Morphological transformation of mouse and rat embryo cells in vitro by an agent from S37 ascites tumour.

When normal rat and mouse embryo cells were treated with a cell free extract of S37 ascites tumour, morphological transformations occurred in both. The transformed cells readily induced lymphosarcoma-type tumours in the mice inoculated when newborn or young adults (8-12 weeks old), but not so readily in rats.Various tests carried out with these cells strongly indicate the presence of an oncogenic and transmissible agent in S37 ascites tumour. This agent appears to be related to mouse sarcoma virus isolated from animals with Moloney leukaemia but differs in only producing characteristic lymphosarcoma-type tumours.

Tumor induction by Moloney sarcoma virus in athymic nude mice.

Athymic nude (nu/nu) mice, normal NMRI (plus/plus) mice, and nu/plus heterozygotes, inoculated at birth with the Moloney strain of murine sarcoma virus (M-MSV), did not differ either in tumor growth rate or incidence or in mortality due to tumor development; neither did nude (nu/nu) and normal NMRI mice inoculated with M-MSV at 30 days of age differ in the initial tumor growth rate. However, tumors induced in adult nude mice grew progressively and eventually killed the hosts, whereas tumors induced in adult normal NMRI mice completely regressed. These results indicated that thymus-dependent immunity, though not active in M-MSV tumor induction, determined whether the tumor, once formed, would regress or kill the host.

The pathology of spontaneously occurring malignant lymphoma in sheep.

The most common locations of tumours in 22 sheep with malignant lymphoma were the lymph nodes (19 sheep), spleen (14), liver (13), kidney (10), small intestine (9) and heart (9). The distribution of the lesions could be classified as multicentric (14 sheep, alimentary (6), skin (1) and thymic (1). Histological examination of malignant lymphomas from 40 sheep showed there were three reticulum cell sarcomas, and 37 lymphosarcomas. The lymphosarcomas were further classified on the basis of cytological features as lymphoblastoid, lymphocytic/prolymphocytic, prolymphocytic and lymphocytic in 14, 6, 11 and six cases, respectively. Viruses were not detected by electron microscopy in tumourous tissue from 10 sheep with malignant lymphoma.

Cancer risk associated with simian virus 40 contaminated polio vaccine.

BACKGROUND: The presence of SV40 in monkey cell cultures used in the preparation of the polio vaccine from 1955 through 1961 is well documented. Investigations have consistently demonstrated the oncogenic behavior of SV40 in animal models. Early epidemiologic studies were inadequate in demonstrating an increase in cancer incidence associated with contaminated vaccine. Recently, investigators have provided persuasive evidence that SV40 is present in human ependymomas, choroid plexus tumors, bone tumors, and mesotheliomas, however, the etiologic role of the virus in tumorigenesis has not been established. MATERIALS AND METHODS: Using data from SEER, we analyzed the incidence of brain tumors, bone tumors, and mesotheliomas from 1973-1993 and the possible relationship of these tumors with the administration of the SV40 contaminated vaccine. RESULTS: Our analysis indicates increased rates of ependymomas (37%), osteogenic sarcomas (26%), other bone tumors (34%) and mesothelioma (90%) among those in the exposed as compared to the unexposed birth cohort. CONCLUSIONS: These data suggest that there may be an increased incidence of certain cancers among the 98 million persons exposed to contaminated polio vaccine in the U.S.; further investigations are clearly justified.
