Studies on the integument of cestodes. VI. Histochemical studies on the fate of structural polysaccharide in the integument of gravid proglottides of Taenia hydatigena.

Histochemical study of the structural polysaccharide in the integument of gravid proglottides revealed that the main cuticle is only feebly positive to PAS reaction when compared with that of other regions of the cestode body. The significance of such change in the PAS reaction has been discussed. It was observed that there is no change in the acid mucopolysaccharide of the outer layer of the integument.

[Cestodes of the genera Gyrocoelia Fuhrmann, 1899 and Infula Burt, 1939, 1939 (Acoleidae) from the snipes of the Soviet Union].

The data are given on the morphology of cestodes of the genera Gyrocoelia Fuhrm,. 1899 and Infula Burt, 1939. It has been found that in the spermatheca of the female of G. perversa there are spermatozoids with an elongated head typical for Plathelminthes. In the spermatheca of the female of I. burhini were found spermatids with long tails. The final formation of spermatozoids in I. burhini takes place in the spermatheca. The taxonomic position of the species of Gyrocoelia found in the waters of the USSR has been made more precise: G. polytestis Saakowa, 1952 and Gyrocoelia sp. II Schaldibin, 1960-1961 are reduced to synonyms of Infula burhini and Gyrocoelia sp. I Schaldibin, 1960-1961 to a synonym of G. perversa. The number and size of the hooks of proboscis in the species of the genus Gyrocoelia vary in wide ranges and cannot be used as a taxonomic character. The number of testes and the structure of the egg membrane are constant for the species of this genus.

[Cytochemical characteristics of the labrocyte-like cells in the capsule of the plerocercoids of Triaenophorus nodulosus (Pallas, 1781) and Diphyllobothrium dendriticum (Nitzsch, 1824) cestodes].

Out of six fish species examined (Esox lucius, Paracottus kessleri, Perca fluviatilis, Leuciscus leuciscus baicalensis, Coregonus autumnalis migratorius, Thymallus arcticus baicalensis) labrocyte-like cells were detected in Esox lucius, Leuciscus leuciscus baicalensis, Coregonus autumnalis migratotius. The cells contain large metachromatic granules. Histochemical methods revealed in them sulfated and carboxylic acid mucopolysacharides. The amount of labrocyte-like cells in the capsules around plerocercoids of Triaenophrus nodulosis from Esox lucits liver depends on the age of the capsule and that of the plerocercoid. It increases when the connective tissue papsule is forming and decreases again under aging and degeneration of the capsule containing plerocercoid. The amount of labrocyte-like cells in the cestodes capsules of Raillietina increases sharply when they locate in an accidental organ (liver). This demonstrates the acuteness of organ reactivity to a nonspecific helminth.

[A new cestode species Paranoplocephala gubanovi sp.n. (Cyclophyllidea: Anoplocephalidae) from the wood lemming Myopus schisticolor of eastern Siberia].

A new species of anoplocephalid cestode, Paranoplocephala gubanovi sp. n. (Cyclophyllidea, Anoplocephalidae), from wood lemmings of Eastern Siberia (Myopus schisticolor) is described. The new species differs from other known species of Paranoplocephala associated with Holarctic lemmings by having unique combination of characters as follows: a few-segmented strobila, superficial suckers sticking out of the scolex, ovary covering practically the whole middle part of the segment, relatively little number of testicles situated in the aporal part of the segment, cirrus bursa crossing the poral excretory vessels, and subspherical spermatheca situated in the middle part of the segment. Comparison of P. gubanovi sp. n. and several closest species, P. fellmani Haukisaimi et Henttonen, 2001, P. serrata Haukisaimi et Henttonen, 2000, and P. arctica (Rausch, 1952) has been carried out. Features distinguishing the new species from Aprostatandrya macrocephala and A. microti have also been studied.

[Final stages of carbohydrate metabolism in a cestode Eubothrium rugosum (Cestoda: Pseudophyllidea)].

Characteristics of final stages of carbohydrate metabolism in a cestode Eubothrium rugosum from the burbot intestine have been studied. It is found out that in the course of in vitro incubation the worms produce lactic acid to the environment with a concomitant acidification of the medium. The rate of pH changes and lactate production eventually fell during the incubation. The medium pH being the result of helminth's metabolism varies within the normal physiological limits of the host intestine pH and pH optima for its basic digestive enzymes.

[Monocercus dokuchaevi sp. n. (Cyclophyllidea: Dilepididae)--a new species of cestodes from shrews of the North-Eastern Asia].

A new cestode species, Monocercus dokuchaevi sp. n. is described from shrews of the genus Sorex from the Middle Kolyma plateau (Magadan Province). The new species is most closer to M. soricis (Neiland, 1953) by having regular alteration of genital atriums in the short strobila (up to 10 mm only). The size of rostellar hooks in M. dokuchaevi is intermedial between those in M. soricis and M. arioni (Sibold, 1850). In Monocercus dokuchaevi, the length of rostellar hooks is 0.038-0.045 mm (average 0.045 mm), in M. arioni, average is 0.05 mm, in M. soricis, limits are 0.027-0.033 mm. Two other species of the genus, M. baicalensis Eltyshev, 1971 and M. estavarensis Euzet et Jourdan, 1968, have much longer rostellar hooks, 0.07-0.08 and 0.092-0.106 mm, respectively, whereas the genital atriums in these species are altered irregularly.

[Spasskylepis tiunovi sp. n. (Cyclophyllidea, Hymenolepididae)--a new cestode species from shrews of the Far East].

The description of a new cestode species with a serial homonomous strobila, Spasskylepis tiunovisp. n. (Cyclophyllidea, Hymenolepididae, Ditestolepidini) from shrews (Sorex spp.) of the Far East is given. The new spesies differs from the type species of the genus, Spasskylepis ovaluteri (Schaldybin, 1964) in having oligomerous strobila and different size and structure of copulative organs. In contrast to S. ovaluteri, S. tiunovi has less than a half the number of proglottids in its strobila (48-64 versus 118-150). The cirrus of S. tiunovi (0.175-0.195 mm long) is twice as large as that of S. ovaluteri (0.080-0.100 mm long), pleated when invaginated, and armed with spines only in its first third. The copulative section of the vagina in S. tiunovi (as compared with S. ovaluteri) is very long, having loops in the median or aporal part of proglottids, and without a sphincter.

[Phenotypic variability of a perch parasite-cestode proteocephalus percae (Müller, 1780) (Proteocephalidea) in different parts of the species range].

Discrete variability of four P. percae characters of the main cestode functional complexes was identified. Phenotypic diversity of P. percae from different parts of the distribution range was analysed. Research revealed low geographic variability and high stability of the dominant variations in combination with morphometric plasticity. The conclusion was made that the patterns of P. percae morphological variability were shaped by the common fate and long-standing co-evolutionary relations between the parasite and the host--the perch Perca fluviatilis.

Cestode parasitic infestation: intracranial and spinal hydatid disease--a clinicopathological study of 29 cases from South India.

In developing countries hydatidosis is both a medical and economic problem related to environmental hygiene and healthy veterinary practice. This cestode parasitic infestation, uncommonly involving the nervous system, presents with varied clinical manifestation, at times causing diagnostic dilemmas. Multiple intracranial and spinal hydatidosis is rare. A series of 29 histologically confirmed cases of hydatidosis of neuraxis (21 intracranial and 8 spinal) from South India are presented. Among the 21 cases of intracranial hydatidosis, 12 cases were in pediatric age, while only 1 spinal lesion was noted in a 5-year-old child. The clinical presentation of intracranial lesions was predominantly that of raised intracranial pressure and visual symptoms, while spinal hydatidosis manifested with severe back pain, weakness and sphincter disturbances. The cranial cysts were usually single and uniloculated (12 cases), multiple in 7 and single but multiloculated in 2. In spinal hydatidosis, the cysts are usually multiple and extradural, rare ones being intramedullary and intradural. Based on clinical features and imaging, the differential diagnosis for intracranial lesions were cystic tumors and arachnoid cyst while metastasis and tuberculosis were considered in cases of spinal hydatidosis because of vertebral bony involvement. The majority of the cysts could be surgically resected totally and some were aspirated under control suction and resected. None of the cases had anaphylactic reaction, with no significant post-operative morbidity and no mortality. One intracranial and 2 spinal lesions caused by fertile cysts recurred to undergo repeated surgery.

Demonstration of the orientation of the Cestodes spermatozoon illustrated by the ultrastructural study of spermiogenesis and the spermatozoon of a Cyclophyllidea: Thysaniezia ovilla, Rivolta, 1874.

Spermiogenesis in Thysaniezia ovilla begins with the appearance of a differentiation zone equipped with peripheral microtubules, surrounded by arched membranes and containing two parallel centrioles surrounded by electron dense granular material. One of the centrioles aborts, the other gives rise to a flagellum which quickly grows in the cytoplasmic extension in which the nucleus migrates and protein granules form. During the nuclear migration crested-like bodies appear among the peripheral microtubules in the differentiation zone. The Th. ovilla spermatozoon is filiform. One of its extremities, which we show to be anterior, exhibits two helicoidal crested-like bodies, surrounded by electron dense material resembling an acrosome. Cortical microtubules run along almost the whole length of the spermatozoon. The cytoplasm has no mitochondria and contains protein granules. The axoneme is of the 9 + '1' pattern. It stops before reaching the posterior extremity of the gamete. The nucleus is wrapped round the axoneme, in the middle region of the spermatozoon. It generally stops before reaching the posterior extremity of the flagellum. This study has enabled us to describe precisely the orientation of the spermatozoon of cestodes in general.

[Histochemical study of the cysticercoids of cestodes of the genus Aploparaksis Clerc, 1903 (Hymenolepididae)].

Hystochemical methods were employed in order to study the distribution of succinic dehydrogenase, glucose-6-phosphate-dehydrogenase, alcaline, acidic phosphatases, lipids, fatty acids, glycogen and neutral. mucopolysaccharides in larvae of three species of cestodes: A. birulai, A. polystictae, A. xemae. A high concentration of succinic dehydrogenase was recorded from the tissue of the tail, scolex and neck. Glycose-6-phosphatdehydrogenase was found in great quantity only in the external parenchyma of the neck of A. polystictae. Here accumulations were observed of lipids which apparently are the products of excretion. The glycogen localization coincides with the distribution of succinis dehydrogenase. The activity of phosphatases is concentration in the structures of the tail and external membranes of cysticercoids, the acidic phosphatase being dominant. It was concluded that aerobic oxidation dominates in larvae and substances are principally transported through the tail area and excretory aperture.

Field trial of Saussurea lappa roots against nematodes and Nigella sativa seeds against cestodes in children.

Antinematodal efficacy of Saussurea lappa roots (Qust-e-Shereen) and anticestodal effect of Nigella sativa seeds (Kalonji) was studied in children infected naturally with the respective worms. The activities were judged on the basis of percentage reductions in the faecal eggs per gram (EPG) counts. The 50 mg/kg single dose of S. lappa and equivalent amount of its methanolic extract produced on days 7 and 15 percentage EPG reduction similar to 10 mg/kg of pyrantel pamoate. Similarly, single oral administration of 40 mg/kg of N. sativa, equivalent amount of its ethanolic extract and 50 mg/kg of niclosamide reduced the percentage of EPG counts not significantly different from each other on the days 7 and 15. Therefore, it is conceivable that these indigenous medicinal plants contain active principles effective against nematodes and cestodes. The crude drugs did not produce any adverse side effects in the doses tested.

Characterisation of taeniid cestodes by DNA analysis.

This review describes how the application of molecular technology is providing improved methods for characterising and identifying taeniid cestodes. Emphasis has been placed on the use of DNA approaches for specific diagnosis of the various taeniid life-cycle stages, unambiguous species and strain discrimination, determination of evolutionary and phylogenetic relationships and the assignment of function to key macromolecules based on inferred amino acid sequences corresponding to cloned cDNA sequences. In addition, the review describes methods for isolating DNA from taeniids, approaches for cloning genomic and mitochondrial DNA from these organisms and the use of DNA methods for strain characterisation and egg detection. Lastly, the review focuses on the polymerase chain reaction (PCR) which can provide the basis for exquisitely sensitive assays diagnostic for individual taeniid species. Coupled with direct sequencing techniques, PCR can also provide a valuable new method for examining genetic variation between and within the various taeniid species.

[Antigenic affinity of cestodes in the genus Taenia].

Antigens of four species of the genus Taenia (T. ovis ovis, T. ovis krabbei, T. hydatigena and T. parenchimatosa) were studied by means of immunodiffusion reaction in agar gel with the use of hyperimmune sera. It has been established that extracts of the studied cestodes contain a great number of antigens, which during parenteral administration cause a synthesis of antibodies in rabbits. In homologous systems the number of recorded antigen-antibody complexes varied from 5 to 10. The most close antigenic affinity was found between T. ovis ovis and T. ovis krabbei, T. ovis ovis and T. hydatigena, as far as the main mass of precipitation bands in the immunodiffusion reaction fused together that suggests the identity of corresponding antigenic components. In all cases when analysing antiserum to T. parenchimatosa extract no differences of species-specific character in heterologous systems were traced.
