Molecular analysis on the chemopreventive properties of resveratrol, a plant polyphenol microcomponent.

As a plant microcomponent, resveratrol is a polyphenolic compound produced by several species and found especially in Polygonum roots, peanuts seeds, berries and also grape and therefore can be present in human diet or beverages (red wine, for instance). Traditional chinese medicine and more recent epidemiological studies strongly suggested that resveratrol may act as a cancer chemopreventive compound. The biochemical mechanism by which resveratrol inhibits cell proliferation was provided by studies in numerous human cell lines including our work in hepatoblastoma HepG2 and colorectal tumor SW480 cells. The results show that resveratrol strongly inhibits cell proliferation at the micromolar range in a time- and dose-dependent manner. Resveratrol appears to block the cell cycle at the transition S to G2/M since there is no inhibition of [3H]-thymidine incorporation observed, while there is an increase of the cell number in S phase. On the other hand, in order to evaluate if the amount of resveratrol taken up during food or drink consumption is sufficient to ensure in the whole body the in vitro described beneficial effects, we evaluated the ratio between plasmatic level of resveratrol and its cell bioabsorption. Our study reports a higher uptake of resveratrol in the human hepatic derived HepG2 cells than in colorectal derived SW480 cells. In contrast, resveratrol is conjugated in these cells and derivatives are released in large amounts in the cell medium. Based on present knowledge, resveratrol appears to be a promising bioactive natural molecule with potential applications in phytotherapy, pharmacology or in nutriprotection (nutraceutic food) area.

The mechanism of Polydatin in shock treatment.

Polydatin is extracted from a traditional Chinese herbal medicine, Polygonum cuspidatum, and has a special effect in shock treatment. The aim of this study is to explain the cellular and molecular basis of Polydatin in shock treatment. The fluorescent probe techniques, patch clamp method, and cellular flow chamber were used to test intracellular variables of vascular smooth muscle cells (VSMC), myocardial cells (MC), endothelial cells (EC), and white blood cell (WBC). It was shown that Polydatin could inhibit ICAM-1 expression in EC stimulated by lipopolysaccharide (LPS), attenuate WBC-EC adhesion, increase [Ca2+]i in MC with enhancement of MC contraction extent, activate KATP channels of VSMC, and decrease pHi value and [Ca2+]i of VSMC in shock. The study suggests that Polydatin has multiple effects on VSMC, MC, WBC and EC, which are related to the enhancement of heart function and improvement of microcirculatory perfusion in shock.

Innovative uses of vegetated drainage ditches for reducing agricultural runoff.

Vegetated agricultural ditches play an important role in mitigation of pesticides following irrigation and storm runoff events. In a simulated runoff event in the Mississippi (USA) Delta, the mitigation capacity of a drainage ditch using the pyrethroid esfenvalerate (Asana XL) was evaluated. The pesticide was amended to soil prior to the runoff event to simulate actual runoff, ensuring the presence of esfenvalerate in both water and suspended particulate phases. Water, sediment, and plant samples were collected temporally and spatially along the drainage ditch. Even with mixing of the pesticide with soil before application, approximately 99% of measured esfenvalerate was associated with ditch vegetation (Ludwigia peploides, Polygonum amphibium, and Leersia oryzoides) three hours following event initiation. This trend continued for the 112 d study duration. Simple modeling results also suggest that aqueous concentrations of esfenvalerate could be mitigated to 0.1% of the initial exposure concentration within 510 m of a vegetated ditch. Observed field half-lives in water, sediment, and plant were 0.12 d, 9 d, and 1.3 d, respectively. These results validate the role vegetation plays in the mitigation of pesticides, and that ditches are an indispensable component of the agricultural production landscape.

[Allelopathy of different plants on wheat, cucumber and radish seedlings].

By means of bioassay in laboratory and field, this paper studied the allelopathy of 18 kinds of plants in Gansu Province on the seedlings of wheat, cucumber and radish. The results showed that the aqueous extract of the stems and leaves of Artemisia annua, Solanum nigrum and Datura stramonium had the strongest allelopathy on test receptor plants, and their synthetic inhibitory effect (SE) was 47.66%, 32.89% and 26.63%, respectively. The SE of Xanthium sibiricum, Portulaca oleraca, Cephalanoplos segetum, and Chenopodium album was 21.71%, 20.93%, 20.83% and 20.2%, respectively, while Vicia amoena (SE 3.5%), Setaria viridis (SE 2.2%), and Cymamchum chinense (SE 1.97%) had a weaker allelopathy. Chenopodium ambrosioides (SE - 1.03%), Polygonum caespitosum (SE - 1.63%) and Avena fatua (SE 5.33%) had no evident allelopathy, but Artemisia annua affected the seedling height and fresh weight of radish, cucumber, wheat and maize, with the SE being 54.07%, 38.46%, 33.35% and 20.88%, respectively. Artemisia annua had a 44.70% of SE on wheat growth, and thus, had a certain value to develop and use.

New bioactive compounds of plant origin.

Crude extracts of Withania somnifera (WS) and Polygonum equisetiforme (PE) as well as the pure compounds isolated therefrom were screened for antibacterial and antifungal activity against 20 bacterial and 17 fungal cultures. The crude extract of PE and WS inhibited the growth of T. mentagrophyte, M. canis and A. boydii at an MIC of 450-500 microg/ml whereas the pure compounds inhibited the growth at MIC of 300-350 micro/ml. Species of Corynebacterium, Bacillus, Streptococcus and Staphylococcus were found to be highly susceptible to both the crude sad the pure compounds. MIC values of both crude extracts for different organisms tested were found to be higher (200-350 microg/ml) than the pure compounds (150-170 microg/ml). The crude extract of PE did not inhibit the growth of Ps. aeruginosa, however, the pure compound was found to be bacteriostatic. Brine shrimp and BALB/c mice lethality test indicated that these extracts maybe toxic at high concentration.

[Studies on the megasporogenesis and microsporogenesis and the development of their female and male gametophyte in Magnolia biloba].

The anther of Magnolia biloba is tetrasporangiate with glandular tapetum, which consists of one or two layers of cells. Cytokinesis during meiosis of its microspore mother cell is modified simultaneous type, and the microspore tetrads are isobilateral. Mature pollen grains are two-celled. Tetrad cells and microspores are irregularly shaped during the microsporogenesis. There were two ovules on the ventral surface of unicarpellate ovary wall. Ovules were anatropous, bitegmnous and crassinucellar. Archesporial cell was one cell and differentiated from cell in the second layer beneath epidermis. The development of the embryo sac conformed to the polygonum type. The embryological characteristics of Magnolia biloba are very similar to those of other species in Magnoliaceae. The megasporogenesis and microsporogenesis and the development of their female and male gametophyte are partially abnormal. Abnormal phenomena in the process of reproduction of Magnolia biloba causing this species to be endangered was discussed.

Cytological and histological studies on reproductive system of hexaploid Bellevalia edirnensis Ozhatay & Mathew (hyacinthaceae).

In this study anther wall, meiosis, sporogenesis and the development of female gametophytes of Bellevalia edirnensis were examined cytologically and histologically. The young anther wall consists of an epidermis, a single layered fibrous endothecium, 1-2 middle layers which disappear early, and a single-layered tapetum. In the glandular tapetum; normal mitosis, the "sticky" type of division and secondary nucleus divisions were observed. In most microspore mother cells the course of meiosis is regular. In some cells irregularities were observed including chromosome bridges and univalents. Cytokinesis is of the successive type and the tetrads are isobilateral. The ovules are anatrapous, bitegmic and crassinucellate. The micropyle is formed by the inner integument. The female archesporium is unicelled and directly functions as the megaspore mother cell. The female gametophyte formed by the functioning chalazal megaspore is monosporic, of the Polygonum type.

Verification of folk medicinal potentiality for some common plants in Jordan.

87 Species belonging to 59 genera and 33 plant families were identified and presented in the area of study. The largest 3 families are: Lamiaceae (9 aquatic species), Asteraceae (7 species), and Salicaceae (7 species). The largest genera are Mentha (6 species), Polygonum (5 species), and Salix (5 species). 63 folk medicinal aquatic species (73.3%) have therapeutic similarities with neighbouring countries, while the 24 remaining species (26.7%) haven't such therapeutic similarity. Emerged species (living with close contact with water body) were the most recorded, while amphibious, submerged or floating species were the least. The folk medicinal importance value of aquatic species recorded was identified according to Friedman parameters. 21 species (24%) have ROP values higher than 50, and therefore; have the highest popularity in folk medicinal potentiality. 26 species (29.9%) have therapeutic effects informed by less than three informants, and therefore, excluded from further consideration. 40 species (46.1%) have ROP values less than 50, and therefore; considered nonpopular medicinal plants.

[Influences of 3,4,5-trihydroxystibene-3-beta-mono-D-glucoside on beat rate and injury of cultured newborn rat myocardial cells].

3,4,5-Trihydroxystibene-3-beta-mono-D-glucoside (PD) from Polygonum cuspidatum 0.05-0.45 mmol/L increased beat rate of myocardial cells from 100% to 112-220%. This action of PD 0.15 mmol/L was agonized by nisoldipine 0.1, propranolol 10 and phentolamine 1 mumol/L. PD (0.05, 0.15 mmol/L) decreased the lactate dehydrogenase (LDH) amount liberated by myocardial cells, from 98 +/- 11 IU/ml of deprivation of oxygen and glucose (DOG) to 55 +/- 16, 68 +/- 14 IU/ml (P less than 0.01) 6 h after injury of DOG; 77 +/- 9, 80 +/- 6 IU/ml (P less than 0.01) 9 h after injury of DOG. PD 0.15 mmol/L decreased the LDH amount liberated by myocardial cells, from 311 +/- 34 to 231 +/- 42 IU/ml (P less than 0.05) 9 h after injury of chlorpromazine (CPZ). It is suggested that PD can protect myocardial cells injured by DOG and CPZ.

Screening of some Nigerian plants for molluscicidal activity.

Methanolic (MEOH), evaporated crude water (ECW) and unevaporated crude water (UECW) extracts of 25 Nigerian plants, used for different medicinal and domestic purposes were screened for molluscacidal activity on laboratory-reared Lymnaea natalensis Krauss. Seven of the plants were not active; extracts from 18 (72 per cent) of the plants, some of which are renowned fish poisons, had molluscicidal activity. These were Acacia nilotica, Aristolochia albida, Balanites aegyptiaca, Blighia sapida, Boswellia dalzielii, Detarium microcarpum, Gnidia kraussiana, Kigelia africana, Nauclea latifolia, Opilia celtidefolia, Parkia clappertoniana, Polygonum limbatum, Pseudocedrela kotschyi, Sclerocarya birrea, Securidaca longipedunculata, Ximenia americana, Vetiveria nigritana and Ziziphus abyssinica. The LC50 of these extracts were determined. It is strongly recommended that the toxic effects of these extracts against fish, cercariae, snail eggs and mammals be further investigated so as to determine the right concentration, especially for use in fish ponds.
