[Studies of a new adsorbent cross-linked agar beads entrapped attapulgite clay for hemoperfusion].

The preparation and properties of a new adsorbent cross-linked agar beads entrapped attapulgite clay (CAA) are reported in detail. Medicinal attapulgite clay made in China was encapsulated with agar, shaped in organic solvent, and cross-linked by epichlorohydrin. The products withstood autoclaving at 121 degrees C for half an hour with no breakage, and had a good selective adsorption ability for some model compounds and medicaments such as methylene blue and phenothiazine drugs. Preliminary investigations and SEM photographs indicate that the CAA as an adsorbent is relatively hemocompatible for hemoperfusion.

The structure of Ap(4)A hydrolase complexed with ATP-MgF(x) reveals the basis of substrate binding.

Ap(4)A hydrolases are Nudix enzymes that regulate intracellular dinucleoside polyphosphate concentrations, implicating them in a range of biological events, including heat shock and metabolic stress. We have demonstrated that ATP x MgF(x) can be used to mimic substrates in the binding site of Ap(4)A hydrolase from Lupinus angustifolius and that, unlike previous substrate analogs, it is in slow exchange with the enzyme. The three-dimensional structure of the enzyme complexed with ATP x MgF(x) was solved and shows significant conformational changes. The substrate binding site of L. angustifolius Ap(4)A hydrolase differs markedly from the two previously published Nudix enzymes, ADP-ribose pyrophosphatase and MutT, despite their common fold and the conservation of active site residues. The majority of residues involved in substrate binding are conserved in asymmetrical Ap(4)A hydrolases from pathogenic bacteria, but are absent in their human counterparts, suggesting that it might be possible to generate compounds that target bacterial, but not human, Ap(4)A hydrolases.

Contribution of intracellular calcium stores to an increase in cytosolic calcium concentration induced by Mannheimia haemolytica leukotoxin.

The contribution of intracellular calcium stores to Mannheimia haemolytica leukotoxin (LKT)-induced increase in cytosolic calcium concentration was studied by pharmacologically inhibiting transport of calcium across the plasma and endoplasmic reticulum membranes of bovine neutrophils exposed to LKT. Active intracellular storage of calcium by sarcoplasmic/endoplasmic reticulum calcium ATPase, influx of extracellular calcium across the plasma membrane, and release of stored calcium via inositol triphosphate receptors and ryanodine-sensitive calcium channels were inhibited using thapsigargin, lanthanum chloride, xestospongin C, and magnesium chloride, respectively. Pre-incubation with thapsigargin attenuated the increase in cytosolic calcium concentration produced by LKT, thus confirming the involvement of intracellular calcium stores. Inhibitory effects of lanthanum chloride, xestospongin C, and magnesium chloride indicated that the increase in cytosolic calcium concentration induced by LKT resulted from both influx of calcium across the plasma membrane and release of calcium from intracellular stores.

Identification and mutational analysis of Mg2+ binding site in EcoP15I DNA methyltransferase: involvement in target base eversion.

EcoP15I DNA methyltransferase catalyzes the transfer of the methyl group of S-adenosyl-l-methionine to the N6 position of the second adenine within the double-stranded DNA sequence 5'-CAGCAG-3'. To achieve catalysis, the enzyme requires a magnesium ion. Binding of magnesium to the enzyme induces significant conformational changes as monitored by circular dichroism spectroscopy. EcoP15I DNA methyltransferase was rapidly inactivated by micromolar concentrations of ferrous sulfate in the presence of ascorbate at pH 8.0. The inactivated enzyme was cleaved into two fragments with molecular masses of 36 and 35 kDa. Using this affinity cleavage assay, we have located the magnesium binding-like motif to amino acids 355-377 of EcoP15I DNA methyltransferase. Sequence homology comparisons between EcoP15I DNA methyltransferase and other restriction endonucleases allowed us to identify a PD(X)n(D/E)XK-like sequence as the putative magnesium ion binding site. Point mutations generated in this region were analyzed for their role in methyltransferase activity, metal coordination, and substrate binding. Although the mutant methyltransferases bind DNA and S-adenosyl-l-methionine as well as the wild-type enzyme does, they are inactive primarily because of their inability to flip the target base. Collectively, these data are consistent with the fact that acidic amino acid residues of the region 355-377 in EcoP15I DNA methyltransferase are important for the critical positioning of magnesium ions for catalysis. This is the first example of metal-dependent function of a DNA methyltransferase. These findings provide impetus for exploring the role(s) of metal ions in the structure and function of DNA methyltransferases.

[Analysis of the composition of 2700 urinary calculi examined by infrared spectrophotometry, 1979-1989].

Two thousand seven hundred samples, received as urinary calculi, in a Turin based laboratory, in the period between March 1979 and February 1989, were examined by infrared spectroscopy to identify components even of difficult characterization: a special procedure was used and it is described. Knowing the composition of each calculus, a comparison was made between the incidence of each compound in the first 1500 samples (1979-1984) and in the remaining 1200 ones (1984-1989). Besides an increase in calcium oxalate as main component, a marked reduction in incidence of struvite was found in the calculi belonging to the second period of observation. The possible interference in calculi composition by various therapy for renal infection is discussed.

Transition-state analogs as inhibitors of human and malarial hypoxanthine-guanine phosphoribosyltransferases.

The proposed transition state for hypoxanthine-guanine phosphoribosyltransferases (HGPRTs) has been used to design and synthesize powerful inhibitors that contain features of the transition state. The iminoribitols (1S)-1-(9-deazahypoxanthin-9-yl)-1,4-dideoxy-1,4-imino-D-ribitol 5-phosphate (immucillinHP) and (1S)-1-(9-deazaguanin-9-yl)-1,4-dideoxy-1,4-imino-D-ribitol 5-phosphate (immucillinGP) are the most powerful inhibitors yet reported for both human and malarial HGPRTs. Equilibrium binding constants are >1,000-fold tighter than the binding of the nucleotide substrate. The NMR spectrum of malaria HGXPRT in the Michaelis complex reveals downfield hydrogen-bonded protons. The chemical shifts move farther downfield with bound inhibitor. The inhibitors are lead compounds for species-specific antibiotics against parasitic protozoa. The high-resolution crystal structure of human HGPRT with immucillinGP is reported in the companion paper.

Inorganic compounds have dual effect on recombinant protein production: influence of anions and cations on serine alkaline protease production.

AIMS: Investigation of concerted effects of cations, i.e. Mg2+ and Mn2+, in combination with their anions, i.e. sulphate, chloride and acetate (Ac), on the physiology of Bacillus licheniformis carrying pHV1431::subC to improve the fermentation medium for serine alkaline protease (SAP) production, whereupon, determination of the acid that can be used in pH control. METHODS AND RESULTS: The cell concentrations increased with the increase in MnSO4 and Mn(Ac)2 concentrations, and the highest values were obtained at Co(MnSO4) = 0.20 mmol l-1 and Co(Mn(CH3COO)2) = 4.0 mmol l-1, as 2.3 and 2.2 g l-1, respectively. However, Co(MnCl2) did not influence biomass concentration. SAP production was inhibited with MnCl2 after Co(MnCl2) = 0.60 mmol l-1, but with MnSO4 SAP production was inhibited drastically. Whereas, at high concentrations of Mn(Ac)2 SAP production increased and the highest activity was obtained as ASAP = 1285 U ml-1 at t = 65 h. With the Mg compounds, cell concentrations increased with the increase in the concentrations of MgSO4, MgCl2 and Mg(Ac)2; and the anions did not show any influence on the cell growth. Similar to the results of Mn compounds, the glucose consumption rate increased with the increase in MgSO4 and MgCl2 concentrations; contrariwise, decreased with the increase in Mg(Ac)2 concentrations, due to the use of acetate as the second carbon source. Co(MgSO4) = 0.40 mmol l-1, Co(MgCl2) = 1.60 mmol l-1 and Co(Mg(Ac)2) = 0.40 mmol l-1 were the optimum concentrations separately, and the highest SAP activity was obtained with Mg(Ac)2 as ASAP = 1338 U ml-1 at t = 47 h. Consequently, ion acetate and its acid HAc appear, respectively, as the superior anion for the essential cations and the control agent for pH control in the bioreactor. Finally, optimum initial concentrations and the concerted effects of Mg(Ac)2 and Mn(Ac)2 were investigated, and the optimum concentrations were found respectively as 0.40 and 0.80 mmol l-1, while the maximum activity was obtained as ASAP = 1010 U ml-1 at a shortened cultivation time of t = 39 h. CONCLUSIONS: Mn(Ac)2 and Mg(Ac)2 together enhanced the cell formation and SAP synthesis rates, moreover, SAP synthesis started at an earlier cultivation time. SIGNIFICANCE AND IMPACT OF THE STUDY: Each inorganic compound with its cation and anion has dual effect on the metabolism. Mg2+ and Mn2+ at their specific concentrations influence the regulation of the pathways that might cause better coupling of supply and demand for the amino acids on the basis of the amino acid composition of the enzyme molecule.

Flexural strength and fracture toughness of Dicor glass-ceramic after embedment modification.

Previous studies on the surface properties of Dicor castable glass-ceramic have shown the formation of a specific crystalline phase at the glass-ceramic/embedment interface. If this phase is not removed by grinding, it leads to an undesirable strength decrease. The aims of this study were: (1) to determine the nature of this surface layer, (2) to promote the formation of a different crystalline phase at the surface with the intention of improving the properties of the glass-ceramic, by modification of the composition of the Dicor ceramming embedment, and (3) to evaluate the fracture toughness and flexural strength of Dicor glass-ceramic after embedment modification. Modifications were made to the embedment by incorporation of 2.5 wt% of lithium fluoride and ceramming at various temperatures. X-ray diffraction was used to determine the crystalline nature of the surface layer. Fracture toughness was investigated by the indentation technique. The maximum bi-axial stresses were calculated after the samples were fractured in water on a ball-on-ring fixture at 0.5 mm/min. With the recommended embedment and ceramming cycle, the crystalline phase constituting the ceram layer was a calcium magnesium silicate CaMg(SiO3)2 (diopside). The crystalline composition of the ceram layer was successfully modified by addition of 2.5 wt% lithium fluoride to the embedment. This promoted the crystallization of mica in the ceram layer and increased the fracture toughness of the glass-ceramic when the ceramming temperature was 950 or 975 degrees C. The flexural strength was significantly increased when the ceramming temperature was 1000 degrees C.

Trimagnesium orthophosphate in renal calculi.

We recently described a new component of calculi, trimagnesium orthophosphate. In a microscopical study of thin sections from 3500 urinary calculi we detected this compound in 14 stones, all of which contained struvite. New data suggest that trimagnesium orthophosphate is a transformation product of struvite. This well-crystallized substance was always detected a few days after the surgical extraction of struvite kidney stones, in contrast to the newberyite that appears on the surface of very old removed stones or after long exposure of stones to the atmosphere.

Agglomeration of struvite crystals.

Struvite crystallisation is widely studied as a way to remove phosphorus from wastewater effluents and simultaneously generates a valuable product for the fertiliser industry. However, to date, some crystallisation processes experimented at either pilot/or full scale face problems linked to the formation of fines. This paper presents results on the investigation of struvite agglomerative properties and the possible application of coagulants and/or flocculants to remove fines. Coagulants investigated were hydrolysing metals salts (Al(3+) and Fe(3+)), calcium compounds and a cationic polymer, polydiallyldimethylammoniumchloride (polyDADMAC). The effects of a natural flocculant (alginate) have also been tested. Results demonstrated that destabilisation of struvite particles by chemical addition was feasible and identified polyDADMAC as a good option for the agglomeration of struvite particles. However, optimisation of its dosage under typical pH conditions for struvite formation showed floc formation to be very pH sensitive.

Lithium fluoride: from LiF:Mg,Ti to LiF:Mg,Cu,P.

Differences and similarities between LiF-based LiF:Mg,Ti and LiF:Mg,Cu,P are discussed, with respect to their dosimetric properties--sensitivity, non-linearity of dose response and heavy charged particle efficiency, as related to the concentration and the individual role of the Mg, Ti, Cu and P dopants. To study further the role of these dopants, the properties of some new, 'hybrid' phosphors: LiF:Mg,Cu,Ti and LiF:Mg,P, specially developed for this purpose, are also discussed. In the glow curve of LiF:Mg,Cu,P with a low concentration of Mg a new peak was found, which appears to be an analogue of peak 4 in LiF:Mg,Ti, Magnesium apparently controls most of the dosimetric properties of LiF-based phosphors. For instance, charged-particle efficiency appears to be anti-correlated with the concentration of Mg, being much less dependent on the content of other dopants. On the other hand, some properties of LiF-based systems seem to be correlated with changes in the emission spectra. It is suggested that Ti hampers the acceptance of any increased amount of Mg into more traps in LiF:MgTi. The absence of Ti, not the presence of P or Cu, is therefore a key to the high sensitivity of LiF:MgCuP.

TLD linearity vs. beam energy and modality.

Thermoluminescent dosimetry (TLD) is considered to be a valuable dosimetric tool in determining patient dose. Lithium fluoride doped with magnesium and titanium (TLD-100) is widely used, as it does not display widely divergent energy dependence. For many years, we have known that TLD-100 shows supralinearity to dose. In a radiotherapy clinic, there are multiple energies and modality beams. This work investigates whether individual linearity corrections must be used for each beam or whether a single correction can be applied to all beams. The response of TLD as a function of dose was measured from 25 cGy to 1000 cGy on both electrons and photons from 6 to 18 MeV. This work shows that, within our measurement uncertainty, TLD-100 exhibits supralinearity at all megavoltage energies and modalities.

A comparative study of the shear bond strengths of four different crystal growth solutions.

Previously, solutions based on polyacrylic acid have been found to initiate crystal growth on the enamel surface. Such crystals have been proposed as being suitable for the attachment of orthodontic brackets via a conventional composite interface, the advantages being improved clean-up characteristics and reduced damage to the enamel. In order to try and improve the reliability of the technique this base solution has been modified by the authors, by the addition of various ionic salts. The strength of the resultant bond was assessed for the base solution of 50 per cent polyacrylic acid and concentrated sulphuric, and for three more solutions modified by the addition of the sulphates of lithium, magnesium, and potassium, respectively. A standard acid/etch method of bracket attachment was also included for the purposes of comparison. The bond strength of each material was assessed by shear testing, performed on human extracted premolar teeth to a standard method. As might have been expected the acid/etch system proved to be the strongest method of bonding brackets. Amongst the crystal solutions, the addition of lithium sulphate provided the highest mean shear strength at 80 per cent of that for acid etch. Therefore, this latter solution provides the most potential for development in the future.

Hydrogenation properties of mechanically milled Mg2Ni0.8Cr0.2-CoO/Al2O3 composites.

Mg2Ni0.8Cr0.2-x wt.% CoO/Al2O3 (x=0.5, 1, 2 and 3) composites were prepared by mechanically milling sintered Mg2Ni0.8Cr0.2 alloy and CoO/Al2O3 compound for 45 h. The addition of CoO/Al2O3 compound resulted in the good kinetics properties of hydriding/dehydriding reaction of the composites. The composite with 1.0 wt.% CoO/Al2O3 catalyst could reach the maximum hydrogen absorption capacity (2.9 wt.%) within 5 min at 393 K under H2 pressure of 4 MPa, and can desorb rapidly at 493 K. The decomposition and synthesis of hydrogen molecule on Mg2Ni0.8Cr0.2 alloy surface was promoted by addition of CoO/Al2O3 catalyst. In addition, the formation of metallic Ni particles, strain and defects during the ball milling process also resulted in the improved hydrogenation performance of Mg2Ni-based alloys.

Unexpected Fe local order in iron oxide coated nanocrystalline magnesium oxides with exceptional reactivities against environmental toxins.

Mg oxide nanoparticles are very reactive materials used to mitigate atmospheric pollution and to sequester polluting molecules. Using Fe K-edge XAFS, we have studied the structure of iron oxide-coated MgO nanoparticles before and after reaction with CCl4. Before reaction, the local structure around Fe is totally different from that in iron oxide coatings on SrO and CaO nanoparticles, although these coated materials were prepared in the same way. In SrO and CaO, the iron oxide coating has been shown to be well separated from the bulk of the nanoparticle, whereas in MgO, Fe was found to mix with MgO. After reaction with CCl4, Fe-Cl bonds can be detected when the coated nanoparticle is saturated. Such Fe-Cl EXAFS signals have not been observed in previously studied nanoparticles.

A new method using a bacterium for dissolution of urinary stones.

PURPOSE: A bacterium, Thiobacillus thiooxidans, oxidizes inorganic sulfur compounds to sulfuric acid to obtain energy for growth. We examined dissolution of urinary stones by the bacteria and studied optimal conditions for the dissolution in human urine. METHODS: Thiobacillus thiooxidans (IFO No. 13 701) was purchased from Hakko Kenkyujyo, Co., Osaka, Japan. The bacteria were grown stationary for 7 days at 30 degrees C in medium. The pH of the medium was changed from pH 4.82 to 1.95 for 28 days. Growth of the bacteria was also examined in human urine by addition of different amounts of sodium thiosulfite. The dissolution of human urinary stones was examined in the bladder of Wistar rat containing the bacteria (1 x 10(8)) for 7 days. 20 ml of 5 % sodium thiosulfite were injected intraperitoneally once a day. Histological changes of urinary tracts and blood test were also examined. As a control, two rats were examined in the same methods without the bacteria. RESULTS: The number of bacteria was increased in human urine depending on the concentration of sodium thiosulfite. Among different kinds of urinary stones, calcium phosphate stone was most dissolved, 78 % reduction of dry weight in the rat bladder. Calcium oxalate, uric acid and magnesium phosphate stones were not well dissolved. But the stone weights in the control rats were increased twice. Slight inflammation was found in the rat bladder. There were no abnormal findings in the blood test. CONCLUSIONS: Thiobacillus thiooxidans can be used for dissolution of urinary stones. This new technique may be useful for dissolution of fragments after ESWL and crystals covered by a urinary stent. Optimal conditions and safety methods should be developed for clinical use.

In vitro prediction of canine urolith mineral composition using computed tomographic mean beam attenuation measurements.

Determination of urolith mineral composition is critical for management of urolithiasis in dogs and cats. Using computed tomography, urolith physical density, and hence chemical composition, can be quantified using mean beam attenuation measurements (Hounsfield units; HU). This study was designed to establish in vitro reference ranges for three types of compositionally pure uroliths retrieved from dogs. Sixty-six canine uroliths (22 uric acid, 21 calcium oxalate, 14 struvite, nine mixed or compound) were placed in a phantom array. Uroliths were scanned at 120 kVp, 200 mA, and 80 kVp, 200 mA. The region of interest (ROI) for mean HU calculation was determined using two techniques, and reference ranges were calculated for each kVp using either ROI technique. HU for urolith types of pure composition were statistically different (Wilcoxon's two-sample test, P < 0.0083 [Bonferonni correction with six comparisons for total P < 0.05]) using both ROI techniques at either kVp. Struvite uroliths were not statistically different from mixed or compound uroliths. The accuracy for determination of composition of pure uroliths ranged from 86% to 93%; the prediction accuracy for each urolith mineral type and for all uroliths in general was highest when the ROI was hand-drawn just within the visible urolith border at 80 kVp. Technique of ROI determination and kVp that yielded the highest sensitivity, specificity, and positive and negative predictive values varied for each urolith type. Therefore, in this study, HU could be used to differentiate three types of uroliths of pure mineral composition in vitro. Further studies are needed to determine the predictive value of HU in vivo.
