Plant regeneration from hairy-root cultures transformed by infection with Agrobacterium rhizogenes in Catharanthus roseus.

Hypocotyl explants of Catharanthus roseus produced hairy roots when cultured on Murashige and Skoog (MS) basal medium after infection by Agrobacterium rhizogenes. Explants gave rise to adventitious shoots at a frequency of up to 80% when cultured on MS medium supplemented with 31.1 microM 6-benzyladenine and 5.4 microM alpha-naphthaleneacetic acid. There was a significant difference in the frequency of adventitious shoot formation for each hairy-root line derived from a different cultivar. Plants derived from hairy roots exhibited prolific rooting and had shortened internodes. Approximately half of the plants had wrinkled leaves and an abundant root mass with extensive lateral branching, but otherwise appeared morphologically normal. Plants with hairy roots that were derived from the cultivar Cooler Apricot developed flowers with petals that were white in the proximal region, whereas the wild-type flower petals are red. PCR and Southern blot analyses revealed that plants derived from hairy roots retained the Ri TL-DNA.

Inorganic salts influence IAA ionization and adventitious rhizogenesis in Pongamia pinnata.

The basal cut end of coppice shoot cuttings of Pongamia pinnata was treated for 24 h with 0 (water treated control) or 5.0 mmol/L of KMnO4, KCI, and KH2PO4 or 2.5 mmol/L of K2HPO4 and K2SO4. Inorganic salts of P, S, Cl and Mn significantly influenced IAA ionization and adventitious rhizogenesis. P and S salts had lower IAA ionization potential, but more pronounced effect on adventitious rhizogenesis than Cl and Mn salts. The linear regression analysis also established negative correlations between salt induced IAA ionization with various characteristics of adventitious rhizogenesis such as sprouting (r = -0.83, p < 0.05), rooting (r = -0.82, p < 0.05), root number (r = -0.95, p < 0.01), and root length (r = -0.80, p < 0.1). The implication of IAA ionization in adventitious rhizogenesis has been discussed and the possible role of inorganic salts therein suggested.

Mevalonate kinase activity during different stages of plant regeneration from nodular callus cultures in white pine (Pinus strobus).

Mevalonate kinase (MK) catalyzes a step in the isoprenoid biosynthetic pathway, which leads to a huge number of compounds that play important roles in plant growth and development. Here, we report on changes in MK activity in white pine (Pinus strobus L.) during plant regeneration by adventitious shoot organogenesis from cotyledons of mature embryos, including nodular callus induction, shoot formation and rooting. Nodular calli were induced from Pinus strobus (PS) embryos by culture in nodular callus induction medium in a 0-, 8- or 16-h photoperiod. Mevalonate kinase activity peaked in nodular calli after three weeks of culture on nodular callus induction medium in a 16-h photoperiod, whereas frequency of nodular callus formation peaked after 4 weeks of culture on nodular callus induction medium in darkness. During adventitious shoot formation, MK activity peaked in shoots derived from dark-grown nodular calli after 3 weeks on bud formation medium, and frequency of shoot formation was highest in dark-grown nodular calli cultured on bud formation medium for 4 weeks. During rooting, MK activity peaked 2 weeks after transfer of adventitious shoots to rooting medium and rooting frequency was highest in adventitious shoots after 3 weeks on rooting medium. Although during nodular callus induction in darkness MK activity was inversely related to frequency of nodular callus formation, MK activity was highly correlated with frequency of shoot formation and with rooting frequency. The observed increase in MK activity preceding rooting suggests that MK could serve as a marker for rooting of white pine shoots in vitro.

Transformation of Antirrhinum majus L. by a rol-type multi-auto-transformation (MAT) vector system.

A total of 11 independent beta-glucuronidase (GUS) positive hairy roots were induced following co-cultivation of leaf explants of Antirrhinum majus L. with Agrobacterium tumefaciens strain GV2260 containing rol-type multi-auto-transformation (MAT) vector pNPI702. A total of 326 adventitious shoots were regenerated from the hairy root lines on 1/2 MS medium without plant growth regulators at 25 degrees C under a 16 h/day photoperiod condition 4 months after infection of the A. tumefaciens GV2260. The absence of the rol genes in five plants was verified by polymerase chain reaction (PCR) and Southern blot analysis. Acclimatized transformants exhibited normal phenotypes in height and in the morphology of leaves and flowers. Furthermore, the GUS gene was strongly expressed in the leaves, inflorescence of the transformed plant, and the progeny. This result demonstrates that the rol-type MAT vector can be used to study gene functions controlling the morphogenesis of Antirrhinum majus plants.

Organogenesis in in vitro cultures of embryonic shoots of Abies balsamea (balsam fir).

Embryonic shoots of 15- to 20-year-old Abies balsamea (balsam fir) trees were soaked in (a) water for 15 min or 24 hr and (b) water with 1000 mg per 1 indolebutyric acid (IBA), N-dimethylaminosuccinamic acid (Alar-85), or 1-phenyl-3-methyl-5-pyrazolone (PPZ), singly or in combination with 100 mg per 1 caffeic acid, for 15 min. After the soaking, the embryonic shoots were transferred to a nutrient medium. Nonsoaked (control) embryonic shoots elongated and often formed a basal callus but never showed organogenesis. The soaked embryonic shoots formed new apical buds, with or without bud scales, adventitious dwarf needles or shoots, and root- and embryo-like structures. One of the embryos germinated and formed an irregular shoot. No differences were found between the various soak treatments, except that the 15-min water soak was ineffective. The 24-hr water soak was as effective as the 15-min growth regulator treatments.

Plant regeneration from protoplasts isolated from embryogenic calli of the forage legume Astragalus melilotoides Pall.

An efficient and reproducible protocol is described for the regeneration of Astragalus melilotoides protoplasts isolated from hypocotyl-derived embryogenic calli. Maximum protoplast yield (11.74 +/- 0.6x10(5)/g FW) and viability (87.07 +/- 2.8%) were achieved using a mixture of 2% (w/v) Cellulase Onozuka R10, 0.5% (w/v) Cellulase Onozuka RS, 0.5% (w/v) Macerozyme R10, 0.5% (w/v) Hemicellulase, and 1% (w/v) Pectinase, all dissolved in a cell protoplast wash (CPW) salt solution with 13% (w/v) sorbitol. First divisions occurred 3-7 days following culture initiation. The highest division frequency (9.86 +/- 0.68%) and plating efficiency (1.68 +/- 0.05%) were obtained in solid-liquid medium (KM8P) supplemented with 1.0 mg/l 2,4-dichlorophenoxyacetic acid, 0.5 mg/l 6-benzylaminopurine (BA), 0.2 mg/l kinetin, 0.2 M glucose, 0.3 M mannitol and 500 mg/l casein hydrolysate. Upon transfer to MS medium with 0.5 mg/l alpha-naphthaleneacetic acid and 1-2 mg/l BA, the protoplast-derived calli produced plantlets via somatic embryogenesis (56.3 +/- 4.1%) and organogenesis (21.6 +/- 0.6%). Somatic embryos or adventitious shoots developed into well-rooted plantlets on MS medium without any plant growth regulators or supplemented with 3.0 mg/l indole-3-butyric acid, respectively. About 81% of the regenerants survived in soil, and all were normal with respect to morphology and growth characters.

[Atresia of the right coronary ostium associated with annulo-aortic ectasia--a case of successful surgical repair].

A case of a 51-year-old male with atresia of the right coronary artery and annulo-aortic ectasia is described. He presented with heart failure and underwent open-heart surgery. Preoperative aortography did not show the ostium of the right coronary artery. At surgery the right coronary ostium was not found on the intimal surface of the aneurismal aortic wall. A fine cord was attached to the adventitial surface of the anterior aortic root. A modified Bentall's procedure was performed to reconstruct the left coronary artery alone. The post-operative course was uneventful. It is speculated that the hypoplastic ostium of the right coronary artery was pulled up and formed a flap-like closure with increased expansion of the aorta.

Enhanced bud and bulblet regeneration from bulbs of Nerine sarniensis cultured in vitro.

Nerine (Nerine sarniensis) cv. Salmon Supreme in vitro-grown bulblets, 7-9 mm in diameter, were cut in half longitudinally and used for adventitious bud initiation following dissection of the roots and two-thirds of the upper part of the bulblets. The terminal apex was injured with a hot, sterile microscope dissecting needle. The highest number of buds formed (seven to nine buds per halved bulblet) on a semi-solid Murashige and Skoog (MS) basal salts medium supplemented with 3% sucrose and either 1 microM 6-benzylaminopurine (BA) and 1 microM alpha-naphthaleneacetic acid (NAA) or 0.5 microM BA and 0.1 microM NAA. Bulblet halves were cultured in the dark for 11-13 weeks with one subculture after 6 weeks. Anatomical studies indicated that the initiation of adventitious buds on the abaxial side of the inner scales of the halved bulblet was adjacent to the basal plate and started from the leaf primordium and a meristematic bulge. Buds developed directly into small bulblets after they were transferred to semi-solid MS basal salts medium supplemented with 6% sucrose, 10 microM indole-3-butyric acid (IBA) and 0.25% activated charcoal. Small bulblets cultured in liquid MS medium supplemented with additional KH(2)PO(4 )(170 mg l(-1)), 6% sucrose and 0.1 microM NAA under a 16/8-h (light/dark) photoperiod for 8 weeks grew into larger bulbs faster than those cultured on semi-solid medium. The bulbs were rooted on a semi-solid medium after 4 weeks and then transferred to the soil. As many as 18 bulblets developed and rooted from one in vitro-grown bulb after 25-27 weeks.

Evidence that direct DNA uptake through cut shoots leads to genetic transformation of Solanum aviculare Forst.

The reporter genes GUS, NPTII and BAR, either separately or in combination, have been exploited to determine if DNA which can directly enter plants, circulate within the plant and enter nuclei, can also integrate into the genome in a manner which will permit gene expression. Feeding of either seed-derived or adventitious cut shoots of Solanum aviculare with the GUS gene followed by rooting of the shoots and growing on, resulted in all tissues of the plant showing GUS activity as detected cytochemically. Southern blot analysis of plants derived from the adventitious shoots confirmed the presence of the reporter gene in roots. Reporter gene expression was observed also in the F1 generation. If GUS and NPTII or GUS, NPTII and BAR were fed together, then in each case it was possible to have both expression and Southern blot confirmation of each of the genes. There was a relatively high rate of transformation of approximately 5% of the fed stems across all experiments conducted during the present study.

Plant regeneration from epicotyl explant of Jatropha curcas.

In vitro epicotyl explants from Jatropha curcas were cultured on MS medium with indole-3-butyric acid (IBA) and 6-benzyladenine (BA). Adventitious buds were directly induced from the surface of epicotyl explants under the condition with the combinations of IBA 0.1 mg/L and BA 0.2-0.7 mg/L. Of which IBA 0.1 mg/L and BA 0.5 mg/L induced the highest regeneration frequency. Shoot regeneration from callus required the combinations of IBA 0.5 mg/L with BA 0.1 mg/L, IBA 0.5 mg/L with BA 0.2 mg/L and IBA 1.0 mg/L with BA 0.5 mg/L, and the most suitable combination was IBA 1.0 mg/L and BA 0.5 mg/L. The health adventitious buds and regenerated shoots could be rooted on growth regulator-free MS medium. Regenerated plants with well developed shoots and roots were successfully transferred to greenhouse, without visible detectable variation.

Genotypic control of high-frequency adventitious shoot regeneration via somatic organogenesis in loblolly pine.

Mature zygotic embryos of 24 genotypes of loblolly pine (Pinus taeda L.) were used as explants to establish an adventitious shoot regeneration system through somatic organogenesis. Callus formation frequencies of 18.2 (genotype 11-1103) -77.7% (genotype 7-100) have been induced from mature zygotic embryos of all genotypes tested on callus induction medium (basal salts) containing 2,4-dichlorophenoxyacetic acid (2,4-D) or alpha-naphthaleneacetic acid (NAA), 6-benzyladenine (BA), and kinetin. Adventitious shoot regeneration via organogenesis with the frequency of 5.4 (genotype 11-1103 and 7-2) -77.2% (genotype 8-1082) was obtained from callus and tissue cultures derived from mature zygotic embryos of 24 genotypes of loblolly pine. The highest mean number of 18 adventitious buds per piece of callus 0.5x0.5 cm(2) in size was obtained from genotype 8-1082. Elongation of adventitious buds was achieved on TE medium supplemented with 0.5 mg/l indole-3-butyric acid (IBA) and 1 mg/l BA. After rooting, regenerated plantlets were established in soil. These results suggested that adventitious shoot regeneration via somatic organogenesis was influenced by the genotypes. The in vitro regeneration procedure established in this investigation could be used for clonal micropropagation of some genotypes of loblolly pine, as well as for establishing a transformation system in coniferous species.

Nod factor requirements for efficient stem and root nodulation of the tropical legume Sesbania rostrata.

Azorhizobium caulinodans ORS571 synthesizes mainly pentameric Nod factors with a household fatty acid, an N-methyl, and a 6-O-carbamoyl group at the nonreducing-terminal residue and with a d-arabinosyl, an l-fucosyl group, or both at the reducing-terminal residue. Nodulation on Sesbania rostrata was carried out with a set of bacterial mutants that produce well characterized Nod factor populations. Purified Nod factors were tested for their capacity to induce root hair formation and for their stability in an in vitro degradation assay with extracts of uninfected adventitious rootlets. The glycosylations increased synergistically the nodulation efficiency and the capacity to induce root hairs, and they protected the Nod factor against degradation. The d-arabinosyl group was more important than the l-fucosyl group for nodulation efficiency. Replacement of the 6-O-l-fucosyl group by a 6-O-sulfate ester did not affect Nod factor stability, but reduced nodulation efficiency, indicating that the l-fucosyl group may play a role in recognition. The 6-O-carbamoyl group contributes to nodulation efficiency, biological activity, and protection, but could be replaced by a 6-O-acetyl group for root nodulation. The results demonstrate that none of the studied substitutions is strictly required for triggering normal nodule formation. However, the nodulation efficiency was greatly determined by the synergistic presence of substitutions. Within the range tested, fluctuations of Nod factor amounts had little impact on the symbiotic phenotype.

Rejuvenation of Sequoia sempervirens by Repeated Grafting of Shoot Tips onto Juvenile Rootstocks in Vitro: Model for Phase Reversal of Trees.

Repeated grafting of 1.5-centimeter long shoot tips from an adult Sequoia sempervirens tree onto fresh, rooted juvenile stem cuttings in vitro resulted in progressive restoration of juvenile traits. After four successive grafts, stem cuttings of previously adult shoots rooted as well, branched as profusely, and grew with as much or more vigor as those of seedling shoots. Reassays disclosed retention for 3 years of rooting competence at similar levels as originally restored. Adventitious shoot formation was remanifested and callus development was depressed in stem segments from the repeatedly grafted adult. The reversion was associated with appearance and disappearance of distinctive leaf proteins. Neither gibberellic acid nor N(6)-beneyladenine as nutrient supplements duplicated the graft effects.

Transformation of ipecac (Cephaelis ipecacuanha) with Agrobacterium rhizogenes.

Transformed root cultures of ipecac (Cephaelis ipecacuanha A. Richard), one of the recalcitrant woody plant species for Agrobacterium-mediated transformation, were established by co-culturing of in vitro petiole segments with Agrobacterium rhizogenes ATCC 15 834. Southern blot analysis of the established roots revealed that only the TL-DNA was integrated into the plant genome without incorporation of the TR-DNA. The transformed roots grew slowly on phytohormone-free solid medium and adventitious shoots were regenerated after over 6 months of culture on HF, half-strength Murashige and Skoog (1/2 MS) medium in the dark. The individually separated transformed shoots developed into plantlets on phytohormone-free solid medium at 25 degrees C under 16 h/day light, and the plants demonstrated wider leaves, shorter internodes and vigorous root growth compared to non-transformed plants. Effects of basal media and auxins on the growth and the ipecac alkaloid production of the transformed roots were investigated either under light or in the dark. The roots cultured in the dark grew well in Gamborg B5 (B5) liquid medium containing 0.5 mg/L IBA and yielded 112 mg/L of cephaeline and 14 mg/L emetine after 8 weeks of culture.

[Construction of fast propagation system of Houttuynia cordata new line].

OBJECTIVE: To construct the fast propagation system of Houttuynia cordata Thunb. new line by selecting the proper type and concentration of plant growth regulators and proper explants. METHOD: The different explants of Houttuynia cordata new line were cultured on MS media with different concentration of different plant grow regulators. RESULT: Stem node over-ground and shoot tip were optimum explants for fast propagation. The stem which node grew on MS medium with KT at 0.5 mg x L(-1) could induce more leaves and grow higher. The shoot tip cultured on MS medium with 1.0 mg x L(-1) 6-BA could induce better callus. Adventitious shoots were better achieved from over-ground nodal explants cultured on MS medium supplemented with 6-BA at 1.0 mg x L(-1) and NAA at 0.2 mg x L(-1), or 6-BA at 0.5 mg x L(-1) and NAA at 0.2 mg x L(-1). MS medium with IAA at 0.2 mg x L(-1) was the best one on inducing roots. CONCLUSION: The propagat coefficient can be highly improved by inducing adventitious shoots through stem node over-ground, and thus plentiful seeds for production can be provided.

Changes in protein kinase C activities are correlated with the metaplastic transformation of Schwann cell precursors of avian embryos into melanocytes.

In previous work, we found that the phorbol ester drug 12-O-tetradecanoyl phorbol acetate (TPA) reverses the developmental restriction of melanogenesis that occurs early in neural crest development, causing Schwann cell precursors to undergo a metaplastic transformation into melanocytes. In this study, we examine whether these effects of TPA may be mediated by changes in endogenous levels of protein kinase C (PKC) activities. We report that low levels of PKC activity are correlated with this adventitious pigmentation in the crest-derived cells of dorsal root ganglia both during normal development and following TPA treatment in culture. These results suggest that regulation of endogenous levels of PKC plays a role in developmental decisions that neural crest cells make during early embryogenesis.

[Neural apparatus of the coronary arteries normal and in ischemic heart disease].

In transversal section of the neural-vascular sonplexes of the left and right coronary arteries stained after Weigert-Pahl taken from 20 men of the second mature (10 healthy, 10 died from the cardiac ischemic disease--CID) and from 20 aged men (10 healthy, 10 died from the CID), the number of nerves forming the adventitial arterial plexus, the number of myelin fibers of three classes in their composition have been calculated. The age involution of the coronary arteries neural connections has proved to be manifested as a decreased number of the nerves, total number of the myelin fibers in their composition, subsidence first of all in the number of middle and thick myelin transmitters. The CID is accompanied with an enhancement of natural (aged) phenomena of the arterial wall denervation. This is expressed as disappearance of highly specialized thick and middle myelin fibers, as rarefication of the cholinergic fibers network in the arterial adventitial plexus, that is ascertained by a comparative morphometry of the film preparations of the adventitia, stained after Gomori and Karnovsky--Roots (from 32 hearts of healthy mature and aged persons and patients with the CID). Changes in the number of the neural elements in the coronary arteries adventitial plexus determines certain new quantitative peculiarities in their innervational connections both in the process of the organism's ontogenesis and under conditions of the CID.
