Rat epitheloid lung cells were transformed with various preparations of SV40 dna using the Ca2+-precipitation technique. The amount of SV40 genetic information integrated into transformed clones was evaluated by DNA-DNA renaturation kinetics. The growth properties on plastic and in soft-agar were examined, as well as the ability to induce tumors in syngeneic new-born animals or in adult nude mice. One particular transformed line, which had received the Hpa II/BamH I A (59 per cent) fragment, was found to contain about 3 integrated copies of this fragment per cell and no significant amount of the Hpa II/BamH I B (41 per cent) fragment. This line which grew to high saturation densities and efficiently formed clones in low serum on plastic, produced tumors in both syngeneic rats and nude mice. Thus the Hpa II/BamH I A fragment, which mainly includes early viral information, was sufficient to impart these properties to rat epitheloid lung cells.
SV40 virion production was enhanced when semipermissive Chinese hamster kidney cells were treated with 5-iodo-2'-deoxyuridine (IUDR), prior to infection either with virus or viral-DNA. Pretreatment did not increase SV40 replication in fully permissive monkey kidney and nonpermissive Syrian hamster or mouse embryo cells.

An ORF located immediately downstream of glnR gene was cloned from Amycolatopsis mediterranei U32 and was named lh3. Sequence analysis revealed that lh3 encodes a putative acetyltransferase, which shows high amino acid sequence similarities to the mycothiol synthase (MshD) from other actinomycetes. For functional analysis, mutation in lh3 gene was generated by gene replacement with an apramycin resistance gene through homologous recombination. Compared with the wild type strain, the resulting mutant was more sensitive to H2O2, apramycin and erythromycin by two- to three-fold. These results suggest that the lh3 gene plays an important role in the course of detoxification in A. mediterranei U32.
Immortalization with simian virus-40 and cloning of immortalized cells from mouse thymus were performed to establish cell lines for characterization of the mode of glycolipid expression in the thymic cells. Among the 25 cell lines obtained, three lines with different morphologies were established, that is, epithelial (IMTH-E), fibroblastic (IMTH-F), and asterisk-like (IMTH-I) cells, and their glycolipids, together with those in the thymus, were determined systematically. The major glycolipids in mouse thymus were the globo- and ganglio-series, both of which, were co-expressed in the three cell lines established. However, the mode of modification of the globo- and ganglio-series was distinct for each cell line. As to the globo-series, the structures with the longest carbohydrate chain for IMTH-E, -F, and -I cells were Gb3Cer, Gb4Cer, and Forssman antigen, respectively, having stepwise shorter carbohydrates at the nonreducing termini. Although the acidic glycolipids in IMTH-E cells comprised GM3 and GM2, and their sulfated isomers, IMTH-F and -I cells expressed GMlb and GDlc for the alpha-pathway, and up to GDI a for the a-pathway of ganglio-series glycolipids. GMlb-GalNAc present in the thymus was not detected in IMTH-F and -I cells, probably due to the lower synthetic activity for the metabolic intermediate Gg4Cer. The results indicate that the immortalization technique is useful for obtaining individual cells having unique glycolipid profiles for analysis of the functional significance and metabolism of glycolipids in the thymus.
