein PO(2) rapidly decreased toward a minimal value with increasing work rate (O(2)) irrespective of the infusion of dobutamine. After reaching its nadir (critical PO(2)), femoral vein lactate began to increase without further decrease in PO(2). Infusion of dobutamine significantly increased femoral vein resting PO(2) (27.4 +/- 4.9 mm Hg vs 32.5 +/- 3.8 mm Hg) and critical PO(2) (20.5 +/- 1.5 mm Hg vs 21.9 +/- 1.7 mm Hg), but not the PO(2) at peak O(2) (22.1 +/- 3.3 mm Hg vs 22.0 +/- 2.9 mm Hg). CONCLUSIONS: Infusion of dobutamine was found to raise the critical PO(2) and LAT but not peak O(2). These findings suggest that some of the acute increase in blood flow induced by dobutamine infusion benefits exercising muscle by increasing capillary PO(2), thereby delaying the onset of lactic acidosis.

Comparison of spontaneous and contraceptive menstrual cycles on a visual discrimination task.

21 women with spontaneous ovulatory menstrual cycles, and 7 women who were regular users of oral contraceptives were investigated over two consecutive menstrual cycles. There two groups were compared on their two flash threshold performance and on mood ratings. It was found that, for women with ovulatory menstrual cycles, visual sensitivity was enhanced during the late follicular phase of the cycle, as ovulation approached. At other phases of the menstrual cycle (paramenstrual and luteal) visual sensitivity remained constant and comparable to the values found in women who were taking to the values found in women who were taking contraceptives. Women who were taking contraceptives showed no significant variation in visual sensitivity with phase of the menstrual cycle. The overall level of anxiety, as reflected by mood ratings was higher in women with spontaneous cycles compared to those taking contraceptives. Neither group, however, showed significant variation in anxiety with phase of the cycle.

3D-reconstruction of coronary arteries in view of flow measurement.

An angiographic method for measurement of absolute coronary blood flow in patients was developed. It is based on 3D-reconstruction of the left or right coronary tree from digitized biplane coronary cineangiograms. The apparatus is presently composed of a 35 mm cinefilm projector with a video camera and a 512 x 512 x 8 bits image array processor controlled by a VAX-11/750 computer. First, the parameters of the two angiographic projections are determined in form of two 4 x 3 matrices from a pair of cineframes showing a 4 cm cube bearing markers. The cube is filmed after the coronary injection, with unchanged geometric configuration. The coronary arteries of interest are then 3D-reconstructed from a pair of cineframes showing them fully opacified. This allows to compute the intravascular volumes needed for flow determination. In vitro experiments showed that the obtained volume are reasonably accurate. For the measurement of coronary flow, the concentration of contrast medium along the involved arteries is computed (in arbitrary units) from two cineframes taken one (or two) cardiac cycle after onset of the injection. This yields a 'concentration-distance' curve per artery. The volume of fluid (contrast medium mixed with blood) which flows into the arteries during this (or these two) cardiac cycle is determined by applying a concentration threshold to the obtained concentration-distance curves. Measurements performed on a constant flow model were satisfying. Preliminary measurements in 12 patients showed that flow values obtained angiographically into the left anterior descending coronary artery correlate well with the values measured simultaneously by thermodilution in the great cardiac vein (Qangio = 0.83 x Qthermo + 16.1 ml, r = 0.87, n = 29).

Do differences between metal body residues reflect the differences between effects for Chironomus riparius exposed to different sediments?

Sediment characteristics are well known to interfere with toxicity, mainly through differences in terms of bioaccumulation. Here, with chironomids exposed to zinc in an artificial and a field sediment, we investigated the differences of zinc accumulation and of effects on the life cycle, at individual and population level. We used biology and energy-based modeling to analyze the data at all the levels of biological organization. This permits a reliable estimation of thresholds values for tissue residues. Differences in zinc tissue residues accounted for most of the differences between the results for both sediments (a factor of 11 for differences from 20 to 100 depending on the parameter which is considered). Taking into account accumulation and background variability, the differences relative to thresholds could be accounted for. However, it appeared that, once the threshold was passed, effects were much more pronounced for organisms exposed to artificial sediment compared to field sediment. We concluded that some sediment characteristics can enhance toxicity, in addition to their influence on the compound accumulation, even if the latter was the major source of differences in our study.

Relationship between the outcomes of assisted reproductive techniques and sperm DNA fragmentation as measured by the sperm chromatin structure assay.

OBJECTIVE: To investigate how moderate and/or high levels of DNA fragmentation (DFI), as measured by the sperm chromatin structure assay (SCSA), affect either IVF or IVF with intracytoplasmic sperm injection (ICSI) fertilization, cleavage, blastulation, implantation, and pregnancy. DESIGN: Retrospective clinical study. SETTING: Academic human reproduction laboratory. PATIENT(S): Eighty-nine couples undergoing IVF with conventional fertilization or ICSI. INTERVENTION(S): Sperm chromatin structure assay testing (SCSA) of semen aliquot taken from ejaculate used for assisted reproductive technology (ART). MAIN OUTCOME MEASURE(S): Related DFI to conventional semen parameters and cycle-specific outcomes after ART. RESULT(S): No patients achieved clinical pregnancy if SCSA values exceeded the DFI (27%, P<.01), moderate DFI (15%, P<.01), or high DFI (15%, P<.05) thresholds. Dividing the DFI sperm population into moderate-fragmentation and high-fragmentation categories did not improve the prognostic value of the SCSA. No coefficient of determination (r(2)) between SCSA parameters and conventional parameters exceeded 0.29. CONCLUSION(S): Sperm chromatin structure assay identified thresholds for negative pregnancy outcome after ART not identified using conventional semen parameters. This is the first study analyzing the clinical value of sperm DFI to [1] include a large number of ART patients (n = 89), [2] perform SCSA analysis on a semen aliquot from the ejaculate used for ART, and [3] examine how the extent (moderate and high DFI) of DFI influenced ART outcomes.

[Effects of swimming with a wet suit on energy expenditure during subsequent cycling].

The aim of this study was to investigate the effects of swimming with a wetsuit on energy expenditure during subsequent cycling. Nine well-trained triathletes underwent three submaximal trials. The first trial (SC) consisted of a 750-m swim realised at a competition pace, followed by a 10-min cycling exercise at a power output corresponding to the ventilatory threshold . The two other trials were composed of the same cycling exercise, preceded either by a 750-m swim with a wetsuit (WSC) or by a cycling warm-up (Ctrl). The main results are that the WSC trial was characterised by significantly lower swimming cadence (-14%), heart rate (-11%), and lactate values (-47%) compared to the SC trial, p < 0.05. Moreover, cycling efficiency was significantly higher in the WSC trial compared to the SC trial (12.1% difference, p < 0.05). The lower relative intensity observed during swimming with a wetsuit suggest the relative importance of swimming condition on the total performance in a sprint triathlon.

FSH and the control of follicular growth.

During 70 days or so from the time of recruitment until just before the beginning of the cycle during which a follicle is destined to ovulate, folliculogenesis is a continuous process dependent on gonadotrophins but independent of the fluctuations in their concentrations occurring during this time. For follicle growth to continue beyond the 2-4 cell antral stage FSH concentrations must rise above a certain threshold level at the correct time. Once threshold levels are exceeded a single follicle becomes dominant suppressing FSH concentrations to subthreshold values. The period during which FSH concentrations are above threshold can be imagined as a gate through which a follicle must pass if it is to ovulate.

Motor unit activity during long-lasting intermittent muscle contractions in humans.

Changes accompanying long-lasting intermittent muscle contractions (30%-50% of the maximal) were investigated by tracing the activity of 38 motor units (MU) of the human biceps brachii muscle recorded from fine-wire branched electrodes. The motor task was a continuous repetition of ramp-and-hold cycles of isometric flexion contractions. During ramp-up phases a significant decline in recruitment thresholds was found with no changes in the discharge pattern. During ramp-down phases the unchanged mean value of derecruitment thresholds during the task was accompanied by increased duration of the last two interspike intervals (ISI). These findings would suggest that during fatigue development the main compensatory mechanism during ramp-up contractions is space coding while for ramp-down contractions it is rate coding. During the steady-state phases the mean value of ISI, as well as the firing variability. had increased by the end of the task in most of the MU investigated . In addition 17 recruited MU were also investigated. These units revealed a lower initial discharge rate and a faster decrease in the mean discharge rate with the development of fatigue. The gradual reduction of the recruitment threshold of already active MU and the recruitment of new units demonstrated an increased excitability of the motorneuron pool during fatigue. A typical recruitment pattern (a first short ISI followed by a long one) was observed during ramp-up contractions in units active from the very beginning of the task, as well as during sustained contractions at the onset of the stable discharge of the additionally recruited MU.

Substrate oxidation during incremental arm and leg exercise in men and women matched for ventilatory threshold.

The aim of this study was to determine the variations in substrate utilization between men and women matched for ventilatory threshold (Tvent) during incremental arm cranking and leg cycling exercise at 70, 85, 100 and 115% of the mode-specific Tvent. Recreationally active men (n=12) and women (n=10) with similar values for percentage of peak oxygen consumption at Tvent participated in the study. Ventilatory equivalence, excess CO2 and modified V-slope methods were used concurrently to determine Tvent. The participants performed 5 min of exercise at each of 70, 85, 100 and 115% Tvent during both arm cranking and leg cycling exercise. The females were tested during the early follicular phase for all trials. A two-way mixed-design analysis of variance was performed to test for differences between the sexes. When carbohydrate and fat oxidation were expressed relative to total fat-free mass, carbohydrate oxidation during arm cranking and leg cycling was significantly higher in men than women at each percentage of Tvent. In contrast, women showed significantly higher fat oxidation across intensities during both arm cranking and leg cycling. Our results suggest that when substrate utilization is expressed relative to total fat-free mass, women appear to maintain a higher rate of fat and lower rate of carbohydrate oxidation than men during both incremental arm cranking and leg cycling exercise relative to Tvent.

Quantitative gene expression of TGF-beta1, IL-10, TNF-alpha and Fas Ligand in renal cortex and medulla.

BACKGROUND: Inflammatory, fibrogenic or apoptotic processes in the kidney are regulated by intra- and intercellular mediators. Intrarenal upregulation of genes may precede structural changes by days and can be examined in extremely small amounts of tissue. With the advent of new quantitative PCR methods results of gene expression are available within few hours after kidney biopsies. METHODS: In order to establish reference values for intrarenal gene expression of TGF-beta1, IL-10, TNF-alpha and Fas Ligand in renal cortex and medulla, we analysed 28 histologically normal kidney samples available after tumour nephrectomy by quantitative real-time PCR. After reverse transcription of isolated RNA, cDNA aliquots were quantified for target genes using the threshold cycle (C(t)) method normalized for the house keeping gene GAPDH. RESULTS: Expression of target genes was lower in cortex as compared to medulla, but the differences were only significant for IL-10 (P=0.0125). TGF-beta1 was found with the highest gene expression about five PCR-cycles (delta C(t)) after GAPDH with markedly lower results for TNF-alpha (delta C(t) approximately 9), IL-10 (approximately 12) and Fas Ligand (approximately 14). CONCLUSIONS: These results are the first reported reference values for human intrarenal gene expression which should facilitate interpretation of data from native or transplant kidneys in future studies.

Skeletal structural adaptations to mechanical usage (SATMU): 4. Mechanical influences on intact fibrous tissues.

This paper proposes that the growth in length of living fibrous tissue structures (tendon, ligament, fascia) responds primarily to circulating systemic rather than mechanical factors. However, growth of the thickness of those structures responds primarily to their mechanical tension loads in the special sense that, when the tissue's typical peak mechanical strains exceed a threshold value, its cells begin to add new collagen to increase its thickness, strength, and tension stiffness. When subsequent peak strains reduce to the threshold value, then further additions of collagen stop. That process defines mechanically controlled modeling of fibrous tissues. The collagen in these tissues can also develop mechanical microdamage (MDx) under repeated tension load-deload cycles. Special maintenance mechanisms normally repair that MDx to prevent accumulations that would threaten structural integrity. As a result, spontaneous complete ruptures of these structures can happen when MDx production exceeds its repair. These maintenance mechanisms also prevent gradual stretching under continuous tension loads, a process the author suggests calling creep compensation. When the creep compensation mechanism becomes incompetent, structures can stretch under continuous loads; when it becomes overactive, contractures can occur. The above meld of fact and inference provides the kernel of a general theory for the responses of the architecture and mechanical competence of intact fibrous tissues to mechanical usage.

Peak oxygen consumption and lactate threshold in full mask versus mouth mask conditions during incremental exercise.

Respirator masks vary in inhalation and exhalation resistance, and in dead volume. It is believed that these factors may contribute significantly to an early anaerobic threshold in mask wearers during maximal exercise. Very little is known concerning the effect of respirator masks on maximal oxygen consumption (VO2max) and the lactate threshold (LT). The purpose of the present study was to assess peak VO2 (VO2peak), LT and the ventilatory threshold (VT) of 14 experienced cyclists performing two maximal cycle exercise protocols while wearing a full respirator mask (FM) (M17 type) and a mouth mask (MM). VO2peak was 10% lower under FM conditions. Peak values for ventilation (VEpeak), respiratory rate (fbpeak) and tidal volume (VTpeak) were all significantly lower under with FM versus MM conditions. Performance time and maximal heart rate (fcpeak) were not different between mask conditions. The LT and VT when expressed in %VO2peak, and the lactate concentration (mmol.l-1) at LT and VT were not significantly different across mask conditions. Bland-Altman plots demonstrated longer inhalation times, decreased Fr values and greater oxygen extraction under FM conditions. Thus, perhaps due to the increased inhalation resistance of the FM condition, subjects were unable to attain their "normal" VO2 despite similar performance times and maximal fc. Furthermore, despite a diminished VO2peak with FM, LT and VT appeared to be the same as with a MM.
