Development of an ELISA for the detection of bromoxynil in water.

For development of an indirect competitive enzyme-linked immunosorbent assay (ELISA) for the nitrile herbicide bromoxynil, the polyclonal antibodies raised against 2,6-dibromo-4-cyano-phenoxyacetic acid (hapten) conjugated to bovine serum albumin (BSA) by the N-hydroxysuccinimide-activated ester method. Antiserum with a sufficiently high titer to provide the determinations of targeted compounds was obtained only 77 days after the primary immunization. Antiserum A2 was applied to the residual analysis of some water samples, under optimized ELISA condition, the quantitative working range was from 10 to 500 ppb with a limit of detection of 5 ppb. Cross-reactivity to structurally similar agrochemicals and related chemicals was determined. The antiserum showed little cross-reactivity with 2,6-dibromophenol and bromoxynil octanoate ester which have a dibromophenol group as common structure, but showed no cross-reactivity with other herbicides. Each water sample (river water, tap water, purified water, and bottled water) had a matrix effect and was investigated by adding Tween20 in the assay buffer. These four kinds of water samples were fortified with bromoxynil at several concentration levels and were directly analyzed with only dilution with an equal volume of antiserum solution, the mean recovery was 102.3%, and the mean coefficient of variation was 5.96%. The proposed ELISA turned out to be a powerful tool for monitoring of residual bromoxynil in water samples at trace level.

Agricultural exposures and gastric cancer risk in Hispanic farm workers in California.

Previous studies have indicated that farm workers may be at increased risk of gastric cancer. Meta-analyses, ecological, case-control, and cohort studies suggest that some aspects of the agricultural environment may be implicated in the elevated risk. Hispanic farm workers in California are exposed to a multitude of potentially toxic substances in the work site, including excessive sunlight, fertilizers, diesel fumes, and pesticides. A previous analysis of a cohort of California farm workers who had been members of a farm labor union, the United Farm Workers of America (UFW) found a proportionate cancer incidence ratio for stomach cancer of 1.69 when using the California Hispanic population as the standard. The aim of the current study was to further evaluate associations between gastric cancer and the types of crops and commodities UFW members cultivate and the associated pesticide use as recorded by the California Department of Pesticide Regulation (DPR). We conducted a nested case-control study of gastric cancer embedded in the UFW cohort and identified 100 cases of newly diagnosed gastric cancer between 1988 and 2003. We identified 210 control participants matched on age, gender, ethnicity, and who were known to be alive and resident in California up to the date of the cases' diagnosis. Both stratified analyses and unconditional logistic regression were used to calculate adjusted odds ratios (OR) and 95% confidence intervals (95% CI). Work in the citrus industry was associated with increased gastric cancer (OR=2.88; 95% CI=1.02-8.12) although no other specific crops or commodities were associated with this disease. Working in areas with high use of the phenoxyacetic acid herbicide 2,4-D was associated with gastric cancer (OR=1.85; 95% CI=1.05-3.25); use of the organochlorine insecticide chlordane was also associated with the disease (OR=2.96; 95% CI=1.48-5.94). Gastric cancer was associated with use of the acaricide propargite and the herbicide triflurin (OR=2.86; 95% CI=1.56-5.23 and 1.69, 95% CI=0.99-2.89, respectively). Gastric cancer in California Hispanic farm workers is associated with work in the citrus fruit industry and among those who work in fields treated with 2,4-D, chlordane, propargite, and trifluin. These findings may have larger public health implications especially in those areas of the country where these pesticides are heavily used and where they may be found in the ambient atmosphere.

Strategies for improving the functionality of an affinity bioreactor.

Heparin employed in extracorporeal blood circulation (ECBC) procedures (e.g. open heart operations) often leads to a high incidence of bleeding complications. Protamine employed in heparin neutralization, on the other hand, can cause severe adverse reactions. We previously developed an approach that could prevent both heparin- and protamine-induced toxic side effects concomitantly. This approach consisted of placing a hollow fiber-based bioreactor device containing immobilized protamine (termed a "protamine bioreactor") at the distal end of the ECBC procedure. This protamine bioreactor would remove heparin after heparin served its anticoagulant purpose in the ECBC device, thereby eliminating heparin-induced bleeding risks. In addition, this protamine bioreactor would prevent protamine from entering the patients, thereby aborting any protamine-induced toxic effects. Both in vitro and in vivo studies have successfully demonstrated the feasibility of this approach. Despite promises, early findings also revealed two shortcomings that must be overcome for the protamine bioreactor to be applied clinically. The first drawback was that the cyanate ester linkages, involved in conjugating protamine to the bioreactor device, were unstable and prone to hydrolysis, resulting in the leakage of a significant amount of protamine into circulation during application of the protamine bioreactor. The second deficiency was that the capacity of the protamine bioreactor in heparin removal was rather low, owing to the limited surface area of the hollow fibers for protamine immobilization and subsequently heparin adsorption. In this paper, we present novel strategies to overcome these two limitations. A new conjugation method based on the use of 4-(oxyacetyl)phenoxyacetic acid (OAPA) as the activating reagent was employed to yield stable linkages, via the abundant arginine residues of protamine, onto the hollow fibers. Results showed that while the amount of protamine immobilized on each gram of fibers was relatively comparable between the OAPA and the previous CNBr activation methods (7.45 mg/g versus 7.69 mg/g fibers), there was virtually no detectable leaching of immobilized protamine from the bioreactor by the OAPA method, comparing to 35% leaching of protamine by the previous CNBr method following 72 h of storage of the bioreactor in PBS buffer at 37 degrees C. To improve the capacity and functionality of the protamine bioreactor, two novel approaches were adopted. Long chain and high molecular weight poly-lysine was linked to the hollow fibers, prior to protamine coupling, to create multiple layers of immobilized protamine for subsequent heparin adsorption. In addition, a poly(ethylene glycol) (PEG) chain was inserted between protamine and the hollow fibers to yield a three-dimensional, free dynamic motion for immobilized protamine. Preliminary observations indicated that a four- to five-fold enhancement in heparin adsorption was attained by utilizing each of these new approaches. Aside from their current use, these new strategies can also be employed generically to improve the functionality of any affinity-type bioreactor. Indeed, efforts have been made recently in utilizing these approaches to develop a clinically usable GPIIb/IIIa bioreactor for the treatment of immune thrombocytopenic purpura (ITP)-an autoimmune disease.

Chromosomal aberrations, micronuclei and nuclear buds induced in human lymphocytes by 2,4-dichlorophenoxyacetic acid pesticide formulation.

Pesticides of worldwide application are used in agriculture in vast amounts each year, of which herbicides are the most prominent class. Phenoxyacetic herbicides constitute one of the largest groups of herbicides sold in the world. Among them, for many years 2,4-dichlorophenoxyacetic acid (2,4-D) has been the one most used. In this study we used Deherban A, a commercial formulation of 2,4-D to determine its possible genotoxic effect on human lymphocytes in vitro by chromosomal aberration analysis and micronucleus assay including the scoring of nuclear buds. Two different concentrations of pesticide formulation were used so that final concentrations of 2,4-D were 0.4 and 4 microg/ml, both in the presence and in the absence of the liver microsomal fraction as metabolic activator. Both concentrations of pesticide caused an increase in chromatid and chromosome breaks, number of micronuclei and number of nuclear buds. Presence of the S9 mix additionally elevated the number of chromatid breaks and micronuclei in treated lymphocytes.

Enhancement of memory consolidation in chicks by beta(3)-adrenoceptor agonists.

The effects of intracranial injection of three beta(3)-adrenoceptor agonists, sodium-4-[-2[-2-hydroxy-2-(-3-chloro-phenyl)ethylamino] propyl]phenoxyacetate (BRL 37344), 2-hydroxy-5(2-((2-hydroxy-3-(4-((1-methyl-4-trifluoromethyl)1H-imidazole-2-yl)-phenoxy)propyl)amino)ethoxy)-benzamide monomethane sulfonate) (+/-)-CGP12177A) and the pro-drug RS-N-(7-carbethoxymethoxyl 1,2,3,4-tetrahydronaphth-2-yl)-2 hydroxy 2-(3-chlorophenyl)ethanamine (SR58611A), were examined on reinforcement of memory in day-old chicks. BRL37344 and CGP12177 facilitated memory, whereas SR58611A had no effect. The dose-response relationships of the beta(3)-adrenoceptor agonists were challenged with the selective beta(3)-adrenoceptor antagonist 3-(2-ethylphenoxy)-1-[(1S)-1,2,3,4-tetrahydronapth-1-ylamino]-2S-2-propanol oxalate (SR59230A) or the beta(2)-adrenoceptor antagonist (-)propranolol. BRL 37344 appeared to act predominantly at beta(3)-adrenoceptors at low doses and at beta(2)-adrenoceptors at higher doses. Facilitation of labile into long-term storage by beta(3)-adrenoceptor agonists appears to be a class action of these drugs.

Functional properties of atypical beta-adrenoceptors on the guinea pig duodenum.

In this study, we attempted to further characterize atypical beta-adrenoceptors on the guinea pig duodenum. (-)-Enantiomers of isoprenaline and noradrenaline were more potent than its (+)-enantiomers. The isomeric activity ratios ((+)/(-)) were less than those obtained in the guinea pig atria and trachea. The concentration-response curves to catecholamines ((-)-isoprenaline, (-)-noradrenaline and (-)-adrenaline), to the selective beta(3)-adrenoceptor agonist, BRL37344 ((R*, R*)-(+/-)-4-[2-[(2-(3-chlorophenyl)-2-hydroxyethyl)amino]propyl]phenoxyacetic acid sodium), and to the non-conventional partial beta(3)-adrenoceptor agonist, (+/-)-CGP12177A ((+/-)-[4-[3-[(1,1-dimethylethyl)amino]-2-hydroxypropoxy]-1,3-dihydro-2H-benzimidazol-2-one] hydrochloride), were resistant to blockade by (+/-)-pindobind, the beta-adrenoceptor alkylating agent. (-)-Noradrenaline and (-)-adrenaline were more potent than dopamine and (-)-phenylephrine, respectively. Selective beta(2)-adrenoceptor agonists possess agonistic activities at atypical beta-adrenoceptors. (+/-)-Propranolol and (+/-)-bupranolol had no agonistic effect, whereas (+/-)-alprenolol, (+/-)-pindolol, (+/-)-nadolol, (+/-)-CGP12177A and (+/-)-carteolol exhibited agonistic activities at atypical beta-adrenoceptors. These results suggest that pharmacological properties of atypical beta-adrenoceptors differ from those of conventional beta(1)- and beta(2)-adrenoceptors on the guinea pig.

Beta2-adrenoceptor-mediated inhibition of field stimulation induced contractile responses of the smooth muscle of the rat prostate gland.

Isolated preparations of rat prostate responded to electrical field stimulation (2 strains every 60 s, 0.5 ms, 10 Hz, 80 V) with contractions. The adrenoceptor agonists adrenaline, isoprenaline and noradrenaline (0.1 nM(-10) x microM) elicited concentration-dependent inhibition of electrical field stimulation-induced contractions of the rat prostate. Phenylephrine had no effect on the amplitude of electrical field stimulation-induced contractions. The rank order of potency was isoprenaline> or =adrenaline=noradrenaline>phenylephrine. Inhibition of electrical field stimulation-induced contractions by isoprenaline was attenuated by propranolol (1 microM). The selective beta1-adrenoceptor agonist (-)-1-(3,4-dimethoxy-phenethylamino)-3-(3,4-dihydroxyphenoxy)-2-propanol)oxalate (RO363) and the selective beta2-adrenoceptor agonist salbutamol (1 nM(-100) x microM) were approximately equipotent in inhibiting electrical field stimulation-induced contractions but the selective beta3-adrenoceptor agonist sodium 4-(2-[2-hydroxy-[3-chlorophenyl]ethylamino]propyl)phenoxyacetate (BRL 37344, 1 nM(-100) x microM) did not inhibit electrical field stimulation-induced contractions. The selective beta2-adrenoceptor antagonist, (+/-)-1-[2,3-(dihydro-7-methyl-1H-inden-4-yl)oxy]-3-[(1-methylethyl)amino]-2-butanol (ICI 118 551, 0.1 microM) attenuated inhibitory responses to isoprenaline and salbutamol, while the selective beta1-adrenoceptor antagonist atenolol (3 microM) did not. Contractions induced by electrical field stimulation were also inhibited by forskolin (10 nM(-3) x mM) but unaffected by sodium nitroprusside (10 nM(-1) x mM) indicating the presence of an inhibitory cAMP mechanism. These data suggest that stimulation of beta2-adrenoceptors can inhibit contractions of the rat prostate induced by electrical field stimulation.

5-HT(7) receptor and beta(2)-adrenoceptor share in the inhibition of porcine uterine contractility in a muscle layer-dependent manner.

To compare the inhibition of uterine contractility mediated by beta-adrenoceptors and 5-HT(7) receptors, the effects of catecholamines and 5-HT on spontaneous contractions were examined in longitudinal and circular muscles isolated from three different regions (cornu, corpus and cervix) of the non-pregnant proestrus porcine uterus. In addition, the distribution of beta-adrenoceptors between muscle layers was characterized by means of adenylate cyclase activity assay, cyclic AMP assay and [(3)H]dihydroalprenolol binding studies. In the cornu, isoprenaline, adrenaline and noradrenaline inhibited the spontaneous contraction of longitudinal and circular muscles but longitudinal muscle was more sensitive to catecholamines than was circular muscle. The inhibitory response to isoprenaline was antagonized by propranolol (300 nM) or (+/-)-1-[2,3-(dihydro-7-methyl-1H-inden-4-yl)oxy]-3-[(1-methylethyl)amino]-2-butanol (ICI 118,551; 100 nM). The rank order of potency was isoprenaline > or =adrenaline > noradrenaline. The beta(2)-adrenoceptor-selective agonist, clenbuterol, was more potent than xamoterol (beta(1)-selective) and (+/-)-4-[2-[(2-(3-chlorophenyl)-2-hydroxyethyl)amino]propyl]phenoxyacetic acid (BRL 37344; beta(3)-selective) to inhibit the spontaneous contraction of longitudinal muscles. Isoprenaline increased adenylate cyclase activity in both muscle layers, but the activity in the longitudinal muscle was greater than that in the circular muscle. Cyclic AMP production by isoprenaline was also more conspicuous in the longitudinal muscle than in the circular muscle. Although both muscle layers contained a single class of [3H]dihydroalprenolol binding sites with similar K(d) values (longitudinal muscle, 3.1+/-0.94 nM, n=4; circular muscle, 2.4+/-0.73 nM, n=4), B(max) in the longitudinal muscle (175.7+/-32.8 fmol/mg protein, n=4) was significantly higher than that in the circular muscle (53.1+/-5.1 fmol/mg protein, n=4). As previously reported [Br. J. Pharmacol. 130 (2000) 79], 5-HT also inhibited the spontaneous contraction of both muscle layers from the cornu and the 5-HT(7) receptor antagonist, 2a-[4-(4-phenyl-1,2,3,6-tetrahydropyridyl)butyl]-2a,3,4,5-tetrahydro-benzo[cd]indol-2(1H)-one (DR4004; 100 nM, n=4) blocked the 5-HT-induced inhibition of spontaneous contractions in the circular muscles, and reversed the less marked inhibition in the longitudinal muscles. In other regions of the uterus (corpus and cervix), 5-HT inhibited the spontaneous contraction of the circular muscles but contracted the longitudinal muscle strips. On the other hand, isoprenaline caused muscle layer-dependent inhibition (longitudinal muscle > circular muscle) in both regions, and the responsiveness tended to increase toward the cervix. In conclusion, beta(2)-adrenoceptors are present heterogeneously in the porcine uterus (longitudinal muscle > circular muscle) and share the inhibition of uterine contractility with 5-HT(7) receptors in a layer-dependent manner (longitudinal muscle: beta(2)-adrenoceptors, circular muscle: 5-HT(7) receptors).

Cyclic AMP-independent relaxation mediated by beta3-adrenoceptors on guinea pig gastrointestine.

In this study, we investigated the signal transduction pathway involved in beta(3)-adrenoceptor-mediated relaxations of guinea pig gastric fundus and duodenum. In the presence of beta1- and beta2-adrenoceptor blockade, the potency (pD2 value) of catecholamines ((-)-isoprenaline, (-)-noradrenaline and (-)-adrenaline) and beta(3)-adrenoceptor agonists ((R*, R*)-(+/-)-4-[2-[(2-(3-chlorophenyl)-2-hydroxyethyl)amino]propyl]phenoxyacetic acid sodium (BRL37344) and (+/-)-[4-[3-[(1,1-dimethylethyl)amino]-2-hydroxypropoxy]-1,3-dihydro-2H-benzimidazol-2-one] hydrochloride ((+/-)-CGP12177A)) to induce relaxation was not affected by the adenylate cyclase inhibitor, 9-(tetrahydro-2-furanyl)-9H-purin-6-amine (SQ-22,536, 100 microM). Catecholamines induced an elevation of cyclic AMP and SQ-22,536 significantly abolished the responses of gastric fundus. However, cyclic AMP levels were unaltered by the beta3-adrenoceptor agonists in gastric fundus and by the five agonists in duodenum. Furthermore, the relaxant responses to catecholamines and to beta3-adrenoceptor agonists were unaffected by the cyclic AMP-dependent protein kinase inhibitor, N-(2-[p-bromocinnamylamino]ethyl)-5-isoquinolinesulfonamide (H-89, 10 microM) in gastric fundus. These results suggest that beta3-adrenoceptor-induced relaxation is mediated through both cyclic AMP-dependent and cyclic AMP-independent pathways in gastric fundus and through a cyclic AMP-independent pathway in duodenum.

Chronic treatment with BRL 35135 potentiates the action of insulin on lipid metabolism.

The effects of a beta 3-adrenoceptor agonist on insulin-induced changes in lipid metabolism were studied in obese male Zucker (fa/fa) rats during euglycaemic clamp. Rats were treated with BRL 35135 (R*, R*-(+/-)-methyl-4-[2-[2-hydroxy-2-(3-chlorophenyl)-ethyl-amino]-propyl] phenoxyacetate hydrobromide) (0.5 mg/kg per day in drinking water) for three weeks before an euglycaemic hyperinsulinaemic clamp was performed. Insulin infusion lowered serum non-esterified fatty acids and plasma glycerol more efficiently in BRL 35135-treated than in control rats although plasma insulin remained significantly lower in the BRL 35135-treated than in the control rats during the clamp. In conclusion, chronic treatment with BRL 35135 potentiates the effect of insulin on lipid metabolism.

Phosphorylation of extracellular signal-regulated kinases 1 and 2 in 3T3-L1 adipocytes by stimulation of beta(3)-adrenoceptor.

Recent studies have revealed that activated extracellular signal-regulated kinases (ERKs) 1 and 2 by the stimulation of beta(3)-adrenoceptors played a critical role in cell survival in brown adipocytes. On the other hand, phosphorylation of ERK1/2 via beta(3)-adrenoceptors and its physiological and pathological significance in white adipocyte has remained uncertain despite the increasing significance of functioning white adipocytes. Accordingly, we here studied phosphorylation of ERK1/2 caused by the stimulation of beta(3)-adrenoceptors in 3T3-L1 adipocytes, and the roles of phosphorylated ERK1/2 in lipolysis. Phosphorylation of ERK1/2 was induced by a selective beta(3)-adrenoceptor agonist, DL-4-[2'-¿2-hydroxy-2-(3-chlorophenyl)ethylamino¿propyl] phenoxyacetic acid sodium salt sesquihydrate (BRL37344), in 3T3-L1 adipocytes in a time- and dose-dependent manner. The phosphorylation of ERK1/2 by BRL37344 was sensitive to the cyclic AMP (cAMP)-dependent protein kinase inhibitor, N-[2-((p-bromocinnamyl)amino)ethyl]-5-isoquinolinesulfonamide (H89). To elucidate the roles of phosphorylated ERK1/2 in lipolysis, the effect of a selective inhibitor of ERK1/2 phosphorylation, 2'-amino-3'-methoxyflavone (PD98059), was examined. This inhibitor did not alter the lipolytic action caused by BRL37344, even at concentrations sufficient to block phosphorylation of ERK1/2, suggesting that ERK1/2 play no role in the lipolysis caused by BRL37344 in 3T3-L1 adipocytes.

Chromatographic approaches to the quality control of chiral propionate anti-inflammatory drugs and herbicides.

The chiral analysis of a range of propionate anti-inflammatory drugs and herbicides was accomplished by gas chromatographic separation of their R-alpha-phenylethylamide diastereomeric derivatives. Using a packed column (3% OV-l on Chromosorb G) good separations were obtained but analysis times were rather long. Examination of the same crystalline derivatives using a capillary column (bonded methylsilicone) resulted in good separations together with much reduced total analysis times. The technique was validated through the analysis of several derivatives prepared on a micro scale and examined directly by gas chromatography without any crystallisation step. Racemic materials all gave virtually 50:50 response ratios while the analysis of optically pure samples produced no evidence of significant racemization. Derivatives of phenoxyacetic acid and myristic acid were also prepared and chromatographed as potential internal standards.

Degradation of 2,4-dichlorophenoxyacetic acid by ionizing radiation: influence of oxygen concentration.

Ionizing radiation has been proved as a promising method for the degradation of the herbicide 2,4-dichlorophenoxyacetic acid (2,4-D). The gamma-radiolytic decomposition and chloride formation follows an apparent first order kinetic, kde/kCl = 3. For complete removal of 500 microM 2,4-D, a dose of 4 kGy is required. Phenolic intermediates (2,4-dichlorophenol, the isomers 2-chloro-4-hydroxy- and 4-chloro-2-hydroxy phenoxyacetic acid and three hydroxylation products of 2,4-D) are formed. Using oxygen saturation during irradiation, they are removed again with a dose of 4 kGy. For the formation of the main organic product acetic acid a reaction mechanism is discussed. Oxygen concentration enhances strongly fragmentation and mineralization. A reduction of 36% TOC could be achieved.

Dioxin and dioxin-like PCB impurities in some Japanese agrochemical formulations.

The profile and amount of dioxin impurity in agrochemicals were studied through detailed analysis of historic Japanese formulations. The chemicals analyzed include pentachlorophenol (PCP), 2,4,6-trichlorophenyl-4'-nitrophenyl ether (chloronitrofen, CNP), 2,4-dichlorophenyl-4'-nitrophenyl ether (nitrofen, NIP), tetrachloro-iso-phthalonitrile (chlorothalonil, TPN), 2-methyl-4-chloro-phenoxyacetic acid (MCP) and 2,4-dichlorophenoxyacetic acid (2,4-D). Among the six, two herbicides, PCP and CNP, produced during the 1960s and 1970s, contained very high concentrations of PCDD/DFs and TEQ. Others contained relatively low concentrations of PCDD/DFs. Dioxin-like PCB concentrations in all chemicals studied were low and their contributions to TEQ were negligible. The total dioxin emissions from the use of agrochemicals in Japan during the past 40 years (1955-1995) were estimated to be about a few hundred thousand kg of PCDD/DFs and 250 kg of WHO-TEQ from PCP and 190 x 10(3) kg of PCDD/DFs and 440 kg of WHO-TEQ from CNP. The major dioxin congeners present in PCP formulations were highly chlorinated PCDD/DFs that can be formed by the coupling of PCP and/or 2,3,4,6-tetrachlorophenol, and those in the CNP formulations were tetra- to hexa-chlorinated PCDD/DFs that can be formed from 2,4,6-trichlorophenol and/or 2,3,4,6-tetrachlorophenol.

Genotoxicological effects of some phenoxyherbicides and their metabolites on Bacillus subtilis M45 Rec- and H17 Rec+ strains.

Because they are used in a number of commercial preparations phenoxyacetic acids and their salts can occur in wastewater. During their degradation genotoxic substances may be created. The results of investigations of biodegradability and genotoxicity of some phenoxyherbicides are presented. Commercial formulations of 2,4-D (Aminopielik 720) and MCPA (Chwastox Extra) were the substrates studied. Biodegradation tests were conducted according to OECD guidelines for testing of chemicals--confirmatory test (OECD Method 303 A). Genotoxicity tests were conducted with Bacillus subtilis strains according to the method of [Chemical Mutagens, vol. 6, Plenum Press. New York, 1980, p. 149]. Genotoxicity of biodegradation products was also studied. Both commercial formulations were biodegradable. Aminopielik 720 was potentially genotoxic but only at great concentrations while Chwastox Extra was not genotoxic. Biodegradation products of neither compound were genotoxic.

Xenobiotics removal from polluted water by a multifunctional constructed wetland.

Removal efficiencies on xenobiotics from polluted water in a twin-shaped constructed wetland consisting of a vertical flow chamber with the crop plant Colocasia esculenta L. Schott and a reverse vertical flow one with Ischaemum aristatum var. glaucum Honda, were assessed by chemical analysis and bioassays. After a four-month period of application, removal efficiencies of the applied pesticides parathion and omethoate were 100% with no detectable parathion and omethoate in the effluent. For the applied herbicides, the decontamination was less efficient with removal efficiencies of 36% and 0% for 4-chloro-2-methyl-phenoxyacetic acid and dicamba, respectively. As shown by toxicity assay with duckweed Lemna minor L., growth retardation may occur if the water treated for herbicide removal is used in irrigation of sensitive cultivars in agriculture or horticulture. In contrast to I. aristatum var. glaucum Honda, the crop C. esculenta L. Schott has a high yield in biomass production as a valuable source of renewable energy.

Formation of chloroacetic acids from soil, humic acid and phenolic moieties.

The mechanism of formation of chloroacetates, which are important toxic environmental substances, has been controversial. Whereas the anthropogenic production has been well established, a natural formation has also been suggested. In this study the natural formation of chloroacetic acids from soil, as well as from humic material which is present in soil and from phenolic model substances has been investigated. It is shown that chloroacetates are formed from humic material with a linear relationship between the amount of humic acid used and chloroacetates found. More dichloroacetate (DCA) than trichloroacetate (TCA) is produced. The addition of Fe(2+), Fe(3+) and H(2)O(2) leads to an increased yield. NaCl was added as a source of chloride. We further examined the relationship between the structure and reactivity of phenolic substances, which can be considered as monomeric units of humic acids. Ethoxyphenol with built-in ethyl groups forms large amounts of DCA and TCA. The experiments with phenoxyacetic acid yielded large amounts of monochloroacetate (MCA). With other phenolic substances a ring cleavage was observed. Our investigations indicate that chloroacetates are formed abiotically from humic material and soils in addition to their known biotic mode of formation.

Retention of organic and inorganic chemicals by the drainage/supply piping material.

A critical issue facing the turfgrass industry is the environmental fate and transport of organic and inorganic chemicals used on golf courses. The fate and distribution of those chemicals are strongly influenced by sorptive interactions with soil and sediment. In this study, the drainage and water supply piping material (used for construction of a prototype encapsulated golf green) was utilized to determine its potential sorption of three organic chemicals [2,4-dichloro-phenoxyacetic acid] (2,4-D), naphthalene and toluene and nitrate. Crushed piping material (small-to-large particle sizes) was evaluated. Isotherms were constructed using a batch equilibration technique. The results showed that the drainage/supply piping material at small particle sizes (<2.5 mm) has higher sorptive ability compared to soil (1.7 for 2,4-D and 13.4 for naphthalene). The K(F) value was 44, 253 and 70 for 2,4-D, naphthalene and toluene, respectively. K(oc) values were much higher than those of peat and soil at lower equilibrium concentrations. However, sorption decreased dramatically with increasing particle size (approaching zero at particle size 10 mm), due to reduction of surface areas and sorption sites. Sorption of NO(3)-N by the piping material was negligible. We concluded that sorption by intact drainage/supply piping material would not affect the recycling efficiency of pesticides and nutrients in the constructed encapsulated green. Conversely, drainage/supply piping material particles smaller than 2.5 mm in diameter can effectively be utilized as a filtering material.
