Microfiches as an aid to teaching histology.

A set of six colour microfiches and an atlas of black-and-white prints of the photomicrographs used in the microfiches have been prepared by the author and have been used for teaching an Histology course to medical students for 4 years. Students reaction to them and their use has been gauged by voluntary questionnaires. Of students who responded, 78% found the microfiches made the course easier for them; 76% like the microfiches because they can use them for home study; 77% find them useful for class discussion; 62% like them because of their colour reproduction. A small group of fifteen students, who failed the Histology course the year prior to the introduction of microfiches and subsequently repeated the course, felt they could revise better with the help of the microfiches. Also they felt the course was clearer. Comparison of the examination results of the students for the 4 years preceding the introduction of microfiches with those of the 4 years since, shows an improvement in mean percentage achieved together with a decrease in the standard deviation (s.d.).

Microcinematographic analysis of the motility of human spermatozoa incubated with caffeine.

The effects of incubation with caffeine at final concentration of 0.5 and 5.0 mM for 30 min at ambient temperature were examined in three normal semen samples and three from asthenozoospermic men. A 50 frames/s microcinematographic analysis of the sperm motility was performed and two characteristic parameters of the trajectories, the progression velocity (Vp) and the amplitude of lateral head displacement (Ah), were measured. At the lower concentration (0.5 mM) no significant effects on the overall mean values (i.e. for all spermatozoa analyzed over the 6 samples) of Vp Ah were found. However, at 5.0 mM a significant reduction in the overall mean Vp was noted. The individual responses were variable between the 6 samples, but only in two of the asthenozoospermic samples and one normal ejaculate were the slow-swimming (less than 20 micrometer/s) spermatozoa accelerated by caffeine treatment. In other samples a reduction in the velocity of faster-swimming spermatozoa was observed. It would seem preferable to reserve caffeine treatment only for severe cases of asthenozoospermia where the distribution of sperm velocities of skewed towards low values.

Infrared color photography: selective demonstration of bacteria.

A microscopic procedure employs infrared color photography to enable visualization of unstained microorganisms in natural habitats. The procedure can be used for either living or dead microorganisms; it enables visualizaion of microbial cells embedded within component materials of the habitat.

Size-separation of yeast mitochondria in the zonal centrifuge.

Mitochondria, released from yeast spheroplasts and subjected to rate separation through sorbitol gradients in the zonal centrifuge, migrated in a wide symmetrical zone. Electron micrographs showed that the mitochondria had been resolved within the zone according to size. The mean mitochondrial diameter at the leading edge was approximately twice that at the trailing edge of the particle zone. Activities of the enzymes cytochrome oxidase, malate dehydrogenase, and reduced nicotinamide adenine dinucleotide- and d-lactate cytochrome c reductases were essentially uniform throughout the mitochondrial zone. Mitochondria from a vegetative-petite mutant had almost the same size distribution as the isogenic wild type, but with somewhat larger mean diameter and either absent or markedly reduced enzyme activities. Mixtures of wild-type and petite mitochondria produced sedimentation profiles showing overlap of particle populations with respect to mean sedimentation rates and mitochondrial diameters, as well as intermediate levels of enzyme activities. Both cristate and noncristate organelles were present throughout the mitochondrial zone from these mixtures. Mitochondria centrifuged in sorbitol density gradients were well-preserved and yielded consistent sedimentation profiles, whereas particles in sucrose density gradients migrated more slowly, produced varied sedimentation profiles, and often showed spurious peaks, presumably due to particle aggregations.

Microfiche as a medium for the long-term storage of laboratory computer records.

The chemical pathology requests on 180 000 patients a year are stored on microfiche, occupying 72 mm of shelf space. They are produced by a sequence of three computer programs which remove data from disc on to magnetic tape using the laboratory's Digital Equipment Corporation PDP 11/34 minicomputer. Processing on to microfiche is performed by a bureau. The magnetic tape is available for retrospective research and management studies in one-month periods.
