Bottled mineral waters polluted by protozoa in Mexico.

A survey of protozoa polluting bottled mineral water in Mexico was carried out using samples obtained form the three best-selling brands of bottled mineral water in the country. The organisms were concentrated through filtration procedures and subsequently cultured in the sterile media. The cultures were observed over four weeks, with identification to the level of genus and species. Most commonly found were the amoebae Naegleria gruberi, Acanthamoeba astronyxis, and Vahlkampfia vahlkampfi (trophic as well as cystic stages) plus one flagellate, Bodomorpha minima. No ciliates were detected. The public health importance of the findings is obvious, since some strains of Naegleria and Acanthamoeba have the potential to cause human disease that may lead to death.

Consumption of fish from polluted waters by WIC participants in east Harlem.

To minimize exposure to neurotoxins such as mercury, polychlorinated biphenyls (PCBs), dioxins, and pesticide residues, the New York State Department of Health issues health advisories about consumption of certain fish and shellfish caught from polluted local waters. Fetal exposure causes cognitive developmental deficits in children. Consumption of fish was assessed. We surveyed 220 WIC (Special Supplemental Nutrition Program for Women, Infants, and Children) participants. Of the participants, 10% ate fish and shellfish caught in local polluted waters. Statistically significant factors associated with eating local, noncommercial fish included male gender and knowledge of the health advisory. Locally caught fish and crabs are consumed; thus, in utero and childhood exposure to these neurotoxins occurs. Interventions to promote safer choices of fish are needed.

Cadmium and zinc response of the fungi Heliscus lugdunensis and Verticillium cf. alboatrum isolated from highly polluted water.

The aquatic hyphomycete Heliscus lugdunensis and the terrestrial fungus Verticillium cf. alboatrum, both isolated from a highly polluted surface water, were investigated for their tolerance against Cd and Zn. Hl-H4 showed a 50% growth inhibition at 0.1 mM Cd, whereas at 0.7 mM Cd the growth of Va-H4 was only reduced by 30%. The fungi also showed a remarkable difference in their Zn-tolerance. The growth of Va-H4 was not inhibited at 1 mM Zn, whereas for Hl-H4 no growth occurred above 0.3 mM Zn. The biosorption and accumulation capacities for Cd or Zn of both fungi differed between the fungal species. In a 0.1 mM Cd-medium Hl-H4 biosorbed 15-fold and accumulated 39-fold more Cd than Va-H4. Exposure to 0.3 mM Zn resulted in a 13-fold higher biosorption and 11-fold higher accumulation for Hl-H4 than Va-H4. As glutathione (GSH) is known to be involved in the phytochelatin synthesis and other stress related processes we investigated its synthesis. Both fungi increased their synthesis of GSH in response to Cd. For Hl-H4 a concentration of 0.0125 mM Cd, corresponding to an intracellular Cd content of 2.1 nmol Cd mg(-1) dw, increased the GSH content, whereas Va-H4 only responded with a higher production of GSH at 1 mM Cd and a concomitant intracellular Cd content of 22.5 nmol Cd mg(-1) dw. An increased GSH synthesis under Zn-stress was only detectable for Va-H4 (20 mM).

Chromosomal effects of newly identified water pollutants PBTA-1 and PBTA-2 and their possible mother compounds (azo dyes) and intermediates (non-ClPBTAs) in two Chinese hamster cell lines.

We performed the in vitro micronucleus (MN) test on 2-[2-(acetylamino)-4-[bis(2-methoxyethyl)amino]-5-methoxyphenyl]-5-amino-7-bromo-4-chloro-2H-benzotriazole (PBTA-1) and 2-[2-(acetylamino)-4-[N-(2-cyanoethyl)-ethylamino]-5-methoxyphenyl]-5-amino-7-bromo-4-chloro-2H-benzotriazole (PBTA-2), which are newly identified water pollutants from the Nishitakase river in Kyoto, Japan, and on their possible mother compounds (AZO DYE) and intermediates (non-ClPBTAs). We tested these compounds in the absence and presence of S9 mix in two Chinese hamster cell lines CHL and V79-MZ and scored MN, polynuclear and karyorrhectic (PN), and mitotic (M) cells. PBTA-2 in the absence of S9 mix induced the strongest responses in both cell lines. It was also a strong inducer of binucleate cells in PN cells in both cell lines, which suggested that it induced polyploidy. PBTA-1 showed clear positive results only in the absence of S9 mix and only in V79-MZ cells, inducing aneuploidy. In CHL cells AZO DYE-1 significantly induced MN cells in the presence of S9 mix, and AZO DYE-2 induced MN and PN cells, including binucleate cells and cells with a multilobed nucleus, in the absence of S9 mix. In V79-MZ cells, AZO DYE-1 and -2 induced primarily M cells in the presence of S9 mix. 9% of the M cells treated with 50 microg/ml AZO DYE-1 showed endoreduplication. AZO DYE-2 at 200 microg/ml condensed the chromatin in 100% of the cells. The non-ClPBTAs were a bit more cytotoxic than the other compounds and induced a slight increase in MN cells in both cell lines. Some of the chemicals tested induced a characteristic karyomorphology that might reflect abnormal cell division. Abnormalities of cell division could be detected in PN and M cells as well as in MN cells. Structure-activity relationships have also been discussed.

[Water pollution and carcinogenic risks].

The main subjects emphasized in this contribution are the following ones: 1 - To investigate the presence of cancerogenic compounds derived directly or by transformations within trophic chains from domestic and industrial effluents. 2 - To evaluate potential cancerogenic health hazards due to polluted a quatic environement. The following substances might be regarded as to be cancerogenic: nitros amins, mycotoxins, complex organic and inorganic compounds and synthetic organicals. Based on epidemiological and epizoonic evidences, as well as human and experimental pathology, the relevant importance of particular group of compounds is evaluated. Presented evaluation is indicating that the water-borne cancerogenesis might be of an important significance. Therefore, the needs for essential increase in relevant research, implemented by advanced methods, which can cope with complexity of ecological processed, are stressed and further efforts recommended.
