Differential expression of CCL19 by DC-Lamp+ mature dendritic cells in human lymph node versus chronically inflamed skin.

De novo formation of lymphoid tissue is one of the characteristic features of chronic inflammation. The formation of T cell-mature dendritic cell (DC) clusters has been previously demonstrated in chronically inflamed skin infected with Candida albicans. A functional similarity was also found between chronic inflammation and the T-cell zone of lymph nodes (LNs), since a substantial fraction of phenotypically mature DCs in both tissues expressed CCL22 (macrophage-derived chemokine; MDC) and were closely surrounded by memory-type T cells expressing its receptor, CCR4. To analyse the nature of T cell-mature DC interactions further in chronically inflamed skin and LNs, the present study focuses on another chemokine system, namely CCL19 (EBI1 ligand chemokine; ELC), CCL21 (secondary lymphoid tissue chemokine; SLC) and their shared receptor, CCR7. RT-PCR analysis revealed expression of CCL19, CCL21, and CCR7 at high levels in LNs and at low levels in inflamed skin. Using immunohistochemistry, the majority of DC-Lamp(+) mature DCs in the T-cell area of LNs expressed CCL19 and were surrounded by CCR7(+) naïve-type lymphocytes, while CCL21 was expressed in reticular stromal cells and vascular endothelial cells. Very few mature DCs in LNs were found to express CCR7. In contrast, the majority of DC-Lamp(+) mature DCs in inflamed skin were totally negative for CCL19 and were surrounded by CCR7(-) memory-type T cells. Furthermore, CCL21 expression in the inflamed skin was detected in dermal lymphatic endothelial cells and rare CCR7(+) mature DCs were mostly seen within the lymphatic vessels. In normal skin, on the other hand, no cells immunoreactive for CCL19, CCL21, or CCR7 were found. The present study thus reveals a striking difference in the function of mature DCs between LNs and chronically inflamed skin.

Uvomorulin, LAMP-1, and laminin are substrates for cell surface beta-1,4-galactosyltransferase on F9 embryonal carcinoma cells: comparisons between wild-type and mutant 5.51 att- cells.

Intercellular adhesions in F9 embryonal carcinoma cells are primarily dependent upon two types of cell adhesion molecules: uvomorulin (Um or E-cadherin), which facilitates intercellular adhesion by homophilic binding, and cell surface beta-1,4-galactosyltransferase (GalTase), which binds terminal N-acetylglucosamine residues on consociate glycoprotein substrates on adjacent cell surfaces. The variant F9 cell line, 5.51 att-, undergoes initial cell aggregation, but fails to form the mature intercellular adhesions characteristic of wild-type F9 cells. The 5.51 att- cells show reduced Um expression; however, previous studies have shown that reduced levels of Um are not responsible for the att- mutant phenotype. Therefore, in this study we determined whether the att- mutant phenotype was the result of altered expression of surface GalTase or of its consociate glycoprotein substrates. Surface GalTase was found to be expressed normally on 5.51 att- cells. Furthermore, the initial intercellular adhesions characteristic of reaggregating 5.51 att- cells were dependent upon surface GalTase, since anti-GalTase antibodies inhibited 5.51 intercellular adhesion. Preliminary studies showed that the interaction of surface GalTase with its consociate glycoprotein substrates was abnormal on 5.51 att- cells. To define the biochemical basis for this observation, specific glycoprotein substrates for cell surface GalTase were identified and their expression and utilization were compared between wild-type and 5.51 att- cells. In wild-type cells, uvomorulin (Um), lysosome-associated membrane protein-1 (LAMP-1), and laminin (Lm) were shown to be substrates for cell surface GalTase, suggesting that they may participate in GalTase-specific adhesions. Moreover, the interaction between GalTase and these glycoproteins exhibited characteristic changes during retinoic acid-induced F9 cell differentiation. In contrast to that seen on wild-type cells, surface GalTase interaction with Um, LAMP-1, and Lm was atypical on 5.51 att- cells, resulting, in part, from increased synthesis of oligosaccharide substrates for surface GalTase, as determined by size exclusion and lectin affinity chromatography. N-Acetylglucosaminyltransferase activity was elevated in 5.51 att- cells and likely responsible for the increased expression of GalTase oligosaccharide substrates. These results suggest that mutant 5.51 att- cells are able to maintain residual intercellular adhesions because of increased expression of oligosaccharide substrates for cell surface GalTase. The impact of aberrant GalTase-specific adhesions on the ability of 5.51 att- cells to form mature intercellular adhesions and to differentiate in response to retinoic acid is discussed.

Forskolin blocks the apical expression of dipeptidyl peptidase IV in Caco-2 cells and induces its retention in lamp-1-containing vesicles.

In a previous work, we showed that the differentiation-dependent expression of dipeptidyl peptidase IV (DPP IV) in Caco-2 cells, a human colon adenocarcinoma cell line, was controlled at the mRNA level (D. Darmoul et al. J. Biol. Chem., 1992, 267, 4824-4833). Whether post-translational events may contribute to the final control of DPP IV cell surface expression was explored here by studying the potential effect of forskolin (FK), a drug known to permanently stimulates adenylyl cyclase and to strongly perturbs glucose metabolism in fully differentiated Caco-2 cells. FK treatment reduces by about 50% the amount of active DPP IV present at the brush border membrane, whereas the total amount of active DPP IV remains unchanged. Biosynthesis and maturation of DPP IV were measured using [35S]methionine labeling and were shown to be essentially unaffected by FK treatment. Pulse-chase experiments demonstrate that up to 50% of the neosynthesized DPP IV do not appear at the apical membrane after FK treatment. To get further insight into this phenomenon, we have used confocal laser scanning microscopy. We demonstrate that the blockade of DPP IV transport is associated with the accumulation of this protein in intracellular vesicles. Double-staining experiments demonstrate that these vesicles are not labeled with a monoclonal antibody directed against the Golgi apparatus but display a strong staining with a polyclonal antibody raised against lamp-1, a lysosomal membrane protein. Using a newly developed image analysis procedure, we have been able to quantitate the relative distribution of lamp-1 and DPP IV labels in both control and forskolin-treated cells. We show that the overlap of the two labels dramatically increases in FK-treated Caco-2 cells. These results suggest that, beside the transcriptional level, post-translational events may be involved in the final control of the apical expression of a differentiation-dependent hydrolase.

On slit-lamp microscopy.

Theoretically the contact lens with a refractive power of -64 dioptres is superior to the concave pre-set lens (Hruby lens) or the convex lens (Bayadi). In practice the results are as follows: With the convex lens the examination of the vitreous is unsatisfactory. The reversed image with the convex lens will become familiar with some experience as one will also get used to the mirror images of the three-mirror contact lens. However, the abnormal curvature of the image is really a nuisance. On the other hand the large field of view is an advantage. The convex lens is only practically useful when it can be firmly attached to the slit-lamp microscope, as otherwise the new adjustment necessary for searching the fundus is too difficult. Attachment to the microscope is also of advantage with the Hruby lens. The concave contact lens of 64 dioptres is best suited for detailed examination of vitreous and fundus and for measurements. When no value is placed on a minute examination and a quick simple orientation suffices, the concave pre-set lens (Hruby lens) has the advantage that the lens does not have to be placed on the eye. If it is centred to the middle axis of the microscope, it facilitates especially quick observation. There will be cases where the pre-set lens will be the only answer, especially shortly after an operation or with very sensitive patients. Furthermore, the Hruby lens is preferable to the Bayadi lens because the vitreous can be better examined. The Hruby lens is also advantageous for examination of the region in the middle, lateral periphery of the fundus, 30 degrees - 60 degrees, and is superior in this area to the simple concave contact lens. The Bayadi lens seems to us only indicated for fundus examination of extremely high myopes. There it definitely offers advantages over and above the concave lens. For the most peripheral fundus, expecially below and above, the three-mirror lens with its modification is so far the best method. Especially for examination of the vitreous good brightness of the slit image is required for stereoscopic examination with as large an angle as possible between microscope and illumination. The lateral parts of the most peripheral fundus cannot be examined with the vertical slit in connection with the three-mirror lens. However, this is possible with the horizontal and tilted position of the slit and intermediate positions with an oblique slit. The slit must form an angle with the microscope in order to examine vitreous and fundus in optical section. With the indentation contact lenses ciliary processes and pars plana are now accessible to slit-lamp microscopy.

Modifications of the slit lamp for digital image processing of the anterior segment of the eye.

The Zeiss slit lamp has been modified in order to extend its use to measurements of lens transparency. Two major modifications have been introduced: (1) a support for a high-sensitivity TV camera that is connected to an image processing system; (2) two potentiometers for recording electrical signals proportional to the rotation angle of the slit-supporting arm with respect to the visual axis and the slit tilting angle. As a result, the slit lamp output consists of three signals; one TV signal for the images and two analog signals for measurement of the angles. With the aid of an image-processing system connected to a minicomputer, software has been developed to enable the operator to acquire reliable digital images of the anterior segment of the eye in real time. The software provides the user with an easy-to-follow menu.

Treatment of superficial basal cell carcinomas using topically applied delta-aminolaevulinic acid and a filtered xenon lamp.

OBJECTIVE: To evaluate the treatment of superficial basal cell carcinomas (SBCC) using photodynamic therapy with topically applied delta-aminolaevulinic acid (ALA) and a filtered short arc xenon lamp as a light source. METHOD: An oil-in-water emulsion containing 20% ALA was applied topically to the lesion sites which were then occluded for 3 h. Irradiation was performed in a single session of 10 min using 125 or 166 mW/cm2 in the wavelength range (620-670 nm, giving a radiation dose of 75 or 100 J/cm2, respectively. Urine samples obtained in connection with the treatment were examined for traces of porphobilinogen and porphyrin. The therapy outcome was determined by histological examination of punch biopsies or by clinical verification. A follow-up scheduled for 5 years was initiated. PATIENTS: A group of 37 patients with histologically verified SBCCs were included. A total of 190 SBCCs were treated. Also included were six patients with 10 nodular basal cell carcinomas and five patients with 18 Mb. Bowen. The urine samples from three patients showed traces of porphyrins. RESULTS: Of the 157 SBCCs followed-up for 6 months, 144 were cleared giving a clearance rate of 92%. Of the nodular basal cell carcinomas and Mb. Bowens, 2 of ten and 11 of 18 were cleared, respectively. CONCLUSION: Photodynamic therapy is well suited to the treatment of SBCCs giving good cosmetic results regardless of lesion size. The filtered xenon lamp proved to be particularly suitable for photodynamic therapy.

Applications of surface analytical techniques for study of the interactions between mercury and fluorescent lamp materials.

Several surface analytical techniques, including electron spectroscopy for chemical analysis (ESCA)(X-ray photoelectron spectroscopy) and sputtered neutral mass spectrometry (SNMS), were used to study the interaction between Hg and other components of fluorescent lamps, a very critical issue in lighting industries. Active sites, responsible for Hg interaction/deposition, can be successfully identified by comparing the x- y distribution (obtained by ESCA mapping) and depth distribution (available through SNMS) of respective lamp components with that of Hg. A correlation in both depth and x- y distribution is strong evidence of site preference for Hg interaction/deposition. A burial mechanism is, however, proposed when only depth distribution, not x- y, is correlated. Other modes of ESCA (high resolution, angle-resolved, etc.) were also helpful. Information about the valence states of the interacted Hg species would help to define the nature of the interaction.

[Pathologically reduced endothelial cell number despite normal slit-lamp microscopic corneal findings. An important result before cataract surgery].

BACKGROUND: A reduced number of endothelial cells increases the risk of corneal decompensation after cataract surgery. It is difficult to quantify the number of endothelial cells using slit-lamp microscopy since normal corneas may also show a reduced number of endothelial cells. MATERIALS AND METHODS: We compared the number of endothelial cells pre- and postoperatively in a group of 500 consecutive patients. RESULTS: Corneas diagnosed preoperatively with corneae guttata by slit-lamp microscopy may reveal more than 1800 endothelial cells/mm2. These corneas may decompensate after surgery. CONCLUSION: We consider the routine use of endothelial microscopy to be a helpful diagnostic tool prior to cataract surgery.

Changes in expression of the lysosomal membrane glycoprotein, LAMP-1 in interdigital regions during embryonic mouse limb development, in vivo and in vitro.

Syndactyly, a failure of the digits to separate into individual units, affects about 8 to 9 per 1000 newborns and results from an aberration of the normal development of the interdigital tissues. Limb digit separation is the result of programmed cell death (apoptosis). Lysosomes play a role in the process of cell self-destruction. Our experiment was designed to test the hypothesis that the intensity of interdigital lysosomes increases during the separation of digits in vivo and in vitro. The primary mouse monoclonal antibody, 1D4B, detects the presence of lysosomes by identifying the LAMP-1 glycoprotein on the lysosome cell membrane. In our experiment this antibody immunodetected interdigital lysosome proteins in serial sections of limbs from Swiss-Webster mouse embryos, gestational ages E12.5 through E15, key developmental stages for digit separation. Digit separation was associated with an increase in intensity of lysosomal protein staining. In E12.5 limbs, the presence of lysosomes was enriched in the distal aspect of the interdigital tissue. However, the number of lysosomes markedly increased in the E13 and E14 limbs, including the entire length and width of the interdigital tissue in the E14 limbs. This lysosomal protein presence in E14 limbs was significant compared to E12.5, E13, and E15 limbs. By day 12.5, the mouse embryo limb is committed to digit separation. Addition of retinoic acid to the culture medium of limbs earlier in development, such as E12, results in induction of the process of digit separation. Cultured E12 limbs that did not receive an addition of retinoic acid, did not show digit separation. We conclude that in the limb development process, the enrichment in interdigit LAMP-1 proteins, may indicate a relationship between lysosomes, apoptosis, and digit separation. We also conclude that retinoic acid has an important role in digit separation in vivo, as shown in limb development, and demonstrated through the addition of retinoic acid to media of cultured tissues.

First report of tomato spotted wilt virus (Orthotospovirus tomatomaculae) and phytoplasma in China aster and development of duplex PCR, LAMP, and qPCR assays for rapid detection.

UNLABELLED: China aster (Callistephus chinensis) is an economically important ornamental crop widely cultivated for cut flowers and landscaping. During field surveys conducted in three districts of Karnataka, India, China aster plants exhibiting chlorotic and necrotic ring spots, leaf deformation, and witches' broom symptoms were collected and analyzed to determine the causal agents. Mechanical inoculation of symptomatic leaf sap onto cowpea (Vigna unguiculata cv. C-152) produced characteristic chlorotic and necrotic ring spots on newly emerging leaves indicating the presence of an infectious viral agent. Serological assay by DAC-ELISA followed by RT-PCR confirmed the presence of tomato spotted wilt virus (TSWV, Orthotospovirus tomatomaculae) in symptomatic plants. Similarly the plants exhibiting witches' broom symptoms tested positive for phytoplasma infection using universal and Nested primers PCR assays targeting the 16S rRNA gene. Sequence analysis of TSWV CP gene revealed more than 97% nucleotide identity with TSWV isolates reported from India and other countries. Based on these results, one representative isolate was selected for complete genome sequencing. The complete sequences of the L, M, S RNA segements were amplified cloned, and sequenced showing more than 97% nucleotide identity with global TSWV isolates available in database. Sequence analysis of 16S rRNA gene of the phytoplasma associated with witches' broom symptoms was identified as 'Candidatus Phytoplasma australasiaticum' belonging to the 16SrII-D subgroup, sharing 99.2% nucleotide identity with previously reported isolates. Phylogenetic analysis further supported the placement of both the TSWV and phytoplasma isolates within their respective taxonomic groups. To facilitate rapid and sensitive diagnosis, quantitative PCR (qPCR) and RT-LAMP assays were developed for TSWV detection. In addition a duplex PCR assay was optimized for simultaneous detection of TSWV and phytoplasma from infected China aster plants in a single reaction. This study represents the first reports of the complete genome characterization of TSWV and phytoplasma infection in China aster in India along with the development of sensitive qPCR, RT-LAMP, and duplex PCR assays for rapid detection of these pathogens providing valuable tools for disease diagnosis, epidemiological studies. Effectiveness estimates for center high mounted stop lamps: a six-year study.

The center high mounted stop lamp is a supplemental brake light mounted at the height of the rear window that has been required on all new cars sold in the U.S. starting with the 1986 model year. It was intended to reduce the number of rear-end collisions by improving the braking signal recognition of following drivers. Insurance claims for vehicle damage were used to compare the rear-end collision rates of model year 1985 and model year 1986 cars over calendar years 1986-91. After adjusting for the difference in vehicle ages, model year 1986 cars were found to have experienced 5% fewer rear-end collisions than would have been expected without the center high mounted stop lamps.

The effect of synchronization on the interpretation of flashing lamp displays.

The question of whether all the lamps on a display indicating the same state should flash in phase with each other or independently is of interest to display designers. A randomized groups experiment was carried out to answer this question. It was found that a synchronized display was interpreted more quickly and with fewer errors than a display with independently flashing lamps.

Mechanisms for the production of DNA damage in cultured human and hamster cells irradiated with light from fluorescent lamps, sunlamps, and the sun.

Light from fluorescent lamps has recently been shown to be toxic and mutagenic to V-79 hamster cells. In this report it is shown that visible light (> 295 nm) from fluorescent lamps, sunlamps and the sun produces DNA single-strand breaks in hamster or human cells irradiated with these sources. Some of the DNA breaks and the mutagenic action of the light can be averted by filtering the light through para-aminobenzoic acid, which filters out wavelengths below 345 nm. The radical scavengers dimethyl sulfoxide, glycerol, and potassium iodide can also protect the cells from some of the DNA breaks, but the scavengers do not protect V-79 cells from the mutagenic action of the light sources. Hydrogen peroxide and riboflavin and tryptophan photoproducts also are shown to produce DNA breaks in V-79 cells; however, these treatments are not mutagenic to V-79 cells. These data suggest that several types of lesions may be produced in the DNA of irradiated cells by several different mechanisms, and that some of the lesions may be related to toxicity and mutagenicity of the light sources, and others may not.

Responses of spring wheat (Triticum aestivum L.) to ozone produced by either electric discharge and dry air or by UV-lamps and ambient air.

The aim of the present study was to examine if ozone produced similar effects on spring wheat growth with and without small amounts of nitrogen oxides. Two methods were used to produce ozone: the first method consisted of dry pressurized air fed to an electric discharge generator generating the byproducts, N2O5 and N2O, the second method consisted of ambient air fed to UV-lamps. Two spring wheat cultivars (Triticum aestivum L. cvs Minaret and Eridano) were exposed in small open-top chambers to charcoal-filtered air, non-filtered ambient air, and non-filtered ambient air with the addition of ozone for 8 h (0900 to 1700 h) daily, for five weeks. Plants were harvested every week. The growth of Minaret was shown to be more sensitive to O3 than that of Eridano. Leaf senescence increased with increasing ozone level in both cultivars. The total above-ground biomass dry weight decreased with increasing ozone concentration in Minaret, but not in Eridano. The Minaret plants reacted with more damaged leaf dry weight and inhibition of growth when O3 was produced by UV-lamps than when O3 was produced by air fed to an electric discharge generator. This could be explained by more nitrogen content per plant but not by increased nitrogen concentration in plant tissue in plants exposed to increased O3 and small amounts of incidental nitrogen oxides.

Detection of p53 and histopathological classification of skin tumours induced by halogen lamps in hairless mice.

Skin lesions induced by exposure of three strains of female hairless mice to the light emitted by uncovered halogen quartz lamps were subjected to histopathological analysis. We examined 170 representative specimens out of a total of 597 skin lesions, i.e. 38 out of 74 SKH-1 mice, 110 out of 472 MF-1 mice, and 42 out of 51 C3H mice. The results provided evidence of various types of alterations, including preneoplastic changes, such as epidermal hyperplasia, and benign tumours, such as papillomas, as well as tumours with an increasing degree of malignancy, i.e., keratoacanthoma-like tumours, appendage/basal tumours, actinic keratoses/carcinomas in situ, and squamocellular carcinomas. SKH-1 was the most sensitive strain to the far-ultraviolet wavelengths delivered by halogen lamps, as shown not only by the shortest latency time and the highest multiplicity of skin lesions but also by the highest frequency of malignant tumours. Some areas of atypical melanocyte proliferation were only detected in C3H pigmented mice. Eighty-two of the lesions excised from MF-1 mice were additionally examined for p53 protein by immunohistochemical methods. Formalin-fixed, paraffin-embedded sections and frozen sections were analyzed in parallel by using polyclonal CM-1 antibody and monoclonal PAb240 antibody, respectively. A positive response for p53 was only observed in squamocellular carcinomas, and was related to the size of cancers; in fact, six out of 10 cancers of 10-30 mm in diameter were positive, whereas all 16 cancers of 2-9 mm in diameter were negative. All six positive squamocellular carcinomas were detected by using the CM-1 antibody, which recognizes both wild-type and mutant forms of p53 protein, and five of them were also positive with the PAb 240 antibody, which only recognizes the mutant form. Thus, p53 mutation appears to be a late event in the development of halogen-induced skin tumours in hairless mice, requiring a severe degree of malignancy and an advanced stage of the neoplastic mass growth.

Detection of gp43 of Paracoccidioides brasiliensis by the loop-mediated isothermal amplification (LAMP) method.

Paracoccidioidomycosis is a deep mycosis caused by the thermo-dependent dimorphic fungus Paracoccidioides brasiliensis and is prevalent in Latin American countries. We detected the species specific gp43 gene of P. brasiliensis by loop-mediated isothermal amplification (LAMP) in 22 clinical and seven armadillo-derived isolates. The amplified DNA appeared as a ladder with a specific banding pattern. The advantage of the LAMP method is speed; only 3 h were necessary for identification of the organism and diagnosis of the disease. We were also able to obtain positive results from DNA extracted from a paraffin-embedded tissue sample of paracoccidioidomycosis, suggesting that this method may achieve clinical application in the near future.

APPI-MS: effects of mobile phases and VUV lamps on the detection of PAH compounds.

The technique of atmospheric pressure photoionization (APPI) has several advantages over electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI), including efficient ionization of nonpolar or low charge affinity compounds, reduced susceptibility to ion suppression, high sensitivity, and large linear dynamic range. These benefits are greatest at low flow rates (i.e., <or=100 microL/min), while at a higher flow, photon absorption and ion-molecule reactions become significant. Under certain circumstances, APPI signal and S/N have been observed to excel at higher flow, which may be due to a nonphotoionzation mechanism. To better understand APPI at higher flow rates, we have selected three lamps (Xe, Kr, and Ar) and four mobile phases typical for reverse-phase, high-pressure liquid chromatography: acetonitrile, methanol, (1:1) acetonitrile:water and (1:1) methanol:water. As test compounds, three polyaromatic hydrocarbons are studied: benzo[a]pyrene, indeno[1,2,3-c, d]pyrene and benz[a]anthracene. We find that solvent photoabsorption cross-section is not the only parameter in explaining relative signal intensity, but that solvent photo-ion chemistry can also play a significant role. Three conclusions from this investigation are: (1) methanol photoionization leads to protonated methanol clusters that can result in chemical ionization of analyte molecule; (2) use of the Ar lamp often results in greater signal and S/N; (3) acetonitrile photoionization is less efficient and resulting clusters are too strongly bound to chemically ionize the analyte efficiently, so that analyte ion formation is dominated by direct photoionization.

Treatment of MSW landfill leachate by a thin gap annular UV/H2O2 photoreactor with multi-UV lamps.

The treatment of leachate from landfills is a major disposal problem for municipal solid waste. The leachate is generally recalcitrant to be treated according to complicated characteristics and high color intensity resulting further threat for environment and human health. In this work, the designed thin gap annular photoreactor with 4-UV lamps in UV/H2O2 process was proposed to decolor and remove chemical oxygen demand (COD) from the landfill leachate for solving this environmental problem. Meanwhile, the operating parameters such as UV dosage, hydrogen peroxide concentration and leachate strength were evaluated. The landfill leachate treated with the maximum dosage of 4-UV lamps and 232.7 mM of hydrogen peroxide concentration achieved 72 and 65% of color and COD removal efficiencies in 300 min. As for less concentrated leachate of 20% strength, 91% of color and 87% of COD were removed within only 120 min. From the experimental results, the UV/H2O2 process in this work was an effective pre-treatment or treatment technology for landfill leachate.
