Solid-phase extraction with supercritical fluid elution as a sample preparation technique for the ultratrace analysis of flavone in blood plasma.

A new sample preparation technique, solid-phase extraction with supercritical fluid elution, was developed for the selective isolation of ultratrace levels of drugs from plasma. Plasma samples spiked with a drug were applied to octadecylsilane cartridges and the cartridges were then washed, briefly dried and directly fitted into cells for subsequent supercritical fluid elution. The absolute recovery was studied by using a radiolabeled model compound. The extraction selectivity was examined by chromatographing the extracts with a reversed-phase high-performance liquid chromatographic method with ultraviolet detection. The effects of extraction pressure and the length of capillary restrictors on drug recovery were examined in order to determine the optimal conditions for supercritical fluid elution. The performance of the method was compared to that of conventional solid-phase extraction in terms of recovery, selectivity, precision and accuracy of analysis. Flavone was used as the model compound and dog plasma as the biological matrix for these studies.

1,4-Dihydropyridine derivatives as calcium channel modulators: the role of 3-methoxy-flavone moiety.

It was earlier recognized that calcium antagonists, and in particular 1,4-dihydropyridines, exhibited distinct cardiovascular profiles. In addition two different splice variants of the L-type calcium channel were found in vascular and cardiac tissues. In this study, novel substituted 1,4-dihydropyridines with a 3-methoxy-flavone moiety were synthesized and structural modifications of the substituents in the dihydropyridine ring of nifedipine were carried out in order to find tissue specific compounds. The negative inotropic, chronotropic and vasorelaxant effects were investigated on guinea-pig left, right atria and aortic strips, respectively. The introduction of an heteroaromatic ring in 4-position of the 1,4-dihydropyridine nucleus led to compounds selective for cardiac tissues. Moreover, different residues in the 1,4-dihydropyridine ring could modulate the chronotropic versus inotropic activity.

Structure-activity relationship study of flavone compounds with anti-HIV-1 integrase activity: a density functional theory study.

Human immunodeficiency virus type-1 integrase (HIV-1 IN) is an essential enzyme for effective viral replication. Flavone compounds have been very much studied due to their activity during the inhibition process of HIV-1 IN. In this study, we employed density functional theory (DFT) using the B3LYP hybrid functional to calculate a set of molecular properties for 32 flavonoid compounds with anti-HIV-1 IN activity. The stepwise discriminant analysis (SDA), principal component analysis (PCA) and hierarchical cluster analysis (HCA) methods were employed to reduce dimensionality and investigate possible relationship between the calculated properties and the anti-HIV-1 IN activity. These analyses showed that the molecular hydrophobicity (ClogP), charge on atom 11 and electrophilic index (omega) are responsible for the separation between anti-HIV-1 IN active and inactive compounds.

Preparative isolation and purification of polymethoxylated flavones from Tangerine peel using high-speed counter-current chromatography.

Several polymethoxylated flavones including nobiletin, 3,5,6,7,8,3',4'-heptamethoxyflavone, tangeretin and 5-hydroxy-6,7,8,3',4'-pentamethoxyflavone were separated from Tangerine peel (Juhong in Chinese) by high-speed counter-current chromatography (HSCCC) with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (1:0.8:1:1, v/v). Then, 26 mg of nobiletin, 6 mg of 3,5,6,7,8,3',4'-heptamethoxyflavone, 35 mg of tangeretin and 11 mg of 5-hydroxy-6,7,8,3',4'-pentamethoxyflavone could be obtained from 150 mg crude extracts and their purities were 98.6%, 95.9%, 99.8% and 96.8%, respectively. All these constituents were identified by EI-MS and 1H NMR.

Simultaneous determination of bioactive flavone derivatives in Chinese herb extraction by capillary electrophoresis used different electrolyte systems--borate and ionic liquids.

In this paper, novel capillary electrophoresis (CE) methods, which used ionic liquids (1-ethyl-3-methylimidazoium tetrafluoroborate, 1-butyl-3-methylimidazoium tetrafluoroborate), were established to separate and determine some bioactive flavone derivatives in Chinese herb Seriphidium santolinum (Schrenk) Poljak. In order to investigate the traits of ionic liquids in CE, borate was also used as electrolyte to compare with. And the excellence of CE, which used ionic liquids as main running electrolyte and beta-cyclodextrin (beta-CD) as modifier, was illuminated as well. As a result of the study, the difference of ionic liquids and borate in CE was discussed and the advantage of CE, which used ionic liquids as electrolytes for separation, was shown. The analysis was obtained within short time (5-6 min). From the result, it was found that the system, which used ionic liquids, was robust because the joule heating was small. The method of CE, which used ionic liquids, has lower detection limits (0.137-0.642 microg/mL) than that of borate (0.762-1.036 microg/mL). And the CE, which used ionic liquids method, has lower limit of linear range (1.100-2.656 microg/mL), while that of CE, which used borate method, was 2.188-5.313 microg/mL.

Specificities of acetoxy derivatives of coumarins, biscoumarins, chromones, flavones, isoflavones and xanthones for acetoxy drug: protein transacetylase.

The earlier work carried out in our laboratory led to the identification of a novel rat liver microsomal enzyme termed as acetoxy drug: protein transacetylase (TAase), catalyzing the transfer of acetyl group from polyphenolic acetates (PA) to functional proteins. In this paper, we have reported the comparison of the specificities of acetoxy derivatives of coumarins, biscoumarins, chromones, flavones, isoflavones and xanthones with special reference to the phenyl moiety/bulky group on the pyran ring of PA. The results clearly indicated that compounds having phenyl moieties, when used as the substrates, resulted in a significant reduction of TAase catalyzed activity. The alteration in TAase catalyzed activation of NADPH cytochrome c reductase and inhibition of benzene-induced micronuclei in bone marrow cells by PA were in tune with their specificities to TAase.

Anti-hyperlipidemic properties of CM108 (a flavone derivative) in vitro and in vivo.

Peroxisome proliferator-activated receptors (PPARs) and liver X receptor alpha are ligand-activated transcription factors that belong to nuclear receptors superfamily and are involved in the regulation of lipid metabolism. PPAR, especially PPAR-alpha, PPAR-gamma agonists and liver X receptor alpha agonists can regulate the expression or biosynthesis of some factors involved in the formation and function of HDL, such as apolipoprotein (apo) A-I and ATP binding cassette transporter A1 (ABCA1). It is well known that HDL plays an important role in the treatment of hyperlipidemia as the carrier of reverse cholesterol transport. In the present study, the anti-hyperlipidemic properties of CM108, a derivative of flavone, 9-Hydroxy-2-mercapto-6-phenyl-2-thioxo-1,3,5-trioxa-2lambda(5)-phospha-cyclopenta[b]naphthalen-8-one, were studied. Through the transactivation assays of in vitro study, it was discovered that CM108 could activate PPAR-alpha PPAR-gamma and liver X receptor alpha at 40-150 microg/ml, which subsequently resulted in activating ABCA1 promoter and enhancing apoA-I and apoA-II production, whereas reducing apoC-III production significantly. Furthermore, after in vivo study that the hyperlipidemic rats were treated with CM108 for 4 weeks, a significant increase was found in HDL cholesterol levels (26.7%, P<0.05) and a significant decrease was also noticed in triglyceride levels (26.3%, P<0.01) at 100 mg/kg CM108 group compared with that of control animals. Meanwhile, the atherogenicity index, represented by total cholesterol/HDL ratio, was significantly reduced (P<0.01). In conclusion, CM108 can effectively elevate HDL levels and lower triglyceride levels in hyperlipidemic rats maybe by regulating a series of genes, receptors and proteins related to HDL.

Tumor inhibition and improved immunity in mice treated with flavone from Cirsium japonicum DC.

The purpose of these experiments is to study the effects of flavone from Cirsium japonicum DC (FLCJ) on tumor activity and on the regulation of the immune response in mice with S180 and H22 tumors. Pectolinarin and 5, 7-dihydroxy-6, 4'-dimethoxyflavone are two major components of FLCJ. FLCJ, pectolinarin and 5, 7-dihydroxy-6, 4'-dimethoxyflavone can inhibit the growth of the implanted tumors S180 and H22 and promote cellular and humoral immune responses. FLCJ is stronger than Pectolinarin and 5, 7-dihydroxy-6, 4'-dimethoxyflavone in inhibiting tumors and promoting immune response significantly. Our results indicated that the tumor inhibition of FLCJ had close relation with its regulation of the immune response Furthermore, this tumor inhibition by FLCJ may activate the innate immune system, which would be expected as a potential anti-tumor drug.

Regioselectivity of 7-O-methyltransferase of poplar to flavones.

POMT-7, an O-methyltransferase from poplar (Populus deltoids) was used to modify a variety of flavonoid compounds. POMT-7 was able to transfer a methyl group to several flavonoids containing a C-7 hydroxyl group. However, POMT-7 showed a higher affinity toward flavonol and flavone such as apigenin, kaempferol, luteolin, and quercetin than flavanone and isoflavone. Based on comparison of HPLC retention times with authentic compounds and corresponding nuclear magnetic resonance spectroscopy data, the methylation position of the reaction products was determined to be at the hydroxyl group of C-7. Biotransformation kinetics indicated that the enzyme converted more than 80% of the apigenin, kaempferol, luteolin and quercetin substrates, which were added at concentration of 70 microM, into corresponding 7-methoxy compounds within 24 h.

Protective effect of total flavones from Elsholtzia blanda (TFEB) on myocardial ischemia induced by coronary occlusion in canines.

This study was undertaken to determine the effect of total flavones from Elsholtzia blanda (Benth.) Benth. (TFEB), a traditional Chinese medicine, on myocardial ischemia induced by coronary occlusion in Beagle dogs. Infarct size of left ventricle, serum activity of creatine kinase-MB (CK-MB) and malondialdehyde (MDA), hemorrheologic variables and homodynamic parameters including mean arterial pressure (MAP), coronary blood flow (CBF), coronary vascular resistance (CVR), end-diastolic pressure of left ventricle (LVEDP), rate of rise and decline of left ventricular pressure (+/-dp/dtmax) were measured in this study. Administration with TFEB produced a dose-dependent reduction in infarct size. TFEB 100 mg/kg exerted notable inhibition in the elevation of serum CK-MB and MDA activity. TFEB significantly reduced MAP and CVR. The decrease in CBF also tended to be smaller in treated dogs. TFEB improved the recovery of myocardial function by depressing the degree of reduction in +/-dp/dtmax and LVEDP. TFEB also showed a capacity to resist ischemic damage through lowering blood viscosity. The results indicated that TFEB kept heart from ischemic damage due to coronary occlusion in Beagle dogs.

Total flavones from Elsholtzia blanda reduce infarct size during acute myocardial ischemia by inhibiting myocardial apoptosis in rats.

Apoptosis is a common pathological feature in acute myocardial infarction (AMI). The infarct size is an important determinant of the prognosis of AMI. In recent years, Chinese medicinal herbs and their extracts have received great attention in prevention of AMI. The aim of this investigation was to evaluate the anti-ischemic effect of total flavones from Elsholtzia blanda (Benth.) Benth. (TFEB), a traditional Chinese medicine and to make clear the mechanism involved in it. Myocardial infarction was induced by coronary occlusion in rats. Apoptosis was measured quantitatively by the terminal transferase UTP nick end-labeling (TUNEL) method and confirmed by DNA laddering on agarose gel. The expression of anti-apoptotic protein, Bcl-2 and pro-apoptotic protein, Bax was visualized by Western blot analysis. TFEB significantly reduced infarct size and TUNEL-positive rate confirmed by disappearance of DNA laddering. Greater Bcl-2 and attenuated Bax expression was found in TFEB treating rats. These results suggest that TFEB reduce infarct size during AMI by inhibiting myocardial apoptosis through modulation of Bcl-2 family.

Citrus polymethoxylated flavones improve lipid and glucose homeostasis and modulate adipocytokines in fructose-induced insulin resistant hamsters.

The present study was undertaken to determine whether supplementation with polymethoxylated flavones (PMFs) could ameliorate the fructose-induced hypertriglyceridemia and other metabolic abnormalities associated with insulin resistance (IR) in hamsters. Following feeding with the fructose diet, hamsters were supplemented orally with PMF-L or PMF-H (62.5 and 125 mg/kg/day) for 4 weeks. Both PMF-treated groups showed a statistically significant (p<0.05) decrease in serum triglyceride (TG) and cholesterol levels compared to the fructose-fed control group. The fructose control group at the end of the study showed elevated serum insulin and impaired insulin sensitivity (glucose intolerance). On the other hand, PMF-supplemented groups showed a reversal in these metabolic defects, including a decrease in insulin level and an improvement in glucose tolerance. PMF supplementation also reduced TG contents in the liver and heart and was able to regulate adipocytokines by significantly suppressing TNF-alpha, INF-gamma, IL-1beta and IL-6 expression and increasing adiponectin in IR hamsters. The mechanism of PMF on the activation of peroxisome proliferator-activated receptors (PPAR) was also explored. PMF-H supplementation significantly increased PPARalpha and PPARgamma protein expression in the liver. This is the first report of positive effects of PMF on adipocytokine production and on PPAR expression in IR hamsters. This study suggests that PMF can ameliorate hypertriglyceridemia and its anti-diabetic effects may occur as a consequence of adipocytokine regulation and PPARalpha and PPARgamma activation.

Total flavones of Hippophae rhamnoides promotes early restoration of ultimate stress of healing patellar tendon in a rat model.

Traditional Chinese herbal medicine has long been used for treatment of tendon injuries. Comparing to the modern way of treatments, Traditional Chinese medicine also stresses on strategies to promote the inherent healing capacity of tendons. Hippophae rhamnoides, known as Shaji, is one of Chinese herbal drugs that are traditionally used to promote tendon and ligament injuries. The total flavones of H. rhamnoides (TFH), with major constituents including quercetin, isorhamnetin and kaempferol, have been demonstrated with most of the bioactive properties of Shaji. In the present study, we evaluated the potential effect of TFH in the restoration of ultimate stress of healing patellar tendon in a well-established gap wound model in rats. A 0.1 mg TFH was injected to wound 1 day after the injury, and the ultimate stress of the healing tendon was measured at day 14 post-injury. The results showed that the ultimate stress of the healing tendon was significantly promoted by injection of TFH, increasing from 30 to 50% as compared to saline control. Excessive fibrotic response was not found in TFH-treated animals, but an enhanced collagen deposition and a better fibre alignment were observed. The results suggest that TFH may improve the ultimate stress of healing tendons at early stages, which implies possible earlier rehabilitation programme and better recovery.

Inhibitory effect of total flavone of Abelmoschus manihot L. Medic on NMDA receptor-mediated current in cultured rat hippocampal neurons.

The modulatory effect of Total Flavone of Abelmoschus manihot L. Medic (TFA) on NMDA-activated current (I(NMDA)) was investigated in cultured rat hippocampal neurons using the whole-cell patch-clamp technique. TFA rapidly and reversibly inhibited the I(NMDA) in a concentration-dependent manner. Furthermore, TFA non-competitively inhibited the I(NMDA) by enhancement of the NMDA receptor desensitization. In addition, intracellular application of TFA did not alter the TFA inhibition of I(NMDA). These results suggest that the inhibition of the NMDA receptor response by TFA could be one of the mechanisms for TFA-mediated neuroprotective actions.

Acylated 5,7,2',6'-oxygenated flavone glycosides from Andrographis alata.

Five acylated 5,7,2',6'-oxygenated flavone glycosides along with the known 5,2',6'-trihydroxy-7-methoxyflavone-2'-O-beta-d-glucopyranoside have been isolated from the whole plant of Andrographis alata. The structures of the compounds were established from spectral (mainly 1D and 2D NMR) and chemical studies.

Biotransformation of the citrus flavone tangeretin in rats. Identification of metabolites with intact flavane nucleus.

The present study was carried out in order to investigate the in vivo biotransformation and excretion of the flavone, tangeretin, found in citrus fruits, by analysing urine and faeces samples from rats after repeated administration of 100 mg/kg body weight/day tangeretin. The formed metabolites were separated and identified by HPLC and the structures elucidated by LC/MS and 1H NMR. Ten new, major metabolites with intact flavonoid structure were identified. The metabolites identified were either demethylated or hydroxylated derivatives of the parent compound and metabolic changes were found primarily to occur in the 4' position of the B-ring. The total urinary excretion of tangeretin metabolites with intact flavan nucleus was about 11% of the administered daily dose.

Linear indices of the "molecular pseudograph's atom adjacency matrix": definition, significance-interpretation, and application to QSAR analysis of flavone derivatives as HIV-1 integrase inhibitors.

This report describes a new set of molecular descriptors of relevance to QSAR/QSPR studies and drug design, atom linear indices fk(xi). These atomic level chemical descriptors are based on the calculation of linear maps on Rn[fk(xi): Rn--> Rn] in canonical basis. In this context, the kth power of the molecular pseudograph's atom adjacency matrix [Mk(G)] denotes the matrix of fk(xi) with respect to the canonical basis. In addition, a local-fragment (atom-type) formalism was developed. The kth atom-type linear indices are calculated by summing the kth atom linear indices of all atoms of the same atom type in the molecules. Moreover, total (whole-molecule) linear indices are also proposed. This descriptor is a linear functional (linear form) on Rn. That is, the kth total linear indices is a linear map from Rn to the scalar R[ fk(x): Rn --> R]. Thus, the kth total linear indices are calculated by summing the atom linear indices of all atoms in the molecule. The features of the kth total and local linear indices are illustrated by examples of various types of molecular structures, including chain-lengthening, branching, heteroatoms-content, and multiple bonds. Additionally, the linear independence of the local linear indices to other 0D, 1D, 2D, and 3D molecular descriptors is demonstrated by using principal component analysis for 42 very heterogeneous molecules. Much redundancy and overlapping was found among total linear indices and most of the other structural indices presently in use in the QSPR/QSAR practice. On the contrary, the information carried by atom-type linear indices was strikingly different from that codified in most of the 229 0D-3D molecular descriptors used in this study. It is concluded that the local linear indices are an independent indices containing important structural information to be used in QSPR/QSAR and drug design studies. In this sense, atom, atom-type, and total linear indices were used for the prediction of pIC50 values for the cleavage process of a set of flavone derivatives inhibitors of HIV-1 integrase. Quantitative models found are significant from a statistical point of view (R of 0.965, 0.902, and 0.927, respectively) and permit a clear interpretation of the studied properties in terms of the structural features of molecules. A LOO cross-validation procedure revealed that the regression models had a fairly good predictability (q2 of 0.679, 0.543, and 0.721, respectively). The comparison with other approaches reveals good behavior of the method proposed. The approach described in this paper appears to be an excellent alternative or guides for discovery and optimization of new lead compounds.
