The microvasculature of human oral mucosa using vascular corrosion casts and India ink injection. I. Tongue papillae.

The microvasculature of human tongue papillae originating from 9 males and 6 females, aged 0.5 to 2 years was studied by scanning electron microscopy (SEM) of vascular corrosion casts and by light microscopy (LM) of India ink injected specimens. All papillae showed a microvasculature characterized by primary, secondary and tertiary capillary loops. In the filiform papillae the loops were generally arranged in a corolla-like pattern with the tertiary loops demonstrating a hair-pin shape. The fungiform papillae showed basically a similar architectural pattern although the loops were somewhat more compact and complex in structure. A small, shallow depression of the tertiary loops at the top of these papillae was found to be occupied by a prominent rete ridge of the surface epithelium. There was a gradual transition from filiform to foliate papillae, the latter appearing as rows of coalesced filiform papillae. The circumvallate papillae easily identified by the surrounding furrow showed a rather complex and compact pattern of capillary loops of which typical hair-pin shaped tertiary loops dominated the periphery of the papilla. Small grooves or depressions in the vascular network were found to be occupied by rete ridges of the overlying mucosal epithelium.

Electron microscopic studies of various cells in the alveolar wall of mice with special reference to spheroid alveolar epithelial cells after intravenous injection of squid-ink (sepia-melanin) solution.

The effect of an intravenous injection of squid-ink (sepia-melanin) solution on adult mouse spheroid alveolar epithelial cells was observed by the electron microscope. Sepia-melanin particles were seen in all alveolar wall cells examined that seems to suggest the entrance of sepia-melanin particles into the spheroid alveolar epithlial cells from the alveolar blood capillary. In cases of large penetrations of sepia-melanin particles into spheroid alveolar epithelial cells, a greater increase was found in the intramitochondrial granules. In addition, the so-called inclusion body believed to be formed by the degeneration of mitochondria had very high electron density and its quantity was abundant. On the contrary, in cases where the quantity of sepia-melanin entrance into the spheroid alveolar epithelial cell was small, neither an increase of intramitochondrial granules, an increase of the electron density nor an increase in the quantity of specific inclusion body was found.

[Reasonable lymph node dissection in radical gastrectomy for gastric cancer: introduction of computer information system and lymphography technique by India-ink].

A computer information system was developed and clinically used in National Cancer Center Tokyo in 1983. Seven data of an individual patient are preoperatively input in the system: sex, age, location, macroscopic type, maximal diameter, depth of invasion, and histological type of biopsy. After the data-analysis of 3,785 patients, the computer outputs three data; expected five year survival rate, each metastatic rate of 16 regional lymph nodes, and types of recurrence. Plan of lymph node dissection can reasonably be decided by this report. 0.5ml of India-ink (CH-40) is injected in the perigastric lymph nodes, and it stains all lymphatic channels from the injection points in black. Using the technique, systematic dissection can be performed easily and completely. After the introduction of these systems, five year survival rate was improved from 90.0% to 95.5% in Stage I, from 76.0% to 81.6% in Stage II, from 51.1% to 56.8% in Stage III, and from 12.9% to 16.6% in Stage IV.

[The effect of heparin, prednisolone and triamterene on the elimination of India ink by the reticulohistiocyte system of the isolated perfused rat liver].

The influence of heparin, prednisolone and triamterene on the elimination of colloidal carbon (India ink) by the reticulohistiocytary system of the isolated perfused rat liver was measured. The drugs were given directly to the perfusion medium. A significant reduction of the phagocytic capacity of Kupffer's cells for colloidal carbon under the influence of heparin, prednisolone and triamterene could be demonstrated.

Lysozyme, intradermal diffusion of india ink and peritoneal capillary permeability in mice.

Hen egg white lysozyme injected intracutaneously into mice inhibits the intradermal diffusion of India Ink. The effect of lysozyme persists in presence of hyaluronidase. Lysozyme injected i.p. into mice inhibits acetic acid (i.p.) induced leakage of Pontamine Sky Blue into the abdominal cavity. The data suggest that lysozyme might be tried for applications outside the microbiological field.

The microvasculature of human infant oral mucosa using vascular corrosion casts and india ink injection. II. Palate and lip.

The microvasculature of human hard and soft palate and lip originating from four infant males and six females, aged 6 months to 2 years was studied by scanning electron microscopy of vascular corrosion casts and light microscopy of India ink injected specimens. The capillary loops of the hard palate mucosa and vermilion border of the lips were found to be tall, numerous and consisted of primary, secondary and tertiary loops. Those of the soft palatal and labial mucosa were short, few in number and demonstrated a simple hair-pin shape originating directly from the subpapillary vascular network. It was concluded that the configuration of capillary loops is not only determined by the shape of the connective tissue papillae in the lamina propria but also influenced by the functional demands characteristic of the different areas of the oral mucosa.

Specific staining of bacilli by means of immuno-indian-ink method and immunofluorescent method.

The possibilities for identification of B. anthracis are investigated by means of the Immuno-Indian-ink method (IIIM) and the immunofluorescent method (IFM) in their direct and indirect modifications. The specifity of sera against noncapsulated cells of B. anthrasis increases by their adsorption with antigenically related strains of B. cereus as their vegetative cells and spores are killed beforehand with performic acid. Repeated uses of the bacillus suspension for adsorption is possible by treating it with hydrochloric acid in order to restore its antigenic properties. The adsorbed sera against noncapsulated cells of B. anthracis in the direct and indirect modifications of IIIM and IFM stain specifically the 19 investigated B. anthracis strains but none of the 152 strains of saprophytic bacilli. In addition to that the possibility for differentiation of B. anthracis from saprophytic bacilli is established also by combination of cultivation on selective medium, facilitating the capsule formation of B. anthracis with IIIM and IFM, performed with sera against capsule antigens of the anthrax agent.

[Experimental glaucoma model in the rat with laser trabecular photocoagulation after intracameral injection of India ink].

We tried to create an experimental glaucoma which is simple and highly reproducible in rodents. India ink was injected into the anterior chamber with a 30-gauge needle. The carbon particles were trapped in the chamber angle and formed a black band along the corneal limbus one week later. This area was then photocoagulated anteroposteriorly with an argon laser. Photocoagulation was repeated until the intraocular pressure was elevated to 25 mmHg or more by pneumotonometry performed at a one-week interval. The above procedure was applied to one eye of 7 rats with the fellow eye serving as control. Chronic elevation of intraocular pressure was obtained in all the 7 treated eyes. The eyes were then enucleated and examined by light microscopy. Carbon particles were phagocytosed by macrophages in the intertrabecular spaces. Peripheral anterior synechias were also present. The optic nerve-head showed remarkable decrease in nerve fibers and cavernous degeneration suggestive of glaucomatous optic nerve damage. This experimental model promises to be of value in further studies of glaucomatous optic nerve damage.
