[Serum IgE levels in atopic diseases in childhood].

The serum IgE levels were determined according to the radioimmunosorbent technique (RIST), in 208 children aged between 1 and 14 years, with atopic diseases alone or in combination. Geometric mean IgE levels were significantly increased and rose progressively from the atopic dermatitis "alone" to the allergic rhinitis, the allergic asthma and the combination of asthma with rhinitis, with the highest values in patients where atopic dermatitis was combined with respiratory allergy. IgE levels rose slowly with age. For the atopy diagnosis in vitro, two criteria were chosen and compared: a fixed IgE-level of 100 U/ml, and one standard deviation below the geometric mean of the atopic children.

[Immunochemical study of the conformational properties of the variable and constant domains of the light chain of mouse MOPC myeloma immunoglobulin].

The antigenic activity of variable (V) and constant (C) domains of mouse myeloma MOPC 21 light (L) chain has been used as a probe of their conformational state. The conformational properties of L-monomers, L-dimers and native IgG1 MOPC 21 protein were investigated by the reaction of inhibition of radioimmunoadsorbtion. Antibodies against the C-domain and different regions of V-domain were used in these experiments. It was shown that V-domain of L-monomers has sharp local conformational disruptions as compared with the native protein, while the conformational state of C-domain remains stable in monomers, dimers and native protein. It was found that the coinformational flexibility of different parts of V-domain of L-monomers is not equal. The antigenic activity of V-domain is partly restored in L-dimers, reaching the conformational state close to that of v-domain in the native protein. The possible interrelation between the conformational properties of V-domains and their functional activity is discussed.

Indication of a certain molecular correlation between rat IgE and human IgE.

The PCA-activity of rat sera sensitized with Ascaris suum antigen can be absorbed by antihuman IgE and by rabbit or sheep antihuman IgE convalently bound to sephadex particles or paper discs respectively. In vitro, rat immune sera show positive results using the human RIST-system abolished by preheating for 2 hours at 56 degrees C and therefore excluding an interaction of IgG 4. In contrast, no rat sera activity was seen using the human PRIST-system. These results agree with the concept of a single common antigenic determinant in human and rat IgE since in the human RIST and PRIST-system the binding of rat IgE to antihuman IgE could only be demonstrated on the principle of competitive inhibition (RIST) but not on that of a sequential binding (PRIST). The possibility of a certain heterogeneity in the IgE population can not be excluded.

Automated measurement of serum thyroxine with the "ARIA II," as compared with competitive protein binding and radioimmunoassay.

Two conventional serum thyroxine assays, run in separate laboratories, one by competitive protein binding and one by radioimmunoassay, were used to evaluate the automated ARIA II (Becton Dickinson Immunodiagnostics) serum thyroxine assay. Competitive protein binding as compared to ARIA II with 111 clinical serum samples gave a slope of 1.04 and a correlation coefficient of 0.94. The radioimmunoassay comparison to ARIA II with 53 clinical serum samples gave a slope of 1.05 and a correlation coefficient of 0.92. The ARIA II inter-assay coefficient of variation for 10 replicates of low, medium, and high thyroxine serum samples was 6.2, 6.0, and 2.9%, respectively, with an inter-assay coefficient of variation among 15 different assays of 15.5, 10.1, and 7.9%. The automated ARIA II, with a 2.2-min cycle per sample, gives results that compare well with those by manual methodology.

Mathematical approach to some methodological difficulties in settling newborn IgE reference levels (study in a Portuguese population).

The authors used statistical procedures to attempt to solve the usual methodological difficulties inherent in the determination of neonatal IgE reference values. The problem of un detectable values was overcome by theoretical estimation of their distribution, through the regression of the probits of the cummulative frequencies of the determinations on the natural logarithms of the known concentrations. Thus, the geometrical mean and 95% confidence limits were established, and population selection was replaced by exclusion of aberrant observations in successive steps, with recalculation of reference values. The final geometrical mean obtained was 0.21 I.U./ml with 95% confidence limits of 0.03 and 1.54 I.U./ml. These results have been compared with the work presented by other authors, observed differences probably reflecting distinct methodologies.
